밀랍지 내에서의 Aspergillus versicolor와 Penicillium polonicum의 생장 특성 연구 (A Study on Growth of Aspergillus versicolor and Penicillium polonicum within the beeswax treated paper)

  • Paper composed mainly of organic materials and they can be used as nutrient by microorganisms. Therefore microorganisms are damaging the important documents and works of art irreversibly. Previous research reported that mycelium or spores of the fungi were observed on the wax of the volumes of King Sejong especially were heavily deteriorated by fungi. Fungal strains isolated from the annals of Joseon dynasty were identified as Biscogniauxia atropunctata, Aspergillus versicolor, Penicillium polonicum, Ceriporia lacerata, Irpex lacteus. Especially Aspergillus and Penicillium are able to grow on the substrates having a 7-8% moisture content, which is much lower than the general storage condition (RH55-60%) of the paper cultural properties. Moreover, they are known as fungi cause paper deterioration and discoloration. In this study, we selected Aspergillus versicolor and Penicillium polonicum among five strains, then biological aging has been executed for 30 days. The growth of Aspergillus versicolor and Penicillium polonicum in wax was lower than wax treated paper and Hanji. The growth of them in beeswax treated paper and Hanji both were excellent.

  • 1997년부터 2003년에 걸쳐 우리나라에서 수확된 보리, 율무, 옥수수, 수수, 벼에서 총 81개의 Penicillium 속균을 분리하였다. 분리된 Penicillium 속균의 배양적 형태적 특징을 이용하여 종을 동정한 결과 P. citrinum, P. chrysogenum, P cyclopium, P. oxalicum, P. polonicum, P. purpurogenum 그리고 P. viridicatum으로 동정되었다. 이 중에서 P. cyclopium, P. polonicum, P. purpurogenum은 국내에서 처음 보고되는 미기록종들이다.

  • Three Penicillium species have been isolated from insect specimens in Korea; Penicillium sp., P. steckii, and P. polonicum. Penicillium sp. (KNU12-3-2) was isolated from Lixus imperessiventris, while P. polonicum (KNU12-1-8) and Penicillium steckii (KNU12-2-9) were isolated from Muljarus japonicas and Meloe proscarabaeus, respectively. The identification was based on the morphological characteristics of the fungi and in internal transcribed spacer analysis. This is the first report on the isolation of these three species of Penicillium from insects in Korea.

산소흡수제 처리가 Aspergillus versicolor와 Penicillium polonicum에 의한 한지의 생물열화 특성에 미치는 효과 (The Effect of Oxygen Absorbent on Aged Characteristics of Hanji during Biological Artificial Aging by Aspergillus versicolor and Penicillium polonicum)

  • 박물관 및 도서관 등에 소장되어 있는 지류 유물은 균류, 곤충, 설치류 등과 같은 다양한 생물학적 열화요인들에 의해 비가역적인 손상을 입을 수 있다. 특히 습도가 높은 조건에서는 균에 의한 열화가 발생되기 쉬우며, 균류는 종이의 주성분인 셀룰로오스 및 사이징제, 전분, 아교 등의 탄수화물과 단백질 성분을 영양원으로 하므로 종이의 노화를 야기하게 된다. 현재까지 박물관과 도서관 등에서 지류 유물에 생물학적 열화를 발생시킬 수 있는 미생물종으로 약 300여 종이 동정되었으며, 그 중에서도 Aspergilli (30%)와 Penicilli (30%)가 셀룰로오스의 주요 열화 균으로 알려져 있다. 균류에 의한 생물학적 손상을 방지하기 위한 가장 좋은 방법으로서 습도, 온도, 산소 등 미생물의 생육 조건을 조절, 억제 또는 차단하는 방법을 들 수 있다. 따라서 본 연구에서는 산소흡수제를 이용하여 미생물의 생육조건 중 산소를 조절하여 Aspergillus versicolor 과 Penicillium polonicum 균에 의한 생물열화 방지 효과를 분석하였다. 연구 결과 산소흡수제 처리가 2종의 균에 의한 생물학적 열화 억제 효과가 있음을 확인하였다.

Two Species of Penicillium Associated with Blue Mold of Yam in Korea

  • During 2007 survey of post-harvest diseases of yam performed in May and June, severe tuber loss caused by blue mold was observed in Iksan, Cheonbuk Province. Two species of Penicillium were isolated from the infected tubers. Based on $\beta$-tubulin gene sequence analysis, and cultural and morphological characteristics, the isolates were identified as Penicillium sclerotigenum and P. polonicum. P. sclerotigenum, which is a novel to Korea, is presently described and illustrated.

