  • In order to survey the prevalence of parasites in soil from playground in a detached dwelling area, 162 samples were taken from playground 54 unit. The survey was carried out during 3 months from july in 2000 to september in 2000. The results were obtained as follows : 1. Eggs of Toxocara canis were detected in 30 samples (18.5%) from 162 cases of playground. 2. The 3 kinds of eggs were isolated from the soil of playground. Those were Stronglyoides spp 23.5%, Toxocara canis 18.5% and Trichuris vulpis 4.3%. 3. It was mixed infestation such as single (74.5%), double (23.7%) and triple(1.6%).

  • In order to survey the prevalence of parasites in soil from playground in a housing development testate, 368 samples were taken from playground 184 unit. The survey was carried out during 2 months from september in 1999 to October in 1999. The results were obtained as follows; 1. Eggs of toxocara canis were detected in 25 samples(6.8%) from 368 cases of playground. 2. The 3 kinds of eggs were isolated from the soil of playground. Those were strongyloides spp 13.9%, toxocara canis 6.8% and toxascaris leonina 2.4%. 3. It was mixed infestation such as single (80.0%) and double (20.0%).

동물 분변으로 인한 모래 바닥재의 오염실태 및 관리 방안에 관한 연구 - 질소분석방법개발 - (A Study on Pollution Conditions and Management of Sand Flooring Related to Animal Feces - Nitrogen Analysis Method Development -)

  • Objectives: Users of parks or children's play facilities have pointed to pets' bowel movements as the most serious problem when using them. In prior studies, a very low detection rate of parasites (eggs) in sand flooring materials has been found. Even though feces have been identified, no parasites (eggs) have been detected. Method: A standard solution of nitrate nitrogen was used to verify the reliability of a new nitrogen analysis method. The linearity, precision, and accuracy of the nitrate nitrogen analysis method were verified. Using this method, the pollution distribution of the sand flooring material and the degree of pollution at each point were investigated. Results: As a result of the verification of the nitrogen analysis method, the linearity was found to be good at r2=0.999 when distilled water is mixed in a standard substance solution. The standard substance additive solution r2=0.968 was found to be good. Precision represented 0.01 to 0.06% RSD for peak height. The recovery rate was 92.4 to 104.0 percent, indicating high accuracy. According to the same method of analysis, the flooring material sand at a general amusement facility with the largest number of concealed spaces was nitrate nitrogen 6.1 times higher than at the entrance of the playground. Also, in a comparison between clean sand and sandy flooring, the average nitrogen concentration of the sand flooring material was 24.4-167 times higher than pure sand. Conclusions: As such, no parasites (eggs) were detected at all points under investigation, but the sand flooring was exposed to animal fecal contamination. Therefore, the management of nitrogenous components should allow accurate identification of animal fecal contamination so that the timing of sand replacement can be managed hygienically and safely.

  • A field survey studying intestinal parasites in humans and microbial pathogen contamination at environment was performed in a Laotian rural village to identify potential risks for disease outbreaks. A parasitological investigation was conducted in Ban Lak Sip village, Luang Prabang, Lao PDR involving fecal samples from 305 inhabitants as well as water samples taken from 3 sites of the local stream. Water analysis indicated the presence of several enteric pathogens, i.e., Aeromonas spp., Vibrio spp., E. coli H7, E. coli O157: H7, verocytotoxin-producing E. coli (VTEC), Shigella spp., and enteric adenovirus. The level of microbial pathogens contamination was associated with human activity, with greater levels of contamination found at the downstream site compared to the site at the village and upstream, respectively. Regarding intestinal parasites, the prevalence of helminth and protozoan infections were 68.9% and 27.2%, respectively. Eight helminth taxa were identified in fecal samples, i.e., 2 tapeworm species (Taenia sp. and Hymenolepis diminuta), 1 trematode (Opisthorchis sp.), and 5 nematodes (Ascaris lumbricoides, Trichuris trichiura, Strongyloides stercoralis, trichostrongylids, and hookworms). Six species of intestinal protists were identified, i.e., Blastocystis hominis, Cyclospora spp., Endolimax nana, Entamoeba histolytica/E. dispar, Entamoeba coli, and Giardia lamblia. Questionnaires and interviews were also conducted to determine risk factors of infection. These analyses together with a prevailing infection level suggested that most of villagers were exposed to parasites in a similar degree due to limited socio-economic differences and sharing of similar practices. Limited access to effective public health facilities is also a significant contributing factor.

