PTP1B Inhibitory Secondary Metabolites from Marine-Derived Fungal Strains Penicillium spp. and Eurotium sp.

  • The selective inhibition of PTP1B has been widely recognized as a potential drug target for the treatment of type 2 diabetes and obesity. In the course of screening for PTP1B inhibitory fungal metabolites, the organic extracts of several fungal species isolated from marine environments were found to exhibit significant inhibitory effects, and the bioassay-guided investigation of these extracts resulted in the isolation of fructigenine A (1), cyclopenol (2), echinulin (3), flavoglaucin (4), and viridicatol (5). The structures of these compounds were determined mainly by analysis of NMR and MS data. These compounds inhibited PTP1B activity with 50% inhibitory concentration values of 10.7, 30.0, 29.4, 13.4, and 64.0 ${\mu}M$, respectively. Furthermore, the kinetic analysis of PTP1B inhibition by compounds 1 and 5 suggested that compound 1 inhibited PTP1B activity in a noncompetitive manner, whereas compound 5 inhibited PTP1B activity in a competitive manner.

  • Quercetin which isolated form the roots of Houttuynia cordata. was determined on the basis of IR, ID and 2D NMR specta by direct comparison with authentic compounds. Protein tyrosine phophatase 1B (PTP1B) is thought to be a negative regulator in insulin signal-transduction pathway. Insulin-resistance by the activation of PTP1B is a hallmark of both type 2 diabetes and obesity. Thus, the compound inhibiting PTP1B can improve insulin resistance and can be effective in treating type 2 diabetes and obesity. Quercetin which measured the inhibitory activity against PTP1B was 92.1% inhibition in the 30 ${\mu}g$/mL, 83.4% inhibition in the 6 ${\mu}g$/mL and 76.5% inhibition in the 3 ${\mu}g$/mL. These results suggest that quercetin retains a potential PTP1B activity.

Screening of Marine Microbial Extracts for Tyrosine Phosphatase 1B Inhibitors

  • Protein tyrosine phosphatase 1B (PTP1B) acts as a negative regulator of insulin signaling, and selective inhibition of PTP1B has served as a potential drug target for the treatment of type 2 diabetes. As part of our searching for PTP1B inhibitors from natural products, the extracts of marine microorganisms were screened for the inhibitory effects on the activity of protein tyrosine phosphatase 1B (PTP1B). Among the tested 304 extracts, 29 extracts exhibited inhibition rate ranging 40.1 - 83.6 % against PTP1B at the concentration level of $30{\mu}g/mL$.

  • Protein tyrosine phosphatase 1B (PTP1B) is predicted to be therapeutic target in treatment of type 2 diabetes and obesity. Thus, in order to search for PTP1B inhibitors, we screened the inhibitory activity of PTP1B in the water extracts of 84 medicinal herbs. Among them, the extracts of Pini Folium, Magnoliae Cortex, Artemisiae asiaticae Herba, Schizonepetae Herba, Menthae Herba, Mume Fructus, Cimicifugae Rhizoma, and Amomi Cardamomi Fructus showed relatively significant (58-68%) inhibitory activity against PTP1B. Especially, the methylene chloride fraction of the methanol extract of Menthae Herba (81% inhibition at 30 ${\mu}g$/ml) showed more potent inhibitory activity against PTP1B than others.

담수산 미세조류로부터 생리활성물질의 탐색 (Screening of Bioactive Materials from Freshwater Microalgae)

  • One hundred and fifty four micro algal strains, newly isolated from nationwide freshwaters in Korea, were screened for their anticancer, ant diabetic, and antibiotic activities. The micro algal strains were cultured with different nutritional conditions that were divided into 4 groups as follows; a normal Allen medium, nitrogen (N)-limited medium, phosphorus (P)-limited medium, and N and P-limited medium. Algal biomass was extracted with a mixture of acetone:H₂O (1:1, v:v) and the extracts were used for the screening of bioactive materials. Anticancer, ant diabetic, and antibiotic materials were screened by the methods of vaccinia Hl-related protein tyrosine phosphates (VHR DS-PTPase) inhibition, protein tyrosine phosphates 1B (PTP1B) inhibition, and paper disk. The inhibition activity of VHR DS-PTPase was observed in 18 strains, having a maximum 79% inhibition from Anabaena affinis and the inhibition activity of PTP1B was observed in 9 strains, having a maximum 97% from Sphaerocystis schroeteri. Microcystis aeruginosa incubated in an N and P-limited medium showed antibiotic activity in 8 species out of 13 pathogenic bacteria. As a whole, it seemed that the stressed condition such as N and/or P limitation increased the production of bioactive materials in micro algae.

