A STUDY OF REGENERATION ENHANCEMENT OF DESTRUCTED PERIODONTAL TISSUE (파괴된 치주조직의 재생촉진에 관한 연구)

  • In order to evaluate the effect of platelet-derived growth factor(PDGF-BB) and guided tissue regeneration(GTR) technique on the regeneration of destructed periodontal tissue,intentional through-and-through furcation defects(4mm in height) were made on both mandibular 2nd and 4th premolars of 8 adult male dogs(30-40lb). Experimental group 1 was composed of the premolars that were treated by only topical application of PDGF-BB with 0.05M acetic acid without any barrier membrane. Experimental group 2 was composed of the premolars that were treated by GTR with expanded polytetrafluoroethylene membrane(ePTFE : Gore-tex periodontal material, USA). Experimental group 3 was composed of the premolars that were treated by GTR with ePTFE after topical application of PDGFBE. Control group was composed of the premolars that were treated by coronally positioned flap operation only without use of PDGF-BB and ePTFE membrane. All ePTFE membranes were carefully removed 4 weeks after regenerative surgery, and all experimental animals were sacrificed 8 weeks after regenerative surgery. The light microscopic findings were as follows ; (1) In experimental group 1, rapid new bone formation along the-root surface with multiple ankylosis and root resorption by multinucleated giant cells, and dense connective tissue in the central portion of the furcation defects were observed. (2) In experimental group 2, it was observed that the furcation defects were filled with newly formed bone, Sharpey's fibers were embedded into new cementum on root dentin of furcation fornix area, but the central portion and the area under furcation fornix were still filled with dense connective tissue. (3) In experimental group 3, the furcation defects were regenerated with newly formed dense bone and regular periodontal ligament with Sharpey's fibers embedded into newly formed cementum and bone underneath fornix area. (4) In control group, unoccupied space, apical migration of epithelium, dense infiltration of inflammatory cells in subepithelial connective tissue in relation to heavy plaque accumulation, and root resorption by inflammatory reaction were shown, but any new cementum formation on resorbed dentin surface could not be observed. The present study demonstrated that the combined therapy of PDGF-BB and GTR could enhance the regeneration of destructed periodontal tissue.

Anti-apoptotic effects of autophagy via ROS regulation in microtubule-targeted and PDGF-stimulated vascular smooth muscle cells

  • Autophagy has been studied as a therapeutic strategy for cardiovascular diseases. However, insufficient studies have been reported concerning the influence of vascular smooth muscle cells (VSMCs) through autophagy regulation. The aim of the present study was to determine the effects of VSMCs on the regulation of autophagy under in vitro conditions similar to vascular status of the equipped micro-tubule target agent-eluting stent and increased release of platelet-derived growth factor-BB (PDGF-BB). Cell viability and proliferation were measured using MTT and cell counting assays. Immunofluorescence using an $anti-{\alpha}-tubulin$ antibody was performed to determine microtubule dynamic formation. Cell apoptosis was measured by cleavage of caspase-3 using western blot analysis, and by nuclear fragmentation using a fluorescence assay. Autophagy activity was assessed by microtubule-associated protein light chain 3-II (LC-II) using western blot analysis. Levels of intracellular reactive oxygen species (ROS) were measured using $H_2DCFDA$. The proliferation and viability of VSMCs were inhibited by microtubule regulation. Additionally, microtubule-regulated and PDGF-BB-stimulated VSMCs increased the cleavage of caspase-3 more than only the microtubule-regulated condition, similar to that of LC3-II, implying autophagy. Inhibitory autophagy of microtubule-regulated and PDGF-BB-stimulated VSMCs resulted in low viability. However, enhancement of autophagy maintained survival through the reduction of ROS. These results suggest that the apoptosis of conditioned VSMCs is decreased by the blocking generation of ROS via the promotion of autophagy, and proliferation is also inhibited. Thus, promoting autophagy as a therapeutic target for vascular restenosis and atherosclerosis may be a good strategy.

THE EFFECT OF PDGF-BB AND IGF-I COMBINATION ON THE HEALING OF ARTIFICIAL PERIAPICAL LESIONS IN BEAGLE DOGS (PDGF-BB와 IGF-I 혼합 투여가 비글견 인공 치근단 병소의 치유에 미치는 영향에 관한 연구)

