Estimation of Residual Biomass, PHB, and Nutrient Concentrations by Supplied Amount of Ammonia Solution in Fermentation of Alcaligenes latus

  • A novel estimation method was investigated for determining the concentrations of residual biomass, poly-3-hydroxybutyrate (PHB), and main nutrients including carbon and nitrogen sources, phosphate, and mineral ions from the supplied amount of ammonia solution used for a pH-control solution and nitrogen source in a PHB fermentation. The estimation equations for a batch culture and a fed-batch culture were derived from the relationship between the growth rate of residual biomass and the feed rate of the pH-control solution, and then were applied to the batch culture and the fed-batch cultures of Alcaligenes latus. This method was successfully applied to estimate the concentrations of residual biomass, PHB, and nutrients.

  • Using PHB biopolymer as polymer matrix, the release mechanism of a model drug, silver sulfadiazine was studied. The release behavior actually conformed to the Higuchi's diffusion controlled model. The release rate was delayed with an increasing proportion of PHB, whereas decreased as glycerine concentration incresed. The release rate was increased as the polymer matrix thickness increased.

  • The characteristics of cell growth and poly-$\beta$-hydroxybutyrate biosynthesis of Alcaligenes latus ATCC 29713 were investigated. The PHB accumulation pattern of A. latus followed a growth-associated type where the cell growth and PHB accumulation were carried out simultaneously. Various intermediate compounds such as metabolites involved in the TCA cycle, amino acids, and saturated and unsaturated fatty acids were added to examine their effect on cell growth and PHB accumulation. Citrate, tyrosine, and palmitic acid showed the most significant increase both on cell growth and PHB accumulation. Maximum PHB concentrations were noticeably increased about 1.4 to 1.6 times higher than that of control, corresponding to 5.54, 6.45, and 6.45 g/l for citrate, tyrosine, and palmitic acid, respectively. The stimulatory effects of the supplemented metabolites were analyzed in terms of the increment of enzyme activities related to sugar catabolism and PHB biosynthesis.

  • The effects of glucose concentration on the production of PHB by fed-batch culture of Ralstonia eutropha were investigated. In the range of glucose concentration of $2.5\;{\sim}\;40\;g/l$, it was found that the high glucose concentration was not favorable for the PHB formation after the phosphate limitation. It was further confirmed by the specific PHB synthesis rates and yields. The PHB concentration decreased much with the increase of glucose concentration. But if the glucose concentration was very low, e.g. 2.5 g/l, the cell growth and PHB synthesis also could be limited because of inadequate glucose supply. Itwould be better to maintain the glucose concentration at about 9.0 g/l to obtain high DCW, PHB concentration and productivity.

  • For the purpose of optimizing poly-l3-hydroxybutyrate (PHB) production from Alcaligenes eutrophus, two-stage fed-batch culture was adopted. In this system, specifk growth rate was maximized during the first stage whereas specific production rate was maximized during the second stage. The optimal concentrations of glucose and ammonium chloride were 16.6 and 0.54 g/I in the growth stage and 20.0 and 0.07 g/l in the production stage, respectively. Proportional feedback control considering time lag was suggested for PHB production process and a simulator was developed for real-time control purpose.

  • The structure of poly-$\beta$-hydroxybutyrate (PHB) in Chlorogloea fritschii was analyzed by FT-IR spectrometry under various conditions (phosphate starves or sufficient conditions). They exhibited characteristic absorption peaks for PHB, such as C=O stretching band at 1700-1800 $cm^-1$and C-H stretching bands at about 2900 $cm^-1$, however, the intensity of C-H stretching peaks, relative to the rest of the spectrum was increased under phosphate starved condition, which suggests that C. fritschii might produce another modified PHB polymer under phosphate starved condition.