Type 3 muscarinic acetylcholine receptor stimulation is a determinant of endothelial barrier function and adherens junctions integrity: role of protein-tyrosine phosphatase 1B

  • The main purpose of this study was to investigate whether type 3 muscarinic acetylcholine receptor (M3R) dysfunction induced vascular hyperpermeability. Transwell system analysis showed that M3R inhibition by selective antagonist 4-diphenylacetoxy-N-methylpiperidine methiodide (4-DAMP) and small interfering RNA both increased endothelial permeability. Using coimmunoprecipitation and Western blot assay, we found that M3R inhibition increased VE-cadherin and ${\beta}$-catenin tyrosine phosphorylation without affecting their expression. Using PTP1B siRNA, we found that PTP1B was required for maintaining VE-cadherin and ${\beta}$-catenin protein dephosphorylation. In addition, 4-DAMP suppressed PTP1B activity by reducing cyclic adenosine monophosphate (cAMP), but not protein kinase $C{\alpha}$ ($PKC{\alpha}$). These data indicate that M3R preserves the endothelial barrier function through a mechanism potentially maintaining PTP1B activity, keeping the adherens junction proteins (AJPs) dephosphorylation.

항당뇨 효능이 있는 천연물의 탐색 및 활성물질의 분석 (Screening of Natural Products for Anti-diabetic Activity and Analysis of Their Active Compounds)

  • 현대인들의 식습관 변화로 인해 대사성 질환의 발생률이 증가하고 있으며, 특히 당뇨병이 가장 큰 문제로 꼽힌다. 하지만 기존의 당뇨병 치료제는 부작용을 동반하고 있어서 예로부터 쓰이던 천연물을 통해 부작용이 적은 항당뇨 활성 물질을 찾고자 하였다. 따라서 본 실험에서는 항당뇨 및 항산화 활성이 있는 천연물을 탐색하기 위하여, DPPH free radical scavenging assay와 α-glucosidase 및 PTP1B inhibition assay를 이용하였다. 항당뇨 효능이 예상되는 12가지의 시료를 메탄올로 추출하고 항당뇨 활성 및 항산화 활성을 측정하였다. 그 중 항당뇨 활성이 우수한 느릅나무, 벌나무 가지, 연 씨앗, 홍화 씨앗 등의 4가지 시료를 대상으로 에틸아세테이트, 부탄올을 이용하여 활성물질을 용매추출하여 항당뇨 활성을 측정하였다. 그 결과, α-glucosidase와 PTP1B 저해활성이 우수한 홍화 씨앗의 에틸아세테이트 분획물(MG-11-E)을 선정하였다. MG-11-E를 대상으로 preparative thin layer chromatography를 수행한 결과, 분획물 #6에서 α-glucosidase 및 PTP1B에 대한 우수한 저해활성을 보였다. 50% 메탄올을 이동상으로 하여 TLC 분획물 #6을 high performance liquid chromatography하여 각각을 분획하고 항당뇨 활성을 측정한 결과, 단일 peak를 보인 RT 4분에서 항당뇨 활성이 확인되었다. 따라서 홍화 씨앗의 추출물의 항당뇨 효능을 나타내는 항당뇨 활성 성분은 PTP1B 보다 α-glucosidase에 대한 저해 활성이 더욱 강하게 나타나 PTP1B를 저해하는 천연물과 조합하여 사용함으로써 당뇨병에 대한 효과를 높일 수 있을 것으로 사료된다.

