SSR 마커를 이용한 산양삼의 유전적 다양성 분석 (Genetic Diversity Analysis of Wood-cultivated Ginseng using Simple Sequence Repeat Markers)

  • Background: Panax ginseng C. A. Meyer is wood-cultivated ginseng (WCG) in Korea which depends on an artificial forest growth method. To produce this type of ginseng, various P. ginseng cultivars can be used. To obtain a WCG similar to wild ginseng (WG), this method is usually performed in a mountain using seeds or seedlings of cultivated ginseng (CG) and WG. Recently, the WCG industry is suffering a problem in that Panax notoginseng (Burk.) F. H. Chen or Panax quinquefolium L. are being sold as WCG Korean market; These morphological similarities have created confusion among customers. Methods and Results: WCG samples were collected from five areas in Korea. After polymerase chain reaction (PCR) amplification using the primer pair labeled with fluorescence dye (FAM, NED, PET, or VIC), fragment analysis were performed. PCR products were separated by capillary electrophoresis with an ABI 3730 DNA analyzer. From the results, WCG cultivated in Korea showed very diverse genetic background. Conclusions: In this study, we tried to develop a method to discriminate between WCG, P. notoginseng or P. quinquefolium using simple sequence repeat (SSR) markers. Furthermore, we analyzed the genetic diversity of WCG collected from five cultivation areas in Korea.

  • The graminicides fluazifop-p-butyl and sethoxydim effectively controlled annual grass weeds in 1,2 and year old ginseng crops during a 2 year study. Ginseng plants in the graminicide treated plots showed an early senescence of the shoots ; and at harvest the average root weight (g/root) and yield (kg/m) were slightly lower in these plants than in the untreated check plots. Roots of the graminicide treated plots remained viable to regenerate shoots. The greenhouse study indicated that shoot growth and seed development in ginseng plants were more sensitive to the graminicide treatment than the roots.

Effects of Soil Moisture on the Growth of American Ginseng (Panax quinquefolium L.)

  • Adequate available soil moisture level is considered to be one the most important components in growing high yields of good quality ginseng. Excessive soil moisture may promote stillborn fungal pathogens and cause serious diseases in ginseng fields. This study showed that soil moisture levels for optimum growth and health of ginseng varied with soil texture. Fifty- percent available moisture for sandy loam and 75% for silty loam are the best moisture levels for good growth and higher yield.

  • Dynamic parameters of biochemistry including the POD (peroxidase) activity, contents of DNA and RNA, isozymes of POD and ES (esterase) in American send (Panax quinquefolium L.) seed are reported in the present paper. The dynamic changes of POD activity proved that the PAS (physiological afterripening stage) is a stage in which some substances are prepared for seed germination. The POD activity correlated with ER (embryo ratio) significantly DNA content changed little only within 0.0036∼0.013 mg/ml, which did not correlate with ER. RNA content changed from 0.1539 to 1.0313 mg/ml and correlated significantly with RE during all of the embryo afterripening. None of the POD isozyme band was obtained in ESGS (embryo slowly growth stage), but five bands in ERGS (embryo rapidly growth stage) and six bands in PAS. Four bands of ES isozymes were obtained in ESGS, but six bands in ERGS, particularly, the content of ES isozymes increased in PAS. All of these may provide some information for understanding the dormancy mechanisms of American song seed.

  • 일복개량의 일환으로 일복의 피복재선택을 위한 기초자료를 얻고자 적색광, 청색광 및 백색광하에서 미국인삼(Panax quinquefolin L.)의 광합성을 측정하였다. 광질별 광합성량은 전반적으로 적색과, 백색광 및 청색광 순으로 많았으며 그 광합성량의 차이는 조도가 증가할수록 감소되었다. 따라서 일복의 피복물 색상은 적색으로 하는 것이 인삼(미국인삼) 생육에 유이할 것임을 시사되었다.

  • Dynamic parameters of physiology including embryo ratio (ER), embryo length to endosperm length, dry weight ratio between embryo and endosperm (DWR) and respiration rate (RR) in American send (Panax quinquefolium L.) seed were investigated. According to the changes of ER during seed stratification, the duration of embryo afterripening could be divided into three stages as \circled1 embryo slow growth stage (ESGS), ER increased from 7.31% to 20.48% (0.16% day-1): \circled2 embryo rapid growth stage (ERGS), ER increased to 80.98% (0.61% day-1) (75G5+ ERGS=morphological afterripening stage (MP,5)) and \circled3pysiological afterripening stage (PAS), ER Increased to 88.50% (0.094 day-1) only. DWR Increased slowly from 0.20% to 2.76clp (0.016% day-1) in MAS and rapidly to 8.81% (0.061% day-1) in PAS. The RR correlated significantly with ER as well (r=0.8934 > rot, 0.6610). The steep increment of both DWR and RR in PAS indicated that the PAS was not a static stage although the ER was not changed too much. All of these may provide some information for understanding the dormancy mechanisms of American sting seed.

