Hematologic and biochemical reference intervals for natural monument Korean goral (Naemorhedus caudatus) (천연기념물 한국 산양의 혈액 및 혈액 화학적 정상표준범위)

  • Korean goral (Neamorhedus caudatus) is registered as a natural monument number 217 by South Korea Cultural Heritage Administration. It is also recognized as the endangered species I by Convention on International Trade in Endangered Species of Wild Fauna and Flora (CITES). In this study, blood samples of Korean goral were collected to make reference intervals. Blood sampling was conducted on 19 numbers of Korean gorals (ten females, nine males), which were reared in Korean Goral Restoration Center. For total samples, the reference intervals of hematological parameters were: white blood cell $7.69{\sim}10.87K/{\mu}{\Lambda}$; hematocrit 36.73~46.18%; red blood cell $10.72{\sim}12.86K/{\mu}{\Lambda}$; hemoglobin 12.79~15.14 g/dL; mean corpuscular volume 33.15~36.75 fL; mean corpuscular hemoglobin 11.53~12.23 pg; mean corpuscular hemoglobin concentration 32.64-35.91 g/dL and red blood cell distribution width 39.2~43.46%. For total samples of each parameters, the following results were obtained for serum biochemistry: glucose 111.81~153.77 mg/dL; blood urea nitrogen 22.35~28.91 mg/dL; creatine 1.22~1.84 mg/dL; phosphate 4.57~6 mg/dL; calcium 8.7~9.1 mg/dL; total protein 6.53~6.92 g/dL; albumin 3.1~3.48 g/dL; globulin 3.26~3.62 g/dL; alanine aminotransferase 56.7~158.56 U/L; aspartate aminotransferase 230.35~473.06 U/L; alkaline phosphatase 178.06~332.47 U/L; gamma-glutamyl transpeptidase 131.6-~181.24 U/L; total bilirubin 1.47~2.12 mg/dL; cholesterol 46.48~71.52 mg/dL; amylase 16.3~26.03 U/L; sodium 150.43~153.88 mmol/L; potassium 3.98~4.6 mmol/L and chlorine 109.48~113.26 mmol/L. The ranges of values were similar campared to previous studies except in the case of RDW value, which showed higher range than the RDW value of a previous study. The reference intervals from this study will be useful data for treatment and management of gorals.

Effect of Subacute Ruminal Acidosis on Plasma Concentrations of Lipopolysaccharide in Dairy Cattle (젖소에 있어 아급성제1위과산증(SARA)의 강도가 혈장 Lipopolysaccharide (LPS)의 농도에 미치는 효과)

  • A study was conducted to determine if subacute ruminal acidosis (SARA) induced by feeding high level of non-structural carbohydrates results in increases in lipopolysaccharides (LPS) of peripheral blood in dairy cattle. In this experiment four Holstein steers, two of which having ruminal canulae with indwelling probes placed for measuring long term pH changes, were alloted into two dietary treatments in a cross-over design, where an acidogenic diet (AD) was formulated by including high amount of non-structural carbohydrates (NSC) based on corn silage and corn flake as TMR ingredients. Data for ruminal pH change and plasma LPS concentration were compared against normal diet (ND) which contained grass hay as forage and low NSC ingredients. Feeding AD for more than 14 days to animals brought about a pH change as low as less than 5.8 for more than 4 hours, which made good contrast to ND fed animals. Decreased ruminal pH also had an effect on LPS concentrations which showed significantly higher level for AD compared with ND. Therefore, plasma LPS concentration may be used as an effective indicator to verify acidosis whether it is acute or chronic. Na-bentonite and Propylene glycol, which are frequently included in dairy TMR as additives, helped decrease ruminal pH by buffering and then ultimately alleviates SARA. Therefore, it could also be helpful to lower the occurrence of laminitis which is often caused by increased blood endotoxin (LPS) concentration.

