Identification of the Maize R Gene Component Responsible for the Anthocyanin Biosynthesis of Kernel Pericarp (옥수수 종피의 안토시아닌 합성을 조절하는 R 유전자 구성요소의 구명)

  • The R-r:standard (R-r:std) allele of maize R gene complex consists of S subcomplex and P component; the S subcomplex regulates anthocyanin pigmentation of seed aleurone layer, and the P component confers pigmentation of the other plant parts. The S subcomplex contains two functional genes, S1 and S2 components. In the presence of Pl gene some alleles of R gene induce anthocyanin pigmentation of pericarp. In the present study, the effects of different R alleles on the anthocyanin pigmentation of pericarp in the presence of Pl gene were analyzed in order to identify the R gene component responsible for pericarp pigmentation. The results show that R-ch and r-ch alleles condition similar degrees of pericarp pigmentation, and that R-r:Ecuador (R-r:Ec) conditions stronger pigmentation. The r-ch allele, which is inferred that its S subcomplex has lost function but the P component is normal, induces pericarp pigmentation in the presence of Pl gene. On the contrary, the R-g:g1111 allele, derived from R-r:Ec and inferred that its S subcomplex functions normal but the P component has lost its function, did not induce pericarp pigmentation in the presence of Pl gene. Moreover, PCR analysis of genomic DNA's of R-ch and r-ch indicate that R-ch maintains both P and S1 components, whereas r-ch lacks for the S1 component. Taken together, The results suggest that the P components of R alleles inducing pericarp pigmentation in the presence of Pl gene are responsible for pericarp pigmentation.

  • The antifungal studies on the triterpenoids from the pericarp of Forsythia viridissima LINDL. were conducted. The pericarp was obtained from the plant which was cultivated at Eusung, Kyungsang-bukkdo, Korea. Substances A and B were isolated by column fractionation and purified by recrystallization. Antifungal actions of substance A, acetyloleanolic acid, show significant inhibition effect against Trichophyton rubrum, T. mentagrophytes, T. tonsurans, Microsporum nanum, M. canis, and M. cookei. Substance B, viridissimic acid, shows negative effect against Candida albicans and shows lower effects against other test fungi.

Comparisons of Pericarp Thickness And Sugar Content on New Waxy Corn Hybrids

  • For waxy corn hybrid development, 23 waxy corn hybrids were planted and surveyed in both pericarp thickness and sugar content. Pericarp thickness showed average 50 to $49{\mu}m$ in field and greenhouse. Results like these advanced than previous other hybrids. Also, sugar content in developed waxy corn evaluated as 13.6 to 15.3 brix. Here, we concluded that edible quality of waxy corn depended on combing of two factors. Especially, CNU11H8 hybrid at field and CNU11H-38 at greenhouse were good in both factors. Accordingly, these hybrids will be planted in next time for productivity and area adaptation.

  • Xanthoxylii Fructus(山椒) is the pericarp of Xanthoxylum piperitum D.C. according to the KP II. The pericarp of Xanthoxylum coreanum $N_{AKAI}$, which is the native plant in Che-ju island is called as Xanthoxylii Fructus in that area. In this paper, the pharmacognostical study on pericarp of Xanthoxylum coreanum $N_{AKAI}$ is investigated to distinguish its morphological from those of pericarps of Xanthoxylum piperitum D.C. and Fagara mandshurica $H_{ONDA}$.

Analysis of Antibacterial Activity against Food Spoilage and Food-borne Pathogens and Cytotoxicity on Human Cancer Cell Lines of Extracts from Pericarp and Seed of Vitis coignetiea (머루 과피와 종자 추출물의 식품 위해성 세균에 대한 항균성 및 인체 암세포주에 대한 cytotoxicity 분석)

