Flow Cytometrical Investigation on Antitumor Activity of Mycelial Culture of Insect-born Fungus Paecilomyces japonica DGUM 32001 (눈꽃동충하초(Paecilomyces japonica DGUM 32001) 균사배양물의 항암 효과에 관한 유세포분석학적 연구)

  • Protein-polysaccharide fractions, PJ-3 and PJ-4, were prepared from mycelial culture filtrate of an insect-born fungus, Paecilomyces japonica DGUM 32001, and subjected to a flow cytometrical analysis for their vivo antitumor and immunomodulating activity in ICR mice. When i.p. injected once daily for semen days at 100 mg/kg, PJ-4 exerted a strong antitumor activity showing the growth inhibition ratio of 85.1% against i.p. implanted sarcoma 180 cells, while PJ-3 showed only a weak activity. Moreover, PJ-4 signiscantly increased the expression level of CD25 (IL-2R $\alpha$-chain) as well as forward scatter (FSC) values of splenic CD8$^{8}$ T cells. It is also noteworthy that PJ-4 strongly induced the peritoneal exudate cells in the same experiment. In an in vitro study, PJ-4 slightly inhibited the growth of sarcoma 180 cells at the concentration of 50$\mu$g/ml or higher. These results strongly suggest that PJ-4 might exert its antitumor activity through immunostimulation as well as direct inhibitory activity on the tumor cells.

Effect of Paecilomyces japonica and Cordyceps militaris Powder on Quality Characteristics of Bread (눈꽃과 번데기 동충하초 첨가가 식빵의 품질특성에 미치는 영향)

  • Paecilomyces japonica and Cordyceps militaris were added to the materials for DongChungHaCho bread, and sensory and mechanical test were performed. Both control and 2% DongChungHaCho added group were acceptable in taste, mouthfeel and odor. Paecilomyces japonica added groups showed higher moisture and volume than control, and Cordyceps militaris stoups showed lower moisture and volume. DongChungHaCho added groups showed lower lightness than control.2% Paecilomyces japonim added group showed similar tendency to control, and it showed the best estimate especially in acceptance, moisture and volume. The optimal addition ratios of DongChungHaCho were evaluated by concentrations of 2% to 4% Paecilomyces japonim and of 2% Cordyceps militaris.

  • This study was carried out to identify the genetic characteristics among isolates of Paecilomyces spp.and Cordyceps spp. by URP-PCR analysis. Twenty URP (universal rice primer) primers of 20 mer which were designed from repetitive sequence of rice, were used for producing PCR DNA fingerprints of the mushrooms. Of them, 5 URP primers, URP2F, URP2R, URP9F, URP4R, and URP17R amplified genomic DNA of the mushrooms with polymorphic PCR patterns. On isolates of Cordyceps militaris, primers URP1F, URP2R, URP6R and URP17R produced PCR polymorphic bands of 4 types. Isolates of Cordypces sp. that are isolated from different area of Korea were identical to isolate of C. militaris, while other species of Cordypces were different to the PCR profiles. However, the URP primers did not identify the polymorphism of PCR profile on isolates of P. japonica.

누에 동충하초(Paecilomyces japonica)를 첨가하여 제조한 고추장의 품질특성 변화에 관한 연구

  • Change in Quality properties of the Kochujang Prepared with the powder of Paecilomyces japonica and extract of p. japonica by different solvents were investigated during 90 days of fermentation at 20$^{\circ}C$. The moisture content was not significantly different, pH of Kochuiang made with p. japonica was lower than control group and decreased continuously according to the fermentation time. Amino nitrogen continuously increased till 60 days and decreased slightly on 90 days. Amino nitrogen of Kochujang made with P. japonica was higher than control group and was highest on 30 days and 60 days by 179.2mg% and 282.2mg% respectively. Value of L, a, b in Color decreased with proportion to fermentation period and L, a, b value of Kochujang made with P. japonica was lower and in particular that of Kochujang made with P. japonica powder was lowest. from the result of sensory evaluation test, the color of control group was similar to“clear red”but that of Kochojang made with powder of P. japonica was close to“dark reddish brown”and that of Kochujang made with extract of P. japonica was darker than that of control group and the preference of dark color was low. Texture was“glossy and smooth”in all and preference was high. In salty taste, the Kochujang by P. japonica addition was stronger and Kochujang made with extract was stronger than that made with powder. In hot taste, the Kochujang made with P. japonica was weaker than control group and the Kochujang made with P. japonica was scored higher in flavor than control group and Kochujang made with powder of P. japonica was the highest in score. In overall preference, the Kochujang made with P. japonica was better than control group like the result of flavor but expecially Kochujang made with water extract of P. japonica among the extract was best one unlike the result of flavor.

  • The production of polysaccharide from Paecilomyces japonica was studied in the shake flask culture. For the cell growth and the polysaccharide production, the optimum synthetic medium was glucose peptone(YMP) medium. The flask culture conditions for the polysaccharide production were $27^{\circ}C$ and 200rpm with the initial pH 9 for 8days cultivation.