발효톱밥돈사에 대한 돼지 내부기생충 조사 (Preliminary survey of swine internal parasites at the sawdust fermentation floor system)

  • Preliminary survey on the prevalence of swine internal parasites was carried out at 91 pens of 4 piggeries being operated by the sawdust fermentation floor system in Kyunggi province. To investigate the effects of viability of eggs., larva and cysts, temperatures at the underlayer of the sawdust floor and rooms of pens were taken three times a day for 8 days. In a pig farm which has been operated by the sawdust fermentation floor system for G years, eggs and larva of Ascaris suum, Trichuris suis, Oesophagostomum sp., Hyostrongylus sp. and Strongyloides ransomi were detected from 42(70.0%) among 60 pens. Cysts and oocysts of coccidia, Balantidium coli, Entamoeba sp. were also detected from 50(83.3%) among 60 pens. In three pig farms which have been operated by the sawdust fermentation floor system for one year, eggs and larva of nematoda were not detected at all, and the contamination rates of protozoan cysts and oocysts was relatively low situation. The change of the temperature in the pig room was variable according to outside condition, whereas that in underfloor was consistently stable between $28^{\circ}C$ to $30^{\circ}C$ which was strictly suitable for the development of eggs, larva and cysts.

팽나무버섯 액체 종균의 접종 전 오염 검사 (The contamination check before inoculation at the liquid Spawn on Flammulina velutipes)

  • 본 연구에서는 배양된 액체종균의 세균 오염 여부를 Giemsa 용액의 단일 염색으로 짧은 시간 안에 정확하게 판정이 가능한지를 조사하였다. Giemsa 용액은 혈액 골수 림프절 말라리아 원충 리케차 세포 등을 염색하는 것으로, 염기성 색소인 methylene azul과 methylene blue, 그리고 산성색소인 eosine을 methyl alcohol-glycerine에 녹여서 제조하였다. 그리고 팽이 액체종균의 폭기 배양액 분취하여 슬라이드에 올리고 Gimesa 용액으로 염색한 후 광학 현미경으로 검사하였다. 이 결과 40~60초 동안의 세균세포를 (팽나무)버섯(류) 균사세포의 부스러기나 잔존 대두박 등과 구별할 수 있었다. 이 Gimesa 용액을 이용한 염색 및 검경방법은 빠르고 간편하며 정확하므로 액체종균을 사용하는 버섯재배 농가에서도 세균 오염을 효과적으로 동정(감지)하는데 이용할 수 있다고 생각한다.

  • In order to survey the prevalence of Toxocara cams in soil of playground in a detached dwelling area, 150 samples were taken from playground 50 unit. The survey was carried out during 3 months from July to September in 2006. The Egg of Toxocara canis were detected in 6 samples (4.0%) from 150 cases of playground. The prevalence of T canis by month was 6.0% in July, 4.0% in August, 2.0% in September, respectively. This study results suggested that the prevalence of T canis were decreased in 2006 compared to 18.5% (30 positive/162 samples) in 2000.

즉석섭취 신선편의 절단 과일 및 채소의 원충류 및 병원성 식중독균의 미생물학적 품질 실태 연구 (Microbial Qualities of Parasites and Foodborne Pathogens in Ready to Eat (RTE) Fresh-cut Produces at the On/Offline Markets)