  • In this study, antibacterial activity and cytotoxicity of the extracts from pericarp and seed of $Vitis$ $coignetiea$, which were extracted with 0.1% HCl-60% ethanol, were analyzed. The antibacterial activity of the extracts was determined by paper disc diffusion method against food spoilage and food-borne pathogens. The pericarp extract showed high antibacterial activity against $Bacillus$ $cereus$, $Escherichia$ $coli$ O157:H7, and $Pseudomonas$ $aeruginosa$, and the seed extract represented the high antibacterial activity against $B.$ $cereus$, $E.$ $coli$ O157:H7, and $Staphylococcus$ $aureus$. The cytotoxicity of the $Vitis$ $coignetiea$ extract against human cancer cells was determined using the MTT assay and SRB assay. The pericarp extract represented strong growth-inhibition activity against G361 and Hep3B cells and the seed extract greatly inhibited the growth of HeLa and G361 cells in the MTT assay. In addition, the pericarp extract displayed a high inhibition activity against the growth of AGS cells and the seed extract greatly inhibited the growth of HeLa, Hep3B, and MCF7 cells in the SRB assay. Especially, the cytotoxicities of the seed extract against HeLa were significantly higher than those of the extract against other cancer cells at all test concentrations. This study demonstrates that the extract from pericarp and seed of $Vitis$ $coignetiea$ possess high antibacterial activity and cytotoxicity.

Analysis of Antioxidative and Antimutagenic Activities of Ethanol Extracts from Pericarp and Seeds of Wild Grape (Vitis coignetiea) (머루 과피와 종자 에탄올 추출물의 항산화 활성 및 항돌연변이 활성 분석)

  • The antioxidative activity and antimutagenic activity of the ethanol extracts from pericarp and seeds of wild grape (Vitis coignetiea) were analyzed in this study. The antioxidative activity of the extracts from wild grape was determined using 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) assay. The antimutagenic activity of the extracts was evaluated on Salmonella typhimurium TA98 and TA100 by Ames test using 4-nitroquinoline 1-oxide (4-NQO) and sodium azide as mutagens. In the antioxidative activity determination, $IC_{50}$ values of the DPPH radical scavenging activity of the extracts from pericarp and seeds were 27.16 ppm and 7.61 ppm, respectively. Additionally, ABTS radical scavenging activities of pericarp and seed extract were 99.75% and 98.87% at 200 ppm, respectively. In the antimutagenic activity determination, pericarp extract at 5 mg/plate inhibited 72.6% and 74.3% of mutagenicity of S. typhimurium TA98 induced by 4-NQO and sodium azaid, respectively. Also, the mutagenicity inhibition rates of seed extract at 5 mg/plate were 77.8% and 74.5% in S. typhimurium TA100 induced by 4-NQO and sodium azaid, respectively. These results demonstrate that the ethanol extract from wild grape has remarkable antioxidant activity and antimutagenicity.

  • Rice (Oryza sativa L.) is one of the major cereal crops for consumption by the world’s population. Recently, various colored rice, such as white, red, brown, green, and black rice, have caught the attention of world consumers. The commercial name ‘black rice’ contains a high amount of anthocyanins in pericarp, which increases nutritional value. Moreover, anthocyanin in black rice possesses biomedical properties, including anti-oxidant, anti-cancer, and anti-inflammatory effects in humans. In genetics, black rice has a dominant PURPLE PERICARP (Prp) trait governed by two genes, Pb and Pp, which are involved in the synthesis of cyanidin-3-O-glucoside (C3G). Since the publication of a report by Nagai at 1921, the genetics and physiological studies of black rice driven by Prp traits are still unable to understand the relevant genes and their roles. However, with the increased demand for anthocyanin-rich black rice as a functional food for human health, it has become urgent to develop highyielding anthocyanin-rich varieties of rice. We explored many years in the genetics of purple pericarp trait, anthocyanin biosynthesis in pericarp during seed development, and, consequently, their products in relation to different physiological and agronomic traits. In this review, we summarized the anthocyanin biosynthesis in pericarp, emphasizing the inheritance pattern of the trait and functions of their products on different physiological and agronomic traits, including the yield of black rice.

