곤충 생리현상의 가소성은 후생유전적 변화와 밀접하게 관련을 지을 수 있다. 이 가설을 증명하기 위해 광식성인 파밤나방(Spodoptera exigua)을 대상으로 상이한 먹이 조건에 따라 이 곤충의 발육과 DNA 메틸화에 영향을 주는 지 분석하였다. 동일한 코호트로 부터 얻은 갓 부화한 유충을 최종령에 이르기까지 세 가지 다른 먹이(대파, 배추, 인공사료)로 섭식 처리하였다. 이 결과 상이한 먹이 조건에 따라 유충발육속도, 용화율 및 우화율에서 뚜렷한 차이를 보였다. 인공사료로 사육된 유충이 가장 빠른 유충발육속도와 높은 용화율 및 우화율을 나타냈다. 반면에 두 자연 기주 가운데는 대파가 배추에 비해 파밤나방 발육에 양호하였다. 이러한 먹이에 따른 변이는 혈림프 단백질 및 혈당에서도 차이가 나타났다. 또한 발육과 연계되었을 것으로 추정되는 인슐린유사펩타이드(SeILP1) 유전자의 발현 정도도 먹이조건에 따라 상이했다. 단일항체를 이용하여 파밤나방 게놈 DNA의 시토신 메틸화를 분석한 결과 이 부위에 DNA 메틸화가 검출되었으며, 메틸화 정도는 먹이 조건에 따라 상이했다. 이 결과들은 동일 집단의 파밤나방이 상이한 먹이 조건에 따라 발육차이를 나타내고 또한 시토신 메틸화에 변이를 보여 이 곤충의 생리적 가소성에 후생유전적 인자가 작용하고 있는 것을 제시한다.
혈액 내 순환시 안정성 향상과 면역원성의 감소를 위해, rhIFN-${\alpha}$-2a은 N-terminus의 ${\alpha}$-아민기에 mPEG aldehyde를 solid-phase PEGylation 시킨다. CM-Sepharose와 같은 양이온 교환수지가 고체 지지체로 사용되었다. Solid-phase PEGylation을 통해 종래의 액상 반응에서 나타날 수 있는 재현성 낮은 반응, 부 반응물 생성, 부 반응물 제거 공정 등의 단점을 극복할 수 있었다. 그러나 solid-phase PEGylation의 문제점인 액상 반응에 비교하여 많은 양의 PEG를 사용하여야 한다는 점은 개선되어야 한다.
Protein B23 is one of the major nucleolar phosphoproteins associated with pre-ribosomal particles, and is localized in the granular region of the nucleolus. Recent studies suggest that protein B23 shuttles between nucleus and cytoplasm and also interacts with HIV Rev. These findings indicate that protein B23 is important in nucleocytoplasmic relationship and viral replication. However, the exact function of protein B23 is not clear yet. In acute nucleolar hypertrophy of rat liver, treated with thioacetamide, there was observed an increase of not only protein B23 but also B23-like protein p45 when anti-B23 monoclonal antibody (MAb) was used for identification. On the basis of the large B23 specific epitope structure composed of 68 amino acids, a hypothesis was formulated to examine that p45 is the pre-B23 resulting from excessive production of B23. In an attempt to investigate the precursor of B23, we analyzed the subcellular fractions and microsomal subfractions. Subsequently, we analyzed the finger printings of B23-like proteins using the tryptic peptide mapping. The results are summarized: 1) Using B23 MAb, we observed the presence of B23-like proteins in nucleolar fraction, nucleoplasmic fraction and microsomal fraction. 2) In the further microsomal subfractionation, we could partially purify B23-like protein in 2M layer of sucrose gradient. 3) When ion exchange chromatography was employed, there were protein species 80kDa(p80), 65kDa(p65) and 60kDa(p60). 4) Based on the tryptic map analysis of $^{125}I$ labeled proteins, the similarity between B23 and p80 was found only in 9 out of 14(B23) and 21(p80) peptides, and difference was found in the remaining peptides. p80 and p60 had 18 common peptides, and all the peptides of p60 were similar to those of p80. From these results, it is proposed that p45 is an abnormal metabolite resulting from carcinogenesis by thioacetamide, and it is not the precursor of B23. In addition, we suggest that p80 may be a precursor of p45.
