Effect of 0.5 mM Dibutyryl cAMP on Meiotic Maturation during Different Incubation Time and Embryonic Development Following In Vitro Fertilization or Parthenogenetic Activation in Porcine Oocytes

  • Presently, the effect of 0.5 mM dibutyryl cAMP (dbcAMP)-supplemented maturation medium during different incubation time on meiotic arrest (germinal vesicle) and resumption (metaphase II) of porcine oocytes and embryonic development of porcine oocytes following in vitro fertilization (IVF) or parthenogenetic activation (PA) was determined. Porcine cumulus oocyte complexes (COCs) were cultured in 0.5 mM dbcAMP for 17, 22, 27, or 42 h, and an additional 22 h without 0.5 mM dbcAMP. The nuclear status was examined at each time point. Oocytes cultured from 39~49 h displayed more than 80% meiotic resumption. More than 85 % of meiotic arrest was presented at 17~22 h. Oocytes were cultured for 22 h with 0.5 mM dbcAMP and additional 22 h without dbcAMP to assess developmental potential following IVF or PA. There were no significant differences in blastocyst rates among the dbcAMPIVF, IVF, dbcAMP-PA, and PA groups, although cleavage rate of IVF group was significantly higher than those of dbcAMP-PA, and PA groups. In conclusion, 0.5 mM dbcAMP influenced meiotic maturation of porcine oocytes depending on incubation time of oocyte, although embryonic development was not improved in both IVF and PA.

Effect of Porcine Serum as Macromolecule on the Meiotic Maturation and Embryonic Development of Porcine Oocytes

  • This study was conducted to establish an in vitro maturation (IVM) system by selection of efficient macromolecule in the porcine in vitro production (IVP) technology. To choose the efficient macromolecules in the development of porcine embryos, the effects of 3 kinds of macromolecules (porcine serum; PS, porcine follicular fluid; pFF, and polyvinyl alcohol; PVA) supplemented in IVM media on the maturation, cleavage, and development rates to blastocyst of parthenogenetic activation (PA) and in vitro fertilization (IVF) embryos were examined. The maturation rates of porcine oocytes in media supplemented with PS were significantly higher than those with pFF and PVA (92.4% vs. 85.4%, 77.1%; p<0.05). In the cleavage and development to blastocyst rates, supplement with PS or pFF in the IVM media was more effective than PA. However, there were no significant differences in cleavage and development to blastocyst between PS and pFF group. From the results of this study, it was demonstrated that PS was optimal macromolecule in the porcine IVM media.

  • Alpha lipoic acid (ALA) is a biological membranes compound. As the antioxidant, it decreases the oxidized forms of other antioxidant substances such as vitamin C, vitamin E, and glutathione (GSH). To examine the effect of ALA on the in vitro maturation (IVM) of porcine oocytes, we investigated intracellular GSH and reactive oxygen species (ROS) levels, and subsequent embryonic development after parthenogenetic activation (PA). Intracellular GSH levels in oocytes treated with 50uM ALA increased significantly (P < 0.05) and exhibited a significant (P < 0.05) decrease in intracellular ROS levels compared with the control group. Oocytes matured with 50 uM of ALA during IVM displayed significantly higher cleavage rates (67.8% vs. 83.4%, respectively), and higher blastocyst formation rates and total cell number of blastocysts after PA (31.6%, 58.49 vs. 46.8%, 68.58, respectively) than the control group. In conclusion, these results suggest that treatment with ALA during IVM improves the cytoplasmic maturation of porcine oocytes by increasing the intracellular GSH levels, thereby decreasing the intracellular ROS levels and subsequent embryonic developmental potential of PA.

