Bisphenol A의 투여가 생쥐의 번식효율에 미치는 영향

  • 내분비계 장애물질 중 관찰물질로 지정된 bisphenol A(BPA)의 투여가 생쥐의 번식 효율에 미치는 영향을 규명하기 위하여 웅성 ICR 생쥐에 무처리구, corn oil, 0.05, 0.5 및 5.0mg BPA/kg b.w의 BPA을 3일 간격으로 5회 투여했으며, 자성 ICR 생쥐도 웅성생쥐와 동일하게 3일 간격으로 5회 투여한 후, 웅성은 투여후 2일, 자성은 7일후에 모든검사를 실시하였다. BPA의 투여가 웅성 생쥐의 번식기관무게에 미치는 영향을 조사한 결과는 체중, 정소상체, 정낭선 및 응고선의 무게는 투여구간에 커다란 차이가 없었으며(P>0.05), 정소의 무게는 5.0mg 투여구가 여타구보다 다소 낮은 무게를 나타냈다. BPA의 투여가 웅성 생쥐의 정액성상에 미치는 영향을 조사한 결과, 생존율과 유효 정자수는 각 투여구간에 커다란 차이가 없었으나 총정자수는 각각 35.50, 35.73, 36.33, 33.13 및 29.28$\times$$10^{7}$ 정자/$m\ell$로서 5.0mg 투여구가 여타구보다 통계적으로 유의하게 낮은 결과을 나타냈다(P<0.05). 기형율은 5.0mg 투여구가 23.15%로서 무처리구(18.13%), 대조구(18.58%) 및 0.05mg 투여구(19.68%)보다 통계적으로 유의하게 높은 기형율을 나타냈다(P<0.05). 웅성 생쥐에 BPA의 투여가 혈액의 일반성분에 미치는 영향을 조사한 결과, RBC, Hb, HT, MCV, MCH 및 MCHC는 각 투여구간에 차이가 없었으며, WBC는 BPA 투여구가 무처리구 및 대조구보다 다소 낮은 수치를 나타냈으며, PLT는 BPA 투여구가 무처리구 및 대조구보다 높은 수치를 나타냈다. BPA의 투여가 자성 생쥐의 번식기관무게에 미치는 영향을 조사한 결과, 체중은 투여구간에 커다란 차이가 없었으며 자궁의 무게는 BPA 투여구가 대조구에 비해 다소 논은 무게를 나타냈다. 오른쪽 난소의 무게는 0.5mg 투여구와 5.0mg 투여구가 각각 0.0059 및 0.0061g으로 대조구(0.0081g)와 0.05mg 투여구(0.0071g)보다 낮은 무게를 나타냈으며, 왼쪽 난소의 무게는 각각 0.0076, 0.0076, 0.0058 및 0.0061g으로 0.5mg 투여구가 대조구와 0.05mg 투여구보다 낮은 무게를 나타냈다(P<0.05). BPA의 투여가 자성 생쥐의 혈구화학치에 미치는 영향을 조사한 결과, RBC, Hb, HT, MCV, MCH 및 MCHC는 각 투여구간에 차이가 없었으며, WBC는 BPA 투여구가 대조구보다 다소 높은 수치를 나타냈다. PLT는 0.5mg 투여구와 5.0mg 투여구가 대조구보다 다소 높은 수치를 나타냈다. 정소, 난소 및 자궁의 조직, 병리학적인 검사에서는 모든 투여구간에 커다란 차이가 없었다.

Analysis of Image for Liver Disease using Blood Test in the Ultrasound Fibroscan (Fibroscan에서의 혈액검사를 이용한 간질환의 영상분석)

  • The liver fibrosis is a disease we often see in clinical medicine, and the persistence and repeatition of inflammation and necrosis of liver cells continue for several years, and it is proceeded to cirrhosis. So decrease of death rate and prevalence rate by complications of cirrhosis and hepatocellular carcinoma is main task of clinical medicine by protection of chronic liver ailment patients from proceeding to cirrhosis and hepatocellular carcinoma. So this study tried to represent the ultrasonic image, blood test, the relationship with liver stiffness of diffuse liver ailment patients as numbers. This study was performed with patients from whom the image was taken by ultrasonic and 141 people who were treated by fibroscan, the basic data for blood test was obtained from the test results at the time when ultrasonic image and liver fibroscan was performed. The statistical analysis was performed by One-way analysis of variance(ANOVA) to verify difference between groups. The value of liver stiffness was increased in the order of normal, chronic liver disease and cirrhosis. As a results, ALT and Albumin have no statistical difference between object groups, and there are statistical differences in the results of ultrasonic decoding at age, AST, ALP, Bilirubin, PLT, PT, and kPa, and they are statistically meaningful(p<0.005). And the value of liver stiffness of chronic liver ailment was presented only as over 12.5kPa in other study, but it was represented as numbers for quantitative diagnosis by presenting average kPa threshold value according to disease in this study. And by presenting relationship of diagnosed results, it is considered that it could be used as first tool to diagnose chronic liver ailment patients according to their disease.