Real-Time PCR Analysis of Metabolic Pathway of PHB in Acidiphilium cryptum DX1-1

  • The time, yield, and related genes expression of PHB accumulation of Acidiphilium cryptum DX1-1 were investigated under four different initial C/N ratios, 1.2, 2.4, 7.5, and 24. The results of time and yield of poly-$\beta$-hydroxybutyrate (PHB) accumulation show that the initial C/N ratio of 2.4 was optimum for strain DX1-1 to accumulate PHB, but both higher and lower initial C/N ratios did not favor that process. Based on the genome of Acidiphilium cryptum JF-5, 13 PHB accumulation related genes in strain JF-5 were chosen and successfully cloned from strain DX1-1. The differential expressions of the 13 functional genes, in different C/N ratios as cited above, were then studied by real-time PCR. The results show that all the 13 genes were most upregulated when the initial C/N ratio was 2.4, and among which the gene Acry_3030 encoding poly-$\beta$-hydroxybutyrate polymerase and Aery_0626 encoding acetyl-CoA synthetase were much more upregulated than the other genes, which proved that they play the most important role for PHB accumulation, and acetate is the main initial substance for PHB accumulation for strain DX1-1. Potential regulatory motifs analysis showed that the genes related to PHB accumulation are regulated by different promoters and that the motif had weak similarity to the model promoters, suggesting that PHB metabolism in Acidiphilium cryptum may be mediated by a different mechanism.

  • This study proposed the PHB mapping model for combining services with DiffServ in the MPLS network capable of high-speed switching, and the effective differential bandwidth service model in the MPLS network on the basis of the proposed mapping model. The result of simulation on the service model using PRI showed that the transmission rate was achieved as much as the assigned bandwidth regardless of the increase of traffic as far as the EF PHB did not exceed the assigned bandwidth at the peak rate when the overall traffic increased.

  • Recently, the growth of Internet and a variety of multimedia services through Internet increasingly demands high-speed packet transmission, the new routing function, and QoS guarantee on conventional routers. Thus, a new switching mechanical called the MPLS(Multi-Protocol Label Switching), was proposed by IETF(Internet Engineering Task Force) as a solution to meet these demands. In addition the deployment of MPLS network supporting Differentiated Services is required. In this paper, we propose the architecture of the MPLS switch supporting Differentiated Services in the MPLS-based network. The traffic conditioner consists of a classifier, a meter, and a marker. The VOQ-PHB module, which combines input Queue with each PHB queue, is implemented to utilize the resources efficiently. It employs the Priority-iSLIP scheduling algorithm to support high-speed switching. We have designed and verified the new and fast hardware architecture of VOQ-PHB and the traffic conditioner for QoS and high-speed switching using NS-2 simulator. In addition, the proposed architecture is modeled in VHDL, synthesized and verified by the VSS analyzer from SYNOPSYS. Finally, to justify the validity of the hardware architecture, the proposed architecture is placed and routed using Apollo tool.

Alcaligenes eutrophus 균주의 성장과 Poly-Beta-hydroxybutyrate 합성에 미치는 포도당과 암모늄농도의 영향

  • The biodegradation of Aroclor 1242 was investigated by the mixed cultivation of the natural bacterial isolates and a genetically engineered microorganism (GEM). The natural strain of MS-1003 degraded the Aroclor 1242 through the ortho-cleavage pathway, while the other strains through the meta-cleavage pathway. When the MS-1003 strain was additionally inoculated into the 1 day culture of the DJ-26 strain and then cultivated for 2 days, the Aroclor was degraded up to 86% and resulted in increase of the meta-cleavage product. But in the MS-1003 culture inoculated with the DJ-26, degradation of the Aroclor was limited to the level of each pure culture. By the mixed cultivation of the DJ-26 strain together with the DJ-12 or its GEM strain of DF-10, which degrades the Aroclor through the meta-cleavage pathway, degradation of the Aroclor as well as production of the meta-cleavage compound were lower than those of each pure culture. The degradation of Aroclor 1242 by the GEM strain was not improved over the parental strain. Therefore, a form of cometaboiism of Aroclor 1242 was found in the mixed culture of the DJ-26 and MS-1003 strains which degrade the Aroclor through the different metabolic pathway, but in the mixed culture of the DJ-26 and DJ-12 strains degrading Aroclor 1242 through the same pathway, a kind of competetion for the substrate was observed.