  • 본 연구에서는 온라인 및 오프라인 마켓에 유통 중인 100건의 신선편의 절단 과일을 계절별로 수집하고 50건의 신선편의 절단 채소와 50건의 밀 키트(월남 쌈 및 무 쌈) 내 절단 채소에 대하여 위생지표세균, 효모 및 곰팡이를 정량적으로 분석하였으며, 5개의 식중독균(B. cereus, S. aureus, L. monocytogenes, Salmonella spp., E. coli O157:H7)을 정성 및 정량적으로 분석하였다. 전체적으로 밀 키트 채소의 일반세균, 대장균군, 효모 및 곰팡이의 평균 오염수준이 신선편의 절단 채소 및 절단 컵 과일보다 높은 것으로 나타났다. 대장균은 두 식품 유형에서 모두 검출되지 않았으며 병원성 식중독균의 경우, S. aureus만 총 200건 중 6건이 검출되었는데(3%), 절단 과일 중 파인애플 1건에서 S. aureus가 0.6 log CFU/g로 검출이 되었고, 분리된 균주는 non-enterotoxin인 것으로 확인되었다. 신선편의 절단 채소 중 절단 채소 셀러리에서 1건, 밀 키트 채소 4건에서 S. aures 정성 검출되었으나, 정량적 분석 결과 밀 키트 채소인 적색 양배추 1건에서만 4.8 CFU/g 오염수준을 확인 하였으나 즉석섭취·편의식품류의 S. aureus 규격(100 CFU/g) 이하임을 확인하였다. 절단 채소에서 검출된 S. aureus의 독소형을 분석한 결과 셀러리와 적색 양배추에서 분리된 균이 각각 seg 와 sei 독소형임을 확인하였다. 따라서 절단 채소 제품 공정에서, 특히 밀 키트 채소에서의 황색포도상구균의 오염에 대하여 더 엄격한 관리방안이 필요한 것으로 확인되었다. 본 연구에서 분석한 50여종의 신선편의 식품과 밀 키트 제품에서는 모두 원포자충 및 작은와 포자충은 검출되지 않았으나 원충류에 의한 식중독 사고를 예방하기 위해 신선편의 식품의 세척 과정에 대한 지속적인 주의가 필요할 것으로 사료된다.

Biological and Genetic Characterization of Cryptosporidium spp. and Giardia duodenalis Isolates from Five Hydrographical Basins in Northern Portugal

  • To understand the situation of water contamination with Cryptosporidium spp. and Giardia spp. in the northern region of Portugal, we have established a long-term program aimed at pinpointing the sources of surface water and environmental contamination, working with the water-supply industry. Here, we describe the results obtained with raw water samples collected in rivers of the 5 hydrographical basins. A total of 283 samples were analyzed using the Method 1623 EPA, USA. Genetic characterization was performed by PCR and sequencing of genes 18S rRNA of Cryptosporidium spp. and $\beta$-giardin of Giardia spp. Infectious stages of the protozoa were detected in 72.8% (206 of 283) of the water samples, with 15.2% (43 of 283) positive for Giardia duodenalis cysts, 9.5% (27 of 283) positive for Cryptosporidium spp. oocysts, and 48.1% (136 of 283) samples positive for both parasites. The most common zoonotic species found were G. duodenalis assemblages A-I, A-II, B, and E genotypes, and Cryptosporidium parvum, Cryptosporidium andersoni, Cryptosporidium hominis, and Cryptosporidium muris. These results suggest that cryptosporidiosis and giardiasis are important public health issues in northern Portugal. To the authors' knowledge, this is the first report evaluating the concentration of environmental stages of Cryptosporidium and Giardia in raw water samples in the northern region of Portugal.

Presence of Cryptosporidium spp. and Giardia duodenalis in Drinking Water Samples in the North of Portugal

  • Cryptosporidium and Giardia are 2 protozoan parasites responsible for waterborne diseases outbreaks worldwide. In order to assess the prevalence of these protozoans in drinking water samples in the northern part of Portugal and the risk of human infection, we have established a long term program aiming at pinpointing the sources of surface water, drinking water, and environmental contamination, working with the water-supply industry. Total 43 sources of drinking water samples were selected, and a total of 167 samples were analyzed using the Method 1623. Sensitivity assays regarding the genetic characterization by PCR and sequencing of the genes, 18S SSU rRNA, for Cryptosporidium spp. and $\beta$, -giardin for G. duodenalis were set in the laboratory. According to the defined criteria, molecular analysis was performed over 4 samples. Environmental stages of the protozoa were detected in 25.7% (43 out of 167) of the water samples, 8.4% (14 out of 167) with cysts of Giardia, 10.2% (17 out of 167) with oocysts of Cryptosporidium and 7.2% (12 out of 167) for both species. The mean concentrations were 0.1-12.7 oocysts of Cryptosporidium spp. per 10 L and 0.1-108.3 cysts of Giardia duodenalis per 10 L. Our results suggest that the efficiency in drinking water plants must be ameliorated in their efficiency in reducing the levels of contamination. We suggest the implementation of systematic monitoring programs for both protozoa. To authors' knowledge, this is the first report evaluating the concentration of environmental stages of Cryptosporidium and Giardia in drinking water samples in the northern part of Portugal.
