Application of Paraffin Oil for Control of Large Patch on Zoysia japonica (들잔디 갈색퍼짐병 방제를 위한 파라핀오일의 살포)

  • Large patch caused by Rhizoctonia solani AG2-2 (IV) is one of the most serious diseases in zoysiagrass. The objectives of this research were to evaluate the in vitro fungicidal effect and in field control of large patch disease of Paraffin oil. In the field experiments, paraffin oil was applied with 1 L/$m^2$ after diluted at 5 ml to 20 ml of oil/1 L of water for the control of large patch on Zoysia japonica in golf course. The same material was tested for inhibition of mycelial growth in vitro. Paraffin oil at 0.5% and 2.0% had a fungicidal effect by 43 to 67% on R. solani AG2-2 (IV) in vitro. In two locations of the field experiments, the paraffin oil provided moderate protection of large patch. It was effective in suppressing large patch development by 48.0% in comparison with a water check after one time application on mid-September in golf course. Paraffin oil may be used as an alternative control agent for environment friendly management of large patch on Zoysiagrass in golf course.

  • A mixed culture of Candida tropicalis and Trichosporon cutaneum was carried out using a n-paraffin medium. The growth of C. tropicalis was markedly enhanced by the mixed culture with T. cutaneum which did not grow on n-paraffin. C. tropicalis extracellularly excreted free fatty acids as metabolic products of n-paraffin in the culture medium. T. cutaneum appeared to assimilate these free fatty acids which were growth inhibitors for C. tropicalis, threreby enhancing the growth of C. tropicalis.

  • Apoptosis can be difficult to detect in routine histological sections. Since extensive DNA fragmentation is an important characteristic of this process, visualization of DNA breaks could greatly facilitate the identification of apoptotic cells. Several techniques for the qualitative and quantitative detection of this process have been established; recently, an in situ nick end-labelling technique based on the detection of DNA fragmentation, which is a molecular characteristic of apoptotic cell death, was described. Applying this method to paraffin sections of rat tissues, sensitivity was observed to be inconsistently low with regard to the expected number of apoptotic cells. I describe a new modified method for formalin-fixed, paraffin-embedded tissue sections, pretense pretreatment to permeate the tissue sections that involves an TUNEL (terminal deoxynucleotidyl transferase-mediated dUTP nick end-labeling) is acknowledged as a method of choice in the rapid identification and quantification of the apoptotic cell fraction in paraffin tissue preparations. TUNEL was performed without apoptosis and with apopotosis samples to each of the three concentrations of proteinase K (10, 25, 40 mg/ml) pretreatments. In this study, I show that chemical pretreatments of the tissue sections in proteinase K (25 mg/ml for 15 min at room temperature) considerably enhances the sensitivity of this nick end labelling technique.

  • The auto-ignition tests of hybrid rockets with the concentrated hydrogen peroxide as an oxidizer were presented. Auto-ignition, restartability, and instant ignition were successfully demonstrated by injecting decomposed gases from $H_2O_2$ into paraffin or polyethylene fuels. In addition, much faster response time was demonstrated in case of a paraffin, which was 13 and 30 ms at ignition delay and rise time respectively.

  • The visualization device for hybrid rocket is fabricated to investigate the combustion phenomena. Visualization device were composed with ignition system, oxidizer supply system, control system and data acquisition system, combustion visualization system. GOX as oxidizer and HDPE, Paraffin-LDPE Blending, Paraffin sd were used. As results, combustion phenomena and fuel droplet entrainment were observed.

  • Generally, a reduced chemical mechanism of n-heptane is used as chemical fuel of a 3-D diesel engine simulation because diesel fuel consists of hundreds of chemical components and various chemical classes so that it is very complex and large to use for the calculation. However, the importance of fuel in a 3-D simulation increases because detailed fuel characteristics are the key factor in the recent engine research such as homogeneous charged compression ignition engine. In this study, normal paraffin, iso paraffin and aromatics were selected to represent diesel characteristics and n-dodecane was used as a representative normal paraffin to describe the heavy molecular weight of diesel oil (C10~C20). Reduced kinetics of iso-octane and toluene which are representative species of iso paraffin and aromatics respectively were developed in the previous study. Some species were selected based on the sensitivity analysis and a mechanism was developed based on the general oxidation scheme. The ignition delay times, maximum pressure and temperature of the new reduced n-dodecane chemical mechanisms were well matched to the detailed mechanism data.

