The Effects of Ostrich Extract Combined with Korean Herb Medicine on the Serum Parameters in Carageenan Induced Paw Edema in Rats (타조 한방 엑기스 제품 급여가 부종 유발 흰쥐의 혈청지질 및 무기질 수준에 미치는 영향)

  • In this study, we investigated the effects of ostrich extract combined with Korean herb medicine(OEKHM) on the serum parameters in carageenan induced paw edema in rats. Carageenan causes enhanced edema formation which contributes to the pathophysiology of inflammation. Male Sprague-Dawley rats were used. There were four groups of treatments: OEKHMs and the control. After treatment with oral doses of 3 mL/180 g BW. 3 kinds of OEKHMs were processed which were ostrich bone with flesh meat with herb medicine(BFH), ostrich bone with flesh meat(BF), and ostrich bone with internal organs(BI). The contents of crude fat of the BFH was lower, but crude protein and crude ash of the BFH were significantly higher than those of the BF and the BI. In case of the levels of minerals(Ca, P, Mg, Fe, Cu and Zn ) of the BFH were significantly higher than those of BF and BI. And the heavy metals(Pb and Cd) were not detected in the OEKHM. The serum level of triglyceride in the BFH treated group decreased compared to that of the edema induced group. The serum levels of HDL-cholesterol concentration in the BFH supplemented group increased compared to that of the other groups. The serum levels of minerals(Ca, Mg, Fe, Cu and Zn) of the BFH treated group significantly increased than those of other groups(p<0.05).

Inhibiting Effects of Hwallak-dan(Huoluo-dan) on the Adjuvant-induced Arthritis in Lewis Rats (활락단(活絡丹)의 Adjuvant 유발 관절염 진행 억제 효과)

  • Objectives : This study was to investigate the antiarthritic effect of Hwallak-dan(Huoluo-dan) on the complete freund's adjuvant(CFA)-induced arthritis in Lewis rats. Methods : Arthritis was induced by intradermal injection of CFA into base of tail. Experimental groups divided into normal(n=10), control(n=10) and treated(n=10) group. Animals of control group received normal saline for twenty days, treated group received extracts Hwallak-dan(Huoluo-dan) for same duration, light mineral oil only injected non-arthritic rats were served as normal group. The incidence of arthritis and arthritic index were observed after treatment. Body weight, paw edema volume and thickness of ankle joint were measured at 0, 11, 14, 17, 20 days after treatment. White blood cell(WBC) counts in blood were analysed at 20 days after treatment and tumor necrosis $factor-{\alpha}(TNF-{\alpha})$, $interleukin-1{\beta}(IL-1{\beta}$), IL-6 contents in paw exudate were analysed by ELISA at 20 days after treatment. Histopathology on the ankle joint were performed at 20 days after treatment. Results : 1. Incidence of arthritis of treated group was 60% and control group was 100% at 20 days after treatment. 2. Arthritic index of treated group was significantly decreased compared with control group at 20 days after treatment. 3. Paw edema volume and thickness of ankle joint of treated group was significantly decreased compared with control group at 20 days after treatment. 4. Total WBC of treated group was significantly decreased compared with control group at 20 days after treatment. 5. Neutrophils of treated group was significantly decreased compared with control group at 20 days after treatment. 6. $TNF-{\alpha}$ content in paw exudate of treated group was significantly decreased compared with control group at 20 days after treatment. 7. $IL-1{\beta}$ content content in paw exudate of treated group was significantly decreased compared with control group at 20 days after treatment. 8. IL-6 content content in paw exudate of treated group was significantly decreased compared with control group at 20 days after treatment. 9. Histopathological arthritic index of treated group was significantly decreased compared with control group at 20 days after treatment. Conclusions : These results indicate that Hwallak-dan(Huoluo-dan) has inhibitory effect on the development and progression of CFA-induced arthritis in rats.

Simultaneous determination of baicalin and glycyrrhizin in Sosiho-tang by HPLC and anti-inflammatory activity (소시호탕에서 baicalin과 glycyrrhizin 동시 분석 및 항염 활성)

