Tumor antigen PRAME is a potential therapeutic target of p53 activation in melanoma cells

  • Upregulation of PRAME (preferentially expressed antigen of melanoma) has been implicated in the progression of a variety of cancers, including melanoma. The tumor suppressor p53 is a transcriptional regulator that mediates cell cycle arrest and apoptosis in response to stress signals. Here, we report that PRAME is a novel repressive target of p53. This was supported by analysis of melanoma cell lines carrying wild-type p53 and human melanoma databases. mRNA expression of PRAME was downregulated by p53 overexpression and activation using DNA-damaging agents, but upregulated by p53 depletion. We identified a p53-responsive element (p53RE) in the promoter region of PRAME. Luciferase and ChIP assays showed that p53 represses the transcriptional activity of the PRAME promoter and is recruited to the p53RE together with HDAC1 upon etoposide treatment. The functional significance of p53 activation-mediated PRAME downregulation was demonstrated by measuring colony formation and p27 expression in melanoma cells. These data suggest that p53 activation, which leads to PRAME downregulation, could be a therapeutic strategy in melanoma cells.

  • In this paper, we propose a new PIV methodology for obtaining a velocity field from a sequence of multiple image data based on a least-square principle (also known as MQD; minimum quadratic difference) for the grey level difference between two neighboring frames of image data. We investigated both the accuracy of the result and the time consumption in the computation. It turns out that the proposed method is not only accurate but fast compared with the conventional correlation PIV techniques. Our method is applied to the spin-up flows and the results show that the method can be a good substitution for the conventional algorithms employed in the existing commercial codes.

  • The facilities of electric power plant which were constructed in and after the 1960's are superannuated in these days. We are expanding them applying an incressary to secure a safe plan for them. In this study, for a feasibility of management plan in electric power structure, a RC test prame was manufactured for monitoring and calculation of variation using close range photogrammetry. On the basis of this data, monitoring and calculating of variation in soil wall building site were carried out. Also a survey and analysis of a influence which occured to a structure near by exactitude site. All of these afford data for a management plan in electric power structure.

Expression of MAGE in the Induced Sputum of Lung Cancer Patients (폐암환자의 유도 객담에서 MAGE의 발현)

  • Background : Tumor associated antigens, which are produced specifically by tumor cells, are promising targets for the early diagnosis and immunotherapy. Among the tumor associated antigens, MAGE (a melanoma antigen), BAGE, GAGE, PRAME and NY-ESO were named as cancer/testis specific antigens they are detected exclusively in the testis or cancer cells If MAGE is easily detectable in the sputum, it would become a convenient method for diagnosing lung cancer. This study was undertaken to investigate MAGE expression in the induced sputum obtained from lung cancer patients. Materials and Methods : In 14 control patients and 30 lung cancer patients, the induced sputum was collected after inhaling 3% saline(5 cc) delivered by nebulizer for approximately 5 minutes after a mouth rinse and bronchodilator inhalation. The induced sputum was placed in a conservative-mixed solution (guanidinium isothiocyanate, Triton X-100). The total cellular mRNA was extracted from the cells and RT PCR and nested PCR were run in 30 and 35 cycles respectively, with two different types of primers specially designed to detect six subtypes of MAGE DNA simultaneously. Results : MAGE expression was not detected in the 14 controls, but in the 30 cancer patients, MAGE was found in 24 patients (80%, p=0.001). In the cancer patients, there were no differences in the expression level according to the tissue types (squamous cell cancer 13/17, adenocarcinoma 7/9, and small cell cancer 4/4, p-0.56). Among the 24 MAGE-positive patients, the tumor was not visible on a bronchoscopy in 11 patients (45.8%). Conclusion : A study of MAGE in induced sputum appears to be a useful and complementary method in the diagnosis of lung cancer. A further prospective study with more patients is recommended.
