An X-linked skeletal disorder, SEDT (spondyloepiphyseal dysplasia tarda) is a genetic disease characterized by a disproportionately short trunk and short stature caused by mutations in the SEDL gene. This gene is evolutionarily conserved from yeast to human. The yeast SEDL protein ortholog, Trs20p, has been isolated as a member of a large multi-protein complex called the transport protein particle (TRAPP), which is involved in endoplasmic reticulum (ER)-to-Golgi transport. The interaction between SEDL and partner proteins is important in order to understand the molecular mechanism of SEDL functions. We isolated several SEDL-binding proteins derived from rat cells by an immobilized GST-fusion method. Furthermore, the SEDL-homologue proteins were identified using motif based methods. Common motifs between SEDL-binding proteins and SEDL-homologue proteins were classified into seven types and 78 common motifs were revealed. Sequence similarities were contracted to seven types using phylogenetic trees. In general, types I-III and VI were classified as having the function of acetyl-CoA carboxylase, glycogen phosphorylase, isocitrate dehydrogenase, and enolase, respectively, and type IV was found to be functionally related to the GST protein. Types V and VII were found to contribute to TRAPP vesicle trafficking.
A new High Frequency Induction Heating (HFIH) process has been developed to fabricate dense $Al_2O_3$ reinforced with Fe-Ni magnetic metal dispersion particles. The process is based on the reduction of metal oxide particles immediately prior to sintering. The synthesized $Al_2O_3$/Fe-Ni nanocomposite powders were formed directly from the selective reduction of metal oxide powders, such as NiO and $Fe_2O_3$. Dense $Al_2O_3$/Fe-Ni nanocomposite was fabricated using the HFIH method with an extremely high heating rate of $2000^{\circ}C/min$. Phase identification and microstructure of nanocomposite powders and sintered specimens were determined by X-ray diffraction and SEM and TEM, respectively. Vickers hardness experiment were performed to investigate the mechanical properties of the $Al_2O_3$/Fe-Ni nanocomposite.

A two-frame PIV (Particle Image Velocimetry) technique is used to investigate the wake characteristics behind a marine propeller with 4 blades at high Reynolds number. For each of 9 different blade phases from $ 0^{\circ} $ to $ 80^{\circ} $, one hundred and fifty instantaneous velocity fields are measured. They are ensemble averaged to study the spatial evolution of the propeller wake in the region ranging from the trailing edge to one propeller diameter (D) downstream location. The phase-averaged mean velocity shows that the trailing vorticity is related to radial velocity jump, and the viscous wake is affected by boundary layers developed on the blade surfaces and centrifugal force. Both Galilean decomposition method and vortex identification method using swirling strength calculation are very useful for the study of vortex behaviors En the propeller wake legion. The slipstream contraction occurs in the near-wake region up to about X/D : 0.53 downstream. Thereafter, unstable oscillation occurs because of the reduction of interaction between the tip vortex and the wake sheet behind the maximum contraction point.

In this study, the chemical compositions of $PM_{2.5}$ samples collected at the Redwood National Park IMPROVE site in California from March 1988 to May 2004 were analyzed to provide source identification and apportionment. A total of 1,640 samples were collected and 33 chemical species were analyzed by particle induced X-ray emission, proton elastic scattering analysis, photon induced X-ray fluorescence, ion chromatography, and thermal optical reflectance methods. Positive matrix factorization (PMF) was used to develop source profiles and to estimate their mass contributions. The PMF modeling identified five sources and the average mass was apportioned to motor vehicle (35.8%, $1.58\;{\mu}g/m^3$), aged sea salt (23.2%, $1.02\;{\mu}g/m^3$), fresh sea salt (21.4%, $0.94\;{\mu}g/m^3$), wood/field burning (16.1%, $0.71\;{\mu}g/m^3$), and airborne soil (3.5%, $0.15\;{\mu}g/m^3$), respectively. To analyze local source impacts from various wind directions, the CPF and NPR analyses were performed using source contribution results with the wind direction values measured at the site. These results suggested that sources of $PM_{2.5}$ are also sources of visibility degradation and then source apportionment studies derived for $PM_{2.5}$ are also used for understanding visibility problem.

