Effect of Bovine Somatotropin (bST) Treatment on Embryo Recovery and Pregnancy Rate in Hanwoo (한우에서 bST 처리가 수정란 회수 및 수태율에 미치는 영향)

  • The purpose of this study was to determine the effect of bST treatment on embryo recovery and pregnancy rate following embryo transfer. Donor cows were superovulated with Folltropin-V and PGF$_2$$\alpha$ combination method and then inseminated with frozen semen 3 times 12 hrs interval. Donor and recipient cows were assigned to control and bST group, of which was given a single injection of bST (500 mg, im) at insemination or estrus detection. Embryo collection of superovulated cows were flushed nonsurgical method at 7 to 8 days after artificial insemination. The percentare and Mean$\pm$S.E. of transferable embryo was not significantly different between control and bST treatment (72.8%/5.9$\pm$4.5 vs. 83.7%15.1 $\pm$ 1.6). The percentage and Mean$\pm$S.E. of transferable embryo in non-summer season was significantly higher than in summer (81.8%/5.4$\pm$2.1 vs. 68.7%14.774.6; P<0.05). The pregnancy rate after embryo transfer in bST treatment was significantly higher than in control (64.0 vs. 47.1%; P<0.05). There was no significant difference in pregnancy rate between summer and non-summer (51.6 vs. 61.5%; P>0.05). The results indicated that InST treatment in recipient cows could improve the efficiency of transferable embryo production and pregnancy rate after embryo transfer, and non-summer season may be better far superovulation treatment and embryo transfer.

Studies on Vaginal Cytology after Induction of Estrus in the Bitch (인공적(人工的) 발정유기견(發情誘起犬)의 질세포상(膣細胞像)에 관(關)한 연구(硏究))

  • The present study was carried out to investigate the estrous behavior and vaginal smear after induction of estrus with exogenous hormones in the premature, metestrous and anestrous bitches. A total of 21 bitches(Mixed breed: 16, Jindo breed: 5), from 10 months to 5 years of age and weighing 8 to 15 kg was studied the change of vaginal smear and the estrous behavior before and after induction of estrus. The results obtained are as follows: 1. In the treatment A(They were given the $PGF_2{\alpha}$, estrone, estradiol-$17{\beta}$, PMSG and HCG) proestrus commenced in $10.16{\pm}1.44$($Mean{\pm}SEM$) days after treatment, The mean duration of proestrus, and estrus was $7.50{\pm}1.44$ and $13.50{\pm}3.44$ days, respectively. In the treatment B(They were given the PMSG and HCG) proestrus commenced in $5.53{\pm}0.59$ days after treatment. The mean duration of proestrus and estrus was $6.60{\pm}0.71$ and $14.60{\pm}1.14$ days, respectively. 2. All of the 6 bitches in the treatment A showed vulval swelling and vaginal discharge, 14 of the 15 bitches in the treatment B showed vulval swelling and vaginal discharge. However, all of the treatment A and B showed male acceptance. 3. The main change of vaginal smear in proestrus and estrus after induction of estrus was a increase in the proportion of anuclear and superficial cells associated with a decrease in small intermediate and parabasal cells. By the estrous behavior and vaginal smear the estrus was induced in all the premature, metestrous and anestrous bitches.

  • As high as 50% of pregnancies are known to fail and the majority of such losses occur during the peri-implantation period. For the establishment of pregnancy in mammalian species, therefore, implantation of the conceptus to the maternal endometrium must be completed successfully. Physiological events associated with implantation differ among mammals. In ruminant ungulates, an elongation of the trophohlast in early conceptus development is required before the attachment of the conceptus to the uterine endometrium. Moreover, implantation sites are restricted to each uterine caruncula where tissue remodeling, feto-maternal cell fusion and placentation take place in a coordinated manner. These unique events occur under strict conditions and are regulated by numerous factors from the uterine endometrium and trophoblast in a spatial manner. Interferon-tau (IFN-${\tau}$), a conceptus-derived anti-Iuteolytic factor, which rescues corpus luteum from its regression in ruminants, is particularly apt to play an important role as a local regulator in coordination with other factors, such as TGF-${\beta}$, Cox-2 and MMPs at the attachment and placentation sites.

