수확후 배 푸른곰팡이병을 일으키는 Penicillium속의 종류 및 특성 (Identification and Characteristics of Penicillium spp. Isolated from Postharvest Decay of Pear)

  • 수확 후 피해를 주는 Penicillium속의 병원체를 분리 동정하기 위하여 여러 특성을 조사하였다. 저장 중 배에서 분리한 198균주는 상처 접종과 무상 접종에서 병반을 형성하여 병원성이 있었으며, 균총 크기와 형태, 색, 배지뒷면 색소와 포자형태 등의 특성에 따라 15개 그룹으로 나눌 수 있었다. 분리한 균주들은 배양·형태적·생리적 특성에 따라 P. expansum, P. solitum, P. crustosum 등 3종으로 동정되었다. 분리된 균주들의 병원성 검정 결과 저온에서 상처접종이 무상접종 보다 병원성이 강하게 나타났으며 P. expansum이 저장 배 과실에 대해 가장 병원성이 강하였고, P. solitum과 P. crustosum은 약한 편이었다.

  • 국내산 사과로부터 분리, 동정한 P. crustosum과 patulin 생성균주인 P. griseofulvum(ATCC 46037)에 대하여 malt extract broth(MEB), sucrose yeast broth(SY) 및 5% glucose, yeast extract, peptone(5-GYEP) broth를 이용하여 생육정도와 독소 생성능을 비교하였다. 또한 균체 생육도와 독소 생성능과의 상관성을 검토하고, 배양액의 pH에 따른 균체 생장과 patulin 생성능을 조사하였다. P. griseofulvum(ATCC 46037)의 균체성장은 SY배지 에서 가장 높았으나 patulin은 SY, MEB, 5-GYEP 배지 모두에서 2,000-3,000 ppm의 높은 생산능을 보여주었다. P. crustosum의 균체생장 역시 SY배지에서 가장 높았으나 patulin 생성능은 5-GYEP broth 에서 배양 3주에 2,794 ppm으로 가장 높게 나타났다. P. crustosum과 P. griseofulvum 모두 균체생장과 patulin 생산능과의 상관관계는 매우 낮은 것으로 나타났다. 배지의 pH에 따른 patulin 생산능은 P. griseofulvum은 경우 pH 3-11의 넓은 범위에서 patulin 생성능을 나타내었으나, P. crustosum은 pH 3-5의 산성조건에서 높은 patulin 생성능을 나타내었다. 탄소원에 따른 patulin 생산능은 P. griseofulvum은 glycerol에서, P. crustosum은 fructose에서 가장 높았다.

  • Patulin is a food mycotoxin which induces genotoxicity and acute intestinal disease in infants. Patulin mainly originates from fruit putrefactive moulds, especially in apples, which necessitates the maintenance of strong safety standards against patulin for fresh and processed apples. To investigate the patulin producing moulds in Korean apples, 16 morphological types of fungi were isolated from Korean apples and a patulin producing fungus was identified based on a sequence analysis of the region of internal transcribed spacers (ITS5-5.8S-ITS4 region, 505 base pair) and the 26 rRNA D1/D2 region (527 base pair). Morphological analyses were also performed. The isolated patulin producing fungus was found to a representative species of Penicillium crustosum. The maximal patulin production ability of the isolated fungus (P. crustosum) and the patulin producing standard strain (P. griseofulvum, ATCC 46037) in an SY broth medium were 0.32 and 2.46 mg/L, respectively.

사과푸른곰팡이병에 관여하는 Penicillium의 계통분석, 형태 및 병원성 (Phylogenetic Analysis, Morphology and Pathogenicity of Penicillium spp. associated with Blue Mold of Apple in Korea)

  • Blue mold is the most important postharvest disease of apples in Korea. Apple fruits with blue mold symptoms were collected from storages in different locations in Korea and were investigated for their association with Penicillium species. A total of sixty five isolates of Penicillium were sampled from the collected apples. Based on DNA sequence analysis of ${\beta}$-tublin gene and ITS and lsu rDNA (ID region) and morphological characteristics, they were identified as P. crustosum, P. expansum, P. italicum, P. solitum and P. sp.. P. sp. which is closely related to P. hirsutum is a new species, not reported before. P. expansum (35%) was predominant species followed by P. crustosum. The phylogenetic tree inferred from combined ${\beta}$-tublin and ID region sequence showed good correlation with species that are defined by morphological characteristics. In pathogenicity test, apples were wound-inoculated with conidial suspension and incubated at $20-22^{\circ}C$. The most severe and destructive species was P. expansum. The species caused a decayed area 42-50mm in diameter after 8-10days. Decayed area caused by P. crustosum and P. sp. was 26-32mm and 20-26mm, respectively. This is the first record of P. crustosum, P. italicum and P. sp. from apple in Korea.

