사향쥐에서 Pasteurella multocida 및 Bordetella bronchiseptica에 의한 bronchopneumonia와 Klebsiellosis의 동시감염 증례 (Coincident finding of bronchopneumonia by Pasteurella multocida, Bordetella bronchiseptica and Klebsiellosis in muskrat)

  • Suppuratives bronchopneumonia was found in a 3-month old domestic muskrat (Ondatra zibethicus). Dead muskrat showed hemorrhagic nasal discharge, severe hemorrhage and consolidation were observed in the lungs in necropsy. Histologically, severe polymorphic neutrophils and alveolar macrophages were infiltrated in the bronchus, bronchioles, alveoli. P. multocida and B. bronchiseptica were identified from the lungs, Klebsiella was isolated from the cecum. We demonstrated those organisms by biochemical test and confirmed P. multocida capsular type A by means of polymerase chain reaction (PCR).

  • Pasteurella multocida is kind of commensal bacteria in the upper respiratory tract of pigs. It is classified toxigenic and nontoxigenic strains based on the production of dermonecrotic toxin. Toxigenic strain is most associated with atrophic rhinitis which brings great economical loss in swine industry. However, toxigenic and nontoxigenic strains do not differ by diagnostic biochemical reaction or morphology. One of recently developed techniques, PCR detects the toxigenic P multocida. Amplification of an 846-nucleotide fragment of toxA gene was developed. The fragment amplified by PCR was detected in P multocida type D not type A. The PCR amplification was as sensitive as it could detect 1 pg of P multocida DNA. We compared the result of the PCR with the enzyme linked immunosorbent assay (ELISA) in a test for 40 swine nasal swabs. All of these isolates were toxin negative based on the ELISA while 2 isolates were detected in the PCR technique. in addition to accuracy, as required for rapid detection from contaminated nasal swabs, toxigenic P multocida was recovered efficiently from contaminated culture without inhibition of the PCR. The results show that the PCR detection of toxigenic P multocida directly form nasal swabs are feasible.

경북지역 돼지 유래 Pasteurella multocida 의 특성 및 항생제 내성양상 (Characteristics and antimicrobial resistance patterns of Pasteurella multocida isolated from swine in Gyeongbuk province)

  • This study was conducted to investigate the species-specific gene detection, capsular serogroup and antimicrobial resistance pattern of Pasteurella multocida isolated from pneumonic lung lesion of swine in Gyeongbuk province. P. multocida isolates were typed for capsular serogroups by polymerase chain reaction. Of the 32 strains, 28 (87.5%) were typed serotype A, 3 (9.3%) were typed serotype D, and 1 strain was unknown (3.1%), respectively. In antimicrobial agents resistance test, almost of strains were susceptible to amoxicillin (100%), enrofloxacin (96.9%), ampicillin (93.8%), florfenicol (90.6%), chloramphenicol (90.6%) and were resistant to streptomycin (71.9%), spectinomycin (56.3%). All strains were resistant to clindamycin, erythromycin and lincomycin.

  • Antimicrobial susceptibility, plasmid profiles and distribution of toxA gene were investigated in Pasteurella multocida isolated from pneumonic lung lesions of swine. The bacteria were highly susceptible to norfloxacin, cabenicillin, enrofloxacin and chloramphenicol, but resistant to colistin, sulfamethoxawle/trimethoprime, bacitracin, streptomycin. Sixty percentage of the isolates was resistant more than 2 drugs used in this experiment and 21 strains (23.6%) were resistant more than 5 drugs. This phenomenon meant that they had highly multi-drugs resistance. In the analysis of plasmid profiles, nineteen strains (47.5%) of 40 P multocida isolates harbored plasmids, ranging from 53.3kb to 2.49kb in size and the plasmid profiles could be classified into 5 groups. However, there was no relationship between the size and the profile of plasmid and the resistance pattern of antimicrobial agents. Thirty strains of 39 P multocida isolates (77%) investigated by PCR harbored toxA gene. This result suggested involvement of the ToxA protein expressed from the gene in pneumonic pasteurellosis of swine.