목적: 본 연구진은 난자성숙 과정의 조절 기작을 규명하기 위하여 생쥐의 미성숙 난자와 성숙난자에서 차이 나게 발현하는 유전자의 목록을 얻은 바 있다. 이들 유전자 중에서 Bcl-2 homolog인 Diva 유전자가 난자에 특이적으로 발현함을 본 연구를 통해 규명하였는데 이러한 Diva의 기능을 밝혀내기 위하여 immunoprecipitation (IP)과 Mass Spectrometry (MS)를 이용하여 Diva와 결합하여 상호작용하는 단백질을 동정하고자 하였다. 연구방법: NIH/3T3 세포주에 Diva를 encoding하는 pCMV-FLAG-Diva를 24 시간 동안 과발현 시키고 대조군으로는 유전자 없는 pCMV-FLAG empty vector를 transfection 하였다. FLAG에 특이적인 항체로 IP하여 Diva와 결합하는 면역복합체를 형성하게 한 후 이를 12% SDS-polyacrylamide gel 상에서 전기 영동하였고 Coomassie Blue 염색을 통해 단백질 발현양상을 관찰하였다. 대조군에서는 관찰되지 않으면서 Diva 유전자가 발현하는 실험군에서만 확인되는 밴드를 오려내어 trypsin을 사용하여 in-gel digestion 한 후 MS 분석을 시행하였다. 모든 mass spectra는 4700 Proteomics Analyzer (Applied Biosystems, Framingham, MA)에 의해 positive reflector mode에서 얻어졌다. 이렇게 얻어진 단백질들은 MASCOT Peptide Mass Fingerprint software (Matrixscience, London)을 이용하여 NCBI nonredundant database를 찾아서 동정하였다. 결과: Diva를 과발현하는 세포주에서만 관찰되는 15개 밴드에 대한 MS/MS 분석 결과, Diva와 결합하는 단백질로서 actin과 그 외에 ${\alpha}$-actinin, tropomyosin, tropomodulin 3 등의 actin-binding 단백질을 동정하였다. Diva를 과발현하는 NIH/3T3 세포주에서 면역 복합체를 형성하는 actin과 tropomyosin이 실제 난소 조직에서도 Diva와 결합하는지 IP와 Western blot을 통해 확인한 결과, actin과 tropomyosin 모두 Diva와 결합함을 확인함으로써, Diva는 이 두 단백질과 난소에서 상호작용함을 알 수 있었다. 결론: 본 연구는 Diva와 결합하여 상호작용하는 단백질들이 cytoskeletal system의 actin filament와 관계 있음을 규명한 최초의 보고이다. Diva가 actin과 tropomyosin과 결합하는 것을 고려해볼때, 난자 특이적인 Diva는 아마도 난자성숙 동안에 cytoskeletal system 을 조절하는 역할을 할 것으로 사료된다.
Background: To evaluate the therapeutic benefits of the treat-to-target (T2T) strategy for Asian patients with early rheumatoid arthritis (RA) in Korea. Methods: In a 1-year, multicenter, open-label strategy trial, 346 patients with early RA were recruited from 20 institutions across Korea and stratified into 2 groups, depending on whether they were recruited by rheumatologists who have adopted the T2T strategy (T2T group) or by rheumatologists who provided usual care (non-T2T group). Data regarding demographics, rheumatoid factor titer, anti-cyclic citrullinated peptide antibody titer, disease activity score of 28 joints (DAS28), and Korean Health Assessment Questionnaire (KHAQ) score were obtained at baseline and after 1 year of treatment. In the T2T group, the prescription for disease-modifying antirheumatic drugs was tailored to the predefined treatment target in each patient, namely remission (DAS28 < 2.6) or low disease activity (LDA) ($2.6{\leq}DAS28$ < 3.2). Results: Data were available for 163 T2T patients and 162 non-T2T patients. At the end of the study period, clinical outcomes were better in the T2T group than in the non-T2T group (LDA or remission, 59.5% vs. 35.8%; P < 0.001; remission, 43.6% vs. 19.8%; P < 0.001). Compared with non-T2T, T2T was also associated with higher rate of good European League Against Rheumatism response (63.0% vs. 39.8%; P < 0.001), improved KHAQ scores (-0.38 vs. -0.13; P = 0.008), and higher frequency of follow-up visits (5.0 vs. 2.0 visits/year; P < 0.001). Conclusion: In Asian patients with early RA, T2T improves disease activity and physical function. Setting a pre-defined treatment target in terms of DAS28 is recommended.