A Study on Various Soil Stiffness Evaluation Methods with Field Test (현장시험을 통한 다양한 지반강성 평가방법에 대한 연구)

  • The plate loading test(PLT) and the field density test are mainly used on the construction of embankments to control the compaction of a limited layer thickness. These two test methods are very time consuming and inefficient, but they are still commonly used as the methods of quality control for soil compaction. In the last 3 decades, many devices such as geogauge, light falling weight deflectometer(LFWD) and dynamic cone penetrometer(DCP) etc., have been introduced into the engineering market with the objective of acquiring in situ stiffness properties of the compacted soil layers. Recently, a new type of sensor, called compactometer, which in mounted on the drum of a roller and measures impact forces continuously with GPS, called as Continuous Compaction Control(CCC), has come into use in many countries such as America, Germany, Japan and so on. The main objective of this paper is to assess the potential use of these new devices as quality control and assurance devices for compacted soil layers. Based on this study, compactometer and the LFWD results werestrongly correlated with the result obtained from the PLT and the field density test.

Effects of Massive Transfusion Protocol Implementation in Trauma Patients at a Level I Trauma Center

  • Purpose: This study was conducted to investigate whether rapid and efficient administration of blood products was achieved and whether clinical outcomes were improved by applying a massive transfusion protocol (MTP). Methods: From January 2016 to September 2019, the medical records of trauma patients who received at least 10 units of packed red blood cells (PRBC) at Pusan National University Hospital (level I trauma center) were retrospectively reviewed. The patients treated from January 2016 to January 2018 were designated as the non-MTP group, and those treated from February 2018 to September 2019 were designated as the MTP group. Results: During the study period, 370 patients received massive transfusions. The non-MTP and MTP groups comprised 84 and 55 patients, respectively. No significant between-group differences were found in the units of PRBC (23.2 vs. 25.3, respectively; p=0.46), fresh frozen plasma (FFP) (21.1 vs. 24.4, respectively; p=0.40), and platelets (PLT) (15.4 vs. 17.0, respectively; p=0.54) administered in the first 24 hours. No statistically significant differences between the non-MTP and MTP groups were found in the FFP-to-PRBC ratio (0.9 vs. 0.94, respectively; p=0.44) and or the PLT-to-PRBC ratio (0.72 vs. 0.72, respectively; p=0.21). However, the total number of cryoprecipitate units was significantly higher in the MTP group than in the non-MTP group (7.4 vs. 15.3 units, respectively; p=0.003) and the ratio of cryoprecipitate to PRBC in the MTP group was significantly higher than in the non-MTP group (0.31 vs. 0.62, respectively; p=0.021). The time to transfusion was significantly reduced after MTP implementation (41.0 vs. 14.9 minutes, respectively; p=0.003). Conclusions: Although no significant differences were found in the clinical outcomes of patients who had undergone severe trauma, rapid and balanced transfusion was achieved after implementing the MTP.

Poor Prognosis Significance of Pretreatment Thrombocytosis in Patients with Colorectal Cancer: a Meta-Analysis