  • The purpose of this study was to determine the effect of rbST treatment on progesterone concentration, volume of luteal tissue and pregnancy rate following embryo transfer. Recipient cows were assigned to control and rbST group, of which was given a single injection of rbST (500 mg. sc) at estrus detection. The concentration of progesterone was not significantly different between control and at 0, 3, 6 days after rbST treatment. However, the concentration of progesterone at 9, 12 days was significantly higher than in control group (4.6 and 6.8 vs. 3.9 and 4.5 ng/ml P4). The pregnancy rate after embryo transfer in rbST treatment was significantly higher than in control group (64.0 vs. 47.1 %; p<0.05). The results indicated that rbST treatment in recipient cows could be improved the efficiency of pregnancy rate after embryo transfer.

  • This presentation firstly is discussed the characteristics of estrus, the time of first postpartum estrus, and the relative accurate of various estrus detection aids and secondly discussed the abnormalities of estrus and ovarian function and its control by treatment of exogenous hormones in cattle and pig. Longer estrus cycles as well as the shorter than 18 day cycles showed the lowered conception rates as compared to the normal cycles of 18 to 25 days. Other characteristics of est겨s such as duration of estrus, intensity of estrus and time of estrus are reviewed to affect fertility. The first postpartum ovulation and estrus in cows usually occurs about 20 to 30 days and 40 to 50 days after parturition, respectively. Irregularities in estrus cycle length have been conducted during early postpartum period. In sows, weaning is followed by ovulation and estrus although there is some individual variation. The most common method of estrus detection is direct visual observation on standing estrus behavior, but various aids of estrus detection have been empolyed with varying degree of effectiveness. The results from heat detector devices are about as accurate as twice-daily observation(about 90%). The abnormal estrus can be classified into three types; irregular or continuous estrus, silent estrus and anestrus. Cystic ovarian disease, follicular cysts and luteal cysts, is a serious cause of reproductive failure in cattle and pig. The follicular cysts are much more common than luteal cysts and the incidence of ovarian cysts in dairy cattle is higher than beef cattle and pig. The occurrences of ovarian cysts have been closely associated with levels of milk production, stages of postpartum period, nutritional levels and seasons. The luteal cysts and persistent corpora lutea are responsive to the luteolytic effects of the recently synthetic analogues of PGF2$\alpha$ in cows and sows and recently GnRH or LH-RH has been successfully used as a treatment for cows and sows with ovarian follicular cysts.

The Effect of Indomethacin on the Production of Eicosanoids and Edema during Ischemia-Reperfusion Injury in Skeletal Muscle

  • During reperfusion of skeletal muscle after ischemia, lipid mediators, mainly eicosanoids, are released and may have a role in the pathogenesis of reperfusion injury. To validate the role of eicosanoids in the ischemia-reperfusion induced functional deficits in skeletal muscle, we compared muscle edema and the changes of eicosanoid concentration in the rat hind limb after ischemia-reperfusion injury by application of tourniquet. After 4 hours of ischemia, reperfusion was established for 4 hours by releasing tourniquet. To assess tissue damage, edema, and wet/dry weight ratios were determined and the eicosanoid concnentrations were measured by the HPLC. The muscle edema and the release of cyclooxygenase metabolites were not induced by the ischemia itself rather they were significantly increased by reperfusion. Indomethacin treatment ameliorated limb edema and decreased the release of $6-keto-PGF_{1{\alpha}},$ thromboxane $B_2,$ and $PGE_2$ inducedby reperfusion. But the inhibitory effect of indomethacin on edema (35%) was relatively low than the inhibitory effect on release of cyclooxygenase metabolites (up to 69%) by reperfusion. These results support the view that cyclooxygenase products may play a significant role in the formation of muscle injury by ischemia-reperfusion and suggest that nonsteroidal antiinflammatory agents might be partially beneficial to the management of acute limb ischemia-reperfusion injury.