Anti-oxidative and Immunomodulating Activities of Solvent Extracts from Broccoli (Brassica oleracea) Sprouts (브로콜리 새싹 용매 추출물의 항산화 및 면역조절 활성)

  • In order to examine the functionality of broccoli sprout (Brassica oleracea, BS), solvent extracts were prepared and their anti-oxidative and immunomodulating activities were compared with those of broccoli (B). EtOH extracts (E) were potently higher than hot-water extracts (HW) in the antioxidant contents and radical scavenging activity. In particular, the total polyphenolic contents in addition to ABTS and DPPH radical scavenging activity were significantly higher in EtOH extract of broccoli sprout (BS-E; 9.15 mg GAE/g, 4.52 mg AEAC/g, and 1.14 mg AEAC/g) compared with that of broccoli (B-E; 7.83 mg GAE/g, 3.63 mg AEAC/g, and 0.97 mg/AEAC/g). Whereas, total flavonoid content was significantly higher in B-E (1.60 mg QE/g) than BS-E (1.43 mg QE/g). Anti-inflammatory activity was investigated using LPS-induced cell line model at a concentration of $10{\sim}100{\mu}g/mL$, in which all solvent extracts of both broccoli sprouts and broccoli were not toxic to RAW 264.7 cell lines. In anti-inflammatory activity of broccoli sprouts, EtOH extracts also showed significantly more potent activity than hot-water extracts in all sample concentrations tested. In addition, BS-E ($100{\mu}g/mL$) inhibited nitric oxide (NO) and IL-6 production to 60.9% and 68.9% compared with the LPS inflammation group (without extracts), whereas B-E inhibited 49.6% and 54.9%. On the other hand, in immunostimulating activity by splenocytes and macrophages, hot-water extract showed significantly higher activity than EtOH extract. Especially, BS-HW stimulated the splenocyte proliferation (1.2-fold against saline group) and IFN-${\gamma}$ production (264.39 pg/mL) at $100{\mu}g/mL$, and the production of IL-6 (1.33-fold), IL-12 (1.09-fold) and TNF-${\alpha}$ (1.49-fold) from macrophages was also significantly enhanced over broccoli. In conclusion, broccoli sprouts showed more potent anti-oxidative, anti-inflammatory and immunostimulating activity than broccoli, suggesting the possibility of using broccoli sprouts as functional food materials.

Type 3 Gastric Neuroendocrine Neoplasm Clinical Features: A Multicenter Study in Korea

  • Background: The aim of this study was to investigate clinicopathologic features of type 3 gastric neuroendocrine neoplasm (NEN) by treatment modality. Methods: The Korean Society of Gastrointestinal Cancer conducted the Korean Gastroenteropancreatic Neuroendocrine Tumor Registry, a retrospective registry database of gastroenteropancreatic neuroendocrine tumors from 16 hospitals in Korea. The normal serum gastrin level range was defined as <100 pg/mL, and gastric NEN patients with normal gastrin level were selected for analysis. Results: Among 358 patients with gastric NEN, 21 (5.9%) patients were classified with type 3 gastric NEN. The median age was 53 years (range 30-74). According to the WHO 2010 classification, 13 (61.9%) patients had grade 1, and 8 (38.1%) patients had grade 2 or 3. Endoscopic treatment was performed in 14 (66.7%) patients, and surgery was performed in 7 (33.3%) patients. The tumor size was smaller in the endoscopic treatment group than in the surgery group (0.6 cm vs 1.3 cm, p=0.006). After treatment, there was one recurrence in the surgery group. Conclusion: In small size Type 3 gastric NEN, endoscopic treatment was associated with a good prognosis, compared to surgery. Thus, endoscopic treatment can be used an alternative modality in selected cases of type 3 gastric NEN.

