고추 탄저병균의 배양형 변이 그리고 병원성 차이 (Red Pepper Anthracnose: Colletotrichum gloeosporioides, It's Cultural Variations and Pathogenicity)

  • 붉은 고추 탄저병균의 우점종인 Colletotrichum gloeosporioides는 C. acutatum에 비해 병원성이 강하였다. C. gloeosporioides는 G와 R계통이 존재하였으며, G계통이 R계통보다 더욱 강한 병원성을 보였다. 붉은 고추 품종간의 병원성 검정 결과, 금세기 품종이 가장 감수성이었고, 파페트 품종이 가장 약한 감수성이었다. 참깨, 홍화, 마, 그리고 딸기와 같은 다른 기주에서 분리되어진 C. gloeosporioides는 붉은 고추에 감염을 일으켰다.

First Report of Leaf Spot of Datura metel Caused by Alternaria tenuissima in Korea

  • In June 2013, we collected leaf spot disease samples of Datura metel from Gangneung, Gangwon Province, Korea. The symptoms observed were small circular to oval dark brown spots with irregular in shape or remained circular with concentric rings. We isolated the pathogen from infected leaves and cultured the fungus on potato dextrose agar. We examined the fungus morphologically and confirmed its pathogenicity according to Koch's postulates. The results of morphological examinations, pathogenicity tests, and the rDNA sequences of the internal transcribed spacer regions (ITS1 and ITS4), glycerol-3-phosphate dehydrogenase (G3PDH) and the RNA polymerase II second largest subunit (RPB2) gene sequence revealed that the causal agent was Alternaria tenuissima. To the best of our knowledge, this is the first report of leaf spot of D. metel caused by A. tenuissima in Korea as well as worldwide.

First Report of Postharvest Gray Mold Rot on Carrot Caused by Botrytis cinerea in Korea

  • In February 2014, gray mold rotting symptoms were observed in carrots in cold storage at Gangneung, Gangwon province, Korea. The typical symptom of gray mold rot showed abundant blackish gray mycelia and conidia was observed on the infected root. The pathogen was isolated from infected root and cultured on PDA for further fungal morphological observation and confirming its pathogenicity according to Koch's postulates. Results of morphological data, pathogenicity test and rDNA internal transcribed spacer (ITS 1 and 4) sequence showed that the postharvest gray mold rot of carrot was caused by Botyrtis cinerea. This is the first report of postharvest gray mold rot on carrot in Korea.

  • Stem rot of cacti was found at major cultivating areas including Koyang, Ansung and Eumsung of Korea in 2000. Bipolaris cactivora was consistently isolated from the lesions. The disease occurred on different species of cactus including Cereus peruvianus, C. neopithahaja f. monstruo년, C. tetragonus, Chamaeceresu silestrii, Ch. silvestrii, f. variegata, Gymnocalcium mihanovichii var. friedrichii. G. denudatum var. pentacantha, Hylocereus trigonus and Isolatocereus dumortier. Major symptoms on the cactus species except H. trigohus were almost identical. A rapid rot of the upper portion of the catus stem appeared, and became blackened and somewhat dry. On H. trigonus, the symptom was initially light yellow, water-soaked lesion, turned into light brown and dried to death. According to pathogenicity test, 10 out of 16 cactus species and varieties tested produced identical symptoms as found in the field. However, the fungi did not show pathogenicity to Notocactus scopa, Echinocactus grusonii, Eriocactus leninghausii, Lobivia nealeana, Mammillaria elongata var. intertexta.

Pathogenicity of viral hemorrhagic septicemia virus (VHSV) isolated from olive flounder Paralichthys olivaceus to masu salmon Oncorhynchus masou

  • The pathogenicity of viral hemorrhagic septicemia virus (VHSV) from olive flounder Paralichthys olivaceus was investigated with masu salmon Oncorhynchus masou fry. The cumulative mortality of fish challenged with FYeosu05 isolate at $10^{6.5}$ $TCID_{50}$/fish was 60%. No mortality was observed in fish challenged with the isolates at $10^{5.5}$ $TCID_{50}$/fish and in mock-challenged fish. The affected fish showed darkening of the body, expanded abdomen, pale gills and enlarged spleen. VHSV from $10^{6.3}$ to $10^{7.8}$ $TCID_{50}$/g-tissue was re-isolated from the dead fish. These results suggest that the VHSV from olive flounder is pathogenic to masu salmon fry, although with low virulence.

