Fusarium속 균주로부터 분리한 Equisetin, Zearalenone 및 8'-Hydroxyzearalenone의 식물병원곰팡이에 대한 항균활성 (Antifungal Activities of Equisetin, Zearalenone, and 8'-Hydroxyaearalenone Isolated from Fusarium Species against Plant Pathogenic Fungi.)

  • 가지에서 분리한 F. equiseti FO-68균주와 벗풀에서 분리한 Fusarium sp. FO-510균주로부터 항균물질을 분리한 후 이들의 식물병원곰팡이에 대한 항균활성을 in vitro및 in vivo에서 조사하였다. FO-68균주의 쌀배양체로부터 하나의 항생물질을 순화하였는데 , 이 물질은 equisetin이라는 물질로 동정되었다. 그리고 FO-510균주의 쌀배양체로부터는 두 개의 항균활성 물질을 분리하였는데, 이들은 zearalenone과 8'-hydroxyzearalenone으로 동정되었다. Equisetin과 zearalenone은 in vitro에서 실험한 식물병원곰팡이 대부분에 대해서 높은 항균활성을 보였지만, 8'-hydroxyzearalenone은 거의 항균활성이 없었다. In vivo assay에서 equisetin은 토마토ㆍ잿빛곰팡이병과 토마토ㆍ역병에 방제효과가 컸으며, zearalenone은 벼ㆍ도열병, 벼ㆍ잎집무의마름병, 토마토ㆍ잿빛곰팡이병 및 토마토ㆍ역병에 대하여 효과를 나타내었다. 하지만 8'-hydroxyzearalenone는 토마토ㆍ잿빛곰팡이병을 제외한 나머지 식물병의 발생은 억제하지 못했다. Equisetin, zearalenone 및 8'-hydroxyzearalenone의 항진균활성은 본 논문에서 처음으로 보고하는 바이다.

장내 유해세균을 억제하는 양돈용 프로바이오틱스 개발을 위한 비피도박테리아 탐색 (Screening of Bifidobacteria for the Development of Probiotics Inhibiting Intestinal Pathogenic Bacteria)

  • 본 연구에서는 동물유래 시료들로부터 양돈 사료첨가용 유산균을 개발하기 위하여 enterotoxigenic E. coli, Salmonella Typhimurium, Clostridium perfringens 등 양돈산업에서 심각한 문제를 일으키는 인수공통 병원성 세균들과 Clostridium jejuni, Listeria monocytogenes, Staphylococcus aureus와 같은 식중독균을 강하게 저해하는 Bifidobacterium 유산균을 분리하고자 하였다. 이를 위하여 소의 반추위 내용물, 닭 창자 내용물, 돼지 분변 등의 시료들로부터 총 65주의 혐기성미생물을 분리하였다. 이 중에서 항병원세균활성이 가장 높은 4주를 선별하였는데, 이들은 16S rDNA 염기서열 분석방법에 의하여 3주의 B. boum과 1주의 B. thermophilum으로 동정되었다. 특히 B. thermophilum는 분리주들 중에서 가장 높은 돼지 장 상피세포에 대한 부착성을 보였고, 여러 인수공통병원세균들과 식중독균들에 대한 높은 항균력, 산과 담즙내성을 보여 양돈용 생균제 후보균주로서 우수한 특성을 나타내었다.

Antifungal Activity of Lactic Acid Bacteria Strains Isolated from Natural Honey against Pathogenic Candida Species