Blood Picture of the Korean Native Goats from Birth to Maturity (한국 재래 염소의 성장에 따르는 혈액상의 변동)

  • Although considerable research has been done on the changes associated with age in the blood picture of domestic and laboratory animals, little work has been made of the changes occurring at different age in the blood picture of goats. And a comprehensive survey of the bood picture of Korean native goats has not been made. The object of the present investigation was to suggest standards for the blood picture of Korean native goats at frequent intervals from birth to maturity. The goats were kept under average farming conditions in this country. Observations were made at the following ages: at birth; 2,4 and 7 days; 2, 3, 4, 5, 6, 7, 8 and 9 weeks; 2.5, 3,6 and 12 months. Blood samples were drawn from the jugular vein. It was taken between 8 and 9 a.m. except those for the at-birth period. Erythrocyte and leukocyte enumerations and, determinations of hemoglobin in blood and hematocrit value were made in the usual manner. Reticulocytes were enumerated per 1,000 erythrocytes in blood smears stained with briIliant cresyl blue and counterstained with Wright's stain. Erythrocytes counts declined from $8.7{\times}10^8/mm^3$ at birth to a low of 7.0 at 4 days of age. These values increased to 11.5 at 5 weeks and reached a maximum of 14.0 at 3 months of age; it then fell to 11.5 at 12 months of age. Concentrations of hemoglobin in blood and hematocrit values were not related to the changes of erythrocyte counts. The values at birth were higher than at any other period during the first year of life. These fell from highs of 12.3 g/100 ml and 38.0 ml/100 ml to lows of 9.2 and 29 at 4 weeks for concentration of hemoglobin in blood and hematocrit value, respectively. There was a common pattern for the hematocrit value and hemoglobin in blood which showed three phases-a fall during the first month, a rise to the third month, and a fall to the mature level at 12 months of age. Mean corpuscular volume and mean corpuscular hemoglobin showed a common pattern. The values were $44.2{\mu}m^3$ and 14.2 pg at birth and fell, at first slowly and then rapidly, to reach adult levels of 24.1 and 7.9 at 6 weeks of age for mean corpuscular volume and mean corpuscular hemoglobin, respectively. Mean corpuscular hemoglobin concentration was little affected by age. Reticulocyte was observed from birth to 4 weeks of age. Percentage of reticulocyte decreased from 0.85% at birth to 0.06% at 4 weeks of age. Total leucocyte counts increased from $5.64{\times}10^3/mm^3$ at birth to a maximum of 13.4 at 3 months; it then fell to 11.5 at 12 months of age. In differential counts myelocyte, juvenile and band form decreased with advancing age. No myelocyte and juvenile were seen after the age of 7 and 9 weeks, respectively, and band forms were rare after the age of 3 months. Percentage of mature neutrophil showed a quick decline from 52.5% at birth to reach a minimum level (34.5%) at 3 months of age; it then rose to 38% at 12 months of age. Percentage of lymphocyte increased from 39.2% at birth to maximum of 59% at 3 month of age; it then fell to 54.9% at 12 months of age. Percentage of monocyte was not affected with advance of age. Percentage of eosinophil and basophil were increased with advance of age to reach a maximum at 2 to 3 month of age. It then fell to adult level at 12 month of age.

Influence of the angles and number of scans on the accuracy of 3D laser scanning (3 차원 레이저 스캔영상 채득 시 스캔각도와 횟수에 따른 정확도)

  • Objective: To investigate whether the accuracy of 3D laser scanning is influenced by the angles and number of scans. Methods: Using a 3D laser scanner, 10 manikins with facial markers were scanned at 7 horizontal angles (front view and at $20^{\circ}$, $45^{\circ}$, and $60^{\circ}$ angles on the right and left sides). Three-dimensional facial images were reconstructed by 6 methods differing in the number and angles of scans, and measurements of these images were compared to the physical measurements from the manikins. Results: The laser scan images were magnified by 0.14 - 0.26%. For images reconstructed by merging 2 scans, excluding the front view; and by merging 3 scans, including the front view and scans obtained at $20^{\circ}$ on both sides; several measurements were significantly different than the physical measurements. However, for images reconstructed by merging 3 scans, including the front view; and 5 scans, including the front view and scans obtained at $20^{\circ}$ and $60^{\circ}$ on both sides; only 1 measurement was significantly different. Conclusions: These results suggest that the number and angle of scans influence the accuracy of 3D laser scanning. A minimum of 3 scans, including the front view and scans obtained at more than $45^{\circ}$ on both sides, should be integrated to obtain accurate 3D facial images.

