Four species of yeast, Saccharomyces cerevisiae, Candida utilis, Saccharomycopsis flbuligera, and Schwanniomyces castellii were evaluated for their ability to bioconvert potato processing waste water into microbial protein and the resulting single-cell proteins were evaluated as protein sources for rainbow trout, using in vitro analyses. The studies indicated that Schwanniomyces castellii, which utilizes starch dircetly and converts it into cell mass efficiently, was suitable for the bioconversion. In the single-stage continuous bioconversion, the yield S. castellii cell mass, which contained approximately 37% protein, was 77%, at dilution rate 0.25 $h^{-1}$. Reduction of total carbohydrate was 81%. During batch fermentations, cell mass yield was about 72% and total carbohydrate reduction was 81%. Among the yeasts tested, S. castellii possessed the most fragile cell wall and had a favorable amino acid profile for salmonid fish; protein score of 86% (Met). In an in vitro pepsin digestibility test 80% digestibility (23~38% above control) was observed when cells were pre-heated in a steam bath for 30 min. Results presented should be regarded as being preliminary in nature because they were derived from single experiments.
Tannins in plant foods and beverages may produce antinutritional or toxic effects although some proteins with high affinity for tannins seem to function as defense mechanism to tannin toxicity. Our objectives were to investigate of tea tannins, iron and protein and to evaluate the role of proteins in tannin effects on iron solubility. Iron solubility in vitro was measured using tea with and without proteins. Mixtures of tea, protein in varying concentrations(either gelatin or bovine serum albumin), and iron(eithe 10 or 50ug/mL) were prepared. Controls contained water in place of tea. Iron bioavailability was assessed by measuring iron solubility in the simulated gastric condition with pepsin digestion. Bound iron was removed by centrifugation and soluble in tea alone. When iron concentratin was 10ug/mL, addition of small amounts of protein to tea dramatically reduced iron solubility, but solubility of iron increased in the tea mixturea as the concentration of protein was increased. The percnetage of iron that precipitated was much greater at 10ug Fe/mL than the values at 50ug Fe/mL suggesting that the iron binding sites on the tea-protein complex was saturated. These results suggest that interactions of iron with tea tannins are influenced by the concentratins of protein and iron.

The physicochemical properties of the $\alpha$-amylase inhibitors from black bean and naked barley is Korea were investigated. Preincubation time for maximum inhibition was 30min and no activity change was seen after that time. Optimum pH of the $\alpha$-amylase inhibitors from the black bean and naked barley was pH 7.0 and the inhibitory activities were stable in the range of pH 6.0~8.0 in both phosphate and Tris-HCI buffer solutions. Both inhibitors maintained more than 50% of activity after incubation for 17 min at 7$0^{\circ}C$. The inhibitors from the black bean and naked barley maintained more than 50% of activities after treatment for 40 min and 30 min with pepsin, and 30 min and 50 min with trypsin, respectively. Both inhibitors functioned via a noncompetitive mechanism and were active against porcine pancreatic and human salivary $\alpha$-amylases. The activities of both inhibitors were linear for the ionic stength ranging from 0 to 0.9. The addition of 70 mM maltose to the reaction mixture caused a maximum increase in the relative activities of both inhibitors, but it did not affect the dissociation of the EI complex. The activities of both inhibitors were significantly enhanced by adding 1mM of K+ or Mg2+.

The protection of dry matter and nitrogen from rumen degradation of dhaincha seed (Sesbania aculeate) meal was studied using nylon bag technique. The dhaincha seed meal was subjected to various heat treatments that included oven drying, autoclaving and boiling. Similar experiment was conducted with fish meal as reference for comparison. The oven-dried dhaincha meal was found to retain more dry matter and nitrogen than was found boiled or autoclaved meal. While autoclaving appeared to improve nitrogen and dry matter retention to some extent, boiling seemed to cause more loss of dry matter from nylon bag. Heat treatment caused high retention of nitrogen by fish meal. The calculated effective protein degradation was 80.4% and 83.2% for the oven dried fish meal and dhaincha seed meal whereas same values were 74.2% and 86.7% for autoclaved fish and dhaincha seed meal respectively at the outflow rate of 4.4% per hour. The in vitro study revealed higher digestibility for heat treated samples by pepsin. The dry matter, nitrogen and ash content of dhaincha seed meal were 85.93%, 5.93% and 7.31% respectively.
A survey on the incidence of metacercariae infection of Clonorchis sinensis in the fresh water fish, Pseudorasbora parva which is well known as the second intermediate host for this fluke was carried out in the upper stream of Phoung-tack stream, Kyungki province during the period of October to November 1997. Forty fish in P. parva were collected by netting, or using the bait in transparent bowls from the two different districts located in Osung-myun, Phoungtack-gun and transported to the laboratory. The body length and weight were measured. They were from 5.0 to 7.6 cm in length and 1.0 to 5.0 g in weight. A total of 40 P. parva were divided 3 groups by the size (70-79, 60-69 and 50-59 mm) and were digested by artificial digestion with pepsin-Hcl solution in a 36$\circ$C incubator and then examined the infection density of metacercariae of the fresh water fish under a binocular dissecting microscope. The incidence of metacercariae infection in P. parva was 406 of 40 examined and the metacercariae detection rate showed 77.5%. The average number of metacercariae detected in P. parva was 10.2. The results of the survey indicate that Clonorchis sinensis infection may occur by consuming raw fresh water fish caught from the upper stream of Phoungtackho, Kyungki province.
