Cyclic Loading Test for Beam-to-Column Connections of Concrete Encased CFT Column (콘크리트피복충전 각형강관 기둥-보 접합부의 주기하중 실험)

  • In this study, the beam-to column connections of concrete-encased-and-filled steel tube columns were tested under cyclic loading. Two specimens using steel beams and two specimens using precast concrete beams were tested. The dimension of the column cross section was $670mm{\pm}670mm$. The beam depths were 488mm and 588mm for the steel beams and 700mm for the precast concrete beams. The longitudinal bar ratios of the precast concrete beams were 1.1% and 1.5%. For the connections to the steel beams, continuity plates were used in the tube columns. For the connections to the PC beams, couplers were used for beam re-bar connections. The test results showed that except for a specimen, deformation capacities of the specimens were greater than 4% rotation angle, which is the requirement for the Special Moment Frame. Particularly, specimens using precast concrete beam showed excellent performances in the strength, deformation, and energy dissipation.

High frequency direct plant regeneration from leaf, internode, and root segments of Eastern Cottonwood (Populus deltoides)

  • Simple, reproducible, high frequency, improved plant regeneration protocol in Eastern Cottonwood (Populus deltoides) clones, WIMCO199 and L34, has been reported. Initially, aseptic cultures established from axillary buds of nodal segments from mature plus trees on MS liquid medium supplemented with $0.25mg\;1^{-1}$ KIN and $0.25mg\;1^{-1}$ IAA. Nodal and internodal segments were found to be extra-prolific over shoot apices during course of aseptic culture establishment, while $0.25mg\;1^{-1}$ KIN concentration played a stimulatory role in high frequency plant regeneration. Diverse explants, such as various leaf segments, internodes, and roots from in vitro raised cultures, were employed. Direct plant regeneration was at high frequency of 92% in internodes, 88% in leaf segments, and 43% in root segments. This led to the formation of multiple shoot clusters on established culture media with rapid proliferation rates. Many-fold enhanced shoot elongation and growth of the clusters could be achieved on liquid MS medium supplemented with borosilicate glass beads, which offer physical support for proliferating shoots leading to faster growth in comparison to semi-solid agar or direct liquid medium. SEM examination of initial cultures confirmed direct plant regeneration events without intervening calli. In vitro regenerated plants induced roots on half-strength MS medium with $0.15mg\;1^{-1}$ IAA. Rooted 5- to 6-week-old in vitro regenerated plants were transferred into a transgenic greenhouse in pots containing 1:1 mixture of vermicompost and soil at $27{\pm}2^{\circ}C$ for hardening and acclimatization. 14- to 15-week-old well-established hardened plants were transplanted to the field and grown to maturity. The mature in vitro raised poplar trees exhibited a high survival rate of 85%; 4-year-old healthy trees attained an average height of 8 m and an average trunk diameter of 25 cm and have performed well under field conditions. The regeneration protocol presented here will be very useful for undertaking genetic manipulation, providing a value addition to Eastern Cottonwood propagation in future.

Effects of the purified extracts from Lycii Cortex Radicis and ginger on lipid statusand serum cytokine levels in rats fed high fat diet (지골피와 생강 정제 추출물이 고지방식이 흰쥐의 체내 지질과 혈청 Cytokine 수준에 미치는 영향)

  • The present study was to investigate the effects of Lycii Cortex Radicis (LCR), the root bark of lycium (Lycium chenese Miller) and ginger (Gin) on body lipid status and serum levels of cytokines. Sprague-Dawley (SD) male rats weighing $193.6{\pm}16.8g$ were divided into five groups, including one low fat (LF) and four high fat groups, i.e. HF-Control, HF-LCR, HF-Gin and HF-LCR + Gin groups. Diets for HF-LCR, HF-Gin and HF-LCR + Gin groups contained purified extracts having 0.2 g LCR tyramine, ginerol and 0.1 g tyramine plus 0.02 g gingerol per kg, respectively. Compared with those of the HF-Control total serum cholesterol level decreased, and HDL-cholesterol level increased in the HF-LCR group and serum triglyceride levels decreased in the three experimental groups fed the purified extracts. Liver cholesterol level was lower in the HF-LCR group than the HF-Control group, but triglyceride levels, which were increased by high fat diets were not changed by significantly by LCR or ginger extracts. Fecal lipid excretion was higher in the HF-LCR and HF-Gin groups, but cholesterol excretion was lower in the HF-Gin group than in the HF-Control group. The activities of liver cytosolic glucose-6-phosphate dehydrogenase and malic enzyme were lower in the HF-LCR + Gin group than in the HF-Control group. Serum adiponectin levels did not differ among the five groups, while leptin level was lower in the HF-Gin group and C-reactive protein levels were lower in the HF-Gin and the HF-LCR + Gin groups than in the HF-Control group. It is concluded that LCR can be utilized as an ingredient for lipid-lowering functional foods in the form of purified extract and addition of small amount of ginger extract would be useful for reducing one of the inflammatory cytokines to help prevent atherosclerosis.

