DNA fingerprinting analysis of maize varieties and parental lines using microsatellite markers (Microsatellite 마커를 이용한 옥수수 품종 및 자식 계통에 대한 DNA Fingerprinting 분석)

  • In the present study, we conducted genetic characterization of 90 commercial maize varieties and parental lines using microsatellite markers. Thirteen microsatellite markers were selected from 100 primer pairs in the maize genome data on the basis of polymorphism information contents (PIC) value and distinct amplification products. These markers detected 5 to 24 alleles, with an average of 13.69. The mean PIC value was 0.865 and ranged from 0.716 to 0.942. The unweighted pair-group method with arithmetical average (UPGMA) analysis was conducted for constructing the dendrogram using Jaccard's genetic similarity coefficient. The genetic similarity varied from 0.07 to 0.824. Thirteen microsatellite markers identified all 90 maize varieties and parental lines. The maize varieties were clustered into 5 major groups consistent with type and pedigree information. The microsatellite profile database of maize varieties could be used to select comparative varieties through genetic relationship analysis between existing varieties and candidate varieties in distinctness tests.

Heritability Estimated Using 50K SNPs Indicates Missing Heritability Problem in Holstein Breeding

  • Previous studies in Holstein have shown 35% to 51.8% heritability in milk production traits, such as milk yield, fat, and protein, using pedigree data. Other studies in complex human traits could be captured by common single-nucleotide polymorphisms (SNPs), and their genetic variations, attributed to chromosomes, are in proportion to their length. Using genome-wide estimation and partitioning approaches, we analyzed three quantitative Holstein traits relevant to milk production in Korean Holstein data harvested from 462 individuals genotyped for 54,609 SNPs. For all three traits (milk yield, fat, and protein), we estimated a nominally significant (p = 0.1) proportion of variance explained by all SNPs on the Illumina BovineSNP50 Beadchip ($h^2_G$). These common SNPs explained approximately most of the narrow-sense heritability. Longer genomic regions tended to provide more phenotypic variation information, with a correlation of 0.46~0.53 between the estimate of variance explained by individual chromosomes and their physical length. These results suggested that polygenicity was ubiquitous for Holstein milk production traits. These results will expand our knowledge on recent animal breeding, such as genomic selection in Holstein.

The Dopamine D4 Receptor Polymorphism Affects the Canine Fearfulness

  • The canine fearfulness is a behavioral trait known to have a genetic basis. This research analyzed genetic effects of the dopamine D4 receptor polymorphism on this behavior by postulating a mixed model of inheritance. Genotyping for the three different repeat polymorphism found in the third exon of the receptor gene was carried out for the population of the Korean native dogs. Four hundred fifty eight dogs with known pedigree were genotyped, and 264 individuals were tested for their fear responses to an experimenter, in which four different behavioral paradigms were adopted. Since the results assessed by principal factor analysis revealed a major factor explaining 69% of the total phenotypic variance, the subsequent analyses were conducted for this quantity. Analyses of the factor scores by estimating their posterior means indicated that there is a fixed effect exerted by the three different repeat polymorphism found in the D4 receptor as well as sex, in addition to unidentified polygenic effects. The phenotypic contribution of the D4 genotype was roughly estimated to be about 2%, which is a fraction of the total genetic effects responsible for more than 20% of the total phenotypic variance.

