Effects of Red Ginseng Extract on Zearalenone Induced Spermatogenesis Impairment in Rat

  • Zearalenone (ZEA) is a phenolic resorcylic acid lactone compound produced by several species of Fusarium. ZEA has toxic effects in the testes of domestic and laboratory animals. Korean red ginseng (KRG), the steamed root of Panax ginseng Meyer, has multiple pharmacological effects such as vasorelaxation, anti-thrombosis, anti-hypertension, etc. In this study, we investigated the effects of KRG extract on testicular toxicity induced by ZEA. Rats were treated with 300 mg/kg oral doses of KRG for 4 weeks every other day. The rats were then treated with a single dose of 5 mg/kg ZEA delivered intraperitoneally, whereas control rats received only doses of the vehicle. As a result, germ cell apoptosis induced by ZEA was decreased by KRG pre-treatment. In addition, Fas and Fas-L expression was reduced in rats that received KRG pre-treatment compared to ones treated with ZEA alone. In conclusion, impaired spermatogenesis resulting from ZEA treatment was prevented by KRG through Fas-Fas L modulating.

Induction of Ginseng Hairy Roots And Their Possible Application To Large Scale Culture

  • Ginseng(Panax ginseng C. A. Meyer) is important medicinal plant but requires 4-year cultivation for root harvest because of slow growth. In contrast, ginseng hairy roots induced by introducing Ri-plasmid of Agrobacterium rhizogenes into genomic DNA of plant cells show vigorous growth, and the hairy roots produce the same or more saponins than natural ginseng roots. Therefore, hairy roots can be used for commercial purposes. The present study was carried out to induce hairy roots with both active growth and high saponin contents. Numerous hairy roots of Panax ginseng were obtained after root disks of three-year old roots were infected with Agrobacterium rhizogenes R1000 A4T in dark condition after one month of culture. About 3 hundred lines of hairy roots were selected according as morphological characters on medium with carbenicillin. After pre-selection of fifteen lines of hairy roots with active growth, KGHR-l and KGHR-8 lines were finally selected which had characters of high content of ginsenoside-Rd and ginsenoside-Re, respectively. The optimum growth of hairy roots was achieved in the culture of 1/2 MS liquid medium in dark (22 $^{\circ}C$) under 60 rpm gyratory shaking. Hairy roots grew well in 5L Erlenmeyer flasks, lL roller drums, 10L jar-fermenters, and especially in 20L air-lift culture vessels.

  • Effect of saponin fraction of ginseng C.A. Meyer on blood serum lipoprotein distribution of high cholesterol fed rabbits for two to four weeks was investigated. A. significant increase of very low density lipoprotein(VLDL) and low density lipoprotein(LDL) occured while high density lipoprotein(HDL) were decreased in the blood of both groups which were fed high cholesterol diet with (test group) and/or without ginseng saponin(control group) for 2-4 weeks. However the degrees of VLDL and LDL increase and HDL decrease were relatively small in ginseng administered group compared with control group. This suggests that the hypocholesterolemic action of the saponin might be brought about by decreasing the raised VLDL and LDL level and increasing the lowered HDL level.

  • Effect of an acidic polysaccharide fraction In Korean white and red ginseng on lipolytic action of Toxohormone-L was studied. Crude acidic polysaccharide fraction was extracted from main and lateral root of Korean white and red ginseng separately and purified several times. Inhibitory effect of crude polysaccharide fraction was determined by unit (1 unit is loft inhibition rate per Is sample). Yields of purified crude polysaccharide fraction of main and lateral root of red ginseng were 2.9 and 2.2 times higher than those of white ginseng, respectively. Inhibitory effects of main root of white and red ginseng, 11.hen final reaction concentrations of sample were 50, 100, 200, 500 $\mu$g/ml, were 37.2가 and 23.7% higher than those of lateral root of white and red ginseng. Inhibitory effect of main root of red ginseng was 2.3 times higher than that of white ginseng.

  • Korean ginseng(Panax gmseng C.A. Meyer) is very difficult to obtain stable production of qualified ginseng roots because of variable stresses in soil environments. In environment stresses, soil condition is the most important factor, among which nutrients, especially inorganic materials such as N, P, K, Ca, Mg, Fe, etc., influence greatly on the ginseng growth. However, present ginseng field soils in Korea contain so much amount of such inorganic materials that a variety of remarkable disorders were noted in many ginseng plantations, resulting in decrease of qualitative ginseng production. Therefore, it is required to search for genetic resources and genes tolerant to salt stress for the development of ginseng cultivars. Selection of stress-tolerant ginseng lines in fields is very difficult because it is almost impossible to control properly the environmental conditions of soil. On the contrary, it can be studied with ease to search for stress-tolerant ginseng lines through in vitro culture because of easy manipulation of stress conditions. Murashige & Skoog(MS) media with 2.5 folds of $KNO_3,\;NH_4NO_3,\;MgSO_4\;7H_2O,\;KH_2PO_4,\;and\;CaCl_2\;2H_2O$ was established for the selection of ginseng lines tolerant to salt stress under the embryo culture.

