원형질체 융합에 사용할 genetic selection marker로써 자외선 조사와 NTG 처리에 의해 Brevibacteriurn lactofermeηturn S SWA(arg trp)와 B. lactofermenturn SWB(met ser)를 분리하였다. 이 균주릎 배양하여 증식기에 penicillin G를 처리한 후, 0.4 M sucrose를 첨가한 lysis fluid에서 lysozyme을 $400\mu\textrm{g}$/ml의 농도로 16시간 처리 하였을 때, B. lactofermenturn SWA는 99.98%, B. lactofermentum SWB는 99.93%의 원형질체 형성률을 나타내었다. 윈형질체 융합에서는 100 mM $CaCl_{2}$, 30%의 PEG 6,000과 fusion fluid릎 15분간 처리하였을때, B. lactofermentum SWA와 B. lactofermentum SWB의 융합빈도는 $2.30\times 10^{-5}$로 나타내었다. 원형질체 융합에 의해 언어진 융합균주의 유전적 분석을 위해 selective media와 non-selective media에서 생장한 colony를 계산함으로써 각 marker간의 재조합주의 형성빈도를 조사하였으며, 이들을 분석하여 유전자간의 순서를 결정하였다.
본 연구에서는 동물세포에서 매우 높은 독성을 갖는 유기인 살충제 coumaphos를 생분해하기 위하여, 재조합 대장균으로부터 발현된 organophosphorus hydrolase (OPH)가 사용되었다. OPH 생산성을 향상시키고자, 균주의 배지에 비이온성계면활성제(Tween 20, PEG 1000)와 유기용매(glycerol, propanol, ethanol)에 대한 효과를 고찰하였다. 0.25% Tween 20과 0.5% glycerol를 배지에 첨가하였을 때, 최대의 OPH가 생산되었다. 초음파를 이용한 균주 파쇄 공정으로부터 생성된 OPH를 사용 하였을 때, 0.2, 0.5, 1.0, 2.0 mM coumaphos는 각각 100, 88, 84, 78%의 생분해효율을 나타내었다. 이 연구에서 얻어진 결과들은 coumaphos가 다양한 농도로 오염된 지역을 생물학적으로 처리할 수 있는 새로운 방법으로서 활용될 수 있을 것이다.
아닐린분해능이 있는 Flavimonas oryzihabitans과 4-chlorobiphenyl (4-CBP)를 분해하는 Pseudomonas sp.를 원형질체 융합하여 아닐린과 4-CBP를 모두 분해하는 융합균주를 개발하고, 융합체의 특성을 조사하였다. F. oryzibhavitans는 융합체 선별표지 위애 NTG처리하여 chloramphenicol 내성 ($Cm^r$)을 유발하였다. Lysozyme-EDTA에 의한 각 모균주의 원형질체 생성율은 약 99%이었고, 원형질체의 재생율은 5.0~6.6%이었다. 원형질체는 40% PEG 6000을 사용하였을 때 효과적으로 융합이 이루어졌으며 융합율은 $3.16{\times}10^{-4} $이었다. 융합균인 F22의 DNA양는 모균주에 비해 약 2배 정도 증가 하였으며, 생화학적 특성은 모균주의 특성이 혼합하여 나타났다. F22는 5 mM 아닐린 최소배지에서는 모균주와 성장능과 분해능이 거의 비슷하였고, 10 mM 아닐린 최소배지에서는 1.5배 빠른 성장속도를 나타내었으나, 4-CBP 분해능은 모균주보다 약간 낮게 나타났다.
폴리에틸렌글리콜(PEGs)과 수산화칼륨(KOH)을 이용하여 서로 다른 온도와 시간의 반응조건에 따라 PBDEs의 화학반응을 수행하였다. PBDEs의 제거효율은 화학반응 전과 후의 농도의 차이로 측정하였다. PBDEs는 25와 $50^{\circ}C$의 낮은 온도에서는 제거되지 않았다. 하지만 온도를 증가시킴에 따라, 바이페닐기에 붙어있는 5-6개의 브롬이 치환되어 있는 PBDEs의 완전제거를 보이면서 ${\sigma}$-xylene에 있는 PBDEs의 제거효율은 점차적으로 증가하였다. 반응조건을 4시간과 $150^{\circ}C$까지 증가시켰을 때 PBDEs의 제거효율은 거의 100%에 도달하였다. 이와 같이 PEGs와 PBDEs의 화학반응 연구를 통해, PBDEs가 PEGs에 의해 브롬이 순차적으로 제거되는 진행에 의해서 탈브롬화 된다는 것을 확인할 수 있었다.
식물원형질체를 이용하여 체세포잡종을 육성하기 기초자료를 얻고자 식물의 엽육부위, Callus에서 효과적인 원형질체의 분리, 적합한 배양조건 및 세포융합에 관한 조건을 조사한 결과를 요약하면 다음과 같다. 1. 배양 융합조건에 적합한 식물원형질체의 분리 방법은 여러 종류의 효소농도에 따라 다르지만 담배 및 완두엽육 원형질체분리에는 Macerozyme 0.5%, Cellulase 2.0%, KDS 0.5%, Mannitol 0.7M의 효소용액에 처리시 활성 있는 원형질체를 얻을 수 있었다. 2. 배양중인 담배 및 대두의 Callus로부터 원형질체분리는 배양기간에 따라 세포활성도가 떨어지거나 쉽게 파괴되는 현상을 보여 양호한 원형질체를 얻을 수가 없었다. 3. 원형질체의 배양은 배양배지에 Plating할 경우 세포농도에 따라 차이가 있었으며 1.0$\times$$10^4$/ml이상이어야 분열이 가능하였고 배양조건은 150Lux 12시간 광암처리후가 세포분열 및 증식이 가장 왕성하였다. 4. 세포융합은 PEG농도가 중요하며 PEG 1500 0.33M에서 CaCl_2농도가 높을수록 융합율이 증가되는 경향이었다.
