Analysis of genetic diversity of cowpea landraces from Korea determined by Simple Sequence Repeats and establishment of a core collection

  • Cowpea might have been introduced from China to Korea and cultivated for several hundred years but it has never been a staple food crop in Korea. In this study, genetic diversity of 492 Korean cowpea landrace accessions that have passport information was estimated using six SSR markers. The mean of Weir's gene diversity was 0.665 from all accessions investigated in the study. Cowpea gene diversity of six local provinces in Korea was ranged from 0.370 in accessions of Gangwon to 0.680 in Jeonra provinces. Low gene diversity of the cowpea genepool of Gangwon province was probably derived from relatively few introductions. Especially SSR markers VM36 and VM39 seem to be good markers to distinguish the Gangwon accessions from others by occurring at a specific locus with higher than 78% of allele frequency. Except for the Gangwon province with the low genetic diversity, gene diversity of cowpea accessions from other provinces was ranged from 0.600 to 0.680 indicating no big differences among provinces. Distribution pattern of the allele frequencies was similar among the other provinces. This may reveal that Korean farmers might exchange cowpea seeds easily with even their neighbors with geographical barriers. A core collection, 100 landraces, ca. 20% of base collection, was developed at the 70% of a similarity coefficient level using random sampling approaches after stratification of the entire landrace collection based on the phenetic dendrogram. The variability of SSR in the base and core collections of Korean cowpea landrace was compared by calculating Weir's gene diversity. The mean of Weir's gene diversity of the core was 0.707 while that of the base collection was 0.665. The higher diversity index in the core collection indicates that it maintains the initial variability and well represents the base collection. The core collection included one of determinate accession (IT 216155) and two of no branching type accessions (IT 103959 and IT 161024). The core collection could be used to guide more efficient management and utilization of the entire collection. This core collection should be revised periodically as additional accessions are collected and further characterization is conducted.

Assessment of the resistance of bacterial soft rot caused by Pectobacterium carotovorum subsp. carotovorum KACC 21701 in Kimchi cabbage genetic resources

  • Bacterial phytopathogen Pectobacterium causes soft rot disease in several vegetable crops globally, resulting in heavy agricultural losses at both the pre and postharvest stages. The present work was carried out to screen Kimchi cabbage genetic resources conserved at the National Agrobiodiversity Center, Rural Development Administration, Korea, for resistance against the soft rot pathogen Pectobacterium carotovorum subsp. carotovorum KACC 21701 over a period of three years (from 2020 to 2022). Infection of the phytopathogen was carried out at four-leaf stage and for each accession, twenty-five plants per germplasm were infected with KACC 21701. Kimchi cabbage cultivars Wangmatbaechu, Seoulbaechu, and CR Kiyoshi were used as control. Seven-days post-infection, the Disease Index (DI) values were manually recorded from zero to four, zero matched perfectly heathy plants and four completely dead plants. The 682 accessions of Kimchi cabbage exhibited varying degrees of disease resistance to KACC 21701 and thirty accessions, exhibiting a DI≤2, were considered for replication studies. During the replication studies, four landrace germplasms (IT102883, IT120036, IT120044, and IT120048) and one cultivar(IT187919) were confirmed to be moderately susceptible to KACC 21701. Results of the preliminary screening as well as replication studies were documented for the all the 682 germplasms. Addition of such information to the passport data of stored germplasms might serve as potential bio-resource for future breeders and researchers to develop resistant varieties or study the mechanisms involved in resistance of plants to such phytopathogen.

  • RFID that provide automatic identification by reading a tag attached to material through radio frequency without direct touch has some specification, such as rapid identification, long distance identification and penetration, so it is being used for distribution, transportation and safety by using the frequency of 125KHz, 134KHz, 13.56MHz, 433.92MHz, 900MHz, and 2.45GHz. Also it is one of main part of Ubiquitous that means connecting to net-work any time and any place they want. RFID is expected to be new growth industry worldwide, so Korean government think it as prospective field and promote research project and exhibition business program to linked with industry effectively. RFID could be used for access control of person and vehicle according to section and for personal certify with password. RFID can provide more confident security than magnetic card, so it could be used to prevent forgery of register card, passport and the others. Active RFID could be used for protecting operation service using it's long distance date transmission by application with positioning system. And RFID's identification and tracking function can provide effective visitor management through visitor's register, personal identification, position check and can control visitor's movement in the secure area without their approval. Also RFID can make possible of the efficient management and prevention of loss of carrying equipments and others. RFID could be applied to copying machine to manager and control it's user, copying quantity and It could provide some function such as observation of copy content, access control of user. RFID tag adhered to small storage device prevent carrying out of item using the position tracking function and control carrying-in and carrying-out of material efficiently. magnetic card and smart card have been doing good job in identification and control of person, but RFID can do above functions. RFID is very useful device but we should consider the prevention of privacy during its application.
