Solution Structure of a Prion Protein: Implications for Infectivity

  • Prions cause neurodegenerative diseases in animals and humans. The scrapie prion protein (PrPSc) is the major-possibly only-component of the infectious prion and is generated from the cellular isoform (PrPC) by a conformational change. Limited proteolysis of PrPSc produces an polypeptide comprised primarily of residues 90 to 231, which retains infectivity. The three-dimensional structure of rPrP(90-231), a recombinant protein resembling PrPC with the Syrian hamster (SHa) sequence, was solved using multidimensional NMR. Low-resolution structures of rPrP(90-231), synthetic peptides up to 56 residues, a longer (29-231, full-length) protein with SHa sequence, and a short here further structure refinement of rPrP(90-231) and dynamic features of the protein. Consideration of these features in the context of published data suggests regions of conformational heterogeneity, structural elements involved in the PrPC\longrightarrowPrPSc transformation, and possible structural features related to a species barrier to transmission of prion diseases.

  • Recently, the construction industry has been changed in such a way that the cost for bridge construction should be optimized or reduced. Therefore, bridges are required be cost-effective in terms of initial construction as well as in the maintenance during service stage. In order to reduce the cost for bridge construction, the Rahmen typed structure, in which the bridge components from superstructure to substructure are integral, has many advantages to reduce the size of structural members including girders, since the loadings from superstructure may be transferred to substructure through the connecting rebars such as stud, etc. This paper studied on the continuous Up and Down Prestressed Concrete (UD PSC) girder bridge in which the reinforced concrete pier cap is integral with the part of girders in superstructure. In previous studies, it is known that the structural behavior of continuous UD PSC girder bridge is quite different compared to the one of the bridges with conventional bearings or shoes to support the loading from girders. Nevertheless, it has hardly been studied about the structural behavior of bridge with UD PSC girder. Therefore, in this study, various dynamic behaviors of continuous UD PSC girder bridge with integral pier cap have been analyzed using numerical method. Furthermore, an equation to evaluate the impact factor is suggested for the UD PSC girder bridge which has two to three continuous spans.

Estimation of Initial Tensile Force Acting on Tendon using the Deformation of a Multi-tendon Anchor Head (멀티 텐던 앵커헤드의 변형을 이용한 텐던의 초기 긴장력 추정)

  • The PSC bridges have recently been widely used in Korea. The PSC bridge is a structure whose performance is improved through the use of tendons and steel bars in deflection and cracking characteristics of the concrete. Therefore, measurement or estimation of the load acting on tendon is important in order to maintain the PSC bridges efficiently and safely. This paper deals with a numerical study on the deformation of a multi-tendon anchor head in order to verify the relationship between the load acting on tendon and the deformation of anchor head. All kinematics, material properties and contact nonlinearity are included for the precise analysis and numerical studies are performed by Abaqus. From the numerical results, it is verified that the hoop strain is most useful in the estimation of the load acting on tendon and strains are affected by various parameters such as friction coefficient, boundary conditions, and arrangement.

An Experimental Study on the Application of Fireproof Panel in Tunnel Duct Slab (터널 풍도슬라브에 사용된 내화패널의 적용성에 관한 실험연구)

  • Purpose: In this study,fire-resistance test were executed to evaluate the effectiveness of the fireproof panel attached to the PSC slab in tunnel. Method: For the fire resistance test, the RWS curve was applied and the furnace of the KICT was used. Result: As a result of the experiment, the maximum temperature measured on the concrete surface of the PSC slab with the fireproof panel was 321.8℃, which was lower than the damage limit temperature of 380℃ for concrete. Also, at the t=25mm, the maximum temperature was 35.2℃, which was lower than the damage temperature of steel, 250℃. The use of precast fire resistance panel(t=30mm) improves fire resistance of PSC structures. Conclusion: As a result of the test, a reinforcement method for attached a fireproof panel in case of fire in a tunnel or an underground roadway is provided to protect a structure from fire. In the future, it is necessary to perform the static performance test of the slab to which the fireproof panel is attached, and to confirm the adhesion performance of the fireproof panel by performing the pull-off test and the fatigue test.

A Development of Torsional Analysis Model and Parametric Study for PSC Box Girder Bridge with Corrugated Steel Web (복부 파형강판을 사용한 PSC 복합 교량의 비틀림 해석모델의 제안 및 변수해석)

  • The Prestressed Concrete (hereinafter PSC) box girder bridges with corrugated steel webs have been drawing an attention as a new structure type of PSC bridge fully utilizing the feature of concrete and steel. However, the previous study focused on the shear buckling of the corrugated steel web and development of connection between concrete flange and steel web. Therefore, it needs to perform a study on the torsional behavior and develop the rational torsional analysis model for PSC box girder with corrugated steel web. In this study, torsional analysis model is developed using Rausch's equation based on space truss model, equilibrium equation considering softening effect of reinforced concrete element and compatibility equation. Validation studies are performed on developed model through the comparison with the experimental results of loading test for PSC box girder with corrugated steel webs. Parametric studies are also performed to investigate the effect of prestressing force and concrete strength in torsional behavior of PSC box girder with corrugated steel web. The modified correction factor is also derived for the torsional coefficient of PSC box girder with corrugated steel web through the parametric study using the proposed anlaytical model.