  • In vitro maturation and activation of oocytes are primary steps towards biotechnological manipulation in embryology. The objectives of the present study were to determine the oocyte recovery rate per ovary, in vitro maturation rates of oocytes and rates of parthenogenetically activation of matured oocytes in Black Bengal goats. All visible follicles were aspirated to recover follicular fluid from individual ovaries (number of ovaries = 456). The immature cumulus oocyte complexes (COCs; n = 1289) were cultured in tissue culture medium (TCM)-199 supplemented with 10% (v/v) fetal bovine serum (FBS) for 27 hours at $39^{\circ}C$ with 5% $CO_2$ in humidified air. The matured oocytes (n = 248) were activated with 5 ${\mu}M$ ionomycin for 5 minutes followed by treatment with 2 mM 6-dimethylaminopurine (6-DMAP) for 4 hours. After activation, oocytes were cultured for another 14 hours in TCM-199 supplemented with bovine serum albumin (BSA) at $39^{\circ}C$ with 5% $CO_2$ in humidified air. The pronucleus formation in activated oocytes was determined by staining with 1% orcein (whole mount technique). Matured oocytes (n = 176) without activation stimuli were used as control. The mean number of oocytes recovered per ovary was $3.5{\pm}0.5$. The proportion of oocytes matured in vitro, confirmed by the presence of first polar body, was $42.1{\pm}4.7%$. Parthenogenetic activation, evidenced by formation of pronucleus, occurred in $37.2{\pm}15.8%$ of matured oocytes. No pronucleus formation was observed in control oocytes. In conclusion, a combination of ionomycin and 6-DMAP induces activation in one third of Black Bengal goats' oocytes.

On the Development of Parthenogenetic Oocytes by Cytochalasin B and Production of Cloned Mice by SCNT

  • This study was conducted to optimize the efficiency of cloning and to produce cloned mice. The majority of cloned mammals derived by nuclear transfer (NT) die during gestation and have enlarged and dysfunctional placentas. In this study, the optimized conditions were established to produce clone mice. The parthenogenetic oocytes were activated after 6 h regardless of cytochalasin B (CB) concentration. CB treatment ($2{\mu}g/ml$) was found second polar body. Lower concentration of CB was decreased the activation rate, but the second polar body was the best highly increased during 6 h incubation. The small fragments were exhibited in the $5{\mu}g/ml$ treatment of CB, but it was not found in lower concentration groups (> $2.5{\mu}g/ml$). To examine effects of $SrCl_2$ on the adult cumulus cells, somatic cell NT oocytes were exposed during 0.5, 1 and 6 hrs. The second polar body was significantly greater in 0.5 h exposure group (6.6%) than 1, 6 hrs. Developmental rate from 2-cell to 4-cell was the lowest in 7.5 mM Strontium chloride ($SrCl_2$) groups (84.1% and 64.3%) than 5, 10 m $MSrCl_2$. The implantation rate was not significantly difference among 5, 7.5 and 10 m $MSrCl_2$ group. Three live fetuses were produced by SCNT. SCNT placentas were remarkably heavier than IVF group (8 fetuses) (0.34, 0.34, 0.33 vs 0.14 g) compared with the placenta weight of IVF and SCNT clones.

Development of Chimeric Embryos Aggregated with Blastomeres from Parthenogenetic and in vitro Fertilized Bovine Embryos (소의 단위 발생란과 체외수정란 유래의 할구 응집에 의한 키메라 수정란(Chimeric Embryo)의 발달)

