Achievement of Korea-China Collaborative Project and the Collaboration with KAHP

  • Congratulations on the 40th Anniversary of KAHP, which has made great contributions to the control of parasitic diseases in Korea, and to the development of collaboration with China and other Asia countries aiming at the common interest. The 40 years of KAH/KAHP and its predecessor, Korea Association of Parasite Eradication has composed a brilliant road map to bring the prevalence of intestinal parasite infection from 84.3% in 1971 to the current prevalence of 2.4%, and to endeavor in the promotion of the health of children, man and woman, and low income families in Korea, as well as to generously share the successful experience in parasite control with China during the last half century. Indeed, the 40th birth day of KAHP is marked with great achievements and bright future.

Development of Lateral Flow Immunoassay for Antigen Detection in Human Angiostrongylus cantonensis Infection

  • Angiostrongyliasis is difficult to be diagnosed for the reason that no ideal method can be used. Serologic tests require specific equipment and are not always available in poverty-stricken zone and are time-consuming. A lateral flow immunoassay (LFIA) may be useful for angiostrongyliasis control. We established a LFIA for the diagnosis of angiostrongyliasis based on 2 monoclonal antibodies (mAbs) against antigens of Angiostrongylus cantonensis adults. The sensitivity and specificity were 91.1% and 100% in LFIA, while those of commercial ELISA kit was 97.8% and 86.3%, respectively. Youden index was 0.91 in LFIA and 0.84 in commercial ELISA kit. LFIA showed detection limit of 1 ng/ml of A. cantonensis ES antigens. This LFIA was simple, rapid, highly sensitive and specific, which opened an alternative approach for the diagnosis of human angiostrongyliasis.

A small-scale survey on the status of intestinal parasite infections in rural villages in Nepal

  • The status of intestinal parasite infections was investigated in two rural villages (Chitrasar, Jerona) in Chitwan District, Nepal in 1999. Stool examination was performed with a total of 300 specimens from schoolchildren by formalin-ether sedinentation technique. The prevalence rate of intestinal parasite infections in the surveyed areas was 44.0%. The prevalence rate in Jerona was slightly higher than that in Chitrasar. The prevalence rate of intestinal parasite infections in female was slighly higher than that in male without statistically significant difference. Entamoeba coli was the most commonly found protozoan parasite (21.0%) followed by Giardia lamblia (13.7%) and others (5.3%). Hookworm was the most prevalent intestinal helminth (13.0%) followed by Trichuris trichiura (3.0%) and others (5.0%). forty-three specimens (14.3%) Showed mixed infections. It is necessary to implement large-scale treatment with anthelminthics, health education and sanitary improvement for intestinal parasite control in the surveyed areas.

A Fluorescent Recombinase Aided Amplification Assay for Detection of Babesia microti

  • Babesia microti is one of the most common causative agents of babesiosis. A sensitive and rapid detection is necessary for screening potentially infected individuals. In this study, B. microti cytochrome c oxidase subunit I (cox1) was selected as the target gene, multiple primers were designed, and optimized by a recombinase-aided amplification (RAA) assay. The optimal primers and probe were labeled with fluorescein. The sensitivity of fluorescent RAA (fRAA) was evaluated using gradient diluents of the cox1 recombinant plasmid and genomic DNA extracted from whole blood of B. microti infected mice. The specificity of fRAA was assessed by other transfusion transmitted parasites. The analytical sensitivity of the fRAA assay was 10 copies of recombinant plasmid per reaction and 10 fg/µl B. microti genomic DNA. No cross-reaction with any other blood-transmitted parasites was observed. Our results demonstrated that the fRAA assay would be rapid, sensitive, and specific for the detection of B. microti.

  • 스트랩다운 탐색기를 탑재한 유도탄의 경우, 비례항법 호밍유도를 위한 관성좌표계 기준 시선각속도 정보를 직접적으로 획득하지 못하기 때문에 유도탄의 자세정보와 탐색기의 지향각 정보를 활용하여 적절히 추정하여야 한다. 하지만, 시선각속도를 추정하는 방법에 따라 획득된 시선각속도 정보에 동체 자세각속도 정보가 포함되어 불필요한 피드백루프(Parasite Loop)가 형성되고, 이로 인해 유도성능이 감소하거나 전체 유도조종루프가 불안정하게 되는 현상(Parasite Effect)이 발생하기도 한다. 본 논문에서는 비례항법 호밍유도를 위한 시선각속도 정보 추정방법과 이에 따라 발생되는 Parasite Loop 원인을 살펴보며, Routh-Hurwitz 안정성 판별법을 이용하여 Parasite Effect를 최소화하는 방안을 제시하도록 한다. 또한 다양한 시뮬레이션을 통해 제안한 방법에 대해 검증하도록 한다.

