Hypoglycemic Effects of Basidiomycetes Mycelia and Cereals Fermented with Basidiomycetes (버섯 균사체 및 배양곡물의 혈당강하 효과)

  • To develop basidiomycetes-fermented cereals with hypoglycemic property, inhibitory effects of basidiomycetes mycelia and basidiomycetes-fermented cereals on postprandial glucose were investigated. In vitro effect of basidiomycetes mycelia on retarding the membrane transport of glucose was compared with pectin. For basidiomycetes mycelia, $13.1{\pm}3.6{\sim}41.8{\pm}8.0%$ of total glucose remained in inner solution of dialysis membrane after dialysis for 120 min, indicating that most of basidiomycetes mycelia might effectively retard membrane transport of glucose. Glucose tolerance of basidiomycetes mycelia and basidiomycetes-fermented cereals was tested on streptozotocin-induced diabetic rats administrated with maltose. Postprandial glucose levels of basidiomycetes mycelia, $389.4{\pm}43.8{\sim}426.3{\pm}49.4mg/dL$, were considerably lower than that of control, $535.3{\pm}78.6mg/dL$, at 30 minutes after maltose administration. Namely, basidiomycetes mycelia showed better postprandial glucose lowering effect than pectin. Brown rice and barley fermented with Paecilomyces japonica showed much lower postprandial glucose level than raw brown rice and barley, especially hypoglycemic effect of barley fermented with Paecilomyces japonica was significant.

Optimization on the Preparation Conditions of Instant Rice Gruel Using Paecilomyces japonica mycelia (동충하초(Paecilomyces japonica)균사체를 이용한 즉석죽 제조조건의 최적화)

  • Cordyceps is very famous of its board biological activities and vital function in traditional medicines. Fourdimensional response surface methodology was used for monitering of instant rice gruels. The organolepitc color of instant rice gruel prepared using mushroom and glutinuous rice showed the maximum score in 38.52% rate of glutinous rice 321.46 mL content of added water and 1.30 g content of P. japonica mycelia. The organoleptic taste of instant rice gruel prepared using mushroom and glutinous rice showed the maximum score in 64.86% rate of glutinous rice 270.66 mL content of added water and 2.32 g content of P. japonica mycelia. The organoleptic flavor of instant rice gruel prepared using mushroom and glutinous rice was maximized in 36.75% rate of glutinous rice, 323.40 mL content of added water and 1.42 g content of P. japonica mycelia. The organoleptic mouth-feel of instant rice gruel prepared using mushroom and glutinous rice was maximized in 65.33% rate of glutinous rice 275.37 mL content of added water and 2.41 g content of P. japonica mycelia. The organoleptic overall palatability of instant rice gruel prepared using mushroom and glutinous rice showed the maximum score in 40.94% rate of glutinous rice 352.79 mL content of added water and 1.27 g content of P. japonica mycelia.

A Study on Antimutagenic Activity of the Extracts from Paecilomyces japonica (동충하초(冬蟲夏草)의 항돌연변이(抗突然變異) 활성(活性)에 관한 연구(硏究))

  • The fruiting bodies of Paecilomyces japonica been used for an anticancer and immuno-stimulating agent as an oriental medicine, Antimutagenecity test with SOS chromotest and antioxidant test with NBT method were carried out using the concentrated culture broth, the extract of mycelia, and that of fruiting bodies. Among the sample extracts tested, the extract of fruiting bodies was most effective to antimutagenecity against the mutagens tested such as MNNG, ethidium bromide(EtBr), 2-aminofluorene(AF) and nitrofluorene(NF). Antimutagenic activity of all the extracts were very effective against mutagen, MNNG. When the extracts were added to certain concentration, antimutagenic activity was enhanced against mutagen, MNNG and NF. Antioxidant activity of the extract from fruiting bodies was highest. However, its activity was very low, compared to ascorbic acid.