  • It is difficult to treat the endodontic apical perforation successfully. In this study, we hypothesized that the application of PDGF-BB and IGF-I into periapical perforation site may accelerate periapical healing and lead to bone deposition. And the specificity of osteonectin in periapical healing was investigated. The experiments were performed on the upper and lower 51 premolar teeth of 4 beagle dogs. The pulp chamber of each tooth was opened and the dental plaque was inserted into the canal for developing the periapical lesion for 5 weeks. Then, the roots were artificially perforated at the apex with the number 4 profile of .06 taper. In each step, standard periapical radiographs were taken to compare the size of lesion each other. The radiographs were scanned and analyzed by image analysis system. The mean and standard deviation of periradicular radiolucency ratios were calculated in each group. ANOVA was used for comparison. 51 premolars were grouped into 3 groups; control group, calcium hydroxide-treated group and calcium hydroxide plus growth factors-treated group. In the control group, the apical perforations were not sealed and obturated with gutta-percha and ZOE sealer by lateral condensation technique. In the experimental groups, the apical perforation were sealed with calcium hydroxide and with/without $4{\mu}g$ of PDGF-BB & IGF-I in cellulose gel and obturated by lateral condensation technique. Fluorescent bone markers were used to measure new bone formation. Following 2, 4, 12 weeks after experiment the dogs were sacrificed and histologic sections were prepared. Each tooth block including periapical lesion was sectioned mesiodistally. One half of the sections were decalcified with 6% nitric acid and processed by standard paraffin embedding technique. The sections were stained by hematoxylin and eosin, and immunostained for osteonectin. Histomorphometrical measurement of neoformed bone was performed using a light microscope. And the other half of the sections were prepared by undecalcified preparation, and confocal laser scanning microscopic investigations were done.

The Effect of Platelet Derived Growth Factor on the Myocardial Revascularization in Implantation of Rabbit Internal Mammary Artery (토끼 내유동맥의 심근이식에서 혈관재형성 촉진에 대한 혈소판 유래 성장인자의 효과)

  • Background: Coronary artery bypases grafting in the old aged is associated with high mortality and morbidity, and it is difficult to perform if the coronary artery is diffusely disease. Recently it has been known that platelet derived growth factor(PDGF), especially PDGF-BB, stimulates angiogenesis. Material and Method: New Zealand white rabbit were used. In an attempt to achieve effevtive cardiac revasculatrization without vascular anastmosis, we divided into three groups(group I : Left anterior descending artery(LAD) was occluded by ligature, group II : Bilateral internal mammary vascular pedicles were dissected and implanted into myocardium, group III : The vascular pedicles were implanted into myocardium and PDGF-BB was injected into the myocardial tissue). Two weeks after IMA implantation, the proximal region of implanted LAD was ligated. Four days after LAD ligation angiogram, triphenyl tetrazolium chloride(TTD) staining and hematoxylin eosin staining were performed. Result: 1. Survival rate in group II was significantly higher than that in group I (P<0.05), and survival rate in group III was signficantly higher than that in group II(53% vs 93%, P<0.01). 2. There were significant differences in the ratio of area of necrosis to area at risk between group I and group II, and between group II and group III (P<0.01). 3. Microangiogram for angiogenic response revealed wide area of extensive revascularization with patent vessels in group III. 4. Histologic findings of three groups showed that polymorphonuclear leukocyte infiltration was minimal in group II and none in group III. Conclusion: PDGF-BB can establish functinal cardiac revasculatization through systemic vessels implanted directly into the myocardium.

A Study on the Expression of Glycosaminoglycans in the Experimental Tooth Movement of Rat and in Cultured Periodontal Ligament Cells (실험적 치아이동시 glycosaminoglycan의 발현에 관한 연구)

  • The purpose of this study was to evaluate 1) in vivo, the expression of chondroitin 4-sulfate (CH-4S), a structural element of glycosaminoglycans(GAGs), in periodontal tissue during the experimental movement of rat incisors, by labelled streptavidine biotin immunohistochemical staining for CH-4S, 2) In vitro, the expression of CH-4S in cultured human periodontal ligament(PDL) cells supplemented with 10ng/ml of $TGF-{\beta}_1$, 20ng/ml of PDGF-BB, 1ng/ml $TNF-\alpha$, or $1{\mu}g/ml$ LPS by western blot analysis. The results of this study were as follows ; 1. The expression of CH-4S was stronger in pulp, PDL, osteoblasts, osteoclasts and osteocytes in experimental group than in control group, but was rare in dentin, and cementum of experimental groups, regardless of the duration of force application, which was not different from that of control group. 2. In experimental group, the expression of CH-4S in pulp began to increase at 1 day after force application and got to the highest degree at 7 days. After 14 days, the expression in CH-4S immunoreactivity was decreased, and became similar to that of control group at 28 days. 3. The expression of CH-4S in PDL was noted in adjacent to alveolar bone. PDL showed higher intensity of immunolabelling after 1 day of orthodontic tooth movement. And the expression was more stronger in the tension side than that of pressure side of PDL at 1 day, but more stronger in the pressure side than that of tension side of PDL at 4 days. After 7 days, a decrease in CH-4S expression was observed. 4. The expression of CH-4S in alveolar bone got to the highest degree at 4 days, and At 7 days, a decrease in CH-4S expression was observed. 5. PDGF-BB notably raised the expression of CH-4S in the PDL cells at 3 days of cultivation 6. The expression of CH-4S of PDL cells was decreased with the application of $TNF-\alpha$ at 1 day. 7. Admixture of $TGF-{\beta}_1$ and PDGF-BB got more expression of CH-4S in PDL as compared to only $TGF-{\beta}1$ or PDGF-BB. A similar decrease of the expression of CH-4S was observed in the case of application of LPS or $TNF-\alpha$.