Analysis of Iridoid Glycoside and GABA Content in the Roots of the Rehmannia glutinosa Cultivars (지황 품종별 뿌리에서 Iridoid 배당체와 GABA 분석)

  • Background: Rehmannia glutinosa is a perennial herb belonging to the family Scrophulariaceae. Its roots have been utilized as a traditional medicine. The aim of this study was to elucidate the basic information of the roots of the R. glutinosa cultivars and their utilization. Methods and Results: The roots of R. glutinosa cultivars were harvested in the end of March. The two iridoid glycosides, aucubin and catalpol, were analyzed by liquid chromatography/mass spectrometry (LC/MS), whereas ${\gamma}-aminobutyric$ acid (GABA) was analyzed by gas chromatography/mass spectrometry (GC/MS). The aucubin content was the highest in the Dakang cultivar, whereas no aucubin was detected in the five cultivars. All cultivars had more than 12 mg/g catalpol content, and the maximum catalpol content was found in Jihwang 1. The GABA content was the highest in Suwon 1, and it was 40 times more than that in the Yeongang cultivar. Conclusions: The highest aucubin, catapol and GABA contents were detected in the Dakang, Jihwang 1, and Suwon 1, cultivars respectively. This study provides the crucial information regarding the versatile utilization and pedigree selection of R. glutinosa cultivars.

  • A 9-year old boy suffering from cleidocranial dysplasia associated with impacted 4 supernumerary teeth and unerupted all permanent teeth is presented with his mother. The pedigree showed autosomal dominant pattern of inheritance, and the raiographic features of them were very similar in clavicle, skull, vertebrae, peivis and extremities. Although almost of the skeleton was involved with this syndrome, they did not recognize any other problem but except dental problem. In mother, who was wearing removable partial dentures leaving 24 impacted teeth in her jaws, the radiographic abnormalities like cystic lesion were not detected. And in the son, who showed impacted 4 supernumerary and all permanent teeth, we have attempted surgical extraction of the supernumerary teeth and periodic surgical opening of the alveolar bone covering the permanent dentition to induce the eruption of permanent teeth at the proper position, Orthodontic treatment has also been combined to correct class III malocclusion state.

Individual Identification using The Multiplex PCR with Microsatellite Markers in Swine

  • The swine is one of the most widespread mammalian throughout the whole world. Presently, many studies concerning microsatellites in swine, especially domestic pigs, have been carried out in order to investigate general diversity patterns among either populations or breeds. Until now, a lot of time and effort spend into a single PCR method. But simple and more rapid multiplex PCR methods have been developed. The purpose of this study is to develop a robust set of microsatellites markers (MS marker) for traceability and individual identification. Using multiplex-PCR method with 23 MS marker divided 2 set, various alleles occurring to 5 swine breed (Berkshire, Landrace, Yorkshire, Duroc and Korea native pig) used markers to determine allele frequency and heterozygosity. MS marker found 4 alleles at SW403, S0227, SWR414, SW1041 and SW1377. The most were found 10 alleles at SW1920. Heterozygosity represented the lowest value of 0.102 at SWR414 and highest value of 0.861 at SW1920. So, it was recognized appropriate allele frequency for individual identification in swine. Using multiplex-PCR method, MS markers used to determine individual identification biomarker and breed-specific marker for faster, more accurate and lower analysis cost. Based on this result, a scientific basis was established to the existing pedigree data by applying genetics additionally. Swine traceability is expected to be very useful system and be conducted nationwide in future.

Estimation of genetic relationships between growth curve parameters in Guilan sheep

  • The objective of this study was to estimate variance components and genetic parameters for growth curve parameters in Guilan sheep. Studied traits were parameters of Brody growth model which included A (asymptotic mature weight), B (initial animal weight) and K (maturation rate). The data set and pedigree information used in this study were obtained from the Agricultural Organization of Guilan province (Rasht, Iran) and comprised 8647 growth curve records of lambs from birth to 240 days of age during 1994 to 2014. Marginal posterior distributions of parameters and variance components were estimated using TM program. The Gibbs sampler was run 300000 rounds and the first 60000 rounds were discarded as a burn-in period. Posterior mean estimates of direct heritabilities for A, B and K were 0.39, 0.23 and 0.039, respectively. Estimates of direct genetic correlation between growth curve parameters were 0.57, 0.03 and -0.01 between A-B, A-K and B-K, respectively. Estimates of direct genetic trends for A, B and K were positive and their corresponding values were $0.014{\pm}0.003$ (P < 0.001), $0.0012{\pm}0.0009$ (P > 0.05) and $0.000002{\pm}0.0001$ (P > 0.05), respectively. Residual correlations between growth curve parameters varied form -0.52 (between A-K) to 0.48 (between A-B). Also, phenotypic correlations between growth curve parameters varied form -0.49 (between A-K) to 0.47 (between A-B). The results of this study indicated that improvement of growth curve parameters of Guilan sheep seems feasible in selection programs. It is worthwhile to develop a selection strategy to obtain an appropriate shape of growth curve through changing genetically the parameters of growth model.