The full-length cDNA of the lumbrokinase fraction 6 (F6) protease gene of Lumbricus rubellus was amplified using an mRNA template, sequenced and expressed in E. coli cells. The F6 protease gene consisted of pro- and mature sequences by gene sequence analysis, and the protease was translated and modified into active mature polypeptide by N-terminal amino acid sequence analysis of the F6 protease. The pro-region of F6 protease consisted of the 44 residues from methionine-1 to lysine-44, and the mature polypeptide sequence (239 amino acid residues and one stop codon; 720 bp) started from isoleucine-45 and continued to the terminal residue. F6 protease gene clones having pro-mature sequence and mature sequence produced inclusion bodies in E. coli cells. When inclusion bodies were orally administrated rats, generated thrombus weight in the rat' venous was reduced by approximately 60% versus controls. When the inclusion bodies were solubilized in pepsin and/or trypsin solutions, the solubilized enzymes showed hemolytic activity in vitro. It was concluded the F6 protease has hemolytic activity, and that it is composed of pro- and mature regions.
The objectives of this study were to identify the histamine-forming bacteria and bacteriocin- producing lactic acid bacteria (LAB) isolated from Myeolchi-jeot according to sequence analysis of the 16S rRNA gene, to evaluate the inhibitory effects of the bacteriocin on the growth and histamine accumulation of histamine-forming bacteria, and to assess the physico-chemical properties of the bacteriocin. Based on 16S rRNA gene sequences, histamine-forming bacteria were identified as Bacillus licheniformis MCH01, Serratia marcescens MCH02, Staphylococcus xylosus MCH03, Aeromonas hydrophila MCH04, and Morganella morganii MCH05. The five LAB strains identified as Pediococcus acidilactici MCL11, Leuconostoc mesenteroides MCL12, Enterococcus faecium MCL13, Lactobacillus sakei MCL14, and Lactobacillus acidophilus MCL15 were found to produce an antibacterial compound with inhibitory activity against the tested histamine-producing bacteria. The inhibitory activity of these bacteriocins obtained from the five LAB remained stable after incubation at pH 4.0-8.0 and heating for 10 min at $80^{\circ}C$; however, the bacteriocin activity was destroyed after treatment with papain, pepsin, proteinase K, ${\alpha}$-chymotrypsin, or trypsin. Meanwhile, these bacteriocins produced by the tested LAB strains also exhibited histamine-degradation ability. Therefore, these antimicrobial substances may play a role in inhibiting histamine formation in the fermented fish products and preventing seafood-related food-borne disease caused by bacterially generated histamine.
In order to investigate the effect of Daewhajungeum on the isolated organs, gastrointestinal tract and C.N.S., some kinds of experiments using mice and rats were done. The results were as follows; 1. Daewhajungeum showed the inhibitory effect on the smooth muscle contraction of the isolated ileum and colon induced by acethylchorine chloride and barium chloride in mice. 2. Daewhajungeum showed inhibitory effect on the contraction of the fundus-strip induced by acethylchorine chloride and barium chloride in rats. 3. The preventive effect on pyloric ulcer, indomethacin induced ulcer and ethanol-HCl induced ulcer of mice was recognized. 4. The effect on the acidity of gastric juice was suppressed, but the effect on the pepsin output was not recognized. 5. The transport ability of small and large intestine was significantly decressed. 6. The analgesic effect and the sedative effect were recognized so that the effect of the C.N.S was suggested.
Angiotensin I-converting enzyme (ACE) inhibitory activities of elk antler hydrolysates prepared with three kinds of proteases, pepsin, trypsin and $\alpha-chymotrypsin$, were investigated. The ACE inhibitory activity of the pepsinolytic hydrolysate was the highest with an $IC_{50}$ value of $9.3\mu g/mL.$ In addition, three kinds of pepsinolytic hydrolysates with relatively high molecular weights (over 10,000 Da), medium molecular weights (5,000 to 10,000 Da), and low molecular weights (below 5,000 Da) were fractionated using an ultrafiltration membrane system. The below 5,000 Da hydrolysate exhibited the highest ACE inhibitory activity. These results indicate that the pepsinolytic hydrolysates of elk velvet antler could be a good source of peptides with ACE inhibitory activity.
The purpose of the present study was to examine the antagonistic activities of Enterococcus faecalis strain PL9003 (PL9003) on Helicobacter pylori. This strain was isolated from infant feces and found to inhibit both the growth of H. pylori and its in vitro adherence to the human gastric cell line MKN-45. The binding of PL9003 to MKN-45 was observed under a light microscope after Cram staining and under a scanning electron microscope. When detected with an FITC-conjugate antibody, both viable and nonviable PL9003 were found to decrease the number of H. pylori bound to MKN-45. When detected by an enzyme-linked immunoabsorbent assay, about 70% of the H. pylori bound on MKN-45 disappeared with the four-1314 addition of viable or nonviable PL9003. The spent culture supernatant (SCS) of PL9003 also decreased the viability of H pylori even after neutralization and pepsin treatment. The above results suggest that PL9003 has a potential as a new probiotic for the stomach.