The Effects of PDGF-BB on the ALP Activity of MC3T3-E1 Cells (MC3T3-E1 세포의 ALP activity에 대한 PDGF-BB의 영향)

  • The ultimate aim of periodontal treatment is periodontal regeneration, which necessiates the regeneration of bone tissues. This paper investigated the effect of growth factor on bone cells. Platelet-derived growth factor(PDGF) is the one of the polypeptide growth factor that has been reported as a biological mediator which regulates activities of the cell proliferation, migration and metabolism of undifferentiated mesenchymal cells. The purpose of this study is to evaluate the effects of PDGF on bone nodule formation and ALP activity of MC3T3-El cells. Cells were seeded at $1{\times}10^5cells/well$ in alpha-modified eagle medium containing 10% fetal bovine serum, lOml beta-glycerophosphate and $50{\mu}g/ml$ of ascorbic acid. PDGF 0, 0.1, 1, 10 ng/ml were added to the cells at a confluent state and cultured for 3, 7, 14, 21, 28 days. We examined bone nodule formation and alkaline phosphatase activity. The results were as follows : There were bone nodule formation at day 21 both in control and all the experimental groups, and at day 28, all the experimental groups showed much more bone nodules than control groups. Compared to control-l group, ALP activity was increased in PDGF O.1ng/ml group and was decreased in 1,10ng/ml PDGF treated groups.{P< 0.05, P< 0.01) Compared to control-2, ALP activity was decreased in all the experimental groups except PDGF 0.1ng/ml in 21 day group. In the time-response effect, ALP activity was increased by the day 14 in all the experimental groups and thereafter ALP activity was decreased.(P<0.05, P< 0.01) In the dose-response effect, ALP activity was decreased as the dose of PDGF was increased, and after 21 day ALP activity was lowest in 1 ng/ml group, ALP activity was highest in the day 7 in control group and 0.1 ng/ml, 14 day experimental group. In conclusion, PDGF is considered more effective in the proliferation than differentiation of osteoblast-like cells, and it may be useful to study the combined effect of PDGF and other growth factors on osteoblast-like cells.

  • This study was to investigate generation of the specific neuronal cell in vitro from the neural progenitors derived from human embryonic stem (hES, MB03) cells. For the neural progenitor cell formation, we produced embryoid bodies (EB: for 5 days, without mitogen) from hES cells and then neurospheres (for 7-10 days, 20 ng/㎖ of bFGF added N2 medium) from EB. And then for the differentiation into neuronal cells, neural progenitor cells were cultured in N2 medium (without bFGF) supplemented with brain derived neurotrophic factor (BDNF, 5 ng/㎖) or platelet derived growth factor-bb (pDGF-bb, 20ng/㎖) for 2 weeks.   • Gynura procumbens (Lour.) Merr. has been used in some parts of Southeast Asia as a folk medicine to treat kidney diseases, diabetes mellitus, and hyperlipidemia. The present work was undertaken to prove the mechanisms of G. procumbens in the management of glomerular diseases. We investigated the effect of an aqueous extract of G. procumbens on cell proliferation, DNA synthesis, and the expressions of $TGF-{\beta}1$, PDGF-BB, CDK1, CDK2, and CDK4 in fetal bovine serum-activated human mesangial cells (MCs). The G. procumbens extract inhibited proliferation, DNA synthesis, expressions of PDGF-BB, CDK1, and CDK2 mRNA, and expression of $TGF-{\beta}1$ protein in MCs. The inhibitory effect of G. procumbens on MC proliferation may be mediated by suppression of PDGF-BB and $TGF-{\beta}1$ expressions and the modulation of CDK1 and CDK2 expression. Therefore, G. procumbens shows promise as an adjunct therapy in preventing progressive renal diseases.

Anti-inflammatory Effect of Portulacae Herba Water Extract on Lipopolysaccharide-activated RAW 264.7 Macrophages (마치현(馬齒莧) 물추출물의 항염효능에 관한 연구)

  • Objectives : The present study aimed to investigate the anti-inflammatory effects of the water extracts of Portulacae Herba (PH).Methods : We measured the effects of the water extracts of Portulacae Herba (PH) on the cell viability of mouse macrophage RAW 264.7 cells, the intracellular calcium production, and the proinflammatory mediators including nitric oxide (NO), vascular endothelial growth factor (VEGF), platelet-derived growth factor (PDGF)-BB, which are induced by the lipopolysaccharides (LPS), and obtained the results shown below.Results : After the cultivation of the PH extracts along with the mouse macrophages, the cell survival rate did not decrease with the MTT assay. However, the PH extracts did significantly suppress the production of NO by the mouse macrophages induced by LPS at the concentrations of 25, 50 and 100 ㎍/mL. The PH extract also significantly suppressed the VEGF, PDGF-BB and intracellular calcium production of the mouse macrophages by LPS at concentrations of 25, 50 and 100 ㎍/mL. As shown in the results above, the PH extracts do not have a toxic effect on the macrophages, but still have an anti-inflammatory effect that significantly reduces the intracellular calcium production as well as the production of NO, VEGF and PDGF-BB at concentrations above 25 ㎍/mL.Conclusions : In conclusion, the inhibitory anti-inflammatory effects of the PH extract can be used for a new treatment of anti-inflammatory diseases.