  • Usually, the Evolutionary Algorithms(EAs) are considered more efficient for optimal, system design because EAs can provide higher opportunity for obtaining the global optimal solution. This paper presents a mechanism of co-evolution consists of the two genetic algorithms(GAs). This mechanism includes host populations and parasite populations. These two populations are closely related to each other, and the parasite populations plays an important role of searching for useful schema in host populations. Host population represented by feedforward neural network and the result of co-evolution we will find the optimal structure of the neural network. We used the genetic algorithm that search the structure of the feedforward neural network, and evolution strategies which train the weight of neuron, and optimize the net structure. The validity and effectiveness of the proposed method is exemplified on the stabilization and position control of the inverted-pendulum system.

Eight cases of canine thelaziosis found in two localities in Korea

  • Thelazia callipaeda, a parasitic nematode that causes thelaziosis in various mammals, including humans, is known to be endemic in Korea. However, life cycle-related information on the parasite, primarily from human infection and a few dog cases, is limited. This study reports additional cases of T. callipaeda infections in dogs from both rural and urban areas in Korea, indicating the potential for transmission to humans and other animals. We collected 61 worms from 8 infected dogs from Paju and Cheongju Cities and observed their morphological characteristics under a light microscope. The findings indicate that T. callipaeda infections in animals in Korea may be underestimated and are distributed close to human environments. Our results contribute to the growing knowledge of the reservoir hosts of T. callipaeda in Korea and highlight the importance of continued surveillance and research to prevent and control this emerging zoonotic disease.

The protective effects of BMSA1 and BMSA5-1-1 proteins against Babesia microti infection

  • The intracellular parasite Babesia microti is among the most significant species causing human babesiosis and is an emerging threat to human health worldwide. Unravelling the pathogenic molecular mechanisms of babesiosis is crucial in developing new diagnostic and preventive methods. This study assessed how priming with B. microti surface antigen 1 (BHSA 1) and seroreactive antigen 5-1-1 (BHSA 5-1-1) mediate protection against B. microti infection. The results showed that 500 ㎍/ml rBMSA1 and rBMSA5-1-1 partially inhibited the invasion of B. microti in vitro by 42.0±3.0%, and 48.0±2.1%, respectively. Blood smears revealed that peak infection at 7 days post-infection (dpi) was 19.6%, 24.7%, and 46.7% in the rBMSA1, rBmSA5-1-1, compared to the control groups (healthy mice infected with B. microti only), respectively. Routine blood tests showed higher white blood cell, red blood cell counts, and haemoglobin levels in the 2 groups (BMSA1 and BMSA5 5-1-1) than in the infection control group at 0-28 dpi. Moreover, the 2 groups had higher serum interferon-γ, tumor necrosis factor-α and Interleukin-17A levels, and lower IL-10 levels than the infection control group throughout the study. These 2 potential vaccine candidate proteins partially inhibit in vitro and in vivo B. microti infection and enhance host immunological response against B. microti infection.

What was the main factor in successful control of ascariasis in Korea?

  • In the l950s, under the legacy of traditional agriculture, Ascaris lumbricoides, spread epidemically in the war-bitten society of Korea. Consensus on the parasite control was drafted in the Parasite Disease Prevention Act, which passed a parliamentary agreement in 1966, and established safe disposal of feces and mass chemotherapy as control strategies. Biannual stool examinations and treating infected schoolchildren were basic scheme of the control activity through which revenue could be secured for organized business. In the 27 years following 1969, a maximum of 16 million stool examinations had been done every year. Cellophane thick smear enabled the task. The infection declined remarkably in the 1970s when industrialization and green revolution proceeded. A population study of A. lumbricoides in the late 1970s helped us better understand its epidemiology. The data also settled down the understandable protest of teachers against the repeated stool examinations. In the 9 years following 1987, the target population was gradually reduced when the egg positive rate was below 0.1%. An article in the Korean Law, stipulating obligatory stool examinations, was made optional. Although the long-term Korean effort of Ascaris control was a success, the effect of mass chemotherapy was not as succinct in terms of lowering reinfection. In the period of control, Korean agricultural technology changed, and the economy grew and supplied sanitary facilities by which the vicious cycle was disconnected. Reduction of morbidity was a benefit of mass chemotherapy, which is the only control method feasible in economically difficult countries. The most important hurdle of parasite control in the 1960s was poverty of general population and limited financial resources in Korea but the society formed a consensus on the priority of intestinal helminthiasis control during the ordeal period. The national consensus in the 1960s was the critical milestone for Ascaris control in Korea. Under the social agreement, application of timely technical and research advancements in parasitology achieved the success of ascariasis elimination. The successful experience of ascariasis elimination in Korea can be a benchmark for countries where neglected tropical diseases are endemically recycled.