Surgical Treatment for Granulosa Cell Tumor in a Thoroughbred Mare (말에서 Granulosa Cell Tumor에 의한 무발정 치료 예)

  • An 8-year-old Thoroughbred mare showing anestrus for longer than 6 months with left ovarian mass was referred to Veterinary Medical Teaching Hospital of Seoul National University. For 6 months, the mare had received 2 times hormonal therapies with $PGF_2{\alpha}$ and progesterone. The mare had not responded to these therapies and not shown estrous behavior. On rectal palpation, the left ovary was enlarged and contralateral ovary was small and inactive. Transrectal ultrasonographic examination of the left ovary revealed a mass composed of multiple, irregularly shaped follicles. Granulosa cell tumor (GCT) was suspected according to her history. Ovariectomy was performed through the left flank laparotomy with a standing position in a simple chute located outside of horse farm. The mass was diagnosed as GCT on histopathology. The mare was recovered without any problem and became pregnant next spring.

Anti-Inhibin Serum 및 Follicular Fluid처리가 한우 난소의 난포발달과 혈중 호르몬변화에 관한 연구

  • 본 연구의 목적은 한우 난포발달에 있어서 난포액 및 anti-inhibin serum의 생리적 역할을 검토하기 위해 수행하였다. Anti-inhibin serum(AI)은 항원으로서 porcine inhibin-$\alpha$-subunit 19~32의 peptide를 사용하여 adjuvant 용액을 1:3의 비율로 혼합하여 앙고라종 토끼 5두(체중 2.5kg)에게 주 2회 간격으로 접종 후 얻어진 항혈청을 사용하였다. 난포액(bFF; bovine follicular fluid)은 도축장에서 도축되는 한우 난소로부터 직경 1.0cm 이하의 난포로부터 회수하여 스테로이드를 제거하기 위해 10% chacoal so lution(50 mg/$m\ell$, Norrit-A, Fisher Sci., USA)을 처리하여 45분간 배양후 원심분리후 상층액을 회수하여 실험에 공시하였다. 공시동물은 1산후 정상적으로 발정주기가 반복되는 한우암소 9두를 난소감정후 황체를 확인하여 PGF2$\alpha$제제(lutylase. USA)를 주사하여 발정을 유기한 다음, 난소의 first wave가 시작되는 시기인 배란직후 12시간째부터 4일간 일일 2회 5 $m\ell$씩 총 8회 40 $m\ell$의 AI와 bFF를 각각 경정맥으로 주사하였으며 대조구로서 생리식염수를 주사하였으며 채혈 및 정맥주사를 용이하게 하기 위해 경정맥에 카테타를 설치하여 6시간간격으로 총 200시간까지 채혈하였으며 초음파진단기를 이용하여 난포의 발달을 검토하였다. 채혈후 혈중Inhibin, progesterone 및 Estrad iol-17$\beta$농도의 분석은 RIA 및 ELISA법으로 분석하여 얻어진 결과는 다음과 같다. 혈중 Progesterone농도는 대조구와 AI처리구에서는 배란후 68시간째부터 유의적으로 증가하기 시작하였으나, bFF처리구에서는 배란후 68시간부터 170시간까지 대조구에 비해 유의적으로 낮은 농도를 나타내었다. 이에 반해 혈중 Estradiol-17$\beta$농도는 대조구의 경우 bFF처리구와 비슷한 수준으로 배란후부터 낮은 농도를 유지하였으나, AI 처리구는 배란후 36시간이후부터 108시간까지 유의적으로 높은 수준을 유지하였다가 그 이후 감소되었다. 한편, 혈중 Inhibin농도는 전 구간에서 배란후 84시간까지 불규칙한 농도를 보이다가 bFF처리구에서는 배란후 84시간부터 유의적으로 증가하였다. 배란후 72시간째에 초음파진단기를 이용하여 난소의 난포발달을 조사한 결과 , 대조구와 bFF처리구에 비해 AI처리구에서 발달난포가 유의적으로 많은 것을 확인하였다. 이상과 같은 결과로, Anti-inhibin serum은 한우 자체에서 분비하는 Inhibin을 특이하게 억제하여 Inhibin에 의해 억제되는 FSH분비가 촉진됨으로써 난포발달과 estrogen의 농도가 촉진되는 것으로 사료되어 anti-inhibin serum이 한우의 과배란유기 효과가 있는 것으로 사료된다.