  • 발생학에서 키메라(chimera)는 2개 이상의 다른 유전자형의 세포, 또는 다른 종의 세포로부터 만들어진 1개의 생물개체를 뜻하는 말로, 이는 초기 수정란의 발달과 포유류의 분화를 연구하는데 이용되고 있다. 키메라를 만드는 방법에는 할구와 내세포괴를 응집시키는 방법과 배반포 내에 여러 종류의 세포를 주입하는 방법이 있다 본 실험에서는 서로 다른 두 가지 방법의 활성화 처리법, 즉, ionomycin 처리 후 Cycloheximide (CHX) 또는 6-Dimetylaminopurine (6-DMAP)에 따른 단위 발생란의 분할과 단계적인 발달율을 살펴 보고자 하였으며, 서로 다른 방법에 의해 생산된 단위발생란 유래의 할구와 체외수정란 유래 할구를 응집하여 키메라 수정란(chimeric embryo)를 만든 후 체외수정란과 발달율을 비교해 보았다. 도축장 유래의 난소에서 난자를 채취하여 체외에서 22~24시간 성숙시킨 후 난구세포를 제거하고 metaphase II 단계의 난자를 5 $\mu$M ionomycin에 4분간 처리한 후, 10 $\mu$g/ml CHX/5 $\mu$g/ml cytochalasin B (CCB)에 5시간 또는 1.9 mM 6-DMAP에 4시간 처리하여 분할율과 배반포기 발달율을 비교 조사하였다. 난자 분할율에서는 체외수정란과 6-DMAP처리 단위 발생란에서 각각가 83.7 와 85.5%로 CHX/CCB 처리 단위발생란의 57.9%보다 유의적으로 높게(P<0.05) 나타났으며, 배반포기 발달율에 있어서는 체외수정란의 발달율이 27.8%로 6-DMAP처리 활성란 12.3%와 CHX/CCB 처리 활성란 5.3%보다 유의적으로 높게 (P<0.05) 나타났다. 키메라 수정란(chimeric embryo)은 서로 다른 두 가지 처리에 의해 생산된 단위발생란의 할구 2개와 체외수정란 유래의 할구 2개를 빈 투명대 내에서 응집시켜 제조하였다 빈 투명대 내에 키메라 수정란(chimeric embryos)의 8 세포기까지의 발달율은, 체외 수정란 할구 2개와 CHX/CCB 처리에 의한 할구 2개를 응집한 그룹은 46.1%, 체외 수정란 할구와 6-DMAP 유래 할구 2개를 응집한 그룹은 52.8% 였으며, handled control은 54.7%로 체외 수정란 77.7%에 비해 유의적으로 낮게(P<0.05) 나타났다. 배반포기까지의 발달율은 체외 수정란과 CHX/CCB에 의해 생산된 키메라 수정란(chimeric embryo)은 12.8%, 체외 수정란과 6-DMAP에 의한 키메라 수정란(chimeric embryo)은 18.8%로 handled control의 21.4%에 비해 유의적으로 낮았으며(P<0.05), 이들 키메라 수정란(chimeric embryos)은 체외 수정란의 34.9%에 비해 유의적으로 낮게(P<0.05)나타났다. 6-DMAP 처리 단위발생이 유기된 수정란 할구 2개와 체외수정란의 할구 2개의 응집에 의한 키메라 수정란(chimeric embryos)의 발달율이 CHX/CCB와 체외수정란의 응집에 의한 키메라 수정란(chimeric embryos)에 비해 다소 높게 나타났으나, 유의적인 차이는 없었다. 본 실험의 결과 서로 다른 방법에 의한 단위 발생란 유래의 할구와 체외 수정란 유래의 할구가 응집에 의한 재조합이 가능하였고 이들을 체외에서 배양하여 배반포기의 수정란까지 발달시켰다.

Effects of (-)-Epicatechin Gallate on porcine oocyte in vitro maturation and subsequent embryonic development after parthenogenetic activation and in vitro fertilization

  • (-)-Epicatechin gallate (ECG) is a polyphenol compound of green tea exhibiting biological activities, such as antioxidant and anticancer effects. To examine the effect of ECG on porcine oocytes during in vitro maturation (IVM), oocytes were treated with 0-, 5-, 15-, and $25{\mu}M$ ECG. After maturation, we investigated nuclear maturation, intracellular glutathione (GSH) and reactive oxygen species (ROS) levels and subsequent embryonic development after parthenogenetic activation (PA) and in vitro fertilization (IVF). After 42 hours of IVM, the $5{\mu}M$ group exhibited significantly increased (p< 0.05) nuclear maturation (89.8%) compared with the control group (86.1%). However, the $25{\mu}M$ group observed significantly decreased (p< 0.05) nuclear maturation (83.5%). In intracellular maturation assessment the 5-, 15-, and $25{\mu}M$ groups had significantly increased (p< 0.05) GSH levels and decreased ROS levels compared with the controls. The 5- and $15{\mu}M$ group showed significantly increased (p< 0.05) embryo formation rates and total cell number of blastocysts after PA (18% and 68.9, 15% and 85.1 vs. 12% and 59.5, respectively) compared with controls. Although the $25{\mu}M$ group observed significantly lower blastocyst formation rates after PA (27.6% vs. 23.2%) than control group, the $5{\mu}M$ group showed significantly increased blastocyst formation rates after PA (37.2% vs. 23.2%) compared to the control group. Furthermore, the $5{\mu}M$ group measured significantly increased blastocyst formation rates (20.7% vs. 8.6%) and total cell number after IVF ($88.3{\pm}1.5$ vs. $58.0{\pm}3.6$) compared to the control group. The treatment of $5{\mu}M$ ECG during IVM affectively improved the porcine embryonic developmental competence by regulating intracellular oxidative stress during IVM.

