Production and Prophylactic Efficacy Study of Human Papillomavirus-like Particle Expressing HPV16 L1 Capsid Protein

  • To perform the prophylactic study of a vaccine derived from human papillomavirus (HPV) using Balb/c mice, we produced virus like particles consisting of HPV capsid protein L1 which has been reported to induce significant humoral and cellular immunity using various animal model systems. In order to produce HPV16 VLPs, the cDNA of L1 capsid protein in HPV type 16, obtained by polymerase chain reaction, was inserted into yeast expression vector, YEG$\alpha$-HIR525 under the control of GAL10 promoter. The transformation of YEG$\alpha$-HPV16 L1 was performed into the yeast Saccharomyces cerevisiae Y2805 by the lithium acetate method and the yeast clone expressing the highest level of L1 capsid protein of human papillomavirus type 16 was selected by Western blot analysis using anti-HPV16 L1 antibody. The purification of HPV16 VLP has been performed by the ultracentrifugation and gel-filtration methods. To validate the vaccine efficacy of the purified HPV16 VLPs and investigate the properties of HPV16 VLPs to induce humoral immunity, ELISA assay was performed. A significantly increased production of anti-HPV16 VLP antibodies was observed in sera from immunized mice. The neutralization activity of antibodies in the sera from the vaccinated mice was demonstrated by a rapid and simple assay to detect hemagglutihation inhibition activity.

  • Recent studies have suggested that integrin (CR3) participates in the signal transduction pathways of certain GPI-anchored phagocytic receptors including $Fc{\gamma}RIIIB$. One consequence of this functional linkage is an inducible association between CR3 and cortical microfilaments that is triggered by $Fc{\gamma}RIIIB$ binding to immobilized immune complexes (IC). That this signaling event requires the co-expression of $Fc{\gamma}RIIIB$ with CR3 was documented by the use of NIH 3T3 transfectants expressing both CR3 and $Fc{\gamma}RIIIB$ (clone 3-23), CR3 alone (clone 3-19), and $Fc{\gamma}RIIIB$ alone (clone 3-15). Pretreatment of 3-23 cells with protein kinase inhibitors such as staurosporine and methyl 2,5-dihydroxycinnamate (MDHC) blocked IC-stimulated CR3 microfilament proximity without affecting the extent to which $Fc{\gamma}RIIIB$ constrains the lateral membrane mobility of a subset of CR3 on the cell surface (as measured in fluorescence recovery after photobleaching experiments). These data support that CR3 and $Fc{\gamma}RIIIB$ molecules are physically and functionally associated and that ligation of FcgRIIIB triggers CR3-dependent signal transduction.

Customers my affairs when small guard security companies, especially the sensitive personal information of customers who need to work, the collected information be leaked or the company's trade secrets, are exposed on the outside, it could be a serious threat to a greater problem cause. Small escort guard companies, however, compared with large companies to build its own security system, due to issues such as the extent of funding, staffing shortages, there are many difficulties. Bio-Derived Poly(${\gamma}$-Glutamic Acid) Nanogels as Controlled Anticancer Drug Delivery Carriers

  • We have developed a novel type of polymer nanogel loaded with anticancer drug based on bio-derived poly(${\gamma}$-glutamic acid) (${\gamma}$-PGA). ${\gamma}$-PGA is a highly anionic polymer that is synthesized naturally by microbial species, most prominently in various bacilli, and has been shown to have excellent biocompatibility. Thiolated ${\gamma}$-PGA was synthesized by covalent coupling between the carboxyl groups of ${\gamma}$-PGA and the primary amine group of cysteamine. Doxorubicin (Dox)-loaded ${\gamma}$-PGA nanogels were fabricated using the following steps: (1) an ionic nanocomplex was formed between thiolated ${\gamma}$-PGA as the negative charge component, and Dox as the positive charge component; (2) addition of poly(ethylene glycol) (PEG) induced hydrogen-bond interactions between thiol groups of thiolated ${\gamma}$-PGA and hydroxyl groups of PEG, resulting in the nanocomplex; and (3) disulfide crosslinked ${\gamma}$-PGA nanogels were fabricated by ultrasonication. The average size and surface charge of Dox-loaded disulfide cross-linked ${\gamma}$-PGA nanogels in aqueous solution were $136.3{\pm}37.6$ nm and $-32.5{\pm}5.3$ mV, respectively. The loading amount of Dox was approximately 38.7 ${\mu}g$ per mg of ${\gamma}$-PGA nanogel. The Dox-loaded disulfide cross-linked ${\gamma}$-PGA nanogels showed controlled drug release behavior in the presence of reducing agents, glutathione (GSH) (1-10 mM). Through fluorescence microscopy and FACS, the cellular uptake of ${\gamma}$-PGA nanogels into breast cancer cells (MCF-7) was analyzed. The cytotoxic effect was evaluated using the MTT assay and was determined to be dependent on both the concentration and treatment time of ${\gamma}$-PGA nanogels. The bio-derived ${\gamma}$-PGA nanogels are expected to be a well-designed delivery carrier for controlled drug delivery applications.

