Characterization of Acetoxyscirpendiol of Paecilomyces tenuipes as Inhibitor of Sodium Glucose Co-transporters Expressed in Xenopus laevis Oocytes

  • Cordyceps possesses numerous health-promoting ingredients including hypoglycemic agents. The mechanism for the reduction of circulatory sugar content, however, is still not fully understand. In this study, 4-beta acetoxyscirpendiol (ASD) was purified from the methanolic extracts from fruiting bodies of Paecilomyces tenuipes. Na+/Glucose transporter-1 (SGLT-1) was expressed in the Xenopus oocytes. The effect of ASD on the oocyte expressed SGLT-1 was analyzed utilizing the voltage clamp and 2-deoxy-D-glucose (2-DOG) uptake studies. ASD was shown to significantly inhibit SGLT-1 activity compared to the non-treated control in a dose- dependent manner. In the presense of its two derivatives (diacetoxyscirpenol or 15-acetoxyscirpendiol), SGLT-1 activity was greatly inhibited similarly as ASD. Between ASD derivatives, 15-acetoxyscirepenol showed inhibition equivalent to that of ASD while diacetoxyscirpenol did less degree of inhibition. Insummary , these results strongly indicate that ASD in P. tenuipes may serve as a functional substance in lowering blood sugar in the circulatory system. ASD and its derivatives can be utilized as inhibitors of SGLT-1.

  • Cordyceps, an entomopathogenic fungus, contains many health-promoting ingredients. Recent reports indicate that the consumption of cordyceps helps reduce blood-sugar content in diabetics. However, the mechanism underlying this reduction in circulatory sugar content is not fully understood. Methanolic extracts were prepared from the fruiting bodies of Paecilomyces tenuipes, and 4-beta acetoxyscirpendiol (4-ASD) was eventually isolated and purified. $Na^+$/Glucose transporter-1 (SGLT-1) was expressed in Xenopus oocytes, and the effect of 4-ASD on SGLT-1 was analyzed utilizing a voltage clamp and by performing 2-deoxy-D-glucose (2-DOG) uptake studies. 4-ASD was shown to significantly inhibit SGLT-1 activity compared to the non-treated control in a dose-dependent manner. In the presence of the derivatives of 4-ASD (diacetoxyscirpenol or 15-acetoxyscirpendiol), SGLT-1 activity was greatly inhibited in an 4-ASD-like manner. Of these derivatives, 15-acetoxyscirepenol inhibited SGLT-1 as well as 4-ASD, whereas diacetoxyscirpenol was slightly less effective. Taken together, these results strongly indicate that 4-ASD in P. tenuipes may lower blood sugar levels in the circulatory system. We conclude that 4-ASD and its derivatives are effective SGLT-1 inhibitors.

Effect of sawdust on cordycepin production from the medicinal fungus Pesilomyces tenuipes in submerged culture

  • Cordycepin (3'-deoxyadenosine) is a nucleoside analog known for its diverse range of biological activities. This study investigated the effect of different types of sawdust on the production of the bioactive compound cordycepin. The results of the study showed that different types of wood sawdust affected the biosynthesis of cordycepin and a significant increase was observed when the conventional SDB medium was replaced with 1% NaOH treated pine sawdust. To optimize cordycepin production from Paecilomyces tenuipes in a medium containing 1% NaOH-pretreated pine sawdust, we employed Response Surface Methodology (RSM) in its Box-Behnken design (BBD) canonical form. The optimal conditions were determined as follows: a particle size of 109.5111-mesh (140 ㎛) for 1% NaOH-pretreated pine sawdust, an input weight of 21.1679 g/L, and an incubation time of 73.8423 hours. According to our model, this combination is expected to yield a maximum cordycepin content of 896.1428 ㎍/mL. Experimental validation of this prediction was performed using the suggested optimal conditions, resulting in an average cordycepin content of 922.6771 ㎍/mL across three replicates, thus confirming the model's accuracy.