The Panax ginseng Flowering Locus T Shows Age Specific Expression Pattern in Ginseng and Increases Root Length in Transgenic Arabidopsis

  • Panax ginseng Meyer is a perennial medicinal plant, the roots of which has been used in the traditional formulations in Oriental countries. To understand its floral transition, we isolated Flowering Locus T (FT) from ginseng, the bioinformatics analysis of PgFT has revealed a considerable homology to the higher plants, with the essential amino acids for FT function are conserved. The phylogenetic analysis has shown that the PgFT is belonged to the shrub classes of plants and closest kin to Jatropha curcas FT. The expression profiling from juvenile (2-year-old) were abundant in leaves as well as in root and was concentrated in the secondary leaflet and stem bottom in adult (4-year-old) ginseng plant tissues, moreover PgFT transcript displayed photoperiod dependent oscillation. The ectopic expression of PgFT in Arabidopsis thaliana, exhibit precocious flowering and several floral pathway integrators were up-regulated, interestingly their root length was increased in the transgenic seedlings. Therefore, we could conclude that PgFT encodes a florigen that acts as a key regulator in the flowering pathway in ginseng and hypothesize that, it might involve in the underground organ development as well. We believe our finding could provoke future studies on the physiology and development in P. ginseng.

Effects of Some Liming Materials Application on the Alleviation of the Symptoms of Mn Toxicity Inculding Marginal Leaf Chlorosis (석회류제의 시용이 엽연형 황피발생 억제에 미치는 영향)

  • Effect of liming material application on the prevention or cure of Mn toxicity symptoms including marginal leaf chlorosis in 3-year-old ginseng plants grown in acidic soils were investigated. It was found that the ratio of Fe to Mn was above about 0.5 and the ratio of Mn to Fe was below about 2.00 in 4-year old ginseng leaves when liming materials were applied in field experiments. It was apparent that the occurrence of marginal leaf chlorosis was decreasing affected by application of Ca, Mg and Fe. The appearance of marginal leaf chlorosis was decreased to about 78 percent in 4-year-old ginseng and to about 69 percent in 5-year-old ginseng, and then the fresh root weight was increased up to 66 percent in 4-year ginseng plants.

  • This study was examined to investigate the time course behaviors of cell growth and sucrose consumption, and effects of various elicitors on ginsenoside production in batch suspension cultures of Panax ginseng Meyer. Suspended cells reached to the stationary phase at 12 days after innoculation. The maximum cell concentration was 14.7 g-DCW/L at 17 days. The highest cell growth rate was 0.59 g-DCW/L d. The sucrose used as a sole carbon source was hydrolysed to glucose and fructose in 4 days, then quickly utilized until the middle-log phase and consumpted completely at 16 days. Various elicitors were app1ied at 8 days from inoculation which is the middle-log phase. Among the elicitors tested, jasmonic acid was the most efficient to increase the ginseneside production, which was 1.5 times higher than control.

Effect of Blue and Red LED irradiation on Growth Characteristics and Saponin Contents in Panax Ginseng C. A. Meyer (청색과 적색 LED 처리가 인삼의 생육 및 사포닌 함량에 미치는 영향)

  • This study was conducted to assess the response of LED (Light-emitting diode) irradiation on the growth characteristics and saponin contents of Panax ginseng C. A. Meyer. LED irradiation showed a positive effect for most of the parameters studied. The content of chlorophyll a in leaves was increased by 4.9$\sim$36.5%, under LED and fluorescent light conditions compared to the control. The content of chlorophyll b was also increased by 44.4$\sim$55.6% under blue and red LED compared to the control except under the red plus blue LED condition. The shoot and root weight were increased by $20\sim60%$ and $14.8\sim59.3%$, respectively under LED and fluorescent light conditions compared to the control. The total saponin content was increased by 1.8% under blue LED compared to the control, while total saponin content was decreased by 8.8$\sim$11.5% under red LED, red plus blue LED and fluorescent light conditions.
