The objective of this research was to classify intact soybeans and foreign objects using near-infrared (NIR) spectroscopy. Intact soybeans and foreign objects were scanned using a NIR spectrometer equipped with scanning monochromator. NIR spectra of intact soybeans and foreign objects in the wavelength range from 900 to 1800 nm were collected. The classification of intact soybeans and foreign objects were conducted by using partial least-square discriminant analysis (PLS-DA) and soft independent modelling of class analogy (SIMCA) multivariate methods. Various types of data pretreatments were tested to develop the classification models. Intact soybeans and foreign objects were successfully classified by the PLS-DA prediction model with mean normalization pretreatment. These results showed the potential of NIR spectroscopy combined with multivariate analysis as a method for classifying intact soybeans and foreign objects.
The objective of this study was the development of a discrimination model for the cultivational origin of paper mulberry bast fiber and Hanji using near infrared (NIR) and mid infrared (MIR) spectroscopy combined with partial least squares discriminant analysis (PLS-DA). Paper mulberry bast fiber was purchased in 10 different regions of Korea, and used to make Hanji. PLS-DA was performed using pre-treated FT-NIR and FT-MIR spectral data for paper mulberry bast fiber and Hanji. PLS-DA of paper mulberry bast fiber and Hanji samples, using FT-NIR spectral data, showed 100 % performance in cross validation and the confusion matrix (accuracy, sensitivity, and specificity). The discrimination models showed four regional groups which demonstrated clearer separation and much superior score plots in the NIR spectral data-based model than in the MIR spectral data-based model. Furthermore, the discrimination model based on the NIR spectral data of paper mulberry bast fiber had highly similar score morphology to that of the discrimination model based on the NIR spectral data of Hanji.
Purpose: We developed a viability evaluation method for cucumber (Cucumis sativus) seed using hyperspectral reflectance imaging. Methods: Reflectance spectra of cucumber seeds in the 400 to 1000 nm range were collected from hyperspectral reflectance images obtained using blue, green, and red LED illumination. A partial least squares-discriminant analysis (PLS-DA) was developed to predict viable and non-viable seeds. Various ranges of spectra induced by four types of LEDs (Blue, Green, Red, and RGB) were investigated to develop the classification models. Results: PLS-DA models for spectra in the 600 to 700 nm range showed 98.5% discrimination accuracy for both viable and non-viable seeds. Using images based on the PLS-DA model, the discrimination accuracy for viable and non-viable seeds was 100% and 99%, respectively Conclusions: Hyperspectral reflectance images made using LED light can be used to select high quality cucumber seeds.

In this study, the feasibility of hyperspectral reflectance imaging technique was investigated for the discrimination of viable and non-viable lettuce seeds. The spectral data of hyperspectral reflectance images with the spectral range between 750 nm and 1000 nm were used to develop PLS-DA model for the classification of viable and non-viable lettuce seeds. The discrimination accuracy of the calibration set was 81.6% and that of the test set was 81.2%. The image analysis method was developed to construct the discriminant images of non-viable seeds with the developed PLS-DA model. The discrimination accuracy obtained from the resultant image were 91%, which showed the feasibility of hyperspectral reflectance imaging technique for the mass discrimination of non-viable lettuce seeds from viable ones.

Geographical classification of A. gigas was performed in the present study using UHPLC-DAD combined with multivariate data analysis techniques. Six active constituents were isolated from A. gigas; nodakenin, marmesin, decursinol, demethylsuberosin, decursin and decursinol angelate. One hundred sixty eight A. gigas samples were simultaneously determined using UHPLC-DAD. A principal component analysis (PCA) and partial least square discriminant analysis (PLS-DA) was used to classify the samples according to geographical origins (Korea and China). The origins of A. gigas from Korea and China were correctly classified by 81.6% and 93.8% using PLS-DA Y prediction. This result demonstrates the potential use of UHPLC-DAD combined with multivariate analysis techniques as an accurate and rapid method to classify A. gigas according to their geographical origin.