Characteristics of A New Flue-cured Tobacco Mutant Line KF 8832-85 (황색종 연초 돌연변이 계통 KF8832-85의 특성)

  • A new flue-cured tobacco mutant line BU 8832-85 was developed at Taegu Experiment Station, Korea Ginseng and Tobacco Research Institute in 1994. KF 8832-85 was resulted from a cross of flue-cured cultivars NC 95$\times$NC 2326, and developed by a pedigree system of breeding ; initial selection was made by plant type and resistance to bacterial wilt(BW) disease(heudomonas solanaceamm) in the F2 generation under the natural field conditions infested with the pathogen. One white flowered plant was occurred by spontaneous mutation in a certain line among the F3 generatioin while the others were pink. Six plants from the seeds by selfing were selected at the field infested with the pathogen among 240 populations with white flowering in the F4, KF 8832-85 was selected based on yield and leaf quality trials among 6 lines in Fs generation. BCF 8832-85 was compared with its Parent for certain agronomic and chemical characteristics at Taegu Experiment Station in 1993 and 1994. The results showed that KF 8832-85 have white flower, the stalk height was approximately that of NC 2326 but averaged about loom taller than NC 95. It produced ground suckers as much as NC 95, and did not breakdown leaf at the same as WC 2326. KF 8832-85 have high resistance to bacterial wilt disease. Yield of KF 8832-85 was 10 and 18% higher then that of NC 2326 and WC 95, respectively. Price per Kg was equal to that of NC 2326. The contents of nicotine and reducing sugar did not differ significantly from NC 95, while total nitrogen was significantly lower than NC 95. Therefore, the new mutant line is genetically stable for agronomic and chemical characteristics and provides a source of bacterial wilt disease resistance for use in breeding resistant flue-cured cultivars.   • A set of definitions for traceability and quick recall service function were urgently needed to facilitate handling of events threatening food safety caused by livestock diseases or germ contaminations. However, the research on the unified management system for both international and domestic livestock products distribution was scarce. The livestock products traceability evaluation model proposed in the paper was composed of four modules: the Forward Transaction Trace (FTT) module, the Backward Record Trace (BRT) module, the Forward Product Trace (FPT) module, and the Origin Pedigree (OP) module. The evaluation indexes for each module was included and finally a pilot system evaluating the proposed management model was suggested and displayed. The result of the paper was expected to be a solution to the distributed traceability system and the proposed traceability management model could be expected to any food traceability.

A New Perilla Cultivar for Edible Seed 'Dayu' with High Oil Content (기름함량이 높은 종실용 들깨 신품종 '다유')

  • 'Dayu', a perilla (Perilla frutescens (L.) Britton) cultivar for edible seed was developed by Department of Functional Crop, NICS, RDA in 2004. It was developed from a cross between a pedigree of Daeyeop as a female and YCPL1846 as a male parent in 1994. 'Dayu' could be characterized by white flower color and brown grain color. Dayu shows 127 cm in plant height, and has lodging tolerance. Maturing date of 'Dayu' was October 6, which was similar with that of 'Saeyeopsil'. This new cultivar has high oil content with 48.5% and high linolenic acid in the fatty acid composition. The grain yield potential of 'Dayu' is about 1.44 MT/ha in the regional yield trial.