시판 건조 인삼류(백삼, 홍삼)의 곰팡이 발생 (Occurrence of Fungal Species in Dried Ginseng Products from Retail Market)

  • 시판 유통 중인 건조 인삼류(백삼 24점, 홍삼 26점) 포장 제품을 수집하여 곰팡이 발생 조사를 수행하였다. 백삼과 홍삼은 각각 50%와 46%의 시료에서 곰팡이가 검출되었고 검출 시료의 평균 곰팡이 오염도는 각각 0.5와 0.2 log10 CFU/g이었다. 백삼에서는 Penicillium polonicum, P. chrysogenum, Rhizopus microsporus가 각각 18.2%로 우점하였으나 홍삼은 Aspergillus spp.이 87.6%로 우점하였으며 이 중 A. chevalieri가 50.0%로 가장 높았다. 이 중 독성 종으로 알려진 P. polonicum, P. chrysogenum, P. melanoconidium, A. chevalieri 균주의 citrinin 독소 생성 가능성을 분석한 결과, 13 균주 모두 독소 유전자가 검출되지 않았다. 이 결과는 조사한 시료의 곰팡이독소 오염 위험은 매우 낮지만, 건조 인삼류에 곰팡이 오염이 가능함을 보여준다.

Geranial, Cineol, Menthol과 편백의 유효성분을 조성물로 하는 손소독제 연구 (Study of a Hand Sanitizer Composed of Active Ingredients Geranial, Cineol, Menthol, and Chamaecyparis obtusa)

  • 수장고 및 서고에서 곰팡이는 문화재와 기록물의 생물학적 열화를 일으키는 주요 원인이다. 또한, 독소를 생성하고 피부알레르기를 유발하여 유물과 기록물 및 도서를 다루는 근무자의 호흡기질환과 피부질환 등을 일으키기도 한다. 특히 근무자의 피부 및 손에 접촉되어 2차 감염을 일으킬 수 있다. 이를 예방하기 위하여 시중에서 쉽게 구입할 수 있는 손소독제는 일반세균에 대하여 소독효과를 보이지만, 곰팡이에는 소독효과가 떨어져 이에 대한 대체 연구가 필요한 실정이다. 본 연구에서는 손소독제 조성물로 친환경적인 Geranial, Cineol, Menthol과 편백(Chamaecyparis obtusa)의 유효성분으로 하는 손소독제를 연구 개발하여 박물관 및 기록관의 서고에서 조사된 곰팡이 10종(Alternaria citrimacularis, Alternaria consortialis, Aspergillus niger, Aspergillus versicolor, Cladosporium sphaerospermum, Coniothyrium aleuritis, Penicillium brevicompactum, Penicillium corylophilum, Penicillium paneum, Penicillium polonicum)에 대하여 소독효과를 시험한 결과, 우수한 소독효과를 나타내는 것을 확인하였다. 이에 Geranial, Cineol, Menthol과 편백의 유효성분을 조성물로 하는 손소독제의 개발로 문화재와 기록물 및 도서를 다루는 근무자의 피부질환과 2차 감염을 예방할 수 있을 것으로 기대한다.

The Origin of Meju Fungi - Fungal Diversity of Soybean, Rice Straw and Air for Meju Fermentation