  • Objectives : To quantitate the main compounds and investigate the biological activity of Sosiho-tang (Xiao-Chai-Hu-Tang, SST), simultaneous determination of baicalin and glycyrrhizin, and anti-inflammatory activity were estimated. Methods : A quantitative analysis was performed using a high performance liquid chromatography (HPLC). Reference compounds were separated on a reversed-phase column using gradient elution with water and acetonitrile each containing acetic acid at a flow rate of 1 mL/min. And the productions of nitric oxide (NO) and prostaglandin $(PE)E_2$ were examined by lipopolysaccharide (LPS)-treated RAW 264.7 cells in the presence of the SST. The anti-inflammatory activity of SST was investigated by carrageenin-induced paw edema in rats. The paw volume was measured at 2 and 4 hr following carrageenin-induced paw edema in rats. Results : The correlation coefficients of the compounds showed good linearity ($r^2$ > 0.9992) over the linear range. The precisions of intra- and inter-day were less than 7.0% of relative standard deviation (RSD) values for baicalin and less than 3.5% of RDS valuse for glycyrrhizin. Recovery rates were within the range of 95.41-101.5%. The contents of baicalin and glycyrrhizin in SST were average 70.52, 6.18 mg/g, respectively. And SST exhibited inhibitory effect on NO production in LPS-treated RAW 264.7 cells but not on $PGE_2$ production. Oral administration of SST (1 g/kg) showed a reduction in carrageenin-induced paw edema on rats. Conclusions : The analytical method was applied successfully to measure the contents of baicalin and glycyrrhizin in SST which exhibited anti-inflammatory activities.

Research on Effects of Cordyceps Sinensis in Collagen Induced Arthritis Mouse-Model. (동충하초 추출물이 콜라겐으로 유발된 관절염 생쥐 모델에 미치는 영향)

  • Objectives : We examined to know the effect of Cordyceps sinensis(CS) on arthritis mouse induced by type II collagen. Methods : To analyse immunomodulatory effects of CS, arthritis index, incidence, hind paw edema, DTH, spleen weight, the number of hemocytes, and surface-receptor expression of CD4+, CD8+ and CD19+ cells in DBA/IJ mice which is experimental model of arthritis induced by collagen were measured in vivo. Results: CS reduced arthritis index, incidence, hind paw edema and DTH significantly as compared with the control group in experimental model of arthritis induced by collagen. CS enhanced the spleen weight significantly as compared with the control group but didn't enhanced the number of leukocytes and thrombocytes in experimental model of arthritis. CS enhanced the number of activating cells and surface-receptor expression of CD4+, CD8+ cells as compared with the control group but didn't enhanced those of CD19+ cells in experimental model of arthritis. Conclusion : We found out that CS may have a suppressing effect againist auto-immune disease and will be need continuous research in looking for the more effective mechanism in the future.

  • Experimental studies were conducted to investigate for the effect of Tongkyu-tang on analgesic, antipyretic, sedative, anti-inflammatory, isolated ileum and anti-histamine actions. Tongkyu-Tang was composed of sixteen crude drugs including Astragali Radix, which has been used for the treatment of common cold, headache, rhinitis, nasal obstruction, rhincospasm, etc. The following results have been obtained; analgesic and antipyretic actions were obtained. Prolonged action against the hypnotic duration of pentobarbital were significantly noted in mice. Anti-inflammatory effects in the paw edema induced by 1% carrageenin and 1% dextran were significantly shown in rats. Spontaneous motilities of isolated ileum of mice were strongly suppressed, and contractions of isolated ileum of mice and guinea-pigs induced by acetylcholine chloride, barium chloride and histamine were remarkably inhibited. Antihistamine actions were noted. Considering the above experimental results, it is suggested that effects of Tongkyu-tang based on the Oriental Medical References were similarly consistent with the actual experimental results.

Sulfuretin, an Antinociceptive and Antiinflammatory Flavonoid from Rhus verniciflua

  • This study was undertaken to evaluate the antinociceptive and antiinflammatory effects of the heartwood extract of Rhus verniciflua (Anacardiaceae) and the two major components, sulfuretin and fustin. The MeOH extract, its EtOAc-soluble portion and sulfuretin showed significant antinociceptive activity in writhing and hot plate test assays and antiinflammaory effects in carrageenan-induced hind paw edema in rats. In particular, treatment of sulfuretin with 10 mg/kg dose (i.p.) reduced writhing frequency by 48.0% (p<0.01) compared to that of a control group. Further, the treatment of sulfuretin (5, 10 mg/kg, i.p.) for 7 days prevented the carrageenan-induced hind paw edema significantly (p<0.01). The antiinflammatory effect of sulfuretin was also confirmed by microscopic observation of mast cell numbers in muscle. In addition, sulfuretin suppressed the cyclooxygenase- 2 (COX-2) activity $(IC_{50}\;=\;28.7\;{\mu}M)$ in lipopolysaccharide-activated macrophage cells. This result indicates that the inhibitory effect of sulfuretin on COX-2 may be one of the antinociceptive/antiinflammatory mechanism.