Migrations and Multiplications of Bursaphelenchus xylophilus and B. mucronatus in Pinus thumbergii in Relation to Their Pathogenicity

  • To evaluate the mechanisms of pathogenicity and nonpathogenicity of Bursaphelenchus xylophilus and B. mucronatus isolated in South Korea, we used 4-year-old P. thunbergii seedlings and 20-cm long one-year-old stem cuttings of 5-year-old Pinus thunbergii, and studied distributions and multiplications of pine wood nematodes after inoculation. The distributions of B. xylophilus in the 20-cm pine stem cuttings were not significantly different from that of B. mucronatus. Conversely, the proliferation rate of B. xylophilus on mycelial mats of Botrytis cinerea was significantly different from that of B. mucronatus. The study using 4-year-old P. thunbergii seedlings also showed that B. mucronatus can migrate to distal portions of the pine seedlings the same as B. xylophilus, but the populations of B. xylophilus remaining in the pine seedlings were relatively larger than those of B. mucronatus. Therefore, we concluded that the pathogenicity of B. xylophilus could be strongly influenced by its ability to multiply.

  • Botrytis cinerea E12 was isolated from the leaves, flowers and fruits of eggplant in the greenhouse in Halrim, Kimhae and Dejeo, Pusan. The leaves infected with the pathogen were appeared initially brown-color, small gray spots at the edge, and finally fall down. The fruit was showed the symptoms of circular or irregular shapes, followed by sunken. When the symptoms were developed, the conidia formed on the surface with gray color. To determine the pathogenicity of B. cinerea E12 against the eggplants, the conidia were suspended with 30% tomato juice, PDB and sterile water, respectively. The result showed that the conidial suspension with 30% tomato juice was highly effective on the pathogenicity as more than 90%. Moreover, the symptoms caused by inoculum were the same as those of wild-type pathogen.

The First Report of Postharvest Stem Rot of Kohlrabi Caused by Sclerotinia sclerotiorum in Korea

  • In March 2014, a kohlrabi stem rot sample was collected from the cold storage room of Daegwallyong Horticultural Cooperative, Korea. White and fuzzy mycelial growth was observed on the stem, symptomatic of stem rot disease. The pathogen was isolated from the infected stem and cultured on potato dextrose agar for further fungal morphological observation and to confirm its pathogenicity, according to Koch's postulates. Morphological data, pathogenicity test results, and rDNA sequences of internal transcribed spacer regions (ITS 1 and 4) showed that the postharvest stem rot of kohlrabi was caused by Sclerotinia sclerotiorum. This is the first report of postharvest stem rot of kohlrabi in Korea.

Rapid Identification of Potato Scab Causing Streptomyces spp. Using Pathogenicity Specific Primers

  • The potato scab is caused by several species of Streptomyces. Among these species, only pathogenic strains were found to produce thaxtomin A characterized by necrotic bioassay and HPLC. In this study, identification of the pathogenic strains of Streptomyces was performed through the polymerase chain reaction (PCR) by using specific pathogenicity primer sets derived from the nec1 gene sequences of Streptomyces scabies. The expected PCR products were obtained approximately 580 bp and confirmed by sequencing. This PCR technique can be used effectively to identify the pathogenic Streptomyces species, that cause scab on potato tubers.

Fusarium Wilt of Korean Blackberry Caused by Fusarium cugenangense

  • Wilt symptoms were frequently observed in Korean blackberry (Rubus coreanus) plants grown in farmers' fields located in Gochang and Jeongeup, Jeonbuk Province, Korea during disease surveys in July 2020. The disease occurred in 10 of the 13 fields surveyed in the two locations. The incidence of diseased plants in the fields was 5-80%. Seven isolates of Fusarium sp. were obtained from the diseased plants and examined for their morphological and molecular characteristics. All the isolates belonged to Fusarium oxysporum species complex based on the morphological characteristics but were identified as F. cugenangense based on the molecular characteristics. Two isolates of F. cugenangense were tested for pathogenicity on Korean blackberry plants by artificial inoculation. Pathogenicity of the two isolates on the plants was confirmed with the inoculation tests, which showed wilt symptoms similar to those observed in the diseased plants in the fields investigated. This is the first report of F. cugenangense causing Fusarium wilt in Korean blackberry.
