Changes in Abscisic Acid and Gibberellin Levels during Stratification in Panax ginseng Roots (인삼근의 휴면타파과정에 있어서 Abscisic acid 함량 및 Gibberellin 활성의 변화)

  • The present study was carried out to get the basic information for clarifying physiological mechanism of breaking dormancy and sprouting in Panax ginseng roots. Changes in Abscisic acid (ABA) content and Gibberellin (GA) activity were investigated in one-year-old root during stratification at 4$^{\circ}C$. 15$^{\circ}C$. and 15$^{\circ}C$ after 60day-treatment at 4$^{\circ}C$. Sprouting rate at 15$^{\circ}C$ was 35% in 30days storage at 4$^{\circ}C$ and 100% in longer than 60days, but there was no sprout in both the constant treatment at 4$^{\circ}C$ or 15$^{\circ}C$ regardless of the treatment period. The longer the period of low temperature treatment. number of days to the first and 50% sprouting was shortened, and number of days to 50% from first sprouting was also shortened. ABA content in the upper part of root(contained bud) was gradually increased at both 4$^{\circ}C$ and 15$^{\circ}C$ as the treatment period was extended. and the degree of increase was higher at 15$^{\circ}C$. In the lower part. it showed a slight increase at 15$^{\circ}C$. while showed little change at 4$^{\circ}C$ throughout the treatment period. In the 15$^{\circ}C$ treatment after 60days at 4$^{\circ}C$, it was greatly increased in the upper part. while rather slightly decreased in the lower part of root. GA activity in the upper part was gradually decreased at both 4$^{\circ}C$ and 15$^{\circ}C$, and the degree of decrease was higher at 15$^{\circ}C$. In the lower part. it was similar tendency to those in the upper part. In the 15$^{\circ}C$ treatment after 60days at 4$^{\circ}C$. it was remarkably increased in both the upper and lower part. The increase was great in the low Rf region, while the decrease appeared relatively in the high Rf region compared to those of 60day-treatment at 4$^{\circ}C$. The above results indicated that the breaking dormancy and sprouting of bud were closely associated with the degree of GA activities in response to temperature condition .during stratification rather than the direct effect associated with the changes in ABA content.

Effect of Ultrasonic treatment on the Isolation of the Chlamydospores of Cylindrocarpon destructans Causing Root rot of Panax ginseng (초음파처리에 의한 인삼 뿌리썩음병균 Cylindrocarpon destructans 후막포자의 분리)

  • Chlamydospores were isolated from hyphae of Cylindrocanon destmctans by homogenization and/or ultrasonic treatment. Rate of the isolated chlamydospores by the homogenization with glass tissue grinder were 9.8% of all total chlamydospores formed in the culture of C. destructans. The length of mycelial fragments after the homogenization was about 400㎛ They were, however, formed in clusters of the chlamydospores and the mycelia The rate of the isolated chlamydospores from additional ultrasonic treatment after the homogenization of the mycelia were 74.3%. The length of mycelial fragments with the ultrasonic treatment was about 20 fm and chlamydospores seemed to be isolated from the mycelial mats and dispersed evenly in the culture. The numbers of chlamydospore in a catena were 1 to 8 cells after the homogenization on potato dextrose agar (PDA). Meanwhile the numbers of them after added ultrasonic treatment were 1 to 4 cells. Germination percentages of the isolated chlamydospores from the ultrasonic treatment were 46.8% after incubation of 2 days on PDA at 20。C and 60.7% after incubation of 13 days at 5。C, respectively. Germination rate of chlamydospores to the total chlamydospores produced by the ultrasonic treatment was 55.8%. However, it was increased to 74% when it was measured in the germinated catenae to the total catenae.

The comparative study of anti-allergic and anti-inflammatory effects by fermented red ginseng and red ginseng (홍삼과 발효홍삼의 항염증 작용 및 항알러지 효과 비교)

  • Red ginseng(RG, steamed and dried root of Panax ginseng C. A. Meyer, family Araliaceae) and fermented red ginseng(FRG, fermented red ginseng by yeast and lactic acid bacteria) are known to show different pharmacological effects by changed composition of saponins through fermentation. We examined the effects of RG and FRG on $\beta$-hexosaminidase secretion, ICAM-1 expression, the mitogen-induced proliferation of lymphocyte from mice in ex vivo systems and HaCaT cell(keratinocyte) proliferation to compare the anti-allergic and anti-inflammatory effects between both groups. RG groups showed inhibition of $\beta$-hexosaminidase secretion and ICAM-1 expression at $1{\mu}g/ml$, $10{\mu}g/ml$ and the same effects were observed at all concentrations in FRG groups. In our study, RG increased LPS-induced B cell proliferation at $1{\mu}g/ml$ and ConA-induced B cell proliferation at $100\;{\mu}g/ml$ but FRG decreased LPS- and ConA-induced lymphocytes at $100\;{\mu}g/ml$. We showed that FRG increased the proliferation of HaCaT at 1, $10{\mu}g/ml$ but not by RG. These findings suggest that RG and FRG might have anti-inflammatory and anti-allergic effects, which can be needed to proper clinical concentration to applied to various allergic diseases and inflammation.

