  • From the Paeoniae Radix, gallic acid methyl ester, (+)-catechin, paconiflorin, albiflorin, paeonol, ${\beta}-sitosterol$ and campesterol, benzoic acid, oleanolic acid, hederagenin and ${\beta}-sitosterol$ 3-O-glucoside were isolated and characterized by means of spectral data. Oleanolic acid and hederagenin have been isolated for the first time from the genus Paeonia.

Studies on Nutritional Analysis and Antioxidant activity of Oriental Medicines with Bloodstream Improvement (혈류개선 효능을 지닌 한약재의 영양성분 분석 및 항산화 활성에 관한 연구)

  • This study investigated applicable values and developments of 5 different kinds of oriental medicine materials as functional supplements or medicinal uses, known to improve blood circulation by means of analyzing nutritional compositions and antioxidant activities. Among general nutritional compositions, protein, iron and copper contents in Polygini Multiflori Radix (20.28%, $132.22{\mu}g$/100 g, $26.54{\mu}g$/100 g), Angelica Gigantis Radix (16.54%, $98.20{\mu}g$/100 g, $32.15{\mu}g$/100 g) and Paeoniae Radix(18.67%, $62.10{\mu}g$/100 g, $20.32{\mu}g$/100 g) were significantly higher than other materials. Angelica Gigantis Radix had the highest of total polyphenols and flavonoid contents, Polygini Multiflori Radix (middle), Paeoniae Radix (the lowest). Antioxidant activities by using DPPH radicals removal showed the highest in Polygini Multiflori Radix, Angelica Gigantis Radix (middle), Paeoniae Radix (the lowest). This study tried to use integrated approaches, including Western and Oriental medicine to improve blood circulation and to set up base to develop products originated from natural compositions.

Isolation of Hyperlipidemic Substances from Methanol Extract of Paeoniae Radix (작약의 메탄올 추출물로부터 항고지혈 활성성분의 분리)

  • We previously showed that a methanol extract of Paeoniae radix decreased total cholesterol level in rats with hyperlipidemia. In order to isolate the active ingredient(s), the methanol extract of Paeoniae radix was fractionated with chloroform/methanol(4:1) solution and isolate into soluble part and insoluble part of the the methanol extract. Above two parts were tested on the experimentally induced hypercholesterolemia in rats for the lowering effect of serum lipoprotein contents. Hyperlipidemia was induced on male Wistar rats by feeding high choleserol diet for 7 days. After oral administration of above samples for 4 weeks, serum lipid profile was verified on these rats by measuring total cholesterol, triglyceride, high density lipoprotein cholesterol and low density lipoprotein cholesterol. The chloroform/methanol(4:1) soluble part and insolule part showed lowering activity of total cholesterol level and triglyceride level at 4 week point significantly(p<0.01 and p<0.05) compare with the control group and the soluble part was more effective.

Inhibitory Effect of Paeoniae Radix Alba Ethanol Extract on Osteoclast Differentiation and Formation (백작약 에탄올 추출물의 파골세포 분화 및 생성 억제 작용)

  • Bone destruction is a pathological symptom of some chronic inflammatory diseases, such as rheumatoid arthritis and periodontitis. Inflammation-induced bone loss of these diseases results from increased number and activity of osteoclasts. Paeoniae Radix Alba has been used in korean traditional medicine to treat disease including inflammation, gynecopathy and various pain. However, these effects have not been tested on osteoclasts, the bone resorbing cells that regulate bone metabolism. Here, we investigated the effects of Paeoniae Radix Alba ethanol extract (PRAE) on receptor activator of nuclear factor-kappa B ligand (RANKL)-mediated osteoclast differentiation and formation. Osteoclast differentiation and formation were measured by tartrate resistant acidic phosphatase (TRAP) staining and TRAP solution assay. The treatment of PRAE on bone marrow derived macrophages (BMMs), which is known as osteoclast precursor cells, inhibited osteoclast differentiation and formation in a dose-dependent manner. In addition, the expression of osteoclast differentiation marker genes was suppressed by PRAE treatment. This inhibitory effect of PRAE resulted from significant repression of c-Fos expression, and subsequent reduction of NFATc1 expression which was previously reported as a master transcription factor for osteoclastogenesis in vitro and in vivo. These results demonstrate that PRAE negatively regulates osteoclast differentiation and formation and suggest that PRAE can be used as a potent preventive or therapeutic candidate for various bone diseases, such as postmenopausal osteoporosis, periodontitis and rheumatoid arthritis.

Review on Original Plane of Oriental Medicines Used in Korea (국내 유통 한약재의 기원식물에 관한 고찰)

  • To clarify the botanical origins of oriental medicines which have been argued or confused for plant origins, species of original plants were investigated through the textural research for oriental medicines and the comparison of Chinese, Korean, Japanese and North Korean Pharmacopoeia. Twenty oriental medicines were studied; Angelicae Gigantis Radix, Rhei Rhizoma, Atractylodis Rhizoma Alba, Aconiti Lateralis Radix Preparata, Acanthopanacis Cortex, Osterici Radix, Cnidii Rhizoma, Saposhnikovae Radix, Magnoliae Cortex, Paeoniae Radix, Liriopis Tuber, Zanthoxyli Fructus, Achyranthis Radix, Sinomeni Caulis et Rhizoma, Polygonati Rhizoma, Cinnamomi Cortex, Visci Herba et Loranthi Ramulus, Fritillariae Thunbergii Bulbus, Pogostemonis Herba, and Curcumae Longae Radix.