  • Entomogenous fungi의 수집된 14균주를 대상으로 종내 그룹의 상호 유연관계를 분석하고자 자실체 및 불완전세대의 형태적, 배양적 특징 관찰과 RAPD 검정을 실시하였다. Paecilomyces tenuipes는 수 없이 많은 흰색 분생포자가 자실체에 불어 눈꽃모양을 나타냈으며, 곤봉형 phialides위에 구형 또는 타원형의 분생포자가 연쇄상을 이루었다. Cordyceps militaris는 담황색 자실체가 방망이 모양으로 번데기에서 형성되었으며, 불완전세대는 Verticillium으로 단생의 phialides에 구형의 분생포자가 Acremonium-type으로 부착되어 있었다. Beauveria bassiana는 자루에서 여러마디가 이어지며, 각 마디에서 1개씩 분생포자가 붙어있는 zigzag type이었다. 또한 이들의 배양에 적합한 배지는 PDA, SDAY이며, 평면과 단면구조로 Paecilomyces는 벨벳과 평탄, Cordyceps는 양모상과 중앙융기로 생육하였고, Beauveria는 벨벳에 밀가루를 뿌린 모양이었다. URP primer를 이용한 rDNA의 RAPD분석 결과 증폭된 산물의 크기는 100bp에서 2.0kb사이로 genomic DNA fragment는 $10{\sim}14$개이었다. UPGMA 분석에 의한 dendrogram 작성 결과 다양한 유전적 분화를 나타내었으며, 2개의 군으로 그룹화 하였다. P. tenuipes 그룹의 $94.8{\sim}100%$, C. militaris와 C. scarabaeicola, B. bassiana 그룹은 $54.3{\sim}$100%의 상동성을 보였다. 또한 완전세대의 자실체를 형성하는 C. militaris 그룹은 불완전세대인 P. tenuipes와 B. bassiana보다 종내의 변화의 폭이 컸으며, 종내의 세분화가 이루어졌다.

Hepatoprotective Effects of Paecilomyces tenuipes Against Carbon Tetrachloride-induced Toxicity in Primary Cultures of Adult Rat Hepatocytes

  • Paecilomyces tenuipes (PT), one of the Ascomycetes family, has been used for medicinal purposes due to its broad pharmacological activities. The present study was undertaken to investigate the hepatoprotective effects of PT water extracts against $CCl_4$-induced hepatotoxicity in primary cultures of adult rat hepatocytes. When the extract of PT was directly added into the culture medium at 1, 2, and 5 mg/ml, the extracts not only reduce the $CCl_4$-induced elevation of aspartate aminotransferase (AST), alanine aminotransferase (ALT), lactate dehydrogenase, and lipid peroxide, but also protect cultured hepatocytes from $CCl_4$-induced reduction of reduced glutathione, glutathione reductase, glutathione-S-transferase, glutathione peroxidase, catalase and superoxide dismutase. In addition, the effects of PT water extracts on cytochrome P450 enzymes were relatively marginal, indicating that the hepatoprotective effects of PT extract against $CCl_4$-induced toxicity might not be due to the inhibition of $CCl_4$ activation. In conclusion, the PT extracts were effective in protecting against $CCl_4$ induced hepatotoxicity in hepatocyte cultures, at least in part, by scavenging free radicals, and by modulating enzyme systems involved in cellular oxidative stress.

  • The caterpillar-shaped Chinese medicinal mushroom (DongChongXiaCao) looks like a worm in the winter and like a grass in the summer. The fruiting body has been regarded as popular folk or effective medicines used to treat human diseases such as asthma, bronchial and lung inflammation, and kidney disease. The fruiting bodies of Paecilomyces tenuipes that formed on the living silkworm (Bombyx mon) host were used in this examination. This study was carried out to investigate the proximate composition, soluble sugar, amino acid and fatty acid profiles, and contents of the bioactive ingredient including adenosine and D-mannitol in the fruiting-bodies. The moisture content was 57.56%. Soluble sugars found were glycerol, glucose, mannitol and sucrose, and the contents exceeded $24\;mgg^{-1}dry$ weight. Total free amino acid content was $17.09\;mgg^{-1}dry$ weight. Arginine, glycine, proline and tyrosine were main amino acids. The content of oleic acid in fatty acids was high. Adenosine was more abundant in fruiting bodies than corpus.