양배추와 무로부터 원형질체를 분리하였고 PEG 처리를 통하여 융합을 하였다. 고농도의 원형질체 혼합물로부터 많은 미소괴를 관찰할 수 있었다. 미소괴는 정상적인 캘러스로 자랐고 이들 캘러스로부터 총 218개의 신초가 재분화 되었다. 재분화된 개체는 순화 과정을 거처 온실에서 재배하였다. 순화된 208개체를 대상으로 마커 검정을 통하여 융합 여부를 확인한 결과, 모두 무의 NWB-CMS에 특이적인 PCR 산물을 확인할 수 없었다. 그러나 ISSR분석을 통하여 208개체 중 3개체에서 양배추와 무의 세포가 융합됨을 확인하였다. 이를 통하여 원형질체의 융합이 성공적으로 일어났음을 증명할 수 있었다. 세포융합이 일어난 3개체는 모두 양배추와 무의 형질을 보이는 중간적인 형태를 가지고 있었다. 이들은 춘화처리를 거쳐 모두 개화 되었으나 꽃의 색깔이 무의 형질과 같은 흰색이었고 이중 한 개체에서만 역교배를 통하여 3개의 종자를 얻었다.
경산 임당유적에서 출토된 목제 갑옷틀은 수침된 상태로 출토된 목제유물로 재질이 매우 취약하여, 대기 중에 노출되면 급속도로 건조가 진행되어 곧바로 수축 변형이 일어나 유물로써 가치를 잃어버리게 된다. 특히 목제 갑옷틀은 오랜 매장기간동안 목질부의 분해가 많이 진행되어 재질이 취약할 뿐 아니라 원통목을 가공한 것이어서 건조 과정에서 수축변형이 발생우려가 높다. 따라서 목제 갑옷틀은 건조변형이 적고 치수안정화가 우수한 처리법을 적용하여야 한다. 따라서 목제 갑옷틀은 t-butanol 용액을 용제로 PEG#4000 40%로 함침 전처리 후 진공 동결건조법으로 보존처리 하였다.

Fouling characteristics of humic substances on tight ultrafiltration (UF) membrane have been investigated. The tight UF membrane was prepared by blending polysulfone (PSf) in N.N-dimethylacetamide (DMAc) with 25%wt of Polyethylene glycol (PEG400) and 4%wt of acetone. Fouling characteristic of the modified PSf membrane was observed during peat water filtration in different trans-membrane pressure (TMP). It was found that the acetone modified membrane provided 13% increase in TMP during five hours of peat water filtration, where a stable flux was reached within 150 minutes. Meanwhile, the increase of TMP from 10 psig to 30 psig resulted in a fouling resistance enhancement of 60%. Furthermore, based on the fouling analysis, fouling mechanism at the first phase of filtration was attributed to intermediate blocking while the second phase was cake formation.
ABA type block copolymers composed of poly($\gamma-benzyl$ L-glutamate) (PBLG) as the A component and poly (ethylene glycol) as the B component were obtained by polymerization of $\gamma-benzyl$ L-gletamate N -carboxyanhydride, initiated by amino groups at both ends of poly(ethylene glycol) . From circular dichroism measurements in ethylene dichloride solution as well as from infrared spectTa measurements in solid state, it was found that the polypep- tide block exists in the a-helical conformation, as in PBLG homopolymer. $Poly-N^5$ (3-hydroxypropyl glutamine) (PHPG)/poly(ethylene glycol)block copolymer hydrogel was obtained by the treatment of PBLG/PBG block copolymer with the mixture of 3-ammine-1-propanol and diamlnooctane. The water content of PHPG/PEG block copolymer hydrogel was about 80wt% when the concentration of crosslinking agent was below 5 mole % per polymer.
Field studies were conducted over two seasons during the summers of 1997 and 1998 to investigate the effects of different spatial arrangements(random or highly aggregated) of sclerotia of Sclerotinia sclerotiorum and alginate pellet types(bran or polyethylene glycol) on colonization of sclerotia by Trichoderma spp. Treatment with alginate pellets increased the mean percentages of sclerotia colonized by Trichoderma spp. in both years. Distribution patterns of sclerotia affected the mean percentage of sclerotia colonized by Trichoderma spp. in both years, indicating that a highly aggregated distribution of sclerotia was more favorable to colonization by Trichoderma spp. The effects of the different pellet types(bran or PEG) were not siginificant in both years(P>0.05). The application of higher densities(200 pellets per 1 $m^2$) of alginate pellets resulted in higher mean percentages of sclerotia colonized by Trichoderma spp. in 1998(P<0.05), but did not in 1997.
