소에 있어서 등장 dextrose액으로 희석한 혈액의 적혈구 침강율(ESR) 측정에 의한 적혈구 침층용적(VPRC)치의 간접계산법 (Indirect calculation for volume of packed red cell(VPRC) by means of erythrocyte sedimentation rate (ESR) of blood diluted with isotonic dextrose solution in cattle)

  • The purpose of this study is to derive indirect values of volume of packed red cell(VPRC), without centrifugation, from erythrocyte sedimentation rate(ESR) of blood diluted with isotonic dextrose solution in cattle. Each of ten blood samples taken from apprently healthy Korean cows and Holstein cows was used to produce eight different mixture of autologous plasma and blood corpuscles such that their values of VPRC lay between 15 to 50ml/100ml, and then each of the blood mixture was again diluted with 5% dextrose solution. The measurements of ESR using 45 degree-angled capillary hematocrit tube, 1.1~1.2mm bore, ($45^{\circ}$-micro-ESR) were practised for the diluted blood of various levels of VPRC under the ambient temperature of $10^{\circ}$, $20^{\circ}$ and $30^{\circ}C$. Reliable values of VPRC on the basis of the correlating linear regressive equation to the ESR could be derived from the values of $45^{\circ}$-micro-ESR/hr in the mixture of one part of whole blood and one part of 5% dextrose solution(1:1). For an aid of practical use, authors suggested a list of the $45^{\circ}$-micro-ESR/hr values of the diluted blood equivalent to VPRC of whole blood.

혐기성 유동층 생물 반응기와 새로운 모델의 AFPBBR에서 유기성폐수 처리시 Biogas 생성과 반응상수에 관한 연구 (A Study on the Kinetics and the Biogas Formation for Organic Wastewater Treatment in Anaerobic Fluidized-Bed Bioreactor and New Model AFPBBR)

  • The anaerobic digestion of organic synthetic wastewater in anaerobic fluidized bed bioreactor (AFBBR) and anaerobic fluidized packed bed bioreactor (AFPBBR) was studied. This study was conducted to evaluate efficiency and reliability of two reactor. Experiment was performed to find the effect of upflow rate with AFBBR and the height of packed bed with AFPBBR. As a result, this program obtained several conclusion. These are given as follows: As applied the upflow rate increased in AFBBR the produced volume of biogas increased, while the gas production and COD removal decreased at above 0.3 m$^3$/h. When a upflow rate is 0.4 m$^3$/h in AFBBR the volatile suspended solid (VSS) became significantly increased. At an organic loading rate from 0.1 to 0.4 of upflow rate in AFBBR, the methane yield was 1.5584 m$^3$CH$_4$/kgCOD removed, and the observed cell yield coefficient was 0.0933 gVSS/gCOD. In case of AFPBBR, the results showed also that 20 cm of height of packed bed was superior to other in the aspect ot biogas production, the content of methane and COD removal. At 20 cm of height, the profile of microorganisms was stable, while at 30 cm the VSS of effluent became higher than AFBBR. Though COD removal of AFPBBR increased with packed bed, COD removal deteriorate with over packing because the loss of pressure became higher in the reactor. At an organic loading rate from 20 to 40 cm of packed bed in-AFPBBR, the methane yield was 2.5649 m$^3$CH$_4$/kgCOD removed, and the observed cell yield coefficient was 0.0506 gVSS/gCOD. Based upon the results obtained, it is suggested that AFBBR and AFPBBR is the most effective conditions at 0.3 m3/h of upflow rate, the 20cm of packed bed, respectively. Can Exogenous Betaine Be an Effective Osmolyte in Broiler Chicks under Water Salinity Stress?

  • A CRD experiment was conducted to evaluate the effects of different exogenous betaine levels (0.000, 0.075, 0.150 and 0.225 percent) on 576 one-day-old male broiler chicks (Ross) under water salinity stress. Different levels of water salinity were made by adding 3 levels of NaCl (0, 1,000 and 2,000 mg/L) to drinking water. Feed and water were available ad libitum. Betaine increased body weight, improved feed conversion ratio, and decreased packed cell volume (p<0.05). Water salinity promoted body weight over the whole period, increased feed intake (11 to 21 and 29 to 42-d) and also improved feed conversion ratio in grower and finisher periods (p<0.01). Breast weight, water consumption (28-d and 42-d) and excreta moisture (28-d) were increased by elevating the level of water salinity (p<0.01). Interaction between dietary betaine and water salinity was significant on plasma osmolarity as well as epithelial osmolarity of the duodenum at 28-d. Epithelial osmolarity was decreased from duodenum to ileum. The data imply that betaine is involved in the protection of intestinal epithelia against osmotic disturbance which can be caused by saline water, but further research is needed to investigate the effects of betaine with higher levels of water salinity.