Effects of High Frequency Electro-acupuncture ($ST_{36}$) on the Adjuvant-induced Arthritis in Rats (족삼리에 시술한 고빈도 전침자극이 류마토이드 관절염 동물모델에 미치는 영향)

  • Objectives: This study was to investigate the effect of high frequency electro-acupuncture (EA) at $ST_{36}$ acupoint on the Freund's Complete Adjuvant (FCA)-induced arthritis in rats. Methods : Arthritis was induced by intradermal injection of FCA into base of tail. Experimental groups were divided into 4 groups; Normal, Control, $ST_{36}$ and Non-Acupoint (NA). $ST_{36}$ group was treated by 120Hz EA at $ST_{36}$ acupoint. Body weight, paw edema volume and ankle joint thickness were measured after treatment. And we investigated the effects of 120Hz EA via WBC count, segmen neurophil, lymphocyte, PGE2 assay and NADPH-d histochemistry. Results and Conclusions : The mean of body weight and ankle joint thickness of $ST_{36}$ group was increased compared with control group. The mean of paw edema volume, WBC count, segment neutrophil and lymphocyte of were not decreased with control group. Suppressive Effects of Potato (Solanum tuberlosum) on Type II Collagen-Induced Arthritis in DBA/1J Mice

  • Collagen-induced arthritis (CIA) is a model for some types of human autoimmune rheumatoid arthritis (RA). In this study, we examined whether ethanol extract of potato (Solanum tuberosum) is efficacious against CIA in mice. Potato extracts (100 and 200 mg/kg) were orally administered to DBA/1J mice once daily for 49 day after initial immunization with type II collagen. Clinical assessment of disease and measurement of paw edema were conducted throughout the study. The production of CIA-related rheumatoid factor, anti-type II collagen antibody, and cytokines were examined in DBA/1J mice. Serum levels of AST, ALT, creatinine, and lipids were measured, and antioxidant enzyme activity in the spleen was also determined. The arthritis score and paw edema were markedly suppressed in the groups treated with potato extract. Levels of rheumatoid factor, anti-type II collagen antibody, interleukin (IL)-1, IL-6, LDL-cholesterol, and malondialdehyde in sera were also reduced by potato extract treatment. The activities of glutathione peroxidase and glutathione reductase were increased in the spleens of CIA mice treated with potato extract. These findings suggest that potato extract has suppressive effects on type II collagen-induced arthritis, an animal model for human RA.

Antibacterial Activity against the Streptococcus mutans and Anti-inflammatory Effect of Lophatheri Herba (담죽엽의 충치균에 대한 항균활성 및 항염효과)

  • Lophatheri Herba of Lophatherum gracile Bronghiart(Gramineae) has long been used for treatment of inflammation, fever and edema in Korea. In order to investigate antibacterial activity of the Lophatheri Herba against Streptococcus mutans ATCC27351, paper disc test and pH check were carried out with 80% ethanol extract of Lophatheri Herba(LEX). The LEX showed significant antibacterial activity. And at the dose of 50 mg/kg, LEX showed signifiant inhibition on the paw edema, vascular permeability and myeloperoxidase activity in rat's paw tissue. These results indicate that LEX has antibacterial activity against the Streptococcus mutans and anti-inflammatory effect.

Antioxidant and anti-inflammatory activities from fruiting body extracts of Lyophyllum decastes

  • Lyophyllum decastes has been used for culinary purpose. The present study was conducted to evaluate the antioxidant and anti-inflammatory effects from methanol, acetone, and hot water extracts of L. decastes fruiting bodies. The acetone and methanol extracts showed the higher 1,1-diphenyl-2-picryl-hydrazy radical scavenging activities than that of the hot water extract at 0.5-2.0 mg/mL and was comparable to the BHT, the positive control. The ferrous ion chelating effects of the mushroom extracts at 0.5-2.0 mg/mL were significantly higher than that of BHT. The reducing power of acetone extract (2.12) was significantly lower than that of BHT (2.73) at 2.0 mg/ mL. The mushroom extracts also showed inhibitory effects on production of nitric oxide (NO), and expression of inducible nitric oxide synthase (iNOS) in lipopolysaccharide-induced murine macrophage cells in a concentration dependent manner. In vivo anti-inflammatory experiment on carrageenan-induced hind-paw edema of rat model, the acetone extract of the mushroom significantly suppressed the carrageenan-induced rat hind paw edema of rats in a dose dependently. The results suggest that the fruiting bodies of Lyophyllum decastes are a good natural resource of antioxidant and anti-inflammation.