Both immunoelectron microscopy(IEM) and immunogold conjugate immunoelectron microscopy (IGC-IEM) techniques were developed for the detection of porcine epidemic diarrhea virus(PEDV) from the feces. Fecal samples were incubated sequentially with anti-PEDV monoclonal antibody(MoAb) and immunogold conjugated goat anti-mouse IgG+IgM. Then negatively stained, mounted on the formvar carbon-coated copper EM grids and observed by the transmission electron microscope. By the direct electron microscopy(DEM), coronavirus particles were observed from 17 cases of total 33 fecal samples of grower pigs and sows. The virons of coronavirus were moderately pleomorphic but mostly spherical, with a diameter ranged from 90 to 190nm. PED virus particles were identified from 15 cases of 17 DEM positive samples by the IEM and IGC-IEM techniques. Aggregates of PED virus coated with specific antibody were seen in fecal samples incubated with homologous anti-PED virus MoAb but not in control samples incubated with anti-TGE virus MoAb. Following incubation with immunogold-conjugated secondary antibody, the gold granules were usually distributed around and among the virus particles and soluble and viral particle-associated antigen. So, IEM and IGC-IEM techniques were proved a rapid and sensitive methods for detection and identification of PED virus from fecal and intestinal contents.

Objectives: The aim of this study is to identify concentration characteristics of indoor and outdoor airborne total fiber particles and asbestos in Gyeongnam Provinces. Methods: This study investigated concentration characteristics of indoor fiber particles from 748 schools and 38 public facilities as well as outdoor particles from 11 sites through PCM (phase contrast microscope). SEM/EDX (scanning electron microscope/energy dispersive using X-ray analysis) was used to obtain physicochemical information of asbestos fiber particles. The study identified asbestos rate in the 15 samples from indoor and outdoor airborne total fiber particles. Results: 1. The average indoor airborne concentrations of total fiber particles were $0.0011{\pm}0007$ f/cc in schools and $0.0015{\pm}0007$ f/cc in public facilities by PCM. Over 90% of the fiber particles were identified as single fibers. 2. The average outdoor airborne concentrations of total fiber particles were $0.0007{\pm}0002$ f/cc, and they were lower than those of indoor airborne concentrations. 3. The results showed that the form of asbestiform was diverse as skein of thread like form and long needle, which was relatively narrower than that of glass fiber and rock wool. 4. The results of SEM/EDX analysis of 15 areas where total fiber particle was relatively high showed that the form was rather similar to that of asbestos, but chemical composition was proven to be non-asbestos. Conclusions: The concentration of indoor and outdoor airborne total fiber particles of Gyeongnam Provinces satisfied the IAQ (Indoor air quality) level of 0.01 f/cc and asbestos was not found in most of the samples by SEM/EDX.
Objective: The present study was to investigate the extraction conditions of dietary fiber from dried cassava pulp (DCP) and cassava distiller's dried grains (CDG) under different NaOH concentrations, and the Fourier transform infrared (FTIR) was used to determine the dietary fiber components. Methods: The dried samples (DCP and CDG) were treated with various concentrations of NaOH at levels of 2%, 4%, 6%, and 8% using a completely randomized design with 4 replications of each. After extraction, the residual DCP and CDG dietary fiber were dried in a hot air oven at 55℃ to 60℃. Finally, the oven dried extracted dietary fiber was powdered to a particle size of 1 mm. Both extracted dietary fibers were analyzed for their chemical composition and determined by FTIR. Results: The DCP and CDG treated with NaOH linearly or quadratically or cubically (p<0.05) increased the total dietary fiber (TDF) and insoluble fiber (IDF). The optimal conditions for extracting dietary fiber from DCP and CDG were under treatment with 6% and 4% NaOH, respectively, as these conditions yielded the highest TDF and IDF contents. These results were associated with the FTIR spectra integration for a semi-quantitative analysis, which obtained the highest cellulose content in dietary fiber extracted from DCP and CDG with 6% and 4% NaOH solution, respectively. The principal component analysis illustrated clear separation of spectral distribution in cassava pulp extracted dietary fiber (DFCP) and cassava distiller's dried grains extracted dietary fiber (DFCDG) when treated with 6% and 4% NaOH, respectively. Conclusion: The optimal conditions for the extraction of dietary fiber from DCP and CDG were treatment with 6% and 4% NaOH solution, respectively. In addition, FTIR spectroscopy proved itself to be a powerful tool for fiber identification.
A time-of-flight and energy (TOF-E) detection system for the measurement of 236U accelerator mass spectrometry (AMS) has been developed to improve the 236U/238U sensitivity at Micro Analysis Laboratory, Tandem accelerator (MALT), The University of Tokyo. With observing TOF distribution of 235U, 236U and 238U, this TOF-E detection system has clearly separated 236U from the interference of 235U and 238U when measuring three kinds of uranium standards. In addition, we have developed a novel method combining kernel-based density estimation method and multi-Gaussian fitting method to estimate the 236U/238U sensitivity of the TOF-E detection system. Using this new estimation method, 3.4 × 10-12 of 236U/238U sensitivity and 1.9 ns of time resolution are obtained. 236U/238U sensitivity of TOF-E detection system has improved two orders of magnitude better than that of previous gas ionization chamber. Moreover, unknown species other than uranium isotopes were also observed in the measurement of a surface soil sample, which has demonstrated that TOF-E detection system has a higher sensitivity in particle identification. With its high sensibility in mass determination, this TOF-E detection system could also be used in other heavy isotope AMS.