To determine whether FT-IR spectroscopy combined with multivariate analysis for whole cell extracts can be used to discriminate major leguminous plant at metabolic level, seed extracts of six leguminous plants were subjected to Fourier transform infrared spectroscopy (FT-IR). FT-IR spectral data from seed extracts were analyzed by principal component analysis (PCA), partial least square discriminant analysis (PLS-DA) and hierarchical clustering analysis (HCA). The PCA could not fully discriminate six leguminous plants, however PLS-DA could successfully discriminate six leguminous plants. The hierarchical dendrogram based on PLS-DA separated the six leguminous plants into four branches. The first branch was consisted of all three Vigna species including Vigna radiata var. radiate, Vigna angularis var. angularis and Vigna unguiculata subsp. Unguiculata. Whereas Pisum sativum var. sativum, Glycine max L and Phaseolus vulgaris var. vulgaris were clustered into a separate branch respectively. The overall results showed that metabolic discrimination system were in accordance with known phylogenic taxonomy. Thus we suggested that the hierarchical dendrogram based on PLS-DA of FT-IR spectral data from seed extracts represented the most probable chemotaxonomical relationship between six leguminous plants.
To determine whether pattern recognition based on metabolite fingerprinting for whole cell extracts can be used to discriminate cultivars metabolically, leaves of nine commercial Orostachys plants were subjected to Fourier transform infrared spectroscopy (FT-IR). FT-IR spectral data from leaves were analyzed by principal component analysis (PCA) and Partial least square discriminant analysis (PLS-DA). The dendrogram based on hierarchical clustering analysis of these PLS-DA data separated the nine Orostachys species into five major groups. The first group consisted of O. iwarenge 'Yimge', 'Jeju', 'Jeongsun' and O. margaritifolius 'Jinju' whereas in the second group, 'Sacheon' was clustered with 'Busan,' both of which belong to O. malacophylla species. However, 'Samchuk', belong to O. malacophylla was not clustered with the other O. malacophylla species. In addition, O. minuta and O. japonica were separated to the other Orostachys plants. Thus we suggested that the hierarchical dendrogram based on PLS-DA of FT-IR spectral data from leaves represented the most probable chemotaxonomical relationship between commercial Orostachys plants. Furthermore these metabolic discrimination systems could be applied for reestablishment of precise taxonomic classification of commercial Orostachys plants.
Purpose: Conventional methods used to evaluate seeds viability are destructive, time consuming, and require the use of chemicals, which are not feasible to implement to process plant in seed industry. In this study, the effectiveness of Fourier transform near infrared (FT-NIR) spectroscopy to differentiate between viable and nonviable watermelon seeds was investigated. Methods: FT-NIR reflectance spectra of both viable and non-viable (aging) seeds were collected in the range of 4,000 - 10,000 $cm^{-1}$ (1,000 - 2,500 nm). To differentiate between viable and non-viable seeds, a multivariate classification model was developed with partial least square discrimination analysis (PLS-DA). Results: The calibration and validation set derived from the PLS-DA model classified viable and non-viable seeds with 100% accuracy. The beta coefficient of PLS-DA, which represented spectral difference between viable and non-viable seeds, showed that change in the chemical component of the seed membrane (such as lipids and proteins) might be responsible for the germination ability of the seeds. Conclusions: The results demonstrate the possibility of using FT-NIR spectroscopy to separate seeds based on viability, which could be used in the development of an online sorting technique.
The Artemisia princeps (Compositae) has been used in traditional Korean medicine for the treatment of microbial infections and inflammatory diseases. Since A. princeps is generally difficult to be discriminated from A. capillaris, A. caplillaris has been misused in place of A. princeps. To solve this problem, a rapid and nondestructive method for discrimination of A. princeps and A. capillaris samples was developed using near infrared spectroscopy (NIRS) in the present study. A principal component analysis (PCA) and a partial least squares discrimination analysis (PLS-DA) were performed to discriminate two species. As a result, with the use of PLS-DA, A. princeps and A. capillaris were clustered according to their genus. These outcomes indicated that the NIRS could be useful for the discrimination between Artemisia princeps and Artemisia capillaris.
To classify Glycyrrhiza species, samples of different species were analyzed by $^1H$ NMR-based metabolomics technique. Partial least squares discriminant analysis (PLS-DA) was used as the multivariate statistical analysis of the 1H NMR data sets. There was a clear separation between various Glycyrrhiza species in the PLS-DA derived score plots. The PLS-DA model was validated, and the key metabolites contributing to the separation in the score plots of various Glycyrrhiza species were lactic acid, alanine, arginine, proline, malic acid, asparagine, choline, glycine, glucose, sucrose, 4-hydroxy-phenylacetic acid, and formic acid. The compounds present at relatively high levels were glucose, and 4-hydroxyphenylacetic acid in G. glabra; lactic acid, alanine, and proline in G. inflata; and arginine, malic acid, and sucrose in G. uralensis. This is the first study to perform the global metabolomic profiling and differentiation of Glycyrrhiza species using $^1H$ NMR and multivariate statistical analysis.