Neuroprotective Effect of $NeuBo153^{\circledR}$ on Transient Focal Cerebral Ischemia in Rats ($NeuBo153^{\circledR}$의 중풍동물 모델에 대한 뇌신경 보호효과)

  • Objectives : The purpose of the present study is to observe the neuroprotective effect of the $NeuBo153^{\circledR}$ on transient focal cerebral ischemia in rats. Methods : $NeuBo153^{\circledR}$ was made by mixing the herbs, mainly the root of Panax ginseng, the root of Rehmannia glutinosa and Poria cocos, the stem bark of Acanthopanax senticosus, the root of Scutellaria baicalensis and Mel, and heating for 96 hours. Transient Focal cerebral ischemia (2 h of ischemia, 22 h of reperfusion) was induced by intraluminal suture method with SD rats. Sensory motor function was tested by rotarod test, prehensile traction test, beam balance test and foot fault test at 24 h after ischemia. The brain slices were stained by 2% 2, 3, 5-triphenyltetrazolium chloride and the infarct volume was measured by graphic analyzer at 24 h after ischemia. Results : $NeuBo153^{\circledR}$ treated group did not show significant differences compared with vehicle treated group in body temperature. Oral administration of $NeuBo153^{\circledR}$ reduced brain infarct volume by 29.7% compared with vehicle treated group. $NeuBo153^{\circledR}$ also showed protective effects on sensory motor functional deficits. Conclusion : $NeuBo153^{\circledR}$ treatment reduced brain damage and improved functional deficits induced by MCAo. It showed neuroprotective effects even when treatment was relayed 2 h after injury. Further research is required to evaluating long term functional recovery am accurate therapeutic range and mechanisms.

Ginsenoside Rg2 Inhibits Lipopolysaccharide-Induced Adhesion Molecule Expression in Human Umbilical Vein Endothelial Cell

  • Vascular cell adhesion molecule 1 (VCAM-1), intercellular adhesion molecule 1 (ICAM-1), P- and E-selectin play a pivotal role for initiation of atherosclerosis. Ginsenoside, a class of steroid glycosides, is abundant in Panax ginseng root, which has been used for prevention of illness in Korea. In this study, we investigated the mechanism(s) by which ginsenoside Rg2 may inhibit VCAM-1 and ICAM-1 expressions stimulated with lipopolysaccharide (LPS) in human umbilical vein endothelial cell (HUVEC). LPS increased VCAM-1 and ICAM-1 expression. Ginsenoside Rg2 prevented LPS-mediated increase of VCAM-1 and ICAM-1 expression. On the other hand, JSH, a nuclear factor kappa B (NF-${\kappa}B$) inhibitor, reduced both VCAM-1 and ICAM-1 expression stimulated with LPS. SB202190, inhibitor of p38 mitogen-activated protein kinase (p38 MAPK), and wortmannin, phosphatidylinositol 3-kinase (PI3-kinase) inhibitor, reduced LPS-mediated VCAM-1 but not ICAM-1 expression. PD98059, inhibitor of mitogen-activated protein kinase kinase/extracellular signal-regulated kinase (MEK/ERK) did not affect VCAM-1 and ICAM-1 expression stimulated with LPS. SP600125, inhibitor of c-Jun N-terminal kinase (JNK), reduced LPS-mediated ICAM-1 but not VCAM-1 expression. LPS reduced IkappaB${\alpha}$ ($I{\kappa}B{\alpha}$) expression, in a time-dependent manner within 1 hr. Ginsenoside Rg2 prevented the decrease of $I{\kappa}B{\alpha}$ expression stimulated with LPS. Moreover, ginsenoside Rg2 reduced LPS-mediated THP-1 monocyte adhesion to HUVEC, in a concentration-dependent manner. These data provide a novel mechanism where the ginsenoside Rg2 may provide direct vascular benefits with inhibition of leukocyte adhesion into vascular wall thereby providing protection against vascular inflammatory disease.