Thin films synthesized by plasma processes have been widely applied in a variety of industrial sectors. The structure control of thin film is one of prime factor in most of these applications. It is well known that the structure of this film is closely associated with plasma parameters and species of plasma which are electrons, ions, radical and neutrals in plasma processes. However the precise control of structure by plasma process is still limited due to inherent complexity, reproducibility and control problems in practical implementation of plasma processing. Therefore the study on the fundamental physical properties that govern the plasmas becomes more crucial for molecular scale control of film structure and corresponding properties for new generation nano scale film materials development and application. The thin films are formed through nucleation and growth stages during thin film depostion. Such stages involve adsorption, surface diffusion, chemical binding and other atomic processes at surfaces. This requires identification, determination and quantification of the surface activity of the species in the plasma. Specifically, the ions and neutrals have kinetic energies ranging from ~ thermal up to tens of eV, which are generated by electron impact of the polyatomic precursor, gas phase reaction, and interactions with the substrate and reactor walls. The present work highlights these aspects for the controlled and low-temperature plasma enhanced chemical vapour disposition (PECVD) of Si-based films like crystalline Si (c-Si), Si-quantum dot, and sputtered crystalline C by the design and control of radicals, plasmas and the deposition energy. Additionally, there is growing demand on the low-temperature deposition process with low hydrogen content by PECVD. The deposition temperature can be reduced significantly by utilizing alternative plasma concepts to lower the reaction activation energy. Evolution in this area continues and has recently produced solutions by increasing the plasma excitation frequency from radio frequency to ultra high frequency (UHF) and in the range of microwave. In this sense, the necessity of dedicated experimental studies, diagnostics and computer modelling of process plasmas to quantify the effect of the unique chemistry and structure of the growing film by radical and plasma control is realized. Different low-temperature PECVD processes using RF, UHF, and RF/UHF hybrid plasmas along with magnetron sputtering plasmas are investigated using numerous diagnostics and film analysis tools. The broad outlook of this work also outlines some of the 'Grand Scientific Challenges' to which significant contributions from plasma nanoscience-related research can be foreseen.

The concentrations of outdoor airborne bacteria measured in Seoul, Korea for one year (Jan. 2008~Dec. 2008) ranged from $500CFU/m^3$ to $7,500CFU/m^3$. In monthly concentration distribution, the level of outdoor airborne bacteria was highest in September and October and lowest in March. In seasonal concentration distribution, the order of level of outdoor airborne bacteria was autumn>winter>spring>summer. In regional concentration distribution, the highest level of outdoor airborne bacteria was generally found in the forest, followed by general area and traffic site. In distribution characteristics according to particle size, outdoor airborne bacteria showed 31% for >$7.0{\mu}m$ (stage 1), 21% for $4.7{\sim}7.0{\mu}m$(stage 2), 15% for $3.3{\sim}4.7{\mu}m$(stage 3), 19% for $2.1{\sim}3.3{\mu}m$(stage 4), 10% for $1.1{\sim}2.1{\mu}m$(stage 5), and 4% for $0.65{\sim}1.1{\mu}m$(stage 6) and its mean respirable fraction was 48%. In concentration distribution by yellow dust, the mean levels of outdoor airborne bacteria were 803 $(\pm479)CFU/m^3$ for period of yellow dust and 691 $(\pm1,134)CFU/m^3$ for period of non-yellow dust. Although the level of outdoor airborne bacteria was higher in period of yellow dust than period of non-yellow dust, there was no significant difference between period of yellow dust and non-yellow dust (p>0.05). In correlation analysis between outdoor airborne bacteria and atmospheric factors (temperature and relative humidity), there was no significant correlation between outdoor airborne bacteria and atmospheric factors. The predominant airborne bacteria were identified to the Bacillus-derived species.