Biochemical Indices of Vitamin E, Ascorbic Acid and Iron Status : Relation to Diet, Supplement Use and Other Lifestyle Variables in Urban and Rural (Amish) Populations

  • The relation of food and supplemental intake of iron, vitamin E and ascorbic acid and other lifestyle variables to packed cell volume (PCV) and serum vitamin levels was studied in urban and rural (71% Amish) communities. Subjects were interviewed (24-h dietary recalls) on three occasions over 18-months, and blood samples were taken (maximum observations = 442). Mean PCV was lower in rural males (43.3) than in urban males (45.4) despite higher man food iron intake (18.7 and 14.4 mg/day, respectively). Mean meal iron availability was higher at lunch and lower at breakfast and dinner for rural than for urban subjects. Smoking was the number one variable in males and females explaining variance in PCV. Supplemental vitamin E and ascorbate intakes explained the most variance in serum vitamin E and ascorbate levels, respectively. Serum vitamin E was also associated with supplemental ascorbate intake (r=0.29). Serum ascorbate was also associated with food ascorbate intake (r=0.28) and body weight (r=-0.24).

  • The Korea chinese chives are a major edible vegetable and they are traditionally been used as a folk medicine. However recently studies on the Allium tuberosum are mainly on the morphological property and the chemical constituent. There are no reports concerning hematological changes in dogs administrated ethyl acetate extracts of Korea chinese chive. This experiment was conducted to investigate the hematological changes in dogs administrated ethyl acetate extracts of the Korea chinese chives($5m{\ell}/mg$) for 6 days. During administration blood samples were collected at 3h, 6h, 12h and then on daily basis until day 6. Blood samples were analyzed red blood cell(RBC), packed cell volume(PCV), hemoglobin concentration(Hb), mean corpuscular volume(MCV), mean corpuscularl hemoglobin concentration(MCHC), reduced glutathion(GSH), and met-hemoglobin concentration. The RBC count was significantly(p<0.01, p<0.05) decreased during experimental periods. Packed cell volume, hemoglobin concentration were not showed significantly difference during experimental periods. The MCV was significantly(p<0.01, p<0.05) increased from 3 hours after administration to day 4. The MCHC was significantly(p<0.05)decreased on day 6. GSH concentration was not showed significant difference during experimental periods and the met-hemoglobin concentration was significantly(p<0.05) increased on day 3.

과량의 부추 생즙 투여가 소형견의 혈액상에 미치는 영향 (Hematological Effect of Administration Excessive Amount of the Korean Native Chinese Chive Extracts in Small Dogs)

  • This study was conducted to observe the hematological effect of the small dogs when administrated excessive amount of the Korean native chinese chive(Allium tuberosum Rottler) extracts for a short period. In this experiment, clinically healthy dogs(n = 10, average weight = 4.17 kg) were used. They were inserted stomach tube and administrated the extracts(5ml/kg) for 7 days. The followings are the result of this experiment. The red blood cell counts, hemoglobin concentration and mean corpuscular hemoglobin concentration were significantly decreased. (P<0.05) The packed cell volume was slightly decreased. The mean corpuscular volume and methemoglobin concentration were significantly increased.(P<0.05) The white blood cell counts and reduced glutathione were slightly increased. Consequently, We determined that administrated excessive amount of the Korean native chinese chive extracts induced hemolytic anemia.

Simple Method for a Cell Count of the Colonial Cyanobacterium, Microcystis sp.

  • The cell counting of colonial Microcystis spp. is a rather difficult and error-prone proposition, as this genus forms irregularly-shaped and irregularly-sized colonies, which are packed with cells. Thus, in order to facilitate a cell count, four methods of dividing the colonies into single cells were compared, including vortexing, sonication, $TiO_2$ treatment, and boiling. As a result, the boiling method was determined to generate the greatest number of single cells from a colony, and all colonies were found to have divided completely after only 6 min of treatment. Furthermore, no significant cell destruction, which might alter the actual cell density, was detected in conjunction with the boiling method (P=0.158). In order to compute the cell number more simply, the relationship between the colony size and the cell number was determined, via the boiling method. The colony volume, rather than the area or diameter was correlated more closely with the cell number ($r^2=0.727$), thereby suggesting that the cell numbers of colonial Microcystis sp. can also be estimated effectively from their volumes.
