Crystallization Behavior of Poly(lactic acid) / Poly($\varepsilon$-caprolactone) Blends (폴리락트산/폴리카프로락톤 블렌드의 결정화 거동)

  • The compatibility of poly(lactic acid)/poly($\varepsilon$-caprolactone) (PLA/PCL) blends as a function of blend composition was studied and triphenyl phosphite (TPP) was applied to PLA/PCL blends as a reactive compatibilizer. Especially the effect of compatibility on the crystallization behavior in both PLA/PCL blends and PLA/PCL blends with TPP was considered. PLA/PCL blends were immiscible based on thermal characteristics of PLA/PCL blends and the miscibility was depend upon the blend composition. The enhancement of compatibility was found in PLA/PCL blends with TPP depend upon its content. The rate of crystallization in PLA/PCL blend varied with blend composition. This was understood as the development of nucleation at the interface of PLA-PCL due to the immiscibility. TPP was acting as a compatibilizer as well as an agent for the acceleration of spherulite growth In PLA. As a result, the crystallization rate increased and the size of spherulite became larger than that of PLA/PCL blend without TPP.

  • In this study, we investigated the effects of electron beam irradiation on the rheological properties of PLA for enhancing processability. The electron beam was irradiated onto the pure PLA, chemically modified PLA by reactive extrusion, and PLA containing functional monomer. The complex viscosity and log G'vs. log G" plot among dynamic rheological properties were chosen for comparison. The complex viscosity of irradiated pure and chemically modified PLA decreased significantly due to de-gradation of PLA molecules with increasing the E-beam dosages. Complex viscosity of irradiated PLA with functional monomer showed maximum value at moderate dosage, while at high dosage the complex viscosity was decreased by a prolonged irradiation.

Development of High-specificity Antibodies against Renal Urate Transporters Using Genetic Immunization

  • Recently three proteins, playing central roles in the bidirectional transport of urate in renal proximal tubules, were identified: two members of the organic anion transporter (OAT) family, OAT1 and OAT3, and a protein that designated renal urate-anion exchanger (URAT1). Antibodies against these transporters are very important for investigating their expressions and functions. With the cytokine gene as a molecular adjuvant, genetic immunization-based antibody production offers several advantages including high specificity and high recognition to the native protein compared with current methods. We fused high antigenicity fragments of the three transporters to the plasmids pBQAP-TT containing T-cell epitopes and flanking regions from tetanus toxin, respectively. Gene gun immunization with these recombinant plasmids and two other adjuvant plasmids, which express granulocyte/macrophage colony-stimulating factor and FMS-like tyrosine kinase 3 ligand, induced high level immunoglobulin G antibodies, respectively. The native corresponding proteins of URAT1, OAT1 and OAT3, in human kidney can be recognized by their specific antibodies, respectively, with Western blot analysis and immunohistochemistry. Besides, URAT1 expression in Xenopus oocytes can also be recognized by its corresponding antibody with immuno-fluorescence. The successful production of the antibodies has provided an important tool for the study of UA transporters.

P3H4 promotes renal cell carcinoma progression and suppresses antitumor immunity via regulating GDF15-MMP9-PD-L1 axis

  • The prolyl 3-hydroxylase family member 4 (P3H4), is associated with post-translational modification of fibrillar collagens and aberrantly activated in cancer leading to tumor progression. However, its role in clear cell renal cell carcinoma (ccRCC) is still unknown. Here we reported that P3H4 was highly expressed in renal cancer tissues and significantly positive correlated with poor prognosis. Knockdown of P3H4 inhibited the proliferation, migration and metastasis of renal cancer cells in vitro and in vivo, and also, overexpression of it enhanced the oncogenic process. Mechanistically, P3H4 depletion decreased the levels of GDF15-MMP9 axis and repressed its downstream signaling. Further functional studies revealed that inhibition of GDF15 suppressed renal cancer cell growth and GDF15 recombinant human protein (rhGDF15) supplementation effectively rescued the inhibitory effect induced by P3H4 knockdown. Moreover, decreased levels of MMP9 caused by inhibition of P3H4-GDF15 signaling constrained the expression of PD-L1 and suppression of P3H4 accordingly promoted anti-tumor immunity via stimulating the infiltration of CD4+ and CD8+ T cells in syngeneic mice model. Taken together, our findings firstly demonstrated that P3H4 promotes ccRCC progression by activating GDF15-MMP9-PD-L1 axis and targeting P3H4-GDF15-MMP9 signaling pathway can be a novel strategy of controlling ccRCC malignancy.

