Assessment of Anti-nutritive Activity of Tannins in Tea By-products Based on In vitro Rumen Fermentation

  • Nutritive values of green and black tea by-products and anti-nutritive activity of their tannins were evaluated in an in vitro rumen fermentation using various molecular weights of polyethylene glycols (PEG), polyvinyl pyrrolidone (PVP) and polyvinyl polypyrrolidone as tannin-binding agents. Significant improvement in gas production by addition of PEG4000, 6000 and 20000 and PVP was observed only from black tea by-product, but not from green tea by-product. All tannin binding agents increased $NH_3$-N concentration from both green and black tea by-products in the fermentation medium, and the PEG6000 and 20000 showed relatively higher improvement in the $NH_3$-N concentration. The PEG6000 and 20000 also improved in vitro organic matter digestibility and metabolizable energy contents of both tea by-products. It was concluded that high molecular PEG would be suitable to assess the suppressive activity of tannins in tea by-products by in vitro fermentation. Higher responses to gas production and $NH_3$-N concentration from black tea by-product than green tea by-product due to PEG indicate that tannins in black tea by-product could suppress rumen fermentation more strongly than that in green tea by-product.

The Effect of Operating Conditions on Cross-Flow Ultrafiltration with using Polyethylene Glycol (Polyethylene Glycol을 이용한 Cross-Flow Ultrafiltration에 있어서 운전조건의 영향)

  • The objective of this study was to investigate the effect of running time, operating pressure, feed concentration and circulation rate on the permeation flux and the rejection rate in cross-flow ultrafiltration of polyethylene glycol(PEG) solution of molecular weight($M_w$) 8000 and 20000. The membranes used for this study were MWCO(Molecular Weight Cut-off) of 6 K and 20 K. The experiments were performed at the operating pressures of 7, 14 and 28 psi, the circulation rates of 1000 mL/min and 2000 mL/min, and the feed concentration of 100 mg/L and 1000 mg/L. At a constant pressure, the permeation flux and the observed rejection($R_o$) appeared to be approximately constant within the range of running time, 0~480 min. The permeation flux increased with increasing the operating pressure, and it increased with decreasing the feed concentration and decreasing Mw of PEG at a given pressure. On the other hand, $R_o$ decreased slightly with increasing the operating pressure. However, $R_o$ increased with increasing the feed concentration and increasing of $M_w$ of PEG at a given pressure. The variation in circulation rates did not cause any significant influence on the permeation flux. Increasing of circulation rate caused the increase of $R_o$, and $\alpha$ was increased substantially with the decrease of $M_w$ of PEG. The dimensionless parameter. permeability ratio($\alpha$), which was used to investigate flux-pressure behavior, was increased with the increase in circulation rate and operating presure. The value of $\alpha$ was less than 1 in all cases. The estimated intrinsic rejection(R). which was obtained from mass transfer coefficient, was decreased with the increase of operating pressure. However R increased with the increase of linear velocity of feed and $M_w$ of PEG.

Process Development for Alcohol Production by Extractive Fermentation (추출 발효에 의한 알콜 제조 공정개발 -PEG/Dx 최적 이상계의 선정-)

  • The quantitative effects of molecular weight and concentrations of two phase-forming polymers-polyethylene glycol and crude dextran on the two phase extractive ethanol fermentation were investigated using a Box-Wilson central composite protocol. The regression model obtained was used in order to determine optimum compositions of aqueous two phase system. In the aqueous two phase extractive ethanol fermentation of Kluyueromyces fragilis CBS 1555 with Jerusalem artichoke juice, it was found from the regression model that the variables influenlcing on ethanol fermentation were PEG concentration, time, Dx concentration, and PEG molecular weight strongly in order. The interaction of PEG concentration and PEG molecular weight was also found, and the effect of PEG concentration decreased with increase in molecular weight of PEG. The ethanol concentration incresed with increase in molecular weight of PEG, and with decrease in concentration of PEG. In conolusion, maximum concentration of ethanol produced was obtained at the following compositions; PEG MW 20000, Dx concentration ranged from 4% to 5%, and PEG concentration ranged from 3% to 7%.

  • The removal properties of Polyethylene glycols (PEGs) known as the important group of synthetic polymers of ethylene oxide were examined by the bio-reactor equipped with a mesh filter module. PEG-1000 and PEG-2000 were fairly removed on the basis of TOC, which were 75.1% and 51.6%, respectively. In the case of PEG-20000, the removal efficiency of TOC was less than 15.2% and the favorable acclimation of microbes was not obtained. It was suggested that this system could effectively maintain microbes for the biodegradation of low molecular weight of PEG and TOC removal was significantly influenced by PEG molecular weight.