Control of MPF Activity and Nuclear Remodeling of Somatic Cell Nuclear Transfer Bovine Embryos by Chemical Treatments (소 체세포 핵이식란의 화학적 처리에 의한 MPF 활성 및 핵의 Remodeling 조절)

  • We attempted to control the maturation promoting factors (MPF) activity and nuclear remodeling of somatic cell nuclear transfer (NT) bovine embryos. Bovine ear skin fibroblasts were fused to enucleated oocytes treated with either 5 mM caffeine for 2.5 h or 0.5 mM vanadate for 0.5 h and activated. The nuclear remodeling type of the reconstituted embryos was evaluated 1.5 h after activation. MPF activity was assessed in enucleated and chemical treated oocytes before the injection of a donor cell. Effect of chemicals on the embryonic development was evaluated with parthenogenetic embryos. MPF activity increased significantly by caffeine treatment, but decreased by vanadate treatment (p<0.05). Caffeine or vanadate had no deleterious effect on the parthenogenetic embryo development. In caffeine treated group, premature chromosome condensation (PCC) was occurred in 72.2% of NT embryos (p<0.05). In contrast, vanadate induced the formation of a pronucleus-like structure (PN) in a high frequency (68.9%, p<0.05) without PCC (NPCC). Blastocyst development of NT embryos increased by treating with caffeine (30.3%), whereas decreased by treating with vanadate (11.4%) compared to control (22.1%, p<0.05). The results indicate that caffeine or vanadate can control of MPF activity and remodeling type of NT embryos, resulting in the increased or decreased in vitro development.

Effect of Insulin Supplement on Development of Porcine Parthenogenetic Embryos

  • This study is performed to evaluate the effect of insulin in the porcine parthenogenetic embryo development. In porcine embryo culture, insulin is helpful factor in the process of embryo development. To identify this, insulin is used in pig embryos development. Therefore, this study was performed to investigate the effect of insulin on early embryonic development in pigs. For that, insulin positive or negative (0, 10 ug/mL) was supplemented in the porcine IVM media and then compared two groups divided by the cytoplasm of the black groups and white ring groups based on the distribution of lipid material of the cell cytoplasm in microscope. In maturation rates of porcine oocytes, significant higher black group rates were shown in the insulin positive groups compared with other groups ($56.0{\pm}2.1$ vs $46.2{\pm}0.3$). In the embryo culture, black groups were showed the significant higher cleavage rates ($82.1{\pm}0.8$, $78.3{\pm}0.1$ vs $63.2{\pm}0.3$, $63.4{\pm}0.0$), and blastocyst formation rates ($15.5{\pm}3.6$, $16.6{\pm}0.4$ vs $11.7{\pm}1.3$, $7.4{\pm}0.2$) regardless of whether the addition of insulin. Also, black groups were showed higher cell number of blastocyst ($33.2{\pm}2.5$, $35.5{\pm}2.6$ vs $31.2{\pm}2.1$, $31.3{\pm}2.2$). In conclusion, supplement of insulin producing black group in vitro maturation, it was effective in vitro maturation and embryonic development of pig embryos.