  • The role of lactic acid bacteria (LAB) in honey as antifungal activity has received little attention and their mechanism of inhibitory of fungi is not fully understood. In this study, LAB were isolated from honey samples from Malaysia, Libya, Saudi Arabia, and Yemen. Twenty-five isolates were confirmed LAB by catalase test and Gram staining, and were screened for antifungal activity. Four LAB showed inhibitory activity against Candida spp. using the dual agar overlay method. And they were identified as Lactobacillus plantarum HS isolated from Al-Seder honey, Lactobacillus curvatus HH isolated from Al-Hanon honey, Pediococcus acidilactici HC isolated from Tualang honey and Pediococcus pentosaceus HM isolated from Al-Maray honey by the 16S rDNA sequence. The growth of Candida glabrata ATCC 2001 was strongly inhibited (>15.0 mm) and (10~15 mm) by the isolates of L. curvatus HH and P. pentosaceus HM, respectively. The antifungal activity of the crude supernatant (cell free supernatant, CFS) was evaluated using well diffusion method. The CFS showed high antifungal activity against Candida spp. especially The CFS of L. curvatus HH was significantly (p < 0.05) inhibited growth of C. glabrata ATCC 2001, C. parapsilosis ATCC 2201, and C. tropicalis ATCC 750 with inhibitory zone 22.0, 15.6, and 14.7 mm, respectively. While CFS of P. pentosaceus HM was significantly (p < 0.05) effective against C. krusei, C. glabrata, and C. albicans with inhibition zone 17.2, 16.0, and 13.3 mm, respectively. The results indicated that LAB isolated from honey produced compounds which can be used to inhibit the growth of the pathogenic Candida species.

  • The cardiovascular drug nifedipine exhibited significant in vitro and in vivo antibacterial activity against 331 strains of bacteria belonging to three Gram-positive and twelve Gram-negative genera. The minimum inhibitory concentration of the drug, as determined both by agar and broth dilution methods, was seen to range from $25\;-\;200\;{\mu}g/ml$ against most test bacteria, including several pathogenic ones, in the in vitro studies. Nifedipine was bacteriostatic in action. in vivo studies with this drug showed that it could offer statistically significant protection (P < 0.001) to mice challenged with a virulent bacterium. Therefore, nifedipine has the potential of an antibacterial agent, which may be developed after further pharmacological studies.

A Moonlighting Protein Secreted by a Nasal Microbiome Fortifies the Innate Host Defense Against Bacterial and Viral Infections

  • Evidence suggests that the human respiratory tract, as with the gastrointestinal tract, has evolved to its current state in association with commensal microbes. However, little is known about how the airway microbiome affects the development of airway immune system. Here, we uncover a previously unidentified mode of interaction between host airway immunity and a unique strain (AIT01) of Staphylococcus epidermidis, a predominant species of the nasal microbiome. Intranasal administration of AIT01 increased the population of neutrophils and monocytes in mouse lungs. The recruitment of these immune cells resulted in the protection of the murine host against infection by Pseudomonas aeruginosa, a pathogenic bacterium. Interestingly, an AIT01-secreted protein identified as GAPDH, a well-known bacterial moonlighting protein, mediated this protective effect. Intranasal delivery of the purified GAPDH conferred significant resistance against other Gram-negative pathogens (Klebsiella pneumoniae and Acinetobacter baumannii) and influenza A virus. Our findings demonstrate the potential of a native nasal microbe and its secretory protein to enhance innate immune defense against airway infections. These results offer a promising preventive measure, particularly relevant in the context of global pandemics.

조류분 및 토양에서 분리된 수종 병원성 진균의 검색 및 병원성에 관항 연구 (Studies on the Distribution of Several Pathogenic Fungi and Its Pathogenicity Isolated from Avian Excreta and Soil)

  • This study was carried out for the identification of several fungi isolated from some different geographic regional soil samples and evaluation of its pathgenicity to the mice. isolates, The results were isolated from 200 soil samples and 11 strains from 25 pigeon excreta. 2. Seventeen of Cryptococcus neoformans, eight of Candida albicans and Microporum gypseum, six of M. coorei, and three of Sporothrix schencrii were identified group. 3. An association of C. neoformans with avian habitats was found. Ten of the 11 isolates came from areas frequently by pigeons such as pigeon nest and pigeon excreta. 4. The inflammatory reaction produced in mice that were inoculated with C. neoformans and C. albicans intraperitoneal route.