The effect of adding ethanolic saffron petal extract and vitamin E on growth performance, blood metabolites and antioxidant status in Baluchi male lambs

  • Objective: This study investigated the effects of the administration of ethanolic saffron petal extract (SPE) and vitamin E (Vit E) on growth performance, blood metabolites and anti-oxidant status in Baluchi lambs. Methods: Thirty-two Baluchi male lambs ($35.22{\pm}5.75kg$) were randomly divided into 4 groups. The 1st (control), 2nd (injectable saffron petal extract [ISPE]), and 3rd (Vit E) groups were respectively injected subcutaneously with either physiological saline (5 mL), SPE (25 mg/kg body weight [BW]) or DL-${\alpha}$-tocopheryl acetate (225 IU) once a week. An oral dose of SPE (500 mg/kg BW) was also administered to the 4th group (oral saffron petal extract [OSPE]). Feed intake and BW were measured for 42 days and blood samples were taken on days 1, 14, 28, and 42. The lambs were slaughtered, and tissue samples were taken. Results: Growth performance and many blood metabolites were not affected (p>0.05) by the treatments. Cholesterol of plasma in the ISPE and Vit E groups was similar and less (p<0.01) than both the OSPE and control groups. Although there was no significant difference between the control and other groups for plasma triglyceride, the ISPE group showed lower (p<0.05) triglyceride than the OSPE and Vit E groups. The highest (p<0.01) plasma glutathione peroxidase (GPx) was detected in the OSPE group, while the ISPE and Vit E groups showed higher (p<0.01) superoxide dismutase (SOD) of plasma than the control. Malondialdehyde of plasma in the ISPE group was lower (p<0.05) than the OSPE. No differences (p>0.05) were observed among the groups for antioxidant status of both longissimus dorsi muscle and liver. However, the activity of GPx in the kidney and heart, as well as SOD activity in the kidney, were influenced ($p{\leq}0.01$) by the treatments. Conclusion: Adding ethanolic SPE improved antioxidant status and lowered lipids oxidation in lambs. The SPE and Vit E demonstrated similar effects on antioxidant status in lambs.

Electrical resistivity characteristics for cement specimens with TiO2 according to activated carbon content (활성탄 함유량에 따른 광촉매(TiO2) 시멘트 시편의 전기비저항 특성)

  • Concrete with activated carbon and titanium dioxide (TiO2) has been used to reduce the particulate matter (PM) in underground structures (e.g., tunnels) due to the high performance of nitrogen oxides (NOx) abatement. Damage (e.g. crack, spalling, or detachment) can be caused by the environmental and ageing effects on the surface of the particulate matter reduction concrete, installed on the tunnel lining. Therefore, it is important to evaluate the existence of spalling on the concrete surface for maintaining performance of NOx reduction. In this study, a basic research was performed for feasibility of spalling evaluation using electrical resistivity characteristics. Given the test results, the electrical resistivity was decreased as the ratios of activated carbon (0~15%) and TiO2 (0~25%) were increased for specimens. Under a dry condition, electrical resistivity of cement specimens, mixed with activated carbon and TiO2, was decreased up to 2.3 times, compared with the normal cement specimen. In addition, under saturation conditions (degree of saturation: 85~98%), electrical resistivity of cement specimens with activated carbon, was decreased up to 3.5 times, compared with the normal cement specimen. Regardless of the condition (dry or saturated), the difference of electrical resistivity values shows the range of 2.3~2.8 times between the mixing specimen (with activated carbon (15%) and TiO2 (25%)) and the normal cement specimen. This study can help to provide basic knowledge for spalling evaluation using the electrical resistivity on the surface of the particulate matter reduction concrete in tunnels.