  • Background: Recently, several studies have reported that elevated platelet counts may be associated with the poor prognosis of colorectal cancer. However, conclusions remain controversial. This meta-analysis was therefore designed to analyze and evaluate the prognostic role of preoperative or pretreatment thrombocytosis in patients with colorectal cancer. Materials and Methods: We searched PubMed, EMBASE, the Cochrane Library and Web of Science to March 29th, 2015. The citation lists of included studies were also hand-searched to identify further relevant trials. To investigate the association between thrombocytosis and prognosis of colorectal cancer, the 1-year, 3-year and 5-year survival of each studies were obtained. The odds ratio (OR) with its 95% confidence interval (CI) was used to evaluate the relation of overall survival (OS) between thrombocytosis and normal platelet counts (PLT). Likewise, disease free survival (DFS) was obtained and evaluated. The analysis was performed and assessed using Review Manager 5.2. Results: A total of 14 studies (N=5,566 participants, 11 including 4,468 for OS, 6 including 1,533 for DFS) were included in this meta-analysis, of which seven (N=3810) defined thrombocytosis as a platelet count ${\geq}400{\times}10^9/L$, and 375 (9.8%) patients exhibited pretreatment thrombocytosis. Thrombocytosis have a close relationship with the poor OS of colorectal cancer compared with normal PLT, with the pooled ORs of 1-year, 3-year and 5-year survival being 0.41 [95% CI 0.34-0.51; P<0.001], 0.28 [95% CI 0.21-0.38; P<0.001] and 0.26 [95% CI 0.20-0.34; P<0.001], respectively. For DFS, the same results were showed as the pooled ORs of 1-year, 3-year and 5-year survival respectively being 0.34 [95% CI 0.24-0.50; P<0.001], 0.31 [95% CI 0.23-0.43; P<0.001] and 0.25 [95% CI 0.18-0.34; P<0.001]. Conclusions: This meta-analysis indicated that thrombocytosis may predict poor prognosis for patients with colorectal cancer, and platelet counts may be a cost-effective and noninvasive marker.

Studies on Effects of Consecutive Lead-Administration on Feed, Water Intake, Weight Gain, Blood Pictures and Mineral Level of Organ in Rats (연속적인 Pb투여가 Rat의 사료와 물섭취량, 증체량, 혈액치 및 장기내 무기물 수준에 미치는 영향)

  • This study was performed to elucidate the patho-physiology of lead acetate-poisoned rats after consecutive oral administrations of the lead. The changes in feed and water intake, gains of body weight, blood pictures and mineral compositions of several organs were observed to measure the effects of lead acetate-poisoning. 1. Compared with control group, every experimental group of which 1,000, 2,000, or 4,000 ppm/kg of lead acetate was administered displayed a gradual decrease in feed intake in dose-dependent manners. 2. After 1,000, 2,000 or 4,000 ppm/kg of lead acetate administration, water intake seemed to decrease in every experimental group in dose-dependent manners. 3. After 1,000, 2,000 or 4,000 ppm/kg of lead acetate administration, body weight decreased in every experimental group in dose-dependent manners. 4. After 1000, 2,000 or 4,000 ppm/kg of lead acetate administration, the PLT values and numbers of RBC and WBC significantly increased after lead acetate administration, but the values of Hb and PCV were lower than those of control group's. 5. After 2,000 or 4,000 ppm/kg of lead acetate administration, the levels of Pb, Mn, Fe, Pb and Zn in the livers were lowered, but the levels of Pb, Mn, Fe, Pb and Zn in the kidneys.

Red Ginseng Ameliorates Place Learning Deficits in Aged Rats Young Rats with Selective Hippocampal Lesions

  • Ameliorating mechanisms of red ginseng on learning deficits were investigated in the following 3 experiments; its effects on 1) place learning deficits in aged rats and in young rats with selective hippocampal lesions (behavioral study), 2) long-term potentiation in the hippocampal formation (neuro- physiological study), and 3) ChAT (choline acetyl transferase) activity in various brain regions of aged rats (pharmacological study). In the behavioral study, first, performance in the place learning tasks were compared among 3 groups of young and aged rats; control young intact rats (10-12 week old) treated with water, aged rats (28-32 month old) treated with water, and aged rats (28-32 month old) treated with red ginseng (100 mghglday) suspended in water. Second, performance in the place learning tasks was compared among 3 groups of young rats; control intact rats treated with water, rats with bilateral hippocampal lesions treated with water, and rats with bilateral hippocampal lesions treated with red ginseng (100 mg/kg/day). Each rat in these 2 behavioral experiments was tested with the 3 types of the place learning tasks in a circular open field using intracranial self-stimulation (ICSS) as reward. The ICSS reward was delivered if the rat (1) moved distance of 100-160 cm (DMT): (2) entered an experiment-determined reward place within the open field, and this place was randomly varied in sequential trials (RRPST); or (3) entered 2 specific places, and did a shuttle behavior between the 2 places (PLT). Performance of the aged rats in the ginseng group was not significantly different from that of control young rats in ICSS (current intensity, bar press rates), DMT and RRPST. However, treatment with red ginseng significantly ameliorated place-navigation learning deficits in aged rats in the PLT. Similarly, red ginseng ameliorated learning and memory deficits in young rats with hippocampal lesions in the same tasks. In the neurophysiological study using young rats, perfusion of hippocampal slices with non-sapon in fraction of red ginseng significantly enhanced magnitudes of the long-term potentiation (LfP) in the CA3 subfield. In the pharmacological study, treatment with red ginseng did not affect ChAT activity in aged rat brain including the hippocampal formation. These results strongly suggest that red ginseng ameliorates learning and memory deficits in aged rats through actions on the CA3 subfield of the hippocampal formation, which were independent of the presynaptic components of the cholinergic system