Penicillium Diversity from Intertidal Zone in Korea

  • Penicillium species are commonly isolated from various outdoor and indoor environments, including marine environments such as sponges, algae and sand. Penicillium is especially important because numerous bioactive compounds have been isolated. Penicillium was the most common species in intertidal zone in Korea, however the diversity and ecological roles of Penicillium in intertidal zone are not clarified. We explored diversity and ecological roles of marine-derived Penicillium from tidal flat and sea sand in Korea. The diversity of marine-derived Penicillium from Korea was investigated using both culture-dependent and culture-independent approach by ${\beta}$-tubulin sequence. In addition, we evaluated optimal temperature, halo-tolerance, and enzyme activity of Penicillium strains, such as extracellular alginase, endoglucanase, ${\beta}$-glucosidase, and protease. For culture-dependent approach, a total of 182 strains of 62 Penicillium species were isolated, with 53 species being identified. The most common species was Penicillium oxalicum, followed by P. crustosum, P. brasilianum, P. koreense, and P. griseofulvum. Species richness and composition were not significantly different by season, substrates, and seaside. For culture-independent approach using Illumina sequencing, 73 OTUSs were detected. The most frequently observed species was P. antarcticum, followed by P. koreense, P. crustosum, and P. brevicompactum. Diversity of Penicillium was higher during winter season than during summer season and in western sea than in southern sea, respectively. Community structure was significantly different by season and sea side. 52 species were detected by both methods. Unique species were isolated from each of methods - 10 from culture methods and 21 from Illumina sequencing. Furthermore, salinity adaption of the Penicillium varied depending on species. Many Penicillium species showed endoglucanase, ${\beta}$-glucosidase, and protease activity. Some species including P. paneum and P. javanicum degraded the polycyclic aromatic hydrocarbons. Thus, our results demonstrate that intertidal zone in Korea harbors diverse Penicillium community and marine-derived Penicillium play important ecological roles as decomposers of organic material in marine environments.

Penicillium crustosum에 의한 감 푸른곰팡이병 발생 (Blue Mold of Persimmon (Diospyros kaki) Caused by Penicillium crustosum)

  • 저장 및 유통중인 과실이 약간 함몰되면서 부패되고 그위에 푸른곰팡이가 심하게 발생하였다. MEA배지에서 균총의 색깔은 처음 흰색에서 푸른색을 띄며 분생포자를 아주 많이 형성하였다. 분생포자의 모양은 타원형 또는 아구형이고 크기가 2.6${\sim}$3.8${\times}$2.4${\sim}$3.8 ${\mu}m$이었다. 분생포자병은 균사표면으로부터 형성되고, Stipes의 크기가 86${\sim}$320${\times}$2.8${\sim}$4.3 ${\mu}m$로 가늘고 길며, Rami의 크기가 7.5${\sim}$32.6${\times}$2.6${\sim}$4.2 ${\mu}m$, Ramuli의 크기가 12.4${\sim}$14.8${\times}$3.2${\sim}$3.8 ${\mu}m$, Matulae의 크기는 8.9${\sim}$13.6${\times}$2.8${\sim}$4.6 ${\mu}m$이고, Phialide는 침상이고 크기가 8.2${\sim}$12.4${\times}$2.3${\sim}$3.6 ${\mu}m$이었다. 균사생육적온은 $25^{\circ}C$였다. 분리된 병원균을 건전한 감에 접종한 결과 동일한 병원성을 나타내었다. 이상의 결과로 이 병을 Penicillium crustosum에 의한 감 푸른곰팡이 병으로 명명할 것을 제안한다.