호흡기 증상을 나타낸 소에서 Pasteurella spp의 분리 및 약제감수성 (Isolation and Antimicrobial Drug Susceptibility of Pasteurella spp from Pneumonic Calves and Cows)

  • The present study was conducted to investigate isolation and antimicrobial drug susceptibility of Pasteurella spp from pneumonic calves and cows (pneumonic lungs) in Gyeongnam south area from January to November 2000. Pasteurella multocida and P haemolytica were isolated 136 (53.8%) and 22(8.7%), respectively, from 158 of 253 pneumonic calves of 1 to 12 months of age and from pneumonic lungs and simultaneous isolation rate of P mutocida and P haemolytica was 4 (1.6%). Seasonal isolation rate of Pasteurella spp in calves and pneumonic lungs varied from 3 to 65.2%, and it was higher in Fall. The majority of biochemical and cultural properties of P multocida and P haemolytica isolated from calves and pneumonic lung were identical to those of the reference strains employed. Pneumonic lesions of the examined lungs were histologically identified as purulent bronchopneumonia and fibrinous pneumonia. All isolates were resistant to penicillin, amikacin, lincomicin, colistin, cloxacilling, sulfonamide, kanamycin, sulfamethoxasoletrimetoprim, and gentamicin(>80%), but some of them were susceptible to amoxicillin, norfloxacin, cephalexin, cefazolin, orbifloxacin, enrofloxacin, and ciprofloxacin(>60%).

돼지 위축성 비염균에 대한 항균성을 나타내는 세균의 분리 및 항균활성 (Isolation of a Bacterium with Potent Antibacterial Activity against Swine Atrophic Rhinitis Bacteria and Antibacterial Activities)

  • 돼지의 만성 호흡기 질환인 위축성 비염을 방제할 수 있는 생물공학적 방법을 개발하고자 먼저 자연계에서 돼지 위축성 비염의 원인균에 대한 항균활성이 강한 세균을 분리하여 동정하고 몇 가지 항균특성을 조사하였다. 자연계에서 항균성 물질 생산 균주로 분리한 1080주의 세균 가운데 돼지 위축성 비염의 원인균인 Bordetella bronchiseptica 와 Pasteurella multocida에 대하여 강한 항균활성을 갖는 Y-5 균주를 최종 선정한 후 동정한 결과 Bacillus sp. Y-5로 추정되었다. Y-5 균주를 pH 6.0으로 조절된 육즙배지에서 $30^{\circ}C$, 21시간 배양하였을 때 Bordetella bronchiseptica에 대하여 가장 높은 항균활성을 보였고, Y-5 균주를 $30^{\circ}C$, pH 6.5에서 18시간 배양 하였을 때 Pasteurella multocida에 대하여 가장 강한 항균활성을 보였다.

오리 및 돈(豚)으로부터 분리(分離)한 Pasteurella multocida 주(株)의 병원성(病原性)에 관(關)하여 (An Observation on Pathogenicity of the Pasteurella multocida Strains Isolated from Pig and Duck)

  • Pasteurella multocida에 대(對)한 각종성장(各種性狀) 및 균주(菌株) 상호간(相互間)의 이동(異同)에 관(關)하여는 많은 연구(硏究)가 있으나 아직까지의 문헌(文獻)에 의(依)하면 자연감염(自然感染에)에 있어서는 일반적(一般的)으로 동종동물(同種動物)에 감염(感染)한다 하여도 각종동물(各種動物)에서 분리(分離)된 Pasteurella multocida는 그 병원성(病原性)이 분리(分離)된 동물(動物)과 명확(明確)한 상관관계(相關關係)를 가지는것이 아니며 그저 생물학적(生物學的) 내지(乃至) 생화학성장(生化學性狀) 혹(或)은 면역학적성장(免疫學的性狀)에 있어서 그차이(差異)로 균형(菌型)을 분류(分類)할수 있는데 불과(不過)하다. 그런데 월지(越智)는 그의 분류(分類)에서 A형균(型菌)(조류(鳥類)에서 분리(分離)한 균주(菌株))은 가토(家兎) mouse 및 조류(鳥類)에 병원성(病原性)이 있고 타(他) 포유동물(哺乳動物)에 병원성(病原性)이 없다고 하였으며 또 B. C. D형균(型菌)(포유류(哺乳類))에서 분리(分離)한것)은 닭, 가압등(家鴨等) 조류(鳥類)에 병원성(病原性)이 없다고 하였다. 그러나 필자(筆者)는 출혈성패혈증(出血性敗血症)으로 죽은 오리 및 돈(豚) cholera와 혼합감염(混合感染)을 이르킨 돈(豚)으로부터 각각(各各) 따로 Pasteurella multocida를 분리(分離)하여 이 병균(病菌)를 가지고 각종동물(各種動物)에 접종시험(接種試驗)을 한바 상술(上述)한 월지(越智)의 견해(見解)는 부합(附合)되지 않았다. 즉 이 양주(兩株)는 돈(豚), 가토(家兎), 오리, 닭등(等)에 대(對)하여 동양(同樣)으로 강(强)한 병원성(病原性)을 가졌으며 또 양주(兩株)는 다같이 면양(緬羊)에 대(對)하여 거이 병원성(病原性)이 없고 Guinea Pig은 양주(兩株)에 대(對)한 저항성(抵抗性)이 다같이 강(强)하였다. 다시말하면 이 양주(兩株)는 조류(鳥類)나 포유류(哺乳類)에 대(對)하여 거의 동일(同一)한 병원성(病原性)이 있음을 인정(認定)하였다.