  • The crystal structure of maltitol, 4-O-${\alpha}$ -D-glucopyranosyl-D-glucitol, has been determined by X-ray diffraction method. The crystal is orthorhombic with cell parameters of a = 8.170(1), b = 12.731(1), c = 13.679(3) ${\AA}$, space group $P2_12_12_1$ and z = 4. The structure was solved by direct methods and refined to R = 0.030 for 1181 observed reflections measured on a diffractometer. The ${\alpha}$-glucose ring has chair conformation. The carbon atom chain of the glucitol residue has the bent, ap, Psc, Psc conformation. The angle at the ring oxygen atom is $112.6^{\circ}$ and the one at the glucosidic oxygen is $117.1^{\circ}$. The molecules are linked by very complicated hydrogen bonds, and there is an intramolecular hydrogen bond between O(1') and O(2').

Reinforcement Effects using V Type External Strands on PSC I Girder Bridges (V자형 배치 외부강선을 이용한 PSC I거더교의 보강 효과)

  • This study intended to analyze reinforcement effects of PSC I girder bridges to which prestresses are introduced using V type of external strands. So that series of bridge loading tests are carried out on existing PSC I girder bridge for the cases of before-reinforcement and reinforcement. The measured results from tests being analyzed and compared with the ones from MIDAS structural analyzing program, the reinforcing effects of the reinforcement system adopted in this study were investigated. It is found out that when the V type systems are applied to the bridge girders, the slope of load distribution factor curves become lower improving soundness of bridge upper structure. And also it is confirmed that the reinforcement system in this study can be taken as helpful for improvement of both flexural and shear ability of PSC I girder bridges, as well as dynamic behavior. Furthermore it is found when the elastic pads are applied to the system, dynamic reinforcing effects are maximized.

Low-Voltage EM(Elasto-Magnetic) Sensing Technique for Tensile Force Management of PSC(Prestressed Concrete) Internal Tendon (PSC 내부 텐던의 긴장력 관리를 위한 저전압 EM 센싱 기법)

  • In this paper, we have verified a low-voltage EM(elasto-magnetic) sensing technique for tensile force management of PSC(prestressed concrete) internal tendon in order to apply the technique to actual construction sites where stable power supply is difficult. From observation of past domestic and overseas PSC structural accident cases, it was found that PS tension is very important to maintain structural stability. In this paper, we have tried to measure the tensile force from a magnetic hysteresis curve through EM sensors according to voltage value by using relation between magnetostriction and stress of ferromagnetic material based on elastic-magnetic theory. For this purpose, EM sensor of double cylindrical coil type was fabricated and tensile force test equipment for PS tendon using hydraulic tensioning device was constructed. The experiment was conducted to confirm relationship between changes of permeability and tensile force from the measurement results of the maximum / minimum voltage amount. The change of magnetic hysteresis curve with magnitude of tensile force was also measured by reducing amount of voltage step by step. As a result, the slope of estimation equation in accordance with magnitude of magnetic field decreases with the voltage reduction. But it was confirmed a similar pattern of change of magnetic permeability for the magnetic hysteresis loop. So, in this study, it is considered that it is possible to manage the tensions of PSC internal tendon using EM sensing technique in low-voltage state.

Nondestructive Damage Detection in PSC Beams : Frequency-Based Method Versus Mode-Shape-Based Method (고유진동수 이용 손상추정법과 모드형상 이용 손상추정법에 의한 PSC 보의 비파괴 손상검색)

  • A methodology to nondestructively locate and estimate size of damage in beam-type structures using a few natural frequencies or a few mode shapes is presented. A damage-localization algorithm to locate damage from changes in natural frequencies and a damage-sizing algorithm to estimate crack-size from natural frequency perturbation are outlined. A damage index algorithm to localize and estimate severity of damage from monitoring changes in mode shapes is outlined. The frequency-based method and the mode-shape-based method are evaluated for several damage scenarios by locating and sizing damage in PS concrete beams lot which a few natural frequencies and mode shapes are generated from finite element models. The result of the analyses indicates that the two methods correctly localize and closely estimate the size of the crack simulated in the test beam.

Durability Evaluation and Defect Pattern Analysis in Railway Bridge Through Field Investigation (현장조사를 통한 철도 고가교 구조물의 내구성 평가 및 결함 패턴 분석)

  • Because of the defect in design, damage in using period, and deterioration in long term exposure to severe environmental condition, degradation of performance in RC (Reinforced Concrete) structures has occurred. This paper contains durability performance evaluation in railway bridges which covers eight districts through field investigation. For the target structures, durability performance is evaluated and the critical problems in use are derived. Additionally, service lifes for the deteriorated structures are evaluated through Durability-Environment index method based on the results from field investigation, and the results are compared with those from the condition assuming the structures without defect, damage, and deterioration. The target structures which consist of RC T girder, PSC girder, RC box, and Rahmen are investigated and the critical damage patterns are derived. They are evaluated to be cracks in PSC girder end, flexural cracks in PSC girder, crack around EPT anchor, and flexural cracks in RC T girder and RC box. The reasons for the critical patterns are also investigated. This study can be utilized for the repair planning considering the different district and the structure types.