Effect of Dietary Cracked Whole Barley on the Meat Compositional Properties of Hanwoo Steer Loin Beef (분쇄 통보리 급여 수준이 거세한우의 등심부위의 성분 조성에 미치는 영향)

  • This study was carried out to investigate the effects of the feeding level of cracked whole barley (CWB) on fatty acid composition, free amino acid content, organic acid content, pH, HDL cholesterol, total phenol and DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity of finishing Hanwoo steers. The dietary treatments were consisted of five types (C: base feed + 0% CWB, T1: base feed + 10% CWB, T2: base feed + 20% CWB, T3: base feed + 30% CWB, T4: base feed + 40% CWB). A total 30 Hanwoo steers(588.6${\pm}$11.8kg) were allocated into 5 feeding groups, and a total of thirty Hanwoo steers raised from 24 to 30 months. Linoleic acid of fatty acid composition was significantly higher for cracked CWB treatments (T1, T2, T3, and T4) than C, and T3 of CWB treatments was the highest than the other treatments (P<0.01). SFA, USFA, MUFA and USFA/SFA did not differ among the treatments. On the contrary, PUFA was significantly higher for CWB treatments(T1, T2, T3, and T4) than C, and T3 was the highest (P<0.05). EAA was higher in the order of T1 > T2 > C > T3 > T4 (P<0.05). NEAA was higher in the order of T2 >T3 >T1 >C >T4, although a statistical significance was not detected. The total organic acid content was the highest for T3 (20.15 mg/100g) and the lowest for T2 (13.19mg/100g). pH and total phenol of all treatments were did not differ. HDL cholesterol was higher in the order of T1 > C > T4 > T2 > T3 (P<0.01). DPPH radical scavenging activity was in order of T1 > T2 >T4 > C > T3 (P<0.01). Based on the above results, T1 treatment compared to other treatments have been shown to improve EAA, DHL, and DPPH radical scavenging activity.

Effects of Ginseng By-Products Supplementation on Performance, Blood Biochemical Profiles, Organ Development, and Stress Parameter in Broiler under Heat Stress Condition (인삼 부산물의 첨가 급여가 고온 스트레스 하 육계의 생산성, 혈액조성, 장기발달 및 스트레스 지표에 미치는 영향)

  • This study was performed to investigate the effects of dietary supplementation with ginseng by-products on growth, organ development, blood biochemical profiles, immune response, and stress parameter of broilers reared in high ambient temperatures. One hundred one-day-old male chicks (Ross 308) were used. At week two, the birds were randomly allocated into five dietary groups; control (CON), 0.5% ginseng berry (GB1), 1.0% ginseng berry (GB2), 0.5% ginseng leaves and stems (GLS1), and 1.0% ginseng leaves and stems (GLS2). The temperature was maintained at 32±1℃from 9 AM to 5 PM. Growth, serum immunoglobulins and corticosterone levels were monitored and analyzed. No significant differences among groups were observed in growth. However, during the finisher period (21~35d) and overall period (7~35 d), body weight gain in all supplemented groups tended higher than CON group. Blood biochemical profiles did not significantly differ among treatment groups except in bilirubin level. Serum immunoglobulins and corticosterone level showed no significant differences among groups. IgM and IgG levels were numerically higher in GLS1 than in other groups, but the difference was not significant. Corticosterone level also tended lower in all supplemented groups than in CON group, and larger decreases were observed in groups with higher ginseng by-product concentration. In conclusion, dietary supplementation of ginseng by-products shows potential to reduce heat stress in growing broilers with no negative effect on productivity.

Preparation of fermentation broth of Sparassis latifolia containing soluble β-glucan using four Lactobacillus species (수용성 β-glucan을 함유한 꽃송이버섯 발효액의 제조)

  • Glucan has been shown to have a significant role in the activation of the immune system, including increased activity of macrophages and so on. Sparassis latifolia (formerly S. crispa) is an edible mushroom abundant in dietary fiber and widely known to contain high levels of ${\beta}$-glucan. In the present study, fermentation broths containing soluble ${\beta}$-glucan were prepared by fermentation with mushrooms with four Lactobacillus species (L. plantarum subsp. Plantarum, L. acidophilus, L. helveticus, and L. delbrueckii subsp. Bulgaricus). After culturing four Lactobacillus spp. in MRS broth, each Lactobacillus was inoculated into MRS broth containing S. latifolia powder 5% (w/v) at $37^{\circ}C$ in an anaerobic incubator for five days. It showed that the ${\beta}$-glucan contents were different in each fermentation sample. The suitable conditions for the preparation of mushroom fermentation broths were investigated and discussed.

