Optimization of the Medium Composition for Heteropolysaccharide-7 Production by Beijerinckia indica L3 Using Response Surface Methodology (표면반응방법을 이용한 Beijerinckia indica L3에 의한 PS-7 생산 최적화)

  • The production of heteropolysaccharide-7 (PS-7) by Beijerinckia indica (B. indica L3) was evaluated in shaker flask culture. The medium optimization was studied using response surface methodology (RSM). A five-level three-factor central composite design was employed to determine the maximum PS-7 yield at optimum levels for whey lactose, glucose and ammonium nitrate contents. The validity of the model could be determined by the regression coefficient, $R^2$. The values of $R^2$ were 0.72, 0.64 and 0.85 in PS-7, DCW and viscosity, respectively. The optimal medium combinations of whey lactose, glucose and ammonium nitrate concentrations on the PS-7 production were whey lactose (2%), glucose (1 %) and ammonium nitrate 5 mM, respectively. The result indicated that PS-7 production was affected significantly by the addition of glucose to whey lactose based on medium and C/N ratio.

Production of Beijerinckia indica HS-2001 in Fed-batch and continuous culture.

  • As a result of broth substitutions when each culture-mediums were difference, whole culture-medium was found to be best feeding solution for production of PS-7 by B. indica. Maximal production of PS-7 was 1$10.0\;g/{\ell}$ and its conversion rate from 2% (w/v) glucose to PS-7 was 50%. After 48 hr, 50%(v/v) medium of working volume began to substitute in 7L jar fermenter. Production of PS-7 increased after 48hr, recovered productivity of PS-7. Following this preliminary culture, the resultant culture was subjected to continuous flow conditions controlled that the dilution rate were $0.01\;{\sim}\;0.04\;h^{-1}$. Production of PS-7 increased at dilution rate $0.0100\;h^{-1}$ whereas productivity of PS-7 decreased gradually in dilution rate $0.0200\;{\sim}\;0.0400\;h^{-1}$. Maximal production of PS-7 was $10.0\;g/{\ell}$ in continuous culture.

Optimization of PS-7 Production Process by Azotobacter indicus var. myxogenes L3 Using the Control of Carbon Source Composition (탄소원 조성 조절을 이용한 Azotobacter indicus var. myxogenes L3로부터 PS-7 생산 최적화)

  • The proteins in whey are separated and used as food additives. The remains (mainly lactose) are spray-dried to produce sweet whey powder, which is widely used as an additive for animal feed. Sweet whey powder is also used as a carbon source for the production of valuable products such as polysaccharides. Glucose, fructose, galactose, and sucrose as asupplemental carbon source were evaluated for the production of PS-7 from Azotobacter indicus var. myxogenes L3 grown on whey based MSM media. Productions of PS-7 with 2% (w/v) fructose and sucrose were 2.05 and 2.31g/L, respectively. The highest production of PS-7 was 2.82g/L when 2% (w/v) glucose was used as the carbon source. Galactose showed low production of PS-7 among the carbon sources tested. The effects of various carbon sources addition to whey based MSM medium showed that glucose could be the best candidate for the enhancement of PS-7 production using whey based MSM medium. To evaluate the effect of glucose addition to whey based media on PS-7 production, fermentations with whey and glucose mixture (whey 1, 2, 3%; whey 1% + glucose 1%, whey 1% + glucose 2% and glucose 2%, w/v) were carried out. Significant enhancement of PS-7 production with addition of 1% (w/v) and 2% (w/v) glucose in 1% (w/v) whey media was observed. The PS-7 concentration of 2% glucose added whey lactose based medium was higher than that of 1% glucose addition, however, the product yield $Y_{p/s}$ was higher in 1% glucose added whey lactose based MSM medium. Therefore, the optimal condition for the PS-7 production from the Azotobacter indicus var.myxogenes L3, was 1% glucose addition to 1% whey lactose MSM medium.