Effect of Aprotinin on Changes in Plasma Thromboxane $B_2$ and Endothelin-1 Concentratin after Extracorporeal Circulation (체외순환후 혈중 Thromboxane $B_2$와 Endothelin-1 농도 변화에 미치는 Aprotinin의 효과)

  • Background: Thromboxane A2 and endothelin-1 are the potent vasoconstrictors affecting pulmonary pathophysiology in response to whole body inflammatin following CPB. Aprotinin, as an antiiflammatory agent, may decrease the release of such vasoactive substance from pulmonary tissues, preventing pulmonary hypertension after cardiopulmonary bypass. Material and Method: Ten mongrel dogs(Bwt. ac. 20kg) were subjected to cardioupulmonary bypass for 2 hours and postbypass pulmonary vascular resistance(0, 1, 2, 3 hours) were compared with prebypass level. The dogs were divided into 2 groups; control group(n-5) and aprotinin group(n=5). In the aprotinin group, aprotinin was administered as follows; 50,000 KIU/kg mixed in pump priming solution, 50,000 KIU/kg prebypass intravenous infusion over 30 minutes, 10,000 KIU/kg/hour postbypass continuous infusion. Prebypass and postbypass 0, 1, 2, 3 hour pulmonary vascular resistance were measured. At prebypass and postbypass 0, 90, 180 minutes, blood samples were obtained from pulmonary arterial and left atrial catherers for the assay of plasma thromboxane B2 a stable metabolite of thromboxane A2, and endothelin-1 concentrations. Result: The ratios of pustbypass over prebypass pulmonary vascular at postbypass 0, 1, 2, 3 hours were 1.28$\pm$0.20, 1.82$\pm$0.23, 1.90$\pm$0.19, 2.14$\pm$0.18 in control group, 1.58$\pm$0.18, 1.73$\pm$0.01, 1.66$\pm$0.10, 1.50$\pm$0.08 in aprotinin group ; the ratios gradually increased in control group while decreased or fluctuated after postbypass 1 hour in aprotinin group. There was statistically significant difference between control group and aprotinin group at postbypass 3 hours(P=0.014). Pulmonary arterial plasma concentration of thromboxane B2(pg/ml) at prebypass, postbypass 0, 90, 180 minutes were 346.4$\pm$61.9, 529.3$\pm$197.6, 578.3$\pm$255.8, 493.3$\pm$171.3 in control group, 323.8$\pm$118.0, 422.6$\pm$75.6, 412.3$\pm$59.9, 394.5$\pm$154.0 in aprotinin group. Left atrial concentrations were 339.3$\pm$89.2, 667.0$\pm$65.7, 731.2$\pm$192.7, 607.5$\pm$165.9 in control group, 330.0$\pm$111.2, 468.4$\pm$190.3, 425.4$\pm$193.6, 4.7.3$\pm$142.8 in aprotinin group. These results showed decrement of pulmonary thromboxane A2 generation in aprotinin group. Pulmonary arterial concentrations of endothelin-1(fmol/ml) at the same time sequence were 7.84$\pm$0.31, 13.2$\pm$0.51, 15.0$\pm$1.22, 16.3$\pm$1.73 in control group, 7.76$\pm$0.12, 15.3$\pm$0.71, 22.6$\pm$6.62, 14.9$\pm$1.11 in aprotinin group. Left atrial concentrations were 7.61$\pm$17.2, 57.1$\pm$28.4, 18.9$\pm$18.2, 31.5$\pm$20.5 in control group, 5.61$\pm$7.61, 37.0$\pm$26.2, 28.6$\pm$21.7, 37.8$\pm$30.6 in aprotinin group. These results showed that aprotinin had no effect on plasma endothelin-1 concentration after cardiopulmonary bypass. Conclusion: Administration of aprotinin during cardiopulmonary bypass could attenuate the increase in pulmonary vascular resistance after bypass. Inhibition of pulmonary thromboxane A2 generation was thought to be one of the mechanism of this effect. Aprotinin had no effect on postbypass endothelin-1 concentration.

