RNase Resistant RNA in the Egg of Xenopus laevis: I. RNA Extraction and in Vitro Labeling

  • RNA was extracted from the eggs of Xenopus laevis to do preliminary experiments before testing the possibility that if RNase resistant RNA molecules exist in the amphibian egg. Chromatography on Sephadex G-100 column indicated 3 peaks consistently. Only high molecular weight RNA species eluted in the first peak were labeled in vitro using $^{3}H$-dimethyl sulfate to eliminate the possible contribution of base paired oligonucleotides from tRNA. By this method, high specific activity could be obtained and the attached methyl groups were quite stable.

Aerodynamic shape optimization of a high-rise rectangular building with wings

  • The present paper is focused on analyzing a set of Computational Fluid Dynamics (CFD) simulation data on reducing orthogonal peak base moment coefficients on a high-rise rectangular building with wings. The study adopts an aerodynamic optimization procedure (AOP) composed of CFD, artificial neural network (ANN), and genetic algorithm (G.A.). A parametric study is primarily accomplished by altering the wing positions with 3D transient CFD analysis using k - ε turbulence models. The CFD technique is validated by taking up a wind tunnel test. The required design parameters are obtained at each design point and used for training ANN. The trained ANN models are used as surrogates to conduct optimization studies using G.A. Two single-objective optimizations are performed to minimize the peak base moment coefficients in the individual directions. An additional multiobjective optimization is implemented with the motivation of diminishing the two orthogonal peak base moments concurrently. Pareto-optimal solutions specifying the preferred building shapes are offered.

  • To elucidate succession of bacterial populations, especially nitrifying bacteria during the composting of cattle manure with apple pomace, fluorescent in situ hybridization(FISH) using rRNA targeted oligonucleotide probes were applied. The density of ammonia-oxidizing bacteria was ranged from $3,3{\times}10^6cells/g$ dw to $13,4{\times}10^6cells/g$ dw with the peak value after 26 composting days whereas that of nitrite-oxidizing bacteria varied between $6.0{\times}10^6cells/g$ dw and $17.2{\times}10^6cells/g$ dw with the peak value after 7 composting days. And the tendency that the numbers of nitrite-oxidizing bacteria were higher than those of ammonia-oxidizing bacteria, and the peak-time of their densities were the same as that of data determined by the ratio of ammonia-oxidizing bacteria and nitrite-oxidizing bacteria to eubacteria. The peak of ammonia-oxidizing bacteria followed the peak of nitrite-oxidizing bacteria, at the late phase of composting process could be probably caused by the depletion of volatile ammonia of composting materials. Besides these results indicate that FISH method is a useful tool for detection of slow growing nitrifying bacteria.

  • Single crystalline I $n_{x}$G $a_{1-x}$ N thin film was grwon by MOCVD on (001) sapphire substrate for the blue light emitting devices. A good quality of I $n_{0.13}$G $a_{0.87}$N/GaN heterostructure grwon above 700.deg. C was confiremed by various characterization techniques of AFM, RHEED and DC-XRD. Through PL measurement at room temperautre for the Si-Zn co-doped I $n_{x}$G $a_{a-x}$N/GaN structure grwon at 800.deg. C to obtain blue wavelength emission, 460-470 nm and 425 nm emission peak were observed, which are believed to be from donor-to-acceptor pair transition and band edge emission of In/x/G $a_{1-x}$ N, respectively. The result of PL measurement of the undoped MQW I $n_{x}$G $a_{1-x}$ N layer at low temperature confirmed that the strong MQW peak was resulted by exciton from the GAN barrier and carrier of DA pair confined into the well layer.ll layer.yer.r.

Biomass Production and Phosphorus Inflow in three Perennial Herb Populations in the Basin of the Mt. Geumoh (금오산분지의 삼종 다년생 초목식물 개체군의 식물량생산과 인의 유입)

  • Seasonal changes in pool size, inflow rates in biomass and phosphorus, and the efficiency of phosphorus use in the stand of three populations (Helianthus tuberosus, Artemisia princeps and Phalaris arundinacea) in the basin of the Mt. Geumoh were investigated. During the early growing period, in the three species populations the relative size of the phosphorus pool of population was larger then that of its biomass pool, but that of the phosphorus pool of belowground part decreased more rapidly than that of its biomass pool. In the A. princeps and P. arundinacea populations, the phosphorus inflow rate was markedly high during the soil thaw in early spring and its seasonal change pattern was different from that of the biomass production rate, showing two peaks in March and June. But in the H. tuberosus population, the two seasonal change patterns were alike. The annual biomass production was 2283 gDM m-2 in the H. tuberosus, 1884 m-2 in the A. princeps and 1879 gDM m-2 in the P. arundinacea population, and the annual phosphorus inflow was 11.35, 9.63 and 7.60 gP m-2, respectively. The P. arundinacea population showed the smallest LAI peak(5.4 in early June), and the largest NAR peak (36.9 gDM m-2wk-1) RGR peak (0.15g g-1 wk-1) among the three species populations. The seasonal change patterns in whole plant EPU of the three species populations showed the bell shape, but the annual EPU values among them were markedly different. It was noticed that the population with the highest RGR showed the highest EPU among the three species populations while the population with the lowest RGR showed the lowest EPU among them.