Usefulness of PCR to Mycobacterium Tuberculous and Nontuberculous Mycobacteria from Paraffin-embedded Tissues

  • The purpose of this study was to evaluate the clinical utility of TB/NTM PCR by comparing the results of TB PCR to detect Mycobacterium tuberculous (MTB) and nontuberculous mycobacteria (NTM) from paraffin-embedded tissue specimens. A total of 60 cases were tested using TB PCR and TB/NTM PCR. The MTB and NTM rate of TB/NTM PCR was 84.2% (16/19), 10.5% (2/19) in TB positive of TB PCR. The NTM rate of TB/NTM PCR was 29.3% (12/41) in TB negative of TB PCR. Fourteen different species of NTM were identified, the common isolate was M. gordonae (21.4%), M. avium (14.3%), M. ulcerans (7.1%), M. interjectum (7.1%), M. gilvum (7.1%), M. fortuitum (7.1%), M. mucogenicum (7.1%). The rare species identified were M. farcinogenes (7.1%), M. tokaiense (7.1%). Therefore, TB/NTM PCR is useful to differentiate MTB and NTM from paraffin-embedded tissue specimens and it is more effective in detecting NTM with TB PCR.

Combustion Characteristics of a Small Hybrid Rocket Using Paraffin-Wax as Fuel (파라핀 연료를 사용하는 소형 하이브리드 로켓의 연소 특성)

  • This study experimentally examines combustion characteristics of a hybrid rocket in which solid paraffin is used as a fuel, while oxidizer is pure oxygen. Especially, the experiment investigates the effects of chamber pressure and configuration of fuel grain. The pressure inside the combustion chamber is varied by changing a flow rate of oxidizer. The regression rate is observed to increase as the chamber pressure does. There also exists the effects of shape of fuel grain on thrust. Characteristic of paraffin hybrid rocket changes with shape of fuel grain. When there is a room near the injector, thrust increases. On the other hand, the room near the nozzle does not contribute to thrust increasement.

A Case Study of Paraffin Double-walled Microencapsulation Preparation Using Acrylic Polymer and Melamine Polymer for Thermal Energy Storage

  • In this study, we investigated the paraffin encapsulation using double-walled encapsulation technique. The first encapsulation used methyl methacrylic acid as the main component in acrylic polymer and the second encapsulation used melamine polymer. Particles size and distribution of the capsules were analyzed using scanning electron microscopy. In the first encapsulation, the stable capsules were obtained at 67% of phase change material ratio to methyl methacrylic acid monomer and the size of the capsule was from 0.2 to $0.3{\mu}m$. In the second encapsulation, the size of the capsules was almost the same with those capsules prepared in the first encapsulation. The particle size of single wall and double wall was about $0.3{\mu}m$. As a result of the encapsulation of paraffin using double-walled encapsulation technique, it was confirmed that the particle size was determined in the process of encapsulating using the acrylic polymer at the first wall material, and the physical and thermal stability of the capsules were imparted using melamine at the secondary wall material.

  • Neospora caninum infections have been associated with neonatal paresis as well as abortion around the world. Bovine abortion induced by N caninum was first reported in 1997 in Korea. Diagnosis of N caninum infection is usually based on histopathology and immunohistochemical detection of organism. However, often the tissues having lesion suggestive of N caninum infection were negative on immunohistochemistry. Here, we describe establishment of PCR-based diagnostic strategy for N caninum infection using DNA extracted from paraffin blocks containing the lesion. PCR was able to amplify N caninum-specific bands from the paraffin blocks containing at least moderate degree of inflammation. Compared to paraffin-blocks, DNA extracted from fresh tissues were less sensitive than that of paraffin blocks. This PCR-based method can be practically applicable for rapid diagnosis of bovine N caninum infection with high specificity and sensitivity. Based on this method, 17% of bovine abortion surveyed during a designated period was associated with N caninum infection.
