Drug evaluation based on phosphomimetic PDHA1 reveals the complexity of activity-related cell death in A549 non-small cell lung cancer cells

  • Cancer cells predominantly generate energy via glycolysis, even in the presence of oxygen, to support abnormal cell proliferation. Suppression of PDHA1 by PDK1 prevents the conversion of cytoplasmic pyruvate into Acetyl-CoA. Several PDK inhibitors have been identified, but their clinical applications have not been successful for unclear reasons. In this study, endogenous PDHA1 in A549 cells was silenced by the CRISPR/Cas9 system, and PDHA1WT and PDHA13SD were transduced. Since PDHA13SD cannot be phosphorylated by PDKs, it was used to evaluate the specific activity of PDK inhibitors. This study highlights that PDHA1WT and PDHA13SD A549 cells can be used as a cell-based PDK inhibitor-distinction system to examine the relationship between PDH activity and cell death by established PDK inhibitors. Leelamine, huzhangoside A and otobaphenol induced PDH activity-dependent apoptosis, whereas AZD7545, VER-246608 and DCA effectively enhanced PDHA1 activity but little toxic to cancer cells. Furthermore, the activity of phosphomimetic PDHA1 revealed the complexity of its regulation, which requires further in-depth investigation.

  • The existed transmissions have PDH and SDH transmission system, which are based on PSTN network. But as the future ATM network is going to be constructed, it is very necessary for ATM network to interwork with PSTN for telephony service. The international standardization organizations like ITU-T, ATM Forum offered CES, VTOA1, VTOA2 and so on for interworking with PSTN service, which spend many network resources and considerable transmission delay. So in this paper, new interworking PSTN for transmission will be proposed to lessen the transmission delay and the network resources.

  • Effect of red ginseng treatment on experimentally induced hyperkeratosis was investigated by light microscopic observation scanning electron microxcope (SEM) examination, epidermal enzyme activities nd lipid contents. Both light microscopic observation and SEM examination showed that hexadecane induced epidermal hyperplasia, hypertrophy and hyperkeratosis by increasing the numbers as well as the sizes of epidermal cells including desquamating horny cells. The superficial horny cells were protruded around the base of hair shaft. Among red ginseng components, only saponin treatment inhibited epidermal hyperplasia and hyperkeratosis by reducing the thickness of epidermis and arranging the cornified cells. Saponin from korean red ginseng inhibited abnormally increased epidermal LDH, ICD and G6PDH activities and reduced the contents of epidermal lipids induced by hexadecane. It seems that red ginseng saponin has preventive effect on experimental hyperkeratosis possibly by controlling the enzyme activities involved in epidermal cellular metabolism, resulting in reduced amounts of abnormal epidermal lipids.

First Report of Gray Mold Disease of Sponge Gourd (Luffa cylindrica) Caused by Botrytis cinerea in Korea

  • In October 2014, an occurrence of gray mold was observed on young fruits of sponge gourd (Luffa cylindrica) in Sachunmun, Gangneung, South Korea. Symptoms included abundant mycelia growth with gray conidia on young fruits and finally rotting the fruits. The fungus was isolated from symptomatic fruits and its pathogenicity was confirmed. Based on the morphological features and sequence analysis of ITS-5.8S rDNA, G3PDH, HSP60, and RPB2 genes, the pathogen was identified as Botrytis cinerea Pers. This is the first report of gray mold caused by B. cinerea on L. cylindrica in Korea.

  • 인터넷 사용자의 급격한 증가로 인해 사용자들은 더욱 다양한 인터넷 서비스들을 제공받기를 원하고 있다. 사용자들의 다양한 요구를 원활히 충족시켜주기 위하여 인터넷 서버의 성능은 날로 증대되고 있으나 서버의 관리는 네트워크의 크기가 증대될수록 점점 더 어려워지고 있다. 또한 기존의 인터넷 서버 시장은 유닉스 계약의 서버가 우위를 차지하고 있었지만, 최근 개인 PC 서버의 증가로 인해 윈도 NT 서버가 전체 서버 시장에서 차지하는 비율이 커지고 있다. 본 논문은 유닉스 서버에 비해 상대적으로 연구가 미약했던 윈도 NT 서버를 관리할 수 있는 모니터링 도구를 제시한다. 순간적으로 변화하는 네트워크 상황에 적응하기 위하여 윈도 NT 서버를 탐지할 수 있는 네트워크 디스커버리 기법을 접목함으로써 관리 대상 서버의 선택을 손쉽게 하고자 하였고, PDH 라이브러리를 사용하여 윈도 NT 서버의 관리를 보다 효율적으로 수행하였다.