Are Head Impacts Safe during Youth Soccer Game Practice? (유소년 축구 경기에서 발생하는 머리 충격은 안전할까?)

  • Objective: This study to identify the mechanism of head impact that occurs during youth soccer game with regard to head injuries in sports. Method: Ten male subjects (age: 10.0±2.0 yrs.) were participated during 10 soccer practices spread out over a time period of 10 weeks. During each soccer game, the participants agreed and wore the X-Patch (wireless accelerometer, gyroscopes). The X-Patch records the head impact mechanics, such as peak linear acceleration (PLA), peak rotational acceleration (PRA), peak rotational velocity (PRV), Head Injury Criterion (HIC), and the location of impact. Results: A total of 501 impacts to the head were measured over the 10 soccer games, PLA 17.8±10.4 g, PRA 3168±2442 rad/s2; PRV 16.1±10.6 rad/s; HIC 11.7±34.2. The severity of impact was classified into 3 ranges; low 10~39 g (482 impacts); medium 40~69 g (17 impacts); and high >69 g (2 impacts). There are no significant differences in PLA and HIC (p=0.08, p=0.15), however PRA and PRV show the differences (p<.05) between each of the participants. For the analysis comparing between the soccer games, there are no significant differences in PLA, PRA, PRV and HIC (p=0.11, p=0.13, p=0.14, p=0.05). Conclusion: Our results indicated that there were significant differences between athletes, especially in terms of rotational acceleration, whereas there were significant differences in linear and rotational based variable between each of the soccer games. Although the vast majority of impacts were below 39 g there were 2 potentially dangerous impacts above 69 g. It is important that future research continuous to measure head impact mechanics during soccer to help understand head injury mechanisms to ensure the safety of athletes.

  • The gas-gun test is a experimental approach employed to validate the operational or structural stability when subjected to the impact energy encountered during launch or target collision. Predicting the outcomes of the gas-gun test has traditionally relied on empirical knowledge, due to numerous factors such as the bird assembly's shape, weight, material, and flight velocity. However, due to the nonlinearity and complex interactions between these variables, numerous tests are necessary to identify the necessary requirements, resulting in significant expense and time consumption during the process. The objective of this study is to forecast the variations in impact energy in future tests by developing a numerical model and analysis that aligns with the test outcomes, utilizing the ABAQUS Explicit. The outcome of the numerical analysis produced a framework that anticipates the peak g and the duration of the actual gas-sun test results, throughout post-processing techniques using FFT and LPF filters.

Modeling Method for the Force and Deformation Curve of Energy Absorbing Structures to Consider Initial Collapse Behaviour in Train Crash (열차 충돌에너지 흡수구조의 초기붕괴특성을 고려하기 위한 하중-변형 곡선 모델링 방법)

  • The Korean rolling stock safety regulation stipulates that the collision deceleration of a car body should be maintained under average 5g and maximum 7.5g during train collisions. One-dimensional dynamic model of a full rake train, which is made up of nonlinear springs/bars-dampers-masses, is often used to estimate the collision decelerations of car bodies in a basic design stage. By the way, the previous studies have often used some average force-deformation curve for energy absorbing structures in rolling stock. Through this study, we intended to analyse how much the collision deceleration levels are influenced by the initial peak force modeling in the one-dimensional force-deformation curve. The numerical results of the one-dimensional dynamic model for the Korean High-Speed Train show that the initial peak force modeling gives significant effect on the collision deceleration levels. Therefore the peak force modeling of the force-deformation curve should be considered in one-dimensional dynamic model of a full rake train to evaluate the article 16 of the domestic rolling stock safety regulations.

A Study on the Thermal Decomposition Characteristics of Nitrophenylhydrazine (니트로페닐하이드라진의 열분해 특성에 관한 연구)

  • For handling and storage of reactive chemicals, the hazard evaluations have been extremely important. In the chemical industry, the most concerns are focused on the thermal harzards such as runaway reactions and thermal decompositions, which are mostly governed by thermodynamics and reaction kinetics or these reactive chemical in the system. This study no investigated the thermal decomposition characteristics of nitrophenylhydrazine isomers by using differential scanning calorimeter(DSC) and accelerating rate calorimeter(ARC). Experimental results showed that exothermic onset-temperatures in nitrophenylhydrazine(NPH) isomers were about 160-$210^{\circ}C$ by DSC and 100-$150^{\circ}C$ by ARC. The decomposition temperature acquired by ARC was about 50-$60^{\circ}C$ lower than that by DSC. Reaction heats were about 40-100cal/g by DSC and 330-750ca1/g by ARC. While ortho isomer of NPH show two distinct exothermic peaks, para isomer shows a single peak in DSC curves. The first exothermic peak for 2-NPH is mainly due to intramolecular dehydration forming 1-hydroxybenzotriazole(HOBT) and the second exothermic peak is mainly due to the decomposition of HOBT formed in the first step of decomposition. The exothermin peak in the DSC curve for 4-NPH is mainly due to dissociation of hydrazino and nitro groups.