  • Meju is a brick of dried fermented soybeans and is the core material for Jang such as Doenjang and Ganjang. Jang is produced by addition of salty water to Meju and is considered the essential sauces of authentic Korean cuisine. Meju is fermented by diverse microorganisms such as bacteria, fungi and yeasts. It is known that fungi play an important role in the Meju fermentation and they degrade macromolecules of the soybeans into small nutrient molecules. In previous study, 26 genera and 0 species were reported as Meju fungi. However, it is not comprehensively examined where the fungi present on the Meju are originated. In order to elucidate the origin of the fungi present on the Meju, the mycobiota of 500 samples soybean kernels, 296 rice straw pieces and air samples of Jang factories was determined in 0, 2 and 7 Jang factories respectively. Forty-one genera covering 86 species were isolated from the soybeans and 33 species were identical with the species from Meju. From sodium hypochlorite untreated soybeans, Eurotium herbariorum, Eurotium repens, Cladosporium tenuissimum, Fusarium fujikuroi, Aspergillus oryzae/flavus and Penicillium steckii were the predominant species. In case of sodium hypochlorite-treated soybeans, Eurotium herbariorum, E. repens and Cladosporium tenuissimum were the predominant species. Of the 4 genera and 86 species isolated from soybeans, 3 genera and 33 species were also found in Meju. Thirty-nine genera and 92 species were isolated from the rice straws and 40 species were identical with the species from Meju. Fusarium asiaticum, Cladosporium cladosporioides, Aspergillus tubingensis, A. oryzae, E. repens and Eurotium chevalieri were frequently isolated from the rice straw obtained from many factories. Twelve genera and 40 species of fungi that were isolated in the rice straw in this study, were also isolated from Meju. Especially, A. oryzae, C. cladosporioides, E. chevalieri, E. repens, F. asiaticum and Penicillium polonicum that are abundant species in Meju, were also isolated frequently from rice straw. C. cladosporioides, F. asiaticum and P. polonicum that are abundant in low temperature fermentation process of Meju fermentation, were frequently isolated from rice straw incubated at $5^{\circ}C$ and $25^{\circ}C$, while A. oryzae, E. repens and E. chevalieri that are abundant in high temperature fermentation process of Meju fermentation, were frequently isolated from rice straw incubated at $25^{\circ}C$ and $35^{\circ}C$. This suggests that the mycobiota of rice straw have a large influence in mycobiota of Meju. Thirty-nine genera and 92 species were isolated from the air of Jang factories and 34 species were identical with the species from Meju. In outside air of the fermentation room, Cladosporium sp. and Cladosporium cladosporioides were the dominant species, followed by Cladosporium tenuissimum, Eurotium sp., Phoma sp. Sistotrema brinkmannii, Alternaria sp., Aspergillus fumigatus, Schizophyllum commune, and Penicillium glabrum. In inside air of the fermentation room, Cladosporium sp., Aspergillus oryzae, Penicillium chrysogenum, A. nidulans, Aspergillus sp., C. cladosporioides, Eurotium sp., Penicillium sp., C. tenuissimum, A. niger, E. herbariorum, A. sydowii, and E. repens were collected with high frequency. The concentrations of the genus Aspergillus, Eurotium and Penicillium were significantly higher in inside air than outside air. From this results, the origin of fungi present on Meju was inferred. Of the dominant fungal species present on Meju, Lichtheimia ramosa, Mucor circinelloides, Mucor racemosus, and Scopulariopsis brevicaulis are thought to be originated from outside air, because these species are not or are rarely isolated from rice straw and soybean; however, they were detected outside air of fermentation room and are species commonly found in indoor environments. However, A. oryzae, P. polonicum, E. repens, P. solitum, and E. chevalieri, which are frequently found on Meju, are common in rice straw and could be transferred from rice straw to Meju. The fungi grow and produce abundant spores during Meju fermentation, and after the spores accumulate in the air of fermentation room, they could influence mycobiota of Meju fermentation in the following year. This could explain why concentrations of the genus Aspergillus, Eurotium, and Penicillium are much higher inside than outside of the fermentation rooms.

기능성 배접지의 보존 특성 분석 -호분지- (Conservation Properties of Chalk Added Functional Lining Papers)

  • Paper cultural heritages experience chemical and physical deterioration due to various factors including preservation environments and the property of paper materials1). Thus, it is important to develop optimal preservation and restoration methods for the damaged paper cultural heritages. Currently, lining is a popular treatment for the restoration of paper cultural heritages in Korea. Since lining paper is a layer of paper directly attached to the inside of cultural heritages for protection, understanding of the preservation properties of lining paper is primarily needed in order to devise the better preservation methods. The main material of lining paper is the paper mulberry, but additives such as chalk and white clay is sometimes used to enhance the preservation properties of lining paper. To date, the properties of the functional lining paper containing these additives have been not fully understood yet. In this study, dry heating aging at $105^{\circ}C$ and biological aging by the Aspergillus versicolor and Penicillium polonicum for the lining paper, which is made from paper mulberry and the chalk, are carried out to evaluate changes in their preservation properties by these aging factors. As a result, it is found that the functional lining paper containing 25.1% of chalk can control the growth of fungi, while the paper containing 32.7% of chalk do not show any protection effect. However, the functional lining paper added by chalk is more aged than the lining paper made from paper mulberry by dry heating accelerated aging.