First Report of Leaf Spot of Datura metel Caused by Alternaria tenuissima in Korea

  • In June 2013, we collected leaf spot disease samples of Datura metel from Gangneung, Gangwon Province, Korea. The symptoms observed were small circular to oval dark brown spots with irregular in shape or remained circular with concentric rings. We isolated the pathogen from infected leaves and cultured the fungus on potato dextrose agar. We examined the fungus morphologically and confirmed its pathogenicity according to Koch's postulates. The results of morphological examinations, pathogenicity tests, and the rDNA sequences of the internal transcribed spacer regions (ITS1 and ITS4), glycerol-3-phosphate dehydrogenase (G3PDH) and the RNA polymerase II second largest subunit (RPB2) gene sequence revealed that the causal agent was Alternaria tenuissima. To the best of our knowledge, this is the first report of leaf spot of D. metel caused by A. tenuissima in Korea as well as worldwide.

저근백피 에탄올 추출물의 혈당강하 작용에 대한 연구 (Antidiabetic Study of ethanol extract on Ailanthi cortex radici)

  • This study was performed to investigate the antidiabetic study effect in ethanol extract of Ailanthicortex(A.C) radici in Streptozotocin(STZ)-induced diabetic rats. Diabetes was induced by intravenous injection of STZ at a dose of 45mg/kg dissolved in citrate buffer. The ethanol extract of A.C radici was orally administrated once a day for 7 days at a dose of 500mg/kg. The content of serum glucose, triglyceride(T.G), total cholesterol were significantly decreased in treated group compared to the those of STZ-control group. The content of hepatic glycogen and activity of glucokinase(GK) were significantly increased in treated group compared to the those of STZ-control group. but activity of glucose-6-phosphate dehydrogenase(G-6-PDH) was significantly increased. These results indicated that ethanol extract of A.C would have antidiabetic effect in STZ-induced diabetic rats.

EPON 장애 대책에 관한 연구 (A Study on the Protection of EPON)

  • 최근까지 통신망의 장애대책은 주로 기간망에 적용되는 개념이었다. 즉, SONET, SDH 등 동기식 전송장치의 점대점, 선형, 링형, 메쉬형 구성 혹은 565 Mbps의 비동기식 전송장치 등이 그 예이다. 그러나 대용량, 광대역 PON 시스템의 도입으로 인해 가입자망에도 보호 및 절체의 필요성이 대두되고 있다. 본 논문에서는 EPON의 경우에 효과적으로 망을 보호할 수 있는 방법에 대해 기술하였다. EPON프레임 및 프로토콜 특성을 이용한 보호절체 방법을 제시하고 이를 구현하여 1ms 이내에 시스템을 보호하는 실험 결과를 보인다.

  • 트래픽의 급격한 증가와 함께 백본을 구성하는 광전송망은 비동기식(PDH) 기반에서동기식(SDH/SONET) 기반으로 발전하여 왔으며, 최근에는 파장분할다중화(WDM) 기술을 적용한 장비가 보급되면서 광전송망을 구성하는 필수 기술로 자리잡고 있다. WDM 기술 적용에 따라 현존하는 SDH/SONET 망과 연동시 유지보수가 되지 않는 등WDM 기술에 SDH/SONET 망을 직접 수용하는 데 어려움이 있어, 이를 해소할 수 있는 새로운 개념의 광전송계위(OTH)의 필요성이 대두되어 관련 기술을 개발하여 표준화가 이루어졌다. OTH 기술의 도입으로 WDM 기반 전송망과 기존 전송망과의 연동이 원활하게 이루어져 유지보수 및 운영상의 문제점을 해소할 수 있을 뿐만 아니라, 기존의 다양한 신호 트래픽과 새로운 서비스 등장에 따른 트래픽 수용 및 전송이 가능하게되었다. 이 논문에서는 WDM 기반의 고속 전송에 필수적인 OTH 신호 전송을 위한 핵심 기술에 관해 분석하고 나아가 관련 기술개발 동향에 대해 기술한다.

하수오 추출물의 혈당강하작용 (Antidiabetic Effect of Ha-Su-O(polygoni radix))
