  • NADPH has been known as a regulating factor the biosynthesis of polyhydroxyalkanote(PHA), and the flux of NADPH for PHA biosynthesis could be enforced by the amplification of zwf gene encoding glucose 6-phosphate dehydrogenase. The recombinant plasmid pCZWF harboring PHA synthase, phbC from R. eutropha and zwf from E. coli were constructed, and were transformed to R. eutropha by electroporation. The biosynthesis of P(3HB-3HV) copolymer were carried out in transformant R. eutropha through the two-stage cultivation method using valerate as a precursor. The biosynthesis rate and PHA content of transformant R. eutropha harboring pCZWF were increased compared with transformant R. eutropha harboring only phbC. Especially, the molar fraction of 3HV was increased from 68% to 74% due to amplification of zwf gene. And the biosynthesis P(3HB-3HV) and P(3HB-4HB) carried out using propionate and ${\gamma}-butyrolactone$ as a precursor, respectively. But the rate, content, and molar fraction of biosynthesis copolymers were not influenced appreciably. This may be due to the reduced availability of NADPH.

  • A batch-type study was conducted to investigate the phosphorus release and uptake under anaerobic and aerobic conditions according to organic loading changes. As organic loading increased, anaerobic P release increased but aerobic P uptake decreased. Where organic carbon contents remain high in aerobic conditions, PHB consumption within the microbial cells diminished, therefore it was found that in order to enhance P uptake rate, it should reach the endogenous growth stage where the entire organic loading was consumed.

  • The effects of transformation capacity on cometabolic degradation of trichloroethene (TCE) were evaluated using TCE-degrading actinomycetes pure and mixed culture under various culture conditions. The TCE transformation capacity of the actinomycetes enrichment culture in a batch test with phenol addition was 1.0 mg of TCE/mg of volatile suspended solids (VSS). The resting cell TCE transformation capacity of the actinomycetes pure culture cell was 0.75 mg TCE/mg VSS, which increased to 2.0 mg TCE/mg VSS when phenol was added as an external substrate. When the pure culture had an internal substrate in the form of poly-β-hydroxybutyrate (PHB) at 19% of the cell mass, the resting cell TCE transformation capacity increased from 0.47 to 0.6 mg TCE/mg VSS. The presence of PHB increased transformation capacity by 57%, whereas, the addition of phenol caused more than two fold increase in transformation capacity. The actinomycetes culture showed the highest transformation capacity.