Effect of split achene pericarp, Giberellin, and germination temperature on the germination of Rosa davurica Pall (생열귀나무 종자 발아에 있어서 개갑처리, 지배렐린, 발아온도의 효과)

  • This study was conducted to investigate the suitable conditions for seed germination of Rosa davurica PALL. In the characteristics of the fruit and seed of Rosa davurica, fruit length and width was 1.3cm and 0.9cm , respectively, and seed number was eighty-nine. Artificial and low temperature storage at 4$^{\circ}C$ increased the rate of split achene pericarp until 46.6% and storage at 15$^{\circ}C$ incubator decreased the rate of split achene pericarp (10.5%). The rate of seed germination of split achene pericarp in control at 15$^{\circ}C$ was 90% and of non-split achene in GA$_3$100ppm at 15$^{\circ}C$ was 36.8%. Average germination day of split achene pericarp seeds in GA$_3$150ppm at 2$0^{\circ}C$ was 4.2 days and non-split achene pericarp in GA$_3$100ppm at 2$0^{\circ}C$ was 7.3 days. Seed germination was not different between various concentrations of GA$_3$ treatments in split achene pericarp seeds but the rate of germination was more reduced in high concentration of 200ppm. Only the treatments of GA$_3$ was increased germination rate at 4$^{\circ}C$ and immature seed of Rosa davurica.

Comparison of Kernel Sample Preparation Methods at Different Grain Filling Periods for Determining Pericarp Thickness in Super Sweet and Waxy Corn Hybrids (시료 준비 방법에 따른 등숙 시기별 초당 및 찰옥수수 교잡종의 과피 두께 비교)

  • Pericarp thickness of vegetable corns such as sweet and waxy corn is one of the crucial traits, contributing to their edible quality. This study was carried out to compare the pericarp thickness of super sweet and waxy corn hybrids measured with kernel samples prepared using different methods at different grain filling periods. The samples comprised excised pericarp from dried, frozen (at $-4^{\circ}C$), and fresh kernels. Analysis of variance performed separately on super sweet and waxy corn hybrids indicated a significant three-way interaction among cultivars, kernel sample preparation methods, and days after pollination (DAP). Dried samples of super sweet corn hybrids presented reasonably stable pericarp thickness measurements during grain filling, while all the sample preparation methods fluctuated less as grains of waxy corn hybrids matured. Waxy corn is best consumed at around 24 days after pollination. Pericarp thickness of waxy kernel samples regardless of preparation methods investigated was the same at 24 DAP with a few exceptions. Overall, the common method of drying kernel samples before pericarp excision can provide reliable data for estimating the tenderness of vegetable corn hybrids.

Study on the Antioxidant and Anti-inflammatory Effects of Extracts of Callus Cultures, pericarp, flesh, fruit of Trapa Japonica (능실 열매의 부위별 추출물 및 캘러스배양 추출물의 항산화, 항염증 효과 연구)

  • The purpose of this study was to evaluate the antioxidant and anti-inflammatory effects of extracts of callus cultures, pericarp, flesh, fruit of Trapa Japonica. The toxicity of extracts from Trapa Japonica pericarp investigated using the RAW 264.7 cell showed 45.8 ± 1.5% of cell survival rate. And the results of investigation using HaCaT cells showed a 51.1 ± 1.0% cell viability at 100 ㎍/㎖ in the pericarp extract and lower cell viability at higher concentrations. The total content of polyphenol pericarp extract was 213.20 ± 15.78 mg/g, while the total content of flavonoid was 29.30 ± 3.24mg. And the total content of polyphenol callus cultures extract was 205.20 ± 18.97 mg/g, while the total content of flavonoid was 237.4 ± 7.43 mg. With a concentration level of 0.005 ~ 1000 ㎍/㎖ extract of Trapa Japonica pericarp the range of removal of 1, 1-diphenyl-2-picryl-hydrazyl (DPPH) radicals was 67.53 ± 1.5 % ~ 75.75 ± 0.5 % respectively and the range of removal of extract of Trapa Japonica callus cultures extract was also 3.1 ± 0.1 % ~ 77.32 ± 0.5 % respectively. As a result of measuring the Nitric Oxide(NO) generation amount of all Trapa Japonica extract 6.25, 12.5, 25, 50 ㎍/㎖ concentration exhibited significant (p < 0.05, p < 0.01) decreases.