제한효소 DNA 분석법에 의한 국내분리 렙토스피라균의 동정 (Restriction Endonuclease DNA Analysis of Leptospiral Field Isolates from Korea)

  • The genomes of leptospiral field isolates from Korea belonging to serogroup Icterohaemorrhagiae (21 strains) and serogroup Canicola (1 strain) were analysed and compared by restriction enzyme analysis with EcoRI and HindIII as digesting enzymes. One isolate belonging to serogroup Canicola showed the same pattern as serovar portlandvere. All 21 isolates belonging to serogroup Icterohaemorrhagiae showed almost same patterns as Leptospira serovar lai from China, But with very slight differences 21 isolates could be classified into 8 subtypes and these grouping seems to reflect the differences in epidemiological niche. And also the geographical data consisted with the grouping into 8 subtypes. According to our results, we concluded that the restriction endonuclease analysis of chromosomal DNA will be an accurate and reliable method to compare and classify pathogenic leptospires.

Genome of Betaproteobacterium Caenimonas sp. Strain SL110 Contains a Coenzyme $F_{420}$ Biosynthesis Gene Cluster

  • To probe the genomic properties of microbes thriving in desert lakes, we sequenced the full genome of a betaproteobacterial strain (SL110) belonging to the understudied genus Caenimonas of the family Comamonadaceae. This strain was isolated from a freshwater lake in the western Gobi Desert, Northern China. Its genome contains genes encoding carbon monoxide dehydrogenase, nitrate reductase, nitrite reductase, nitric oxide reductase, and sulfur oxidation enzymes, highlighting the potentially important contribution of this group of bacteria to the cycling of inorganic elements in nature. Unexpectedly, a coenzyme $F_{420}$ biosynthesis gene cluster was identified. A further search for $F_{420}$ biosynthesis gene homologs in genomic databases suggests the possible widespread presence of $F_{420}$ biosynthesis gene clusters in proteobacterial genomes.

  • Jeotgal, salt- fermented sea food, is a kind of Korean traditional foods. We carried out hazard analysis on Changran (stomach and intestine of Alaska pollack) Jeotgal and squid Jeotgal through the whole processing procedures at H Co. located at Guryongpo from April 2000 to September 2001. During this studying period, we educated employee regularly and analyzed hazards on the whole process and then could conclude that pathogenic bacteria and metal particles were most important hazards in Jeotgal. Metal particles in end-product will be eliminated by laser detector before packaging. But bacteria must thoroughly manage through the whole process. Bacteriological qualities of the end products were much improved after education for the employees and by effort for HACCP (Hazard Analysis and Critical Control Point) program introduction. Pathogenic bacteria such as pathogenic Escherichia coli, Salmonella spp., coagulase positive Staphylococcus aureus and Vibrio parahaemolyticus were not detected from not only raw materials but also end products. The falling bacteria in the places such as thawing area, packaging area, seasoning area, fermenting room, subsidiary materials room and storage room were less than 30 CFU per plate for 30 minutes during working time. But those were increased more than 10 times during the resting time. It means that special measures are needed during the break time such as lunch time or exchanging working teams.

  • This study was attempted to see more clear relationships among the enterobacteria, especially between the intestinal normal flora and pathogenic bacteria. It has been known that some intestinal normal flora produce the bactrial metabolites that are harmful to other enteric bacteria. One of the metabolites is known as colicin, the protein fraction, which possesses certain degree of inhibitory effect against other bacterial growth fraction, whih possesses certain degree of inhibitory effect against other bacterial growth. As a preliminary study for a colicin purification, the antagonistic effect of E, coli to groups of Salmonella and Shigella has been studied by means of in vitro quantitative culture method. 1. E.coli showed definite inhibitory effects aganist both Salmonella and Shigella groups in the mixture of two organisms. 2. The inhibitory effects of E.coli in the E.coli-Salmonella and the E.coli-Shigella mixture occurred from 4 hours incubation following the inoculation. 3. Even the complete inhibition of pathogenic enteric bacterial growth was noticed in the E.coli-Salmonella mixture at overnight incubation. 4. Among the diluted mixtures, 1:100, 1:1,000, and 1:10,000, survival rate of pathogenic enteric bacteria in the mixtures with E.coli showed least affected at the 1:1,000 dilution. 5. It was found that the antagonistic effect aganist groups of Salmonella-shigella was depending upon the groups of the genera.