Extractive Ethanol Fermentation Characteristics of K.fragilis in an Aqueous Two Phase System (수성이상계에서 K.fragilis의 에탄올 추출발효 특성에 관한 연구)

  • Fermentation characteristics of Kluyveromyces fragilis CBS 1555 with Jerusalem artichoke juice, in extractive ethanol fermentation in aqueous two phase systems composed of polyethylene glycol 20000 (PEG) and crude dextran(Dx), were investigated as a function of initial sugar concentrations, concentrations of ethanol formed, or fermentation time. Both specific ethanol production rate increased with decrease in concentrations of PEG and Dx in two-phase systems. Without being related to the compositions of aqueous two-phase system, maximum specific cell growth rate and maximum specific ethanol production rate were showed in the initial sugar concentration fo $80g/\ell$ and $120g/\ell$, respectively. The inhibition effects of ethanol on specific cell growth rate and specific ethanol production rate decreased with decrease in PEG concentration and in the range of 2.5 to 5% Dx. Specific cell growth rate and specific ethanol production rate was fitted as an exponential function and a hyperbolic function, respectively, of the concentrations of ethanol formed. Overall ethanol productivity increased with increase in initial sugar concentrations, and also the required time for the maximum productivity was so. Ethanol production rate by the elapsed fermentation time showed the maximum value in the initial sugar concentration of $160g/\ell$.

Optimization of Conditions for Extractive Ethanol Fermentation in an Aqueous Two Phase System (수성이상계 에탄올 추출발효 조건의 최적화에 관한 연구)

  • This study was undertaken with objective of optimizing the conditions of fermentation in an aqueous two-phase system which is composed of polyethylene glycol (PEG) 20000 and crude dextran (Dx). The data were obtained and analyzed using the Box-Wilson's experimental design protocol and the response surface methodology. To reach this end a multilinear polynomial regres- sion model was developed, which can be utilized for the purpose of optimizing the extractive fermentation. Optimum conditions for batch fermentation with aqueous two phase system were found to be at 4.2~5.4% PEG/3.2~4.2% Dx range. The composition of the center was 4.8% PEG/ 3.6% Dx. Optimum operating conditions for initial sugar concentration and fermentation time were approximately 160 g/l, and 21~22 hr, respectively. Fermentation in the aqueous two phase system composed of 5% PEG/4% Dx showed increase of 23% in ethanol concentration, of 9.5% in ethanol yield, and of 19% in ethanol productivity as compared to the case of fermentation of neat Jerusalem artichoke juice.

Fluorescence Microscopy of Condensed DNA Conformations of Bacterial Cells

  • Cellular DNA in prokaryotes is organized in nucleic acid-protein self-assemblies referred to as the nucleoid. The physical forces responsible for its stability inside the poor solvent properties of the cytoplasm and their functional implications are not understood. Studies on the organisation and functioning of the cytosol of cells largely rely on experimental protocols performed in highly dilute solutions using biochemically purified molecules, which is not a reliable substitute for the situation existing in vivo. Our current research interest is focused on the characterization of biological and physical forces determining the compaction and phase separation of DNA in Escherichia coli cytoplasm. We have emphasized the effect of excluded volume in solutions with high macromolecular concentrations (macromolecular crowding) upon self-association patterns of reactions. The prokaryotic cytosol was simulated by addition of inert polymer polyethylene glycol (PEG) (average molecular weight 20000), as an agent which afterwards facilitates the self-association of macromolecules. Fluorescence microscopy was used for direct visualization of nucleoids in intact cells, after staining with DAPI (4',6-diamidino-2-phenylindole dihydrochloride). Addition of the crowding agent PEG 20,000, in increasing concentrations generated progressively enhanced nucleoid compaction, the effect being stronger in the presence of 0.2 M NaCl and 5 mM MgCl$\_$2/. Under these conditions, the nucleoids were compacted to volumes of around 2 ㎛$\^$3/ or comparable sizes with that of living cells.

  • The effects of paste components on the properties of porous $TiO_2$ film electrodes prepared through screen-printing technique were investigated in order to efficiently control and optimize the main fabrication step of the dye-sensitized solar cells (DSC). The screen-printed porous $TiO_2$ films were characterized by ultraviolet-visible (UV-Vis) spectroscopy and scanning electron microscopy (SEM), and applied as a part of the DSC for the energy conversion. The fabricated DSC were evaluated by a solar simulator. The experimental results indicate that the microstructural characteristics of the printed films and the performances of the DSC are dependent on the paste compositions. As a result that the efficiency of DSC prepared by manufactured paste was 0.5%~1% higher than existing paste.