Effect of Paecilomyces japonica on the Microbiological Quality and Shelf-life of Jeungpyun (눈꽃동충하초(Paecilomyces japonica)를 첨가한 증편의 미생물학적 품질특성 및 저장성)

  • The purpose of this study was to investigate the effect of Paecilomyces japonica mycelia(PJM) on pH, titrable acidity and microbiological qualify of Jeungpyun(fermented rice cake). Jeungpyun prepared with $0\~\%$ of PJM stored at $5^{\circ}C\;and\;20^{\circ}C$ for 4 weeks and 7 days respectively. Before fermentation of Jeungpyun dough, viable cells of total bacterial counts(TBC), yeasts and lactic acid bacteria(LAB) were $6.0\~9.8\times10^6,\;5.3\~9.0\times10^6,\;5.4\~8.5\times10^6\;CFU/g$, respectively. During the fermentation of dough, viable cells of TBC, yeasts and LAB increased $0.3\~0.4$ log cycle and pH was decreased whereas acidity increased as the progress of fermentation. Total viable cells in Jeungpyun before storage were $5.0\times10^1\;CFU/g$. During storage of Jeungpyun, TBC, yeasts and LAB of control group increased 2.6, 2.4, 2.1 log cycle at $5^{\circ}C$ and 4.8, 4.6, 4.5 log cycle at $50^{\circ}C$, respectively, when reached at maximum level. Major microflora of Jeungpyun was composed of yeasts and LAB during fermentation of dough and storage at $5^{\circ}C\;and\;20^{\circ}C$. Addition of PJM, inhibited the growth of microorganisms, the changes of PH and titrable acidity of Jeungpyun during storage at both of $5^{\circ}C\;and\;20^{\circ}C$. From these results, the addition of PJM extended the shelf-life of Jeungpyun during storage at $5^{\circ}C\;and\;20^{\circ}C$.

A Study on the Method of Culture for Paecilomyces japonica Using an Egg (계란을 이용한 눈꽃동충하초 재배방법에 관한 연구)

  • This study was conducted to established method of culture for Paecilomyces japonica using an egg. Mycelia grew favorably at the temperature of 22~26$^{\circ}C$ on eggs. 5.1g of dry matter basis(average 7.2cm of longer and 199.6 of numbers) of artificial fruiting bodies were harvested at 60 days after inoculation from one of egg. Commercial fruiting bodies of Paecilomyces japonica from silkworms was used for comparative nutriental contents. Cordycepin contents of fruit bodies of Paecilomyces japonica cultivated on eggs and silkworms were not significantly different. Crude fat contents of fruiting bodies of Paeilomyces japonica cultivated from eggs was significantly higher than from silkworms(P<0.05). Mn and Cu contents of fruiting bodies of Paecilomyces japonica cultivated from silkworms were significantly higher than from eggs(P<0.05), but Na, Mg, Fe and Zn contents were significantly higher from eggs(P<0.05). Glycine, Arginine and Proline contents in the fruiting bodies of Paecilomyces japonica cultivated from silkworms were tend to higher than from eggs, but Serine, cystein. methionine, isoleucine and phenylalanin were tend to higher from eggs. These results were made possible that possible mass production of artificial fruiting bodies of Paecilomyces Japonica cultivated on eggs.

Effect of Growth and Culture Conditions of Paecilomyces japonica and Cordyceps militaris on the Formation of Bioactive Substance

  • To investigated the effect of liquid culture conditions and nutrient sources on the formation with bioactive substance of Paeilomyces japonica and Cordyceps militaris cultivated in the country, the result are as follow; The growth temperature of two mycelia is 25$^{\circ}C$ and the proper temperature for cordycepin growth is around 20$^{\circ}C$. The formation amount of bioactive substance by nutrient sources reached its peak with using 2% glucose and 1% galactose in case of carbon sources and 0.4% inorganic compound in case of nitrogen sources. Also, the ratio of C/N was optimal with 3% glucose 1% peptone. For a natural medium, most grains were sufficient but the soybean oil was superb. The formation amount of protein-binding polysaccharide that are used for anticancer substance was in proportion to the growth rate of mycelium, had lots of aeration and showed a trend of increasing when the acidity lower. and the content of structural protein showed a trend of increasing when the acidity lower. However, the content of the structural hexosamin did not get a great the effect of culture conditions and nutrient sources. The constitution of monosaccharide that organizes a protein-binding polysaccharide greatly changed in proportion to carbon sources. When Paecilomyces japonica cultured in a silkworm larvae for 30 days, the content of cordycepin was 204.5 mg/100mL as a dry weight in the fruiting body, 41.8 mg/100mL in mycelium and larva, and the content for each bottle was average 29.5 mg/100mL. In case of Cordyceps militaris for 45days, the fruiting body was 563.5 mg/100mL, the larva and the mycelium was 86.1 mg/100mL, and the content for each bottle was average 65.0 mg/100mL.