The Preventive Effect of Bacillus polyfermenticus KJS-2 and Bacillus mojavensis KJS-3 on Triton WR-1339-induced Hyperlipidemia (Triton WR-1339로 유도된 고지혈증에 대한 Bacillus polyfermenticus KJS-2와 Bacillus mojavensis KJS-3의 예방효과)

  • The purpose of this study was to evaluate the possibility that administration of Bacillus polyfermenticus KJS-2 (BP2), Bacillus mojavensis KJS-3 (Moja3), and their mixtures could control serum lipid levels. We observed changes in the blood cell level, metabolic function evaluation, and blood lipid levels after two weeks of oral administration of these microbial strains to hyperlipidemia-induced rats. Measurements of major cell changes in the white blood cells (WBC) indicated no significant effects due to the administration of the microbial strains. Platelet (PLT) levels decreased by 18.4% in the Triton WR-1339-treated group (NCON) and recovered to the control (CON) group levels in the positive control (PCON) group and the microbial strain-administered groups (p<0.05). No functional changes were observed in red blood cells (RBC) by Triton WR-1339-induced hyperlipidemia. The blood AST, ALT, BUN, and creatinine levels did not indicated effects on liver and kidney function, and all rats administered the microbial mixture recovered. The blood lipid levels in the microbe-treated groups indicated reduced levels of triglyceride (TG) and total cholesterol (TC), and increased levels of serum HDL cholesterol. The HMG-CoA inhibition rate of 7-O-succinyl macrolactin A (SMA) produced by BP2 showed similar activity at a concentration of 1,000 times lower than that achieved with atorvastatin. The administration of the microbial strains to the Triton WR-1339-induced rat model of hyperlipidemia resulted in reduced weight gain without affecting the food and water intake. Thus, blood circulation can be improved by controlling serum lipid levels by the combined administration of the BP2 and Moja3 microbial strains.

Evaluation of Compaction Impact According to Compaction Roller Operating Conditions through CMV Analysis (CMV 분석을 통한 다짐롤러 운용 조건에 따른 다짐 영향 평가)

  • The compaction process using vibrating rollers in road construction is essential to increase soil stiffness in earthworks. Currently, there is no clear standard for the operation method of the compaction roller during compaction. Although simple quality inspection techniques have been developed, plate load test (PLT) and field density test (FDT) are the most frequently used test methods to evaluate the degree of compaction during road construction as the most frequently used quality inspection methods. However, both inspection methods are inefficient because they cannot perform quality inspection in all sections due to time and cost reasons. In this study, we analyzed how the operating conditions of vibrating rollers affect the compaction quality. An intelligent quality management system, which is a currently developed and commercialized technology, was used to obtain quality inspection results in all sections. As a result of the test, it was analyzed that the speed and vibration direction of the compaction roller had an effect on the compaction degree, and it was found that the compaction direction had no effect on the compaction degree.

Load-Settlement Behavior of Rock-socketed Drilled Shafts by Bi-directional Pile Load Test (양방향 말뚝선단재하시험에 의한 암반근입 현장타설말뚝의 하중-침하거동 분석)

  • Load settlement behaviors and load transfer characteristics of rock-socketed pile subjected bi-directional load at pile tip were investigated using bi-directional pile load tests (BD PLT) performed on ten large-diameter drilled shafts at four sites. Based on test results, additional pile-toe displacement ($w_{bs}$) by coupled soil resistance was analyzed, and thus equivalent top loaded load-settlement curve of pile subjected bi-directional load was proposed by taking into account the coupled soil resistance. Through comparisons with field case studies, it is found that for test piles there exists effect of coupled soil resistance, which is represented by wbs, and thus an equivalent curve obtained by existing uncoupled methods can overestimate bearing capacity of piles by BD PLT. On the other hand, the analysis by the proposed method with soil coupling effect has a considerably larger settlement when compared with the results by uncoupled load transfer method and estimates reasonable load-settlement behaviors of test piles. In case of pile socketed in high strength rocks, however, effects of coupled soil resistance can be neglected.