Effects of Paeoniae radix alba(PRA) on Skin whitening and Elasticity using Melanoma cells (백작약(白芍藥)이 피부 미백 및 주름에 미치는 영향)

  • Objective : Paeoniae radix alba(PRA) can enrich the blood and regulate menstruation, astringe yin and arrest sweating, calm the liver and arrest pain. This study was designed to investigate effects of PRA on skin whitening and elasticity using melanoma cells. Methods : In this experiment, effect of PRA on cell viability, inhibition of melanin synthesis and inhibitory effect on tyrosinase and elastase. Results : 1. More than $1,000\;{\mu}g/ml$ of PRA treated group showed lowered proliferation rates significantly compared to non-treated control group. 2. All of treated groups were lower levels of melanin synthesis respectively. 3. PRA did not show inhibitory effect on tyrosinase activities in vitro. But, PRA suppressed tyrosinase activities in B16F10 cells significantly. 4. PRA suppressed elastse type 1 activities in dose-dependent manner in vitro. But, PRA slightly suppressed elastase type 4 activities in vitro, and PRA also slightly suppressed elastase activities in vivo. Conclusion : These results suggest that PRA can inhibit melanin synthesis through ihhibitory action on tyrosinase activity and inhibt elastase activity, and also suggest that these results can be used for the study on maintaining skin whitening or elasticity.

Studies on the Heavy Metal Contents in Cultivated Herbal Medicines and Their Soils at North Kyungbuk Area (경북북부지역 재배한약재와 토양의 중금속 함량연구)

  • The concentration of heavy metal were studied for the estimation of quality in herbal medicines which is Bupleuri Radix, Paeoniae Radix, Dioscoreae Rhizoma and Astragali Radix in products of north Kyungbuk area. And we compared with heavy metal concentration of herbal medicines in products of Kyungbuk and the other area of Korea. Also, we collected soils from several area in Kyungbuk, and determined heavy metal concentration of soils by ICP. We compared with herbal medicines in products of Kyungbuk and their soils. The results were as follow : Kyungbuk area appear to be more proper to cultivated land than other area. For the heavy metals contents of Kyungbuk area, in Gong Jung Ri, represented the higher level, it may be due to a near the road. Correlation coefficients of heavy metal contents in cultured herbal medicines and their soils did not give reproducible results.

The Protective Effect of Paeoniae Radix Extract against 1-methyl-4-Phenylpyridium-induced Apoptosis on SK-N-MC Neuroblastoma Cells

  • Background: There are increasing neuro-degenerative disorders with aging. Paeoniae Radix(PR) possesses various pharmacological effects such as sedative, analgesic, anti-inflammatory, anti-stress and neuro-protective actions. Also antiaging and anti-cancer effects of PR were reported. Our purpose was to investigate whether PR is useful on the treatment of Parkinson's disease, one of the neuro-degenerative disorders. Objective: We investigated whether Paeonia Radix possesses a protective effect against 1-methyl-4 phenylpyridine(MPP+)-induced cytotoxicity in neuronal cells. Methods: 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT) assay, flow cytometry, DNA fragmentation assay, reverse transcription-polymerase chain reaction(RT-PCR), and Western blotting were performed on SK-N-MC neuroblastoma cells. Results: Cells treated with MPP+ exhibited several apoptotic features, while cells pre-treated with Paeonia Radix prior to MPP+ exposure showed s decrease in the occurrence of apoptotic features. Conclusions: These results suggest that Paeonia Radix may exert a protective effect against MPP+-induced apoptosis in SK-N-MC neuroblastoma cells.

Skin Wrinkle Improvement Effect of Paeoniae radix and processed Paeoniae radix Through inhibition of Advanced glycation end products (AGEs) (작약 및 포제작약의 최종당화산물 억제를 통한 피부 주름 개선 효과)

  • Objectives : Collagen decrease of Skin appears through various path ways. One of causes may be the Advanced glycation endproducts (AGEs) that combine formation of glucose and protein. The aim of this study was to explore the prevent wrinkle formation of Paeoniae radix (PR) and heated Paeoniae radix (HPR) via AGEs path way. Methods : AGEs formation inhibitory activities of PR and HPR measured using bovine serum albumin, glucose, and fructose. To evaluate the protective effects of PR and HPR in diabetic rats induced with streptozotocin (STZ) and methyl glyoxal (MGO), SD rat were distributed into four groups. Normal rats (Nor), AGEs-induced rats (Con), AGEs-induced rats treated with 100 mg/kg PR(PR), AGEs-induced rats treated with 100 mg/kg HPR (HPR). To induce AGEs, streptozotocin (50 mg/kg) was administered intraperitoneally and after 3 days administrated 100mM methyl glyoxal for 3 weeks. Results : The oral administration of HPR inhibited AGEs in skin tissues compared with PR. The increased reactive oxygen species (ROS) levels in the serum were diminished by HPR treatment. The analyses of kidney and skin tissues proteins indicated that HPR treatment effectively reduced AGEs related protein levels as compared to that by PR treatment. Also, HPR decreased anti-oxidant related protein levels in skin tissues such as catalase, glutathione peroxidase. Moreover, it inhibited the reduction of COL1A2 by decreasing MMP-1. Conclusion : Based on these results, it was suggested that PR and HPR could have Improving effects on wrinkle formation. These evidences provide useful information for the development wrinkle formation treated agent.