Effects of Shiryung-tang Extract on the Liver Injury induced by Ethanol in Rats (시령탕(柴苓湯)이 에탄올 투여로 유발된 흰쥐의 간손상에 미치는 방어효과)

  • Alcoholic liver disease (ALD) is a major cause of morbidity and mortality around the world. Although much progress has been made in understanding the pathogenesis of ALD, there remains no effective therapy for it. Accumulated evidence indicates that oxidative stress is the main pathological factors in the development of ALD. Ethanol administration causes accumulation of reactive oxygen species (ROS), including superoxide, hydroxyl radical, and hydrogen peroxide. ROS, in turn, cause lipid peroxidation of cellular membranes, and protein and DNA oxidation, which results in hepatocyte injury. In addition to pro-oxidants formation, antioxidants depletion caused by ethanol administration also results in oxidative stress. The objective of this study is to investigate the effects of Shiryung-tang extract on the chronic alcoholic liver injury induced by EtOH. Male Sprague Dawley rats were used in this study. All rats were maintained under standard laboratory conditions ($23{\pm}1^{\circ}C$, 12h light/12h dark cycles). All animals (n=30) were randomly divided into following groups: (1) Normal group, treated with distilled water (n=10); (2) Control group, treated with ethanol (n=10); (3) Sample group, treated with ethanol + pharmacopuncture (n=10). For oral administration of ethanol in Control and Sample group, the ethanol was dissolved in distilled water in concentrations of 25%(v/v). Throughout the experiment of 8 week, the rats were allowed free access to water and standard chow. Sample group were administrated by Shiryung-tang extract daily for 8 weeks. Control group were given normal saline for same weeks. As a results, the oral administration of ethanol for 8 weeks leads to hepatotoxicity. The levels of hepatic marker such as HDL-cholesterol, triglyceride, aspartate aminotransferase and alanine aminotransferase were altered. The ethanol also increased lipid peroxidation and depletion of antioxidant enzyme activities as well as hepatic tissue injury. However, the treatment of Shiryung-tang extract prevented all the alterations induced by ethanol and returned their levels to near normal. These data suggest that Shiryung-tang extract could have a beneficial effect in inhibiting the oxidative damage induced by chronic ethanol administration. Therefore, Shiryung-tang extract can be a candidate to protect against EtOH-induced liver injury.

Detection of Expressed IL-32 in Human Stomach Cancer Using ELISA and Immunostaining

  • Interleukin (IL)-32 is a recently identified proinflammatory cytokine that is one of the IL-18 inducible genes, and plays an important role in autoimmune and inflammatory diseases. We produced antibodies against IL-32 and studied the expression of IL-32 in human stomach cancer. We detected IL-32 secreted from K-562 cells which were stably transfected with IL-32 and in the sera of stomach cancer patients by a sandwich ELISA using a monoclonal antibody KU32-52 and a polyclonal antibody. In order to optimize a sandwich immunoassay, recombinant IL-32a was added, followed by the addition of a biotinylated KU32-52 into microtiter plate wells precoated with a goat anti-IL-32 antibody. The bound biotinylated KU32-52 was probed with a streptavidin conjugated to HRP. This sandwich ELISA was highly specific and had a minimal detection limit of 80 pg/ml (mean${\pm}$SD of zero calibrator) and measuring up to 3,000 pg/ml. This ELISA showed no cross-reaction with other cytokines such as hIL-1$\alpha$, hIL-1$\beta$, hIL-2, hIL-6, hIL-8, hIL-10, hIL-18, and hTNF-$\alpha$. Intra-assay coefficients of variation were 18.5% to 4.6% (n=10), and inter-assay coefficients were 23% to 9% (n=10). The average IL-32 level in the sera of 16 stomach cancer patients (189 pg/ml) was higher than that of 12 healthy control men (109 pg/ml). Our results indicate that serum IL-32 level can be detected by using an established ELISA, and that this immunoassay and mAb KU32-09 specific for immunohistochemistry can be used in the detection of expressed and secreted IL-32 in stomach cancer patients.