Anti-obesity and hypolipidemic effects of Rheum undulatum in high-fat diet-fed C57BL/6 mice through protein tyrosine phosphatase 1B inhibition

  • Protein tyrosine phosphatase 1B (PTP1B) is important in the regulation of metabolic diseases and has emerged as a promising signaling target. Previously, we reported the PTP1B inhibitory activity of Rheum undulatum (RU). In the present study, we investigated the metabolic regulatory effects of RU in a high-fat diet (HFD) model. RU treatment significantly blocked body weight gain, which was accompanied by a reduction of feed efficiency. In addition, it led to a reduction of liver weight mediated by overexpression of PPAR${\alpha}$ and CPT1 in the liver, and an increase in the expression of adiponectin, aP2, and UCP3 in adipose tissue responsible for the reduction of total and LDL-cholesterol levels. Chrysophanol and physcion from RU significantly inhibited PTP1B activity and strongly enhanced insulin sensitivity. Altogether, our findings strongly suggest that 2 compounds are novel PTP1B inhibitors and might be considered as anti-obesity agents that are effective for suppressing body weight gain and improving lipid homeostasis.

전통 처방의 Protein Tyrosine Phosphatase 1B 저해 활성 검색 및 작감황신부탕(芍甘黃辛附湯) 처방 분석 (Screening of Korean Traditional Prescriptions with Inhibitory Activity against Protein Tyrosine Phosphatase 1B and Analysis of Jakgamhwangsinbu-tang (芍甘黃辛附湯) Prescription)

  • In order to search for protein tyrosine phosphatase 1B (PTP1B) inhibitors as therapy of type 2 diabetes and obesity from Korean traditional prescriptions, we selected 58 traditional prescriptions based on a review of the Korean traditional medicine books. The hot water extracts of Korean traditional prescriptions were screened for the inhibitory activity against PTP1B. Among the tested extracts, water extracts of Jakgamhwangsinbu-tang, Seonbanghwalmyung-eum, and Takreeonjoong-tang showed relatively good inhibitory activity against PTP1B at the concentration of $30{\mu}g/ml$. Additionally, we evaluated PTP1B inhibitory effect for each herbal ingredient and composition in Jakgamhwangsinbu-tang (芍甘黃辛附湯). Of the tested ingredients from this herbal medicine, water extracts of Paeoniae Radix rubra and Rhei Rhizoma, and ethanol extracts of Paeoniae Radix alba, Rhei Rhizoma, Asiasari Radix, and Aconiti Tuber showed good PTP1B inhibitory effect. Herbal compositions composed of these active herbal ingredients exhibited significant activity for PTP1B inhibition over 70% at $7.5{\mu}g/ml$.

3${\beta}$-Hydroxy-12-oleanen-28-oic Acid 유도체들의 PTP-1B저해활성에 대한 CoMSIA분석 (CoMSIA Analysis on The Inhibition Activity of PTP-1B with 3${\beta}$-Hydroxy-12-oleanen-28-oic Acid Analogues)

  • 기질 화합물로써 3${\beta}$-Hydroxy-12-oleanen-28-oic acid 유도체(1-30)들과 그들의 protein tyrosine phosphatase(PTP)-1B 저해활성에 관한 비교분자 유사성 지수분석(CoMSIA)보델을 유도하였다. QSAR 모델의 통계 값은 CoMFA>CoMSIA${\geq}$HQSAR>2D-QSAR 모델의 순서로 양호하였다. 최적화된 CoMSIA F1 모델은 grid 3.0${\AA}$과 field fit 정렬조건에서 가장 족은 예측성과 상관성($r^2_{cf}$=0.754 및 $r^2_{ncv}$=0.976)을 나타내었다. 저해 활성에 관한 CoMSIA상의 기여비율(%)은 수소결합 받게장(48.9%), 입체장(25.8%) 및 소수성장(25.4%)의 순서이었다. 그러므로 기질 화합물의 PTP-1B에 대한 저해활성은 $R_4$-치환기의 수소결합 받게 장(A)에 의존적이었다. 등고도 분석 결과로부터 $R_1$-치환기는 수소결합 받게장이 작고 $R_3$-치환기는 입체장이 작으며 그리고 $R_4$-치환기는 수소결합 받게장, 소수성 및 입체장이 큰 치환기가 저해활성을 증가시킬 것으로 예측되었다.