Influence of Service Marketing-Mix(7Ps) on Consumers' Risk Perception of Eating at Family Restaurants in Seoul (서울지역 패밀리레스토랑의 서비스마케팅믹스(7Ps)성과가 고객의 구매위험인지에 미치는 영향)

  • The purpose of this article was to study how the 7Ps influence consumers' risk perception of eating at family restaurants in Seoul. In this study, frequency analysis, reliability analysis, factor analysis and path analysis (SEM) of the data were performed. First, reliability analysis confirmed that the 7Ps performance and risk data could be used in this investigation. Path analysis showed that the 7Ps significantly influenced customers' risk perception of eating at restaurants in Seoul. According to the results, product negatively influenced performance risk (p<0.05) and financial risk (p<0.001); price negatively influenced performance risk (p<0.001), financial risk (p<0.001), and time risk (p<0.01); place negatively influenced performance risk (p<0.01) and time risk (p<0.001); promotion negatively influenced financial risk (p<0.05) and time risk (p<0.001); process negatively influenced performance risk (p<0.001) and time risk (p<0.001); physical evidence negatively influenced performance risk (p<0.05) and financial risk (p<0.001); and people negatively influenced performance risk (p<0.05), financial risk (p<0.001), and time risk (p<0.001). As a result, we confirmed that 7Ps were an effective marketing tactic for reducing consumers' risk perception of eating at restaurants. Therefore, family restaurant companies are recommended to administer the 7Ps without additional cost.

  • Maximal production of heteropolysaccharide-7(PS-7) with a batch culture for 48 hr was $10.0\;g/{\ell}$ and its conversion rate from 2% (w/v) glucose to PS-7 was 50%. After substitution of media, production of PS-7 continued and reached its maximal production. The highest productivity occurred when the fresh medium, which contained all ingredients, for the production of PS-7 was substituted. Higher production of PS-7 was maintained at a dilution rate of 0.0125, which was established as the optimal dilution rate for the production of PS-7 by B. indica HS-2001.

  • Beijerinckia indica was cultured in mineral salts medium (MSM) medium with various carbon and nitrogen sources to improve the production yield of heteropolysaccharide-7 (PS-7). At high C/N ratio, the high concentration of PS-7 was produced until 40 h of the culture, whereas most of the glucose as a carbon source was used for the cell growth at low C/N ratio. However, at the high C/N ratio, PS-7 accumulation stopped at 48 h of the culture due to the increasing viscosity of the culture broth would inhibit the cell growth. Therefore, the optimized value of C/N ratio was 33.3 (20 g/L glucose, 7.5 mM $NH_{4}NO_3$) for the high production of PS-7. In the culture with various carbon sources, B. indica effectively used the hexoses or glucose-generating sugars for PS-7 formation. Especially, sucrose was the best carbon source for the high production of PS-7 (6.96 g/L) with a high viscosity (40772 cp). In the culture of B. indica with MSM medium containing 20 g/L glucose and 7.5 mM $NH_{4}NO_3$ in a 51 fermentor, the highest cell concentration was 2.5 g/L and the highest concentration of PS-7 was 7.5 g/L (35174 cp). The additional nitrogen sources of 7.5 mM $NH_{4}NO_3$, glutamine and glutamate at 12 h of the culture after exhaustion of a nitrogen source regulated the metabolism of carbon sources, therefore the nitrogen sources could control PS-7 synthesis.

  • A bacterial strain producing highly viscous extracellular polysaccharide was isolated from soil. Through morphological, physiological and chemotaxonomical studies, it was identified as a Bacillus sp. and named Bacillus sp. PS-12. The extracellular polysaccharide, named PS-12 was purified by ethanol precipitation, cetylpyridinium chloride (CPC) precipitation and gel permeation chromatography. The purified polysaccharide was found to consist of glucose, mannose, galactose, and fucose, with a molar ratio of approximately 7:3.2:2:1, respectively. PS-12 was investigated for its immunostimulating activity on murine macrophage RAW264.7 cells using an ELISA assay. PS-12 stimulated the production of TNF-${\alpha}$ to a level 50 times greater than the control and also induced 1L-6 secretion in a dose-dependent manner. The cytotoxicity on RAW264.7 cells by PS-12 was relatively low with 10% cytotoxicity at 2 ${\mu}g$/ml. These results indicate that PS-12 is less cytotoxic to immune cells and possess immunomodulating activity in which it can produce cytokines including TNF-${\alpha}$ and 1L-6 from macrophages.