Comparison of Estrous Behavior and Ovulation Time in Dairy Cows and Heifers (젖소 경산우와 미경산우의 발정 행동과 배란 시간의 비교)

  • The objective of this study was to investigate the characteristics of various estrous behavior and ovulation time in dairy cows and heifers. In total, 73 ovulations and 61 estrous detection were observed in 89 Holstein cows. Various estrous behavior were observed during 72 hours from two days after $PGF_2{\alpha}$ injection and their relation with the time of ovulation(ultrasound examinations at 4-h intervals) was investigated. In estrous periods, the rate of sniffing, chin resting, mounting and standing heat was 81%, 78%, 78% and 56%, respectively in cows. In heifers, the rate of sniffing, chin resting, mounting and standing heat was 61%, 68%, 82% and 76%, respectively. Ovulation in cows and heifers occurred $25.58{\pm}7.94\;and\;25.55{\pm}5.72h$ after onset of estrus, and $13.42{\pm}7.14\;and\;7.48{\pm}7.41h$ after end of estrus, respectively. Interval between onset of estrus and ovulation time was significantly (p<0.05) shorter for standing heat ($17.33{\pm}5.83\;h$) than for mounting, sniffing and chin resting ($23.58{\pm}5.12\;h,\;24.25{\pm}6.09\;h,\;23.42{\pm}6.04\;h$) in cows but not significantly different in heifers. Interval between end of standing heat and ovulation time was significantly (p<0.05) shorter for heifer($6.38{\pm}4.80$) than for cows($13.05{\pm}4.53$). Our results show that characteristics of estrous behavior and ovulation in dairy heifers are different to that of cows.

Relationship between Various Estrous Behavioral Signs and Ovulation Time in Dairy Cows (젖소의 다양한 발정 행동 징후와 배란 시간과의 관계)

  • The objective of this study was to investigate the relationship between various estrous behavior and ovulation time, and to determine which estrous behavior could predict ovulation time more accurately. In total, 37 ovulations and 28 estrous detection were observed in 51 Holstein-Friesian dairy cows. Various estrous behavior were observed during 72 h from two days after $PGF_2{\alpha}$ injection and their relation with the time of ovulation (ultrasound examinations at 4-h intervals) was investigated. In estrous periods, the rate of sniffing, chin resting, mounting, standing heat was 81%, 78%, 78% and 56%, respectively. Ovulation occurred $25.6{\pm}7.9h$ after onset of estrus (ranging between 7 and 37h) and $13.4{\pm}7.1h$ after end of estrus (ranging between 1 and 28h). Interval between onset of estrus and ovulation time was significantly (p<0.05) shorter for standing heat $(17.33{\pm}5.83h)$ than for mounting, sniffing and chin resting $(23.58{\pm}5.12h,\;24.25{\pm}6.09h,\;23.42{\pm}6.04h)$. In 88% of the animals that displayed mounting, ovulation occurred between $16{\sim}28h$ after onset of mounting. Onset of standing heat, sniffing and chin resting occurred between $10{\sim}22(81%)h,\;16{\sim}28(79%)h\;and\; 19{\sim}31(79%)h$ before ovulation respectively. Sniffing and chin resting were displayed during the non-estrous period and are therefore, not useful predictors of ovulation time. The standing heat and mounting can be a good predictor for time of ovulation but the disadvantage of using standing heat is that only a limited number of cows display standing heat. Thus, it is concluded that mounting behavior could be the best predictor for time of ovulation.