A Study on the Characteristics of Design and Acculturation of Planting of 'Guǐ(槐)' in Chirinjeong Wonlim of Pohang (포항 칠인정원림(七印亭園林)의 조영특성과 '괴목(槐木)' 식재(植栽)의 문화변용(文化變容))

  • The purpose of this study is to illuminate historical sensitivity and design in the days of the Joseon era and examines the phenomenon of acculturation as revealed in the planting of Sophora japonica and Zelkova serrata in the garden grove by looking into the purpose underlying the construction, formative intention and the process of change in Pohang's Chirinjeong wonlim. Chirinjeong, also called Sanggaejeong(雙槐亭) after the fact that two Zelkova serrata trees were planted there, is a hermit pavilion in Sa-il village, Chogok-ri, built in the 9th year of King Taejong(1409) by Jang, Pyo(張彪, 1349~?) of an officer of bureaucratic origin toward the end of the Goryeo Dynasty, the Chirinjeong garden grove is an element essential to Chogok, the 3rd Gok of Sanggaegoogok(雙溪九曲). The wonlim of Chirinjeong is divided into the pavilion section composed of the entrance area, Chirinjeong, composed of Zelkova serrata and Sophora japonica, and the pond section composed of a quadrangular pond and island(方池方島), and Lagerstroemia indica. In view of the intent of the name Chirinjeong or of the fact that the pavilion was named based on that the royal seal string was hung on the Sanggwaesoo(雙槐樹) or two Zelkova serrata trees, it is reasonable to view it as Zelkova serrata trees having been planted early in the days of the Joseon Dynasty and those additionally planted later on that symbolizes the position of the three nobles and the Sophora japonica. In addition, in spite of the fact that the type of trees supplementarily planted in the 21st year of King Yeongjo(1745) is known as Sophora japonica, it is impossible to rule out the possibility of the tree planted outside of Chirinjeong being Zelkova serrata. In short, the three Zelkova serrata trees planted along with Sophora japonica must certainly be evidence that Zelkova serrata planted along with Sophora japonica in Chirinjeong wonlim indicates that the off spring of the Indong Jang family could tell the difference between Sophora japonica and Zelkova serrata. In the recognition process of Zelkova serrata trees finding their way into Korea, it was known as Zelkova serrata on the one hand and as Sophora japonica on the other, and the former, which enjoys a comparative advantage over in terms of the easiness with which to purchase, growth speed and possibility of growing into a long-lived tree was interchanged with Zelkova serrata, a case of acculturation as manifested in the planting of Sophora japonica in the Chirinjeong garden grove.

Study on the Present Condition and Type Analysis of Rock Korean Chessboards in Korean Landscape (암각장기판의 형상 및 입지 특성에 관한 연구)

  • The objective of this study is to investigate the value of rock Korean chessboards as a corroborative evidence that go, the play culture naturalized from the past, was also performed under the pavilion of the mountain stream, and the old tree. This study executes analysis on the location and form of rock Korean chessboards, physical environment such as the position of surrounding space, the origin of the nomination, surrounding landscape, and historical significance through the research analysis of comprehensive present conditions, and highlights the position types, landscape significance, and preservation value of the rock Korean chessboards of Korea from diverse angles. The result is as follows. 1. The styles of Korean chess (將棋), which identically propagated in the representative countries of Eastern culture, Korea, China, and Japan, and modified in accordance with each country's idea and native tendency, were organized, and confirmed the Korean Go as a naturalized native culture. 2. Out of 15 rock Korean chessboards confirmed through this study, 9 (60%) were categorized as rock Korean chessboards, and 6 (40%) were categorized as stone Korean chessboards. Also, the average size of the go boards were $51cm{\times}46.6cm$, which demonstrated not much difference with present day universal go boards. The Pearson correlation coefficient between bed rock and go boards were 0.647, demonstrating a relatively high correlation; the research subject of rock go boards have been constructed in consideration of rationality, convenience, and the value of promotion of public welfare. 3. The results of analysis of location patterns of rock Korean chessboard showed that villages (87%), mooring (73%), plains (60%) and mountains (47%). The most frequent location pattern is mooring and village, and these two factors played the most important role in determining the location of the rock Korean chessboard.
