Purification of Cellulase Produced from Cellulomonas sp. YE-5 (Cellulomonas sp. YE-5가 생산하는 Cellulase의 정제)

  • An extracellular cellulase producing bacterium YE-5 was isolated from soil, and identified as a Cellulomonas sp. by its taxonomical characteristics. The maximal activities of avicelase (0.35 units/ml), CMCase (3.18 units/ml), FPase (0.315 units/ml) and $\beta$-glucosidase (0.882 units/ml) were obtained when this strain was cultured for 48 hrs at $30^{\circ}C$ in a medium containing 0.8% (w/v) Solka floc, 0.06010 (wlv) urea, 0.1% (w/v) $K_2HP0_4$, 0.1% (w/v) $MgS0_4.7H2_0$, 0.2% (w/v) bacto peptone, 0.2% (w/v) yeast extract and pH 6.5. The cellulase was purified by ammonium sulfate fractionation, DEAE-Sepharose column chromatography and Sephadex 6-100 column chromatography from culture filtrate of Cellulomonus sp. YE-5. The molecular weights of purified avieelase, CMCase I, and CMCase II were estimated to be about 95,000 ~ 105,000, 46,000 ~ 47,000 and 120,000 ~ 125,000, respectively.

  • A filamentous microorganism was isolated from completely rotten wood for the production of cellulolytic enzyme. The Trichoderma sp. FJ1 produced a large amount of cellulolytic enzymes, such as CMC, xylanase, ${\beta}-glucosidase$, and avicelase. For the production of the enzymes, when cellulolsic wastes were used as carbon sources of strain FJ1, rice straw showed higher enzyme activities than sawdust and pulp. The activities of CMC, xylanase, ${\beta}-glucosidase$, and avicelase were 2.95, 5.89, 0.45, and 0.12 U/ml in use of rice straw, respectively. To enhance production of the enzymes, the mixture substrate of rice straw and commercial cellulosic materials was investigated as carbon sources. The highest activities of CMCase, ${\beta}-glucosidase$, and avicelase were found in the mixture of rice straw and avicel, particularly rice straw:avicel (50:50), and the highest xylanase was obtained in the mixture ratio of 71:29. Bacto peptone addition of 0.1% showed enhanced production of the cellulolytic enzymes in which the activities of CMCase, xylanase ${\beta}-glucosidase$, and avicelase were 19.23, 27.18, 1.28, and 0.53 U/ml, respectively. The production of the enzymes using rice straw was efficiently induced in present of avicel and pulp containing high content of cellulose. Consequently, the filamentous microorganism, strain FJ1 utilized various cellulosic wastes as carbon sources and cellulases productivities were excellent compared to those of others strains reported previously, suggesting that the strain FJ1 will be expected as a favorable candidate for biological saccharification of cellulosic wastes in further.

  • This study was attempted to select an antagonist against Phytophthora blight of red-pepper caused by Phytophthora capsici. The three strains, A-35, A-67 and A-183 were isolated from the rhizosphere in soil where red-pepper had been cultivated continuously for a long time, and the strain A-83 was estimated to be the strongest antagonist against P. capsici. The A-183 strain was identified as a strain of Pseudomonas sp., showing the maximum antifungal activity, when cultured at $30^{\circ}C$ for 5 days in the potato extract medium(pH 6.5) containing 2.0% mannitol and 0.3% peptone.

  • A bacterial strain, producing extracellular inulin fructotransferase which converts inulin into di-D-fructofuranose 1,2':2,3' dianhydride(DFA III), was isolated from soil and presumed as Bacillus sp.. The highest production of the enzyme was obtained by using medium containing Jerusalem artichoke extract as carbon source, peptone as organic nitrogen source, and $NH_4H_2PO_4$, as inorganic source. Under optimum condition, the enzyme activity of the culture broth supernatant reached maximal 2.61 units/ml after cultivation for 45 hrs.

  • A cyclodextrin glycosyltransferase-producing bacterium was newly isolated from soil using alkaline pH medium containing 1% $Na_2CO_3$. The isolated strain was identified as Bacillus brevis by morphological and biochemical characteristics, and fatty acid composition and designated Bacillus brevis CD162. The strain showed the best enzyme production of 0.9 unit/ml after 96 hrs of culture at $30^{\circ}C$ in a medium of 2.0% soluble starch, 0.75% yeast extract, 0.5% bacto peptone, 0.2% $K_2HPO_4$ 0.05% $MgSO_4{\cdot}7H_2O$, and 1.5% $Na_2CO_3$ at initial pH 10.2.