강원 동해안 지역 돼지 폐렴에서 분리한 puteurella multocida 에 대한 조사 (A Survey of Puteurella Multocida Isolated from Pigs Affected with Pneumonia in Eastern Kangwon)

  • The present study was conducted to investigate the incidence of pasteurella multocida(p. multocida) infection and some properties of the isolated organisms from the swine herds in Eastern Kangwon during the periods from March 1993 to November 1993. The results obtained were summerized as follows: 1. The lungs of 180 slaughtered pigs were sampled and p. multocida was isolated from 38 pigs (21.1%) and cultured positive. 2. The majority of biochemical md cultural properties of the p. multocida isolates were identical to those of the standard strains. 3. We investigated the capsular serogroup and drug susceptibility of 38 Isolates of p. multocida from pigs with pneumonic lesions 4. p. multocida isolateds were typed for capsular serogroupes A by hyaluronidase inhibition of capsule and D by acriflavin auto agglutination. Most isolates(55.3%) were type A, 15.8% were type D, and the remaining 28.9% were untypable 5. In antimicrobial susceptibility test these isolates of p. multocida were susceptible in order of colistin(94.7%), ampicillin(94.7%), cepalothin(92.1%). gentamicin(92.1%), amikacin(89.5%), but the majority of them were resistant in order of neomycin(26.3%), teracycline(23.7%), streptomycin( 15.8%)

Antigenicity of Partial Fragments of Recombinant Pasteurella multocida Toxin

  • Pasteurella multocida serogroup D strain, which produces P. multocida toxin (PMT), is a widespread and harmful pathogen of respiratory diseases such as pneumonia and progressive atrophic rhinitis (PAR) in swine. Vaccination has been considered the most desirable and effective approach for controlling the diseases caused by toxigenic P. multocida. To investigate the antigenicity and immunogenicity of partial fragments of recombinant PMT, recombinant proteins of the N-terminal (PMT-A), middle (PMT-B), C-terminal (PMT-C), and middle-C-terminal (PMT2.3) regions of PMT were successfully produced in an Escherichia coli expression system. The molecular masses of PMT-A, PMT-B, PMT-C, and PMT2.3 were ca. 53, 55, 35, and 84 kDa, respectively, purified by nickel-nitrilotriacetic acid (Ni-NTA) affinity column chromatography. All the recombinant proteins except for PMT-A showed immune responses to antisera obtained from a swine showing symptoms of PAR. Moreover, high titers of PMT-specific antibodies were raised from mice immunized with each of the recombinant proteins; however, the immunoreactivities of the antibodies to authentic PMT and heat-inactivated whole bacteria were different, respectively. In the protection study, the highest protection against homologous challenge was shown in the case of PMT2.3; relatively poor protections occurred for the other PMT fragments.

Simultaneous infection with rabbit hemorrhagic disease virus and Pasteurella multocida in rabbits

  • Rabbit infectious hemorrhagic fever has been reported in rabbits worldwide. The disease is also frequently reported on Korean rabbit farms, and the pathological study of 9 rabbits on such disease-occurring farms was attempted to identify the pathogen. Clinical signs were torticollis and ear ulceration. Most rabbit died with bloody nasal discharges. At necropsy, multiple hemorrhages and inflammation were observed in heart, lung, liver and uterus. The main histopathologic features were hemorrhagic suppurative meningoencephalitis, fibrinous bronchointerstitial pneumonia, bacteremia, liver cell necrosis, multifocal hemorrhages in kidney and disseminated intravascular coagulopathy. The viral VP60 gene of RHDV was identified by Reverse Transcriptase PCR. Pasteurella multocida organisms were cultured, identified by biochemical test and serotyped as A by multiplex capsular typing PCR. In conclusion, the fatal hemorrhagic disease was due to combined infection with both RHDV and P. multocida in rabbits. To our knowledge, this is the first case report about co-infection with both RHDV and P. multocida in rabbits in Korea.