  • Post-harvest decay, caused by Penicillium spp. is a serious problem of fruits worldwide. Morphological characteristics and molecular markers were used to characterize 22 Penicillium isolates from apples, 18 isolates from pears, 60 from oranges and 18 from grapes and 23reference isolates representing related Penicillium spp. to assess their diversity and resolve their taxonomy. Based on morphological and physiological characteristics, the isolates were grouped as identical or very similar to P. digitatum, P. italicum, P. ulaiense or very similar to P. crustosum, P. expansum, P. solitum and unidentified Penicillium spp. Based on sequence comparisons of ITS region, variable site were presented within and among the species, but there variation were not correlated with the species. Cluster analyses of AP-PCR fragment patterns using UP and L45 primer and the -tubulin gene sequence, the Penicillium species were segregated into distinct groups. Particularly. the -tubulin partial sequence data provided support for species concepts based on morphological and physiological characteristics.

  • A total of 65 isolates of Penicillium were isolated from decayed post-harvest fruits of apple pear citrus and grape. The Penicillium species isolated from the apple were idnetified as P. aurantiogriseum and P. expansum those from the pear were P. crustosum and P. expansum and those from the grape were P. aurantiogriseum and P. expansum, From decayed citrus fruits. P. digitatum and P. italicum were isolated. Citrinin and patulin from these species in the YES(yeast extract sucrose) broth were extracted with ethyl acetate and purified by thin-layer chromatography(TLC) and high performance liquid chromatography(HPLC) Among 51 isolates of Penicillium from apple pear and grape 7 isolates produced citrinin 13 isolates produces patulin and 12 isolates produced citrinin and patulin also. All 14 isolates of Penicillium from citrus produced only patulin.

Diversity of Aspergillus, Penicillium, and Talaromyces Species Isolated from Freshwater Environments in Korea

  • In order to elucidate the fungal diversity and community structure in freshwater environments, numerous fungal strains were isolated from freshwater, submerged soils, twigs, dead insects, etc. Among them, the present study has focused specifically on Aspergillus, Penicillium, and Talaromyces species, which produce diverse useful metabolites in general. Twelve strains of Aspergillus isolated were identified as A. japonicus (n = 5), A. tubingensis (3), A. niger (2), and A. flavus (2), 10 strains of which belong to Aspergillus section Nigri, named black Aspergillus. Eight strains of Penicillium were identified as P. brasilianim (n = 3), P. oxalicum (2), P. crustosum (1), P. expansum (1), and P. piscarium (1). Two different strains of Talaromyces were identified as T. pinophilus and T. versatilis. Thus far, Penicillium piscarium and Talaromyces versatilis have been unrecorded in Korea, for which we provide detailed morphological and molecular characteristics.

저장 배에서 분리한 Penicillium속의 배양적 특성과 RAPD에 의한 유연관계 분석 (Analysis of Genetic Relationship and Cultural Characteristics of Penicillium species Isolated from Postharvest Decay of Pear by Random Amplified Polymorphic DNA)

  • 저장배에서 분리한 15그룹 3종을 KCTC, KCCM, 충남대에서 분양받은 16균주를 대상으로 상호 유연관계를 분석하고자 배양적 특성을 관찰하고 RAPD 검정에 의한 진단 가능성을 검토하였다. 병원균의 배양특성을 조사한 결과 배지상 생육형태, 색, 속도 등에 있어서 균주간 차이를 보였으며 배양적 특성과 RAPD에 의한 유연관계 분석 결과를 비교 분석하기 위하여 배에서 분리한 15개 균주와 16개 표준 균주의 종간 다형성을 관찰하였다. RAPD을 이용하여 유전적 다양성을 분석하기 위하여 5개의 URP primer를 이용하여 genomic DNA를 증폭시킨 결과 $0.1{\sim}2.0kb$ 크기의 총 744개 band들이 증폭되었다. cluster 분석결과 전체 유사도는 47.7%로 31 균주들은 4개의 genomic DNA RAPD 집단으로 분류되었다. 배에서 분리한 P. expansum 균주간에는 87.4% 이상의 상동성을 보였고, P. solitum 균주간에는 97.5%, P. crustosum 균주 간에는 95.1% 이상의 상동성을 보였다. 배에서 분리된 동일한 종내에서는 배양적 특성에 의한 동정결과와 RAPD에 의한 분석결과와 일치하였다.