  • This study was carried out to improve the liquid culture methods of Paecilomyces japonica. The results show that the size of granular mycelium is smaller when the shaking speed is increased. Especially, the granular mycelium is the smallest at the shaking speed of 150rpm under the photoperiod of 12L-12D. Dry weight of mycelium was averagely 1.216 g in the Sikworm larva (SL) medium, and the weight was 2 times heavier than in the Potato dextrose (PD) medium. By adding 6 g of 6 mmbeads in the SL medium, the dry weight is increased to 1.332 g. The optimal addition of silkworm larval powder to the culture medium for gest harvest was 1.360$\pm$0.67 g in dry weight.

  • This study was carried out to investigate the growth inhibition of Propionibacterium acnes by mycelial culture broth of Paecilomyces japonica in the Mulberry leaf extract. The growth inhibition effect of P. japonica mycelial culture broth against P. acnes in various concentration of Mulberry leaf extract was the most effective in 3% Mulberry leaf extract. The inhibition effect of P. japonica mycelial culture broth against P. acnes according to the culture time was the moust effective after 25 days mycelial cultivation. As the treating amount of the mycelial culture broth was increased, the growth inhibition effect against P. acnes was increased gradually. The growth inhibition effect of mycelial culture broth against P. acnes according to the time of heat treatment was active by 45min at $100^{\circ}C$, while it was inactive more than 60min at $100^{\circ}C$. This result suggested that the antibacterial materials are possible to be glycoside or peptides. Taken together P. japonica mycelial culture in the Mulbarry leaf extract has a possibility to be an element of skin-care cosmetics regulating the acnes.

Effects of Inoculated Diet with Paecilomyces japonica on Broiler Performance (눈꽃동충하초균주(Paecilomyces japonica) 접종사료 급여가 육계의 생산성에 미치는 영향)

  • This study was carried out to investigate the effects of the inoculated diet with Paecilomyces japonica on weight gain, feed efficiency and cordycepin contents in the breast and thigh meats of broilers. One hundred and twenty broiler chicks were divided into four dietary groups ; 1) control (basal diet), 2) T1 (substituted with 5% inoculated diet), 3) T2 (substituted with 10% inoculated diet) 4) T3 (substituted with 20% inoculated diet) during 4 weeks. Cordycepin contents of inoculated diet with Pacilomyces japonica was significantly higher than before inoculation (P<0.05). Body weight gain and feed efficiency were highest in T1 group when fed starter diet (P<0.05) but these were lowest in T3 group fed when fed finished diet (P<0.05). Body weight gain and feed efficiency were the lowest in T2 group for overall period than other groups. Cordycepin contents of breast and thigh meats were significantly higher in Pacilomyces japonica groups than control at 5 weeks of age (P<0.05). However, it was similar among the T1, T2 and T3 groups. These results indicated that body weight gain and feed efficiency tended to increase in birds fed replaced 5% inoculation diet Cordycepin contents of breast and thigh meats would be increased significantly in birds fed inoculated diet (P<0.05).

  • Carbohydrate-binding proteins have been isolated from various sources, including plants, animals, fungi, and bacteria, and they have been used extensively in the detection, localization, and isolation of glycoconjugates. Many carbohydrate-binding proteins are purified from mushrooms, however, only a few proteins with sialic acid-binding specificity have been reported. In the present study, a novel sialic acid-binding protein, designated PJA, has been purified from the mushroom Paecilomyces japonica. followed by extraction and affinity chromatography. (omitted)

Effect of Paecilomyces japonica on the Microbiological Quality and Shelf-life of Jeungpyun (눈꽃동충하초(Paecilomyces japonica)를 첨가한 증편의 미생물학적 품질특성 및 저장성)

  • The purpose of this study was to investigate the effect of Paecilomyces japonica mycelia(PJM) on pH, titrable acidity and microbiological qualify of Jeungpyun(fermented rice cake). Jeungpyun prepared with $0\~\%$ of PJM stored at $5^{\circ}C\;and\;20^{\circ}C$ for 4 weeks and 7 days respectively. Before fermentation of Jeungpyun dough, viable cells of total bacterial counts(TBC), yeasts and lactic acid bacteria(LAB) were $6.0\~9.8\times10^6,\;5.3\~9.0\times10^6,\;5.4\~8.5\times10^6\;CFU/g$, respectively. During the fermentation of dough, viable cells of TBC, yeasts and LAB increased $0.3\~0.4$ log cycle and pH was decreased whereas acidity increased as the progress of fermentation. Total viable cells in Jeungpyun before storage were $5.0\times10^1\;CFU/g$. During storage of Jeungpyun, TBC, yeasts and LAB of control group increased 2.6, 2.4, 2.1 log cycle at $5^{\circ}C$ and 4.8, 4.6, 4.5 log cycle at $50^{\circ}C$, respectively, when reached at maximum level. Major microflora of Jeungpyun was composed of yeasts and LAB during fermentation of dough and storage at $5^{\circ}C\;and\;20^{\circ}C$. Addition of PJM, inhibited the growth of microorganisms, the changes of PH and titrable acidity of Jeungpyun during storage at both of $5^{\circ}C\;and\;20^{\circ}C$. From these results, the addition of PJM extended the shelf-life of Jeungpyun during storage at $5^{\circ}C\;and\;20^{\circ}C$.