Characteristics of CMP-PLA Heatsink Materials with Carbon Nanotube Contents (탄소나노튜브 양에 따른 CMP-PLA 방열 소재의 특성)

  • In this study, we proposed CMP-PLAs to replace the Al heat sinks as heat sink materials, and investigated heat dissipation characteristics of the LED lighting devices using them. The crystallinity of the proposed CMP-PLA heat sinks decreased with increasing carbon nanotube contents in CMP-PLA. However, the thermal conductivity was improved with the increase of the carbon nanotube contents. The heat dissipation characteristics of the LED lighting devices using CMP-PLA heat sinks was improved with increasing carbon nanotube contents in CMP-PLA. For the LED lighting devices using CMP-PLA heat sinks with 40% carbon nanotube contents, the initial temperature measured at the heat sink plate was $27^{\circ}C$, which increased as time, and it was saturated around $56^{\circ}C$ after an hour. The LED lighting devices using CMP-PLA heat sinks are expected to be functional materials that can reduce their weight and improve their electric properties, compared to those using existing Al heat sinks.

  • Product Line Engineering (PLE) has been widely accepted as a representative software reuse methodology by using core assets. Product line architecture (PLA) is a key element of core assets. However, current research works on designing PLA do not provide sufficient and detailed guidelines of defining PLA and reflecting variability in the architecture. In this paper, we present a reference model of PLA and propose a process to design PLA with detailed instructions. Especially architectural variability is codified by describing decision model depending variation points and traced through PLA activities. The proposed process would make it feasible to apply PLE to practice areas.

Rheological Properties of Poly(lactic acid) Modified by Electron Beam Irradiation (전자선 조사로 개질된 PLA의 유변학적 물성)

  • Poly(lactic acid)(PLA) has been modified by electron radiation in the presence of 5 phr glycidyl methacrylate (GMA) to enhance the melt strength of PLA. The modified PLA was prepared by varying the dose of irradiation and was characterized by observing the thermal properties, the melt viscoelastic properties and the gel fraction. The irradiated PLA with 300 kGy in the presence of 5 phr GMA showed drastically improved complex viscosity and storage modulus properties: a complex viscosity of about 210 times higher and a storage modulus of 14500 times higher than those of virgin PLA when measured at a frequency of 0.1 rad/s. Gel fraction study revealed that a branching reaction was more dominant than a crosslinking reaction when the PLA was irradiated with less than 200 kGy.

Fundamental Properties of Electrospun Polylactic Acid/Cellulose Nanocrystal Composite Mats (전기방사를 이용한 PLA/CNC 복합 매트의 기초 특성)

  • In this study, nanocomposite mats consisting of cellulose nanocrystals (CNCs) and poly(lactic acide) (PLA) were electrospun from a suspension mixture consisting of tetrahydrofuran at room temperature. Morphology study showed that fibers of electrospun composite mats were aligned in three dimensional surface along the fiber long-axis. Average diameter of the electrospun fibers decreased with an increase in the CNC loading level. Tensile strength of the electrospun fibers mat decreased with an increase in the CNC loading level because of bead formation in the formed fibers and low interfacial bond strength between PLA and CNC. Meanwhile, thermal stability of the electrospun nanocomposite mats was effectively improved as the amount of CNC increased.

  • Poly-Lactic Acid(PLA) fiber is an eco-friendly material and is biodegradable, so it can be utilized for manufacturing porous construction materia ls with interna l pore connection. In this study, domestic PLA fiber products(0.5mm india meter, 1.0mm in length, 10mm in length) were tested for melting at high temperatures and high alkality, and they were incorporated with FA-based geopolymer. Compressive strength was obtained through high temperature curing and alkali activator, however the complete melting of the PLA fiber was not ensured. The previous study handling PLA fiber with 0.003mm in diameter was completely dissolved, but 0.5mm and 1.0mm in diameter showed 42.5% and 33.3% of dissolution ratio, respectively. In addition, the increasing fiber volume led floating fibers during curing, which had a negative effect on its workability and solubility. Although the properties of PLA fiber may vary depending on the raw materials and production conditions, PLA fiber with 0.1mm or less diameter is recommended for porous construction material.

Study on Isothermal Crystallization Characteristics of PLA Film by Adding APP as a Nucleation Agent (APP 핵제를 첨가한 PLA 필름의 등온결정화 특성에 관한 연구)

  • In this paper, it was studied on the crystallization characteristics of PLA film by adding ammonium phosphate (APP) as a nucleation agent. Crystallinity and crystallite size of PLA film were determined by Scherrer equation. Crystallization rate constant of PLA film was calculated through Avrami equation. Film samples in the study were prepared by two steps. PLA films were prepared by adding 1, 5, and 10 wt%, respectively, at first and was secondly annealed at 130, 140, and $150^{\circ}C$. Crystallinity of pure PLA film was average 4.6% and those of PLA film with adding 1, 5, and 10 wt% APP were 12.2, 47.7, and 50.0%, respectively. Crystallite size of PLA film was average 28.0 nm and those of PLA film with adding 1, 5, and 10 wt% APP were 26.8, 24.0, and 19.0 nm, respectively. Crystallization rate constants of PLA film with 1 wt% APP were 2.12, 3.86, and 0.27 by annealing at 130, 140, and $150^{\circ}C$, respectively, where was higher than pure PLA film and those with adding 5 and 10 wt% APP, respectively.