Effect of Hot Water Boiling and Autoclaving on Physicochemical Properties of American Ginseng (Panax quinquefolium L.)

  • This study evaluates changes in the chemical composition and bioactivities of American ginseng (Panax quinquefolius L.) processed by boiling in water, $75^{\circ}C$ for 10, 20, 30, and 40 min, and autoclaving at high temperatures, $115^{\circ}C$ for 30 and 60 min and $130^{\circ}C$ for 90 and 120 min. Total ginsenoside contents of boiled ginseng remained relatively unchanged, whereas the contents of autoclaved ginseng samples significantly decreased with an increase of both time and temperature. Compared to unheated ginseng (control), the color of both boiled and autoclaved ginseng decreased in lightness and increased in redness. The acidic polysaccharide contents, the total phenolic contents and the antioxidant capacity of boiled and autoclaved ginseng were higher than the untreated ginseng, with the highest values being exhibited by the autoclaved samples. In particular, the antioxidant capacity of unheated ginseng increased about 2.5 times ($285.7{\pm}14.03\;mg$/100g to $777.2{\pm}26.4\;mg$/100g) when ginseng was autoclaved at $130^{\circ}C$ for 120 min as compared to the control. It was concluded that as American ginseng was processed at a high temperature, especially steam-heated in an autoclave, its chemical constituents changed and, in particular, acidic polysaccharides, total phenolics and antioxidant capacity were considerably increased.

  • Dynamic parameters of biochemistry including the contents of soluble carbohydrate (SC) , crude fat (CF) , fatty arid (FA) and soluble protein (SP) in American send (Panax qufnquelolium L.) seed are reported in the present paper. When embryo ratio (ER) increased from 7.31 to 20.48%, the SC content decreased from 4.08 to 1.16%. After that, SC content changed little. The CF content decreased successively from 34.50 to 14.40% from the beginning to the end during the embryo afterripening. The changes of FA content did not correlate with that of ER and the changing range of SP content was not large during the seed stratification. All of these may provide some information for understanding the dormancy mechanisms of American seng seed.

Comparison of Seed Oil Characteristics from Korean Ginseng, Chinese Ginseng (Panax ginseng C.A. Meyer) and American Ginseng (Panax quinquefolium L.)

  • The chemical characteristics of seed oils of Asian ginseng (Panax ginseng C.A. Meyer) at different ages grown in Korea (3, 4 and 5-year old) and China (5-year old), and American ginseng (Panax quinquefoliu L., 5-year old) grown in China were compared. Total fatty acid composition showed a significantly higher oleic acid content in American (87.50%) than in Korean (68.02~69.14%) and Chinese ginseng seed oils (61.19%). At the sn-2 position, the highest oleic acid (81.09%) and lowest linoleic acid (15.77%) were found in American ginseng seed oil. The main triacylglycerol species in ginseng seed oils were triolein (OOO) and 1,2-dioleoyl-3-linoleoyl-glycerol (LOO)/1,3-dioleoyl-2-linoleoyl-glycerol (OLO). In addition, the seed oils possessed an ideal oxidative stability showing 16.55~23.12 hr of induction time by Rancimat test. The results revealed that ginseng seed oil could be developed as a new healthy edible oil, and that the oil's chemical characteristics were strongly associated with the ginseng species and habitats.

고려인삼과 미국삼의 수삼 및 홍삼품질 비교 (Comparison of Quality on the Raw and Red Ginseng in Korean and American Ginseng)

  • 고려인삼과 미국삼을 공시하여 Panax속 종간의 원료수삼 및 홍삼 등급에 기초하여 품질을 평가분석하였다. 1. 미국삼은 고려인삼에 비하여 근중이 적고 부정형 지근이 동체에서 많이 발생하는 관계로 체형이 불량하였다. 2. 홍삼수율은 고려인삼이 30.4%,미국삼이 33.8%로 미국삼이 높았고 홍삼의 본삼수율은 고려인삼이 80.4%, 미국삼이 72.2로 고려인삼이 높았다. 3. 원료수삼 1등급은 고려인삼이 미국삼에 비하여 2배정도 많았으나 2등급은 미국삼이 고려인삼에 비하여 많았다. 한편 고급홍삼인 천지삼율은 고려인삼이 미국삼에 비하여 높았다. 4. 홍삼 품질의 저하 원인은 고려인삼 미국삼 모두 내공과 내백 발생에 있었고 특히 미국삼은 내공에 비해 내백이 3배 가량 많이 발생하였다. 5. 홍삼의 지별 분포에서 고려인삼은 20지 이상의 대편급이 많았고 미국삼은 20지 이하의 소편급이 많았다.