Effects of Removal Times of Transplanted Pituitary Gland on Induction of Ovulation and Levels of Estrogen and Progesterone in Immature Rats (미성숙 쥐에 이식한 뇌하수체의 경시별제거가 배란유도와 Estrogen 및 Progesterone수준에 미치는 영향)

  • The present work was designed to understand the mechanism of superovuiation and the cause of early embryo loss and Implantation failure in the superovulating immature female rats which were elaborated by a pituitary gland transplantation. A pituitary gland obtained from the orchidectomized rats was transplanted under the right kidney capsule of 28 day old female rats (PGT group) on the starting day of experiment which was designated as Day 2. The grafted pituitary glands were removed at 6h (RPGT 6h group), 12h (RPGT 12hgroup) and 24h (RPGT 24h group) after the transplantation. Control rats were treated with 41U PMSG on Day 2 (PMSG group). The estrous cycle and the levels of plasma progesterone and estradiol-17$\beta$ were observed on Day 0, Day 5, respectively. The implantation sites, the weights of ovary and uterus, and the number of corpora lutea were examined in all group on Day 8. The resuft obtained were summarized as follows: 1. The percentages of the number of the rats in proestrus and estrus were 93.0%, 82.6%, 0%, 90.7% and 89.5% in PMSG, PGT, RPGT6h, RPGT12h and RPGT24h group, respectively. The synchronization of estrus cycle was dchieved in all groups. 2. The mating rates of each group were 80.2, 75.0, 0, 56.4, 57.8% in PMSG, PGT, RPGT6h, RPGT12h, RPGT24h group, respectively. 3. The numbers of copora lutea on Day 8 were 47.1 i 4.9, 18.1 $\pm$0.5, 14.1 i 0.3 and 8.9 $\pm$ 0.3 in PGT, RPGT24h, RPGTl2h and PMSG group, respectively. There were signIficantly difference between all groups (P<0.05). 4. The numbers of implantation sites (18.1 +- 4.0) in PGT group on Day 8 were higher than those of PMSG (8.5 $\pm$ 2.5), RPGT 12h (9.8 i 0.2) and RPGT 24h group (10.8 i 0.2) (P<0.05). 5. The ovarlan weights in PGT (95.2 $\pm$ 14.3mgIlOOg BW), RPGT 12h (51.7 $\pm$ 0.6mgIlOOg BW), and RPGT24h (57.9 $\pm$ 0.9mg/l00g 8W) groups were significantly higher than those of PMSG group (30.4 $\pm$7.4mg/l00g BW) (P<0.05). 6. The uterine weights in PMSG (672.4 $\pm$ 4.7mg/l00g 8W), and PGT (660.7 $\pm$7.8mg/l00g BW) groupswere greater than those of RPGT 12h (403.0 $\pm$ 1.lmg/lOOg BW) and RPGT 24h (490.1 $\pm$ 0.9mg/l00g BW) group (P<0.05). 7. The plasma progesterone levels in PGT groups (15 lng/ml) on Day 5 were higher than those of PMSG (83ng/ml), RPGT 12h (S7ng/ml), RPGT24h (8lng/ml) group (P<0.05). 8. The plasma estradiol-17$\beta$ levels in PMSG group (lBSpg/ml) on Day S were higher than those of RPGT 24h (l3pg/ml) group (P<0.05). But estradiol-17$\beta$ levels in PGT and RPGT 12h group were too low to discuss.