Accelerating effect of some photosensitizers on photodegradation of the herbicide quinclorac in aqueous solution and soil (감광제에 의한 수용액 및 토양 중 제초제 quinclorac의 광분해 촉진효과)

  • In order to artificially reduce the quinclorac residue in aqueous solution and soil, six potential photosensitizers were screened for their effectiveness in enhancing the photodegradation. The degraded amount of quinclorac in distilled water by sunlight was minor compared to that in the dark, indicating that there was little direct photolysis. The photodegradation ratio of quinclorac in methanol was 40.3%. Whereas, the ratios in the presence of photosensitizers PS-1 (aromatic ketone), PS-3 (polycyclic quinone), and PS-6 (inorganic semiconductor) were 96.6, 72.7, and 95.7%, respectively, showing the most photosensitizing effects. In sand, PS-3 was more effective than any other photosensitizer PS-1 (19.6%), PS-3 (64.1%) and PS-6 ($17.9{\sim}19.4%$). five photoproducts of quinclorac in methanol were identified by GC-MS and quinclorac added with the photosensitizer PS-1 gave three photoproducts. Photoproducts with an aldehyde group formed in methanol were confirmed by the reduction of sodium 3,5-dinitrosalicylate in the Lindsay's method. E. crus-galli war. oryzicola was not controlled by the quinclorac residues photodegraded at tile concentrations higher than 30 ppm of the photosensitizer PS-3 in a flooded rice paddy soil. These results indicate that the quinclorac residues in aqueous solution and soil can be degraded efficiently by tile photosensitizers PS-1, PS-3, and PS-6.

Delivery of Ti Plasmid into Nicotiana sanderae Protoplasts via Liposomes (Liposome을 이용한 Ti Plasmid의 꽃담배 원형질체내 도입)

  • Ti plasmid of A. tumefaciens was labeled with $^3H-thymidine$, purified and encapsulated into phosphatidylserine (PS) and PS-cholesterol (Chol; 1 : 1 molar ratio) liposomes by lyophilization-rehydration method. PS was supplemented with 1 mole percent octadecyl rhodamine B for fluorometric measurement of PS. Liposomes entrapping $^3H-Ti plasmid$ were fused with Nicotiana sanderae protoplasts by treating with 5 mM $CaCl_2$ and 10% PEG. The fusion was evidenced by fluorescence microscopic technique. The amounts of Ti plasmid and PS associated with protoplasts were assayed by the radioactivity of $^3H-Ti plasmid$ and by the fluorescence of rhodamine B. About 7.9% of the PS liposome and 7.2% of PS-Chol liposome were fused with protoplasts. During the fusion process, about 30% of the liposomal contents of PS-Chol liposome was leaked, in contrast to about 60% leakage of its contents in PS liposome. Accounting the number of liposomes fused with protoplasts together with the encapsulation efficiency and the leakage of liposomal contents, it was calculated that ca. 1,700 Ti plasmid was transfered into one protoplast by the present method. This result may indicates that the present method transfers enough Ti plasmid into plant protoplast to elicit genetic transformation of plants.

  • To investigate the antibacterial activities of 53 species of mushrooms for Gram-positive bacteria, bioactive substances from mushrooms were extracted with petroleum ether, 80% ethanol, and distilled water. The 14 extracts of 12 species of mushrooms, largely with 80% ethanol, showed antibacterial activities against 6 species of Gram-positive bacteria selectively. Especially, the extracts of Thylopilus PS-5, Pulveroboletus PS-7, and Russula PS-9 showed various antibacterial activities against several bacteria. Minimal inhibitory concentration (MIC) of the 80% ethanol extracts from Pulveroboletus PS-7 and Boletus PS-8 was $300\;{\mu}g/ml$ against Bacillus subtilis, and that of the 80% ethanol extracts from Trichaptium PS-3 was $500\;{\mu}g/ml$ against Micrococcus luteus. MIC of other extracts with antibacterial activities was above $1000\;{\mu}g/ml$.