  • To reducing offensive odor form compost UIW-6 mutant obtained by UV treatment from sulfur-oxidizing bacteria, Thiobacillus sp. IW. The UIW-6 mutant was found 1.6 times faster in growth than the parent strain on thiosulfate medium (TM) at 36 h after incubation. Initial pH, temperature and agitation for the optimum growth of UIW-6 were 6.5, $35^{\circ}C$ and 200 rpm, respectively. The UIW-6 mutant growth was two times higher than parent strain at 6 h culture in TM liquid medium containing 50 mM sodium thiosulfate. The UIW-6 mutant used fructose and sucrose as carbon sources and yeast extract> tryptone> peptone as nitrogen ones. It was found that the growth of UIW-6 was increased in addition of 0.2% yeast extract.

  • Nutritional characteristics of mycelial growth and fruit-body formation of Flammulina velutipes in synthetic media were investigated. The results are summarized as follows: 1. Among sugar substances, mannitol gave rapid mycelial growth and formation of fruit-body with higher yield, and among organic acids only succinic acid showed the fruit-body but poorly. Ethyl alcohol and glycerol comparatively promoted the formation of the fruit-body with higher yield. 2. Among nitrogen sources, peptone resulted in rapid mycelial growth and fruit-body formation with higher yield. and among amino acids glycine gave fast fruit-body formation with higher yield. However. nitrite nitrogen, lysine and methionine showed no mycelial growth at all. 3. The concentration of substances tended to affect the fruit-body formation and yield. Lower concentration of mannitol and peptone prompted fruit-body formation but with low yields, and higher concentration delayed the fruit-body formation with increased yields.

Isolation and production of soymilk-clotting enzymes from Bacillus sp. K-324-7 (대두유 응고효소 생산에 관한 연구)

  • A bacterial strain which was capable of producing extracellular soymilk-clotting enzyme was isolated from soil samples during the course of screening test. The characteristics of the isolated strain K-324-7, indicated that the strain belonged to species of Bacillus cereus. The crude purification of this enzyme was precipitated by salting out with ammonium sulfate of 0.8 saturation. The optimal pH for the enzyme activity was at $6.1{\sim}7.0$ and below $50^{\circ}C$. The optimal culture medium for the production of soymilk-clotting enzyme were consisted of 0.2% glucose, 0.2% peptone, and 0.5% $KH_2PO_4$ with initial pH value of 6.5. The activity of enzyme was maximum when the microbe was cultured for 3 days at $35^{\circ}C$.

  • In order to increase the production and secretion rate of mouse salivary $\alpha$-amylase from Saccharomyces cerevisiae, various experiments were attempted. A plasmid pCNNinv (AMY) was constructed by the substitution of ADCl promoter and native signal sequence of mouse salivary $\alpha$-amylase cDNA gene with PRBI promoter and yeast invertase leader sequence, which resulted in 25% increase in the production of $\alpha$-amylase in the culture medium. The respiratory deficient transformant carrying pCNNinv (AMY) were obtained by treating yeast cells with ethidium bromide, and the $\alpha$-amylase activities in the culture brothes of the respiratory-deficient transformants were 5-8 times higher than that of parental wild type strain. $\alpha$-Amylase activity was also increased 3 times when the 0.015% (w/v) of 2-mercaptoethanol was added to the culture medium.

A STUDY ON THE MYCELIA CUL TURE AND GENETIC CHARACTERISTICS IN Lepista nuda (민자주방망이버섯(Lepista nuda)의 균사배양 및 유전적 특성에 관한 연구)

  • This study was carried out to investigate the cultural characteristics and the genetic characteristics in Lepists nuda. The mycelia cultural characteristics which include the specificity for pH, carbon source, nitrogen source and medium were studied using petridish culture and liquid media culture respectively. The genetic characteristics were also investigated using random amplified polymorphic DNA (RAPD) analysis. To find out the mycelial growth rate by various medium, mycelia was cultured for 25 days in PDA, YM, MEA, GPB and Yamanaka. The Yamanaka was superior to the other media in supporting the mycelial growth. pH6 produced the best result in the test of an optimal pH. In experiment for optimal carbon source, starch showa 46.8$\pm$1.7mm of diameter of mycelial colony as the best and for nitrogen source, yeast extract showed a good effect as well. In investigation for genetic characteristics, Lepists nuda was amplified by 7 primers among 10 primers and Pleurotus ostreatus was also amplified by 7 primers. From the RAPD analysis between species, the band patterns were different.