The Therapeutic Effect of Angiotensin II Receptor Antagonist in Idiopathic Pulmonary Fibrosis (특발성 폐섬유화증 환자의 치료에서 Angiotensin II Receptor Antagonist의 치료효과)

  • Background : There have been several studies showing that the angiotensin II and angiotensin converting enzyme(ACE) contributes to the apoptosis of alveolar epithelial cells in idiopathic interstitial pneumonia and the activation of fibroblasts during the process of pulmonary fibrosis. These results suggest that the pulmonary fibrosis can be inhibited by the angiotensin II receptor antagonist(AGIIRA). This study was performed to identify the therapeutic effect of AGIIRA in idiopathic pulmonary fibrosis(IPF). Method : Thirteen patients with IPF, who were diagnosed with an open lung biopsy(6 patients) and furfilling the ATS criteria(7 patients) between March 1999 and October 2001 at the Gachon medical center, were enrolled in this study. Of these patients, eight patients were treated with a regimen including AGIIRA(AT group), and five were treated without AGIIRA(NT group). The pulmonary function tests and dyspnea(ATS scale) were measured at diagnosis and 1 year after treatment. All the data was collected to analyze the therapeutic effect of AGIIRA on the patients with IPF. Results : The AT group contained 8 patients(M:F=4:4) and the NT group contained 5 patients(M:F=3:2). There was no significant difference in the serum angiotensin II level between the two groups($202.5{\pm}58.5$ vs $163.7{\pm}47.3pg/ml$, p>0.05). The AT group showed an upward trend in TLC(+3%), FVC(+4%), FEV1(+3%) and DLco(+2%) compared to the NT group(TLC(-14%), FVC(-3%), FEV1(-4%) except for DLco(+5%)). The dyspnea score in the AT group improved significantly but not in the NT group. Conclusion : These results suggest that the angiotensin II receptor antagonist may have an effect on stabilizing IPF.

Hypocalcemic Convulsion in Formula Feeding Young Infants (영아에서 경련을 동반한 저칼슘혈증에 관한 연구)

  • Purpose : Hypocalcemia is one of the major causes of convulsion in infants. The causes of hypocalcemia are hypoparathyroidism, deficiency and metabolic abnormalities of vitamin D, or increased uptake of inorganic phosphorous, etc. We analyzed the causes, symptoms and signs, treatment, and clinical courses of hypocalcemia as, recently, there were many clinical experiences of hypocalcemic infants under age of 6 months in the department of pediatrics, Kyungpook University Hospital. Objects and Methods : The authors observed 11 infants with hypocalcemia who had been admitted to the department of pediatrics, Kyungpook University Hospital, during the period of February 1992 to April 1997. Various clinical and laboratory data concerning causes, clinical courses and treatment of hypocalcemia were analyzed retrospectively Results : (1) The sex incidence revealed male predominance with male to female ratio 4.5 : 1. The mean age at onset of symptoms was $2.2{\pm}1.1$ months old. (2) The causes of hypocalcemia were vitamin D deficiency in 8 cases and excessive inorganic phosphate intake in 3 cases. (3) All eleven patients manifestated convulsion which was generalized tonic-clonic in 9, and focal clonic in 2 cases. (4) Serum calcium concentrarion increased from $6.3{\pm}0.9$ mg/dL to $9.9{\pm}1.7$ mg/dL after therapy of $1,25(OH)_{2}D_{3}$ with or without calcium(P=0.0008), and serum ALP concentration decreased from $1,418{\pm}864$ U/L to $772{\pm}503$ U/L (P=0.0112). Serum iPTH levels were high in all 11 patients initially. All showed decreased $25(OH)D_3$ levels initially. (5) All patients were treated successfully with $1,25(OH)_{2}D_{3}$ and/or calcium supplement. Conclusions : Vitamin D deficiency should be considered as one of the causes of hypocalcemia even in formula(known as vitamin fortified) feeding infants. Fortunately, they were successfully treated with $1,25(OH)_{2}D_{3}$.