한국자생 동충하초의 채집 및 자원보고 (Report on the Genus Cordyceps and Its Anamorphs Collected in Korea)

  • 국내 자생 동충하초균의 발생환경과 그 발생빈도를 조사하고 수집된 표본의 형태적 특성에 의해 동정하였다. 국내 산지 19개소에서 1999년부터 2002년까지 수집된 동충하초는 5속 17종이었으며(Cordyceps gracilioides, C. japonica, C. longissima, C. martialis, C. militaris, C. myrmeopila, C. nutans, C. pruinosa, C. sphecocephala, C. tricentri, Hirsutella nutans, Paecilomyces cicadae, P. farinosus, P. tenuipes, paecilomyces sp., Shimizuomyces paradoxa, Tilachidiopsis nigra), 그 외 중국도입 C. sinensis균 특성ㄷ 추가조사하였다. 국내수집 균주는 계곡 및 저습지, 이끼지역, 활엽수림 등에 주고 분포하였으며, 제한된 조사지역과 채집시기에 비하여 다양한 종이 발견되었으나, 종별 채집 수는 10여 점으로서 발생빈도는 대체로 낮았다. 제주도는 내륙과 격리된 남쪽에 위치하므로 비교적 온난하며 한라산은 기후의 수직분포가 커서 종의 분포가 다양하며 특이한 것으로 평가된다. 수집종은 온도가 높고 습기가 많은 8월 초순에 많이 채집되었으며, C. nutans, C. militaris, P. tenuipes의 발생빈도가 현저히 높았고 그 중에서 C. nutans는 667념 중 65%로서 우점종이었다. Cordyceps는 숙주개체 당 1~2개의 자좌를 형성하며, 자낭각 배열과 이차포자 발달에는 종내 변이가 없으나, 자좌의 수, 형태, 색상 드은 발생환경 및 숙주에 따른 변이가 인정되었다.

눈꽃동충하초 재배상 내에 발생된 오염균의 동정 (Identification of Infectious Fungus in the Artificial Cultivation Rooms of Paecilomyces tenuipes)

  • 전라북도 동충하초 재배농가 오염균은 Paecilomyces farinosus(Holm ex S. F. Gray) Brown & Smith로 동정되었으며 특성은 다음과 같다. 분리$.$균은 감자한천배지 상에서 24$\pm$1$^{\circ}C$, 14일간 배양한 결과 배지 앞면은 흰색, 뒷면은 노란색을 띠고 직경 19mm로 생장하였다. 균사 직경은 1.2-1.9$\mu\textrm{m}$이며 분생자병은 투명하며 직경 1.3-1.5$\mu\textrm{m}$이다. phialides는 윤생형으로 분생자병으로부터 2-4개가 형성되며 기부는 부풀어 있고 상단부로 갈수록 폭이 좁아지는 호리병형으로 3.8-5.8${\times}$1.5-1.8$\mu\textrm{m}$이다. phialides의 neck은 직경 0.6-0.8$\mu\textrm{m}$이며, 분생 포자는 분생자형성세포로부터 연쇄상으로 발생하며 타원형 혹은 방추형이며 크기 1.8-2.4${\times}$l.1-1.4$\mu\textrm{m}$이다. 균 농도 1${\times}$$10^{8}$ conidia/$m\ell$로 경피 접종된 누에의 감염률은 41.3%이었으며, 분생자병속 형성률은 23.7%이었다. 균 접종 5-9일 후 누에피부 상에 검은 반점이 형성되었으며, 보호기간 3-4일 후 감염누에 표피는 흰색의 균사에서 점차 노란색 균사로 덮이며, 분생자병속이 발생되었다 분생자병속은 노란색, 선단부로 갈수록 흰색을 나타내었으며 4-6mm 길이로 표피에 한 개에서 다수가 형성되었다. 또한 성숙된 분생자병속의 끝부분에는 분지 후 흰색의 분생포자가 다량 발생하였다.
