To investigate the effect of protein kinase on melanin production via cAMP-dependent pathway, we measured the melanin amount and tyrosinase activity in B16 melanoma cells stimulated by alpha-melanocyte stimulating hormone (MSH), forskolin and 8-Br-cAMP. MSH, forskolin and 8-Br-cAMP significantly increased both melanin production and tyrosinase activity in B16 cells. Melanin production and tyrosinase activity by MSH are significantly inhibited by cyclic AMP-dependent protein kinase inhibitor (KT5720) and protein kinase C down-regulation treated with PMA. Bisindolmaleimide (1μM), protein kinase C inhibitor, significantly inhibited melanin production and tyrosinase activity stimulated by MSH, forskolin and 8-Br-cAMP with the following order of potency: MSH>forskolin>8-Br-cAMP. Tyrosine kinase inhibitor, genistein and DHC, significantly inhibited both, but the inhibitory effect was more potent in 8-Br-cAMP-stimulated B16 cells than MSH-stimulated cells. NFkB inhibitor (parthenolide) significantly inhibited melanin production and tyrosinase activity. Neither melanin production nor tyrosinase activity induced by MSH, forskolin and 8-Br-cAMP were affected by KN-62 (calmodulin-dependent protein kinase II inhibitor), PD098059 (mitogen-activated protein kinase inhibitor, MAPKK) and worthmannin (phosphatidylinositol 3-kinase inhibitor). These results suggest that both protein kinase C and tyrosine kinase are involved in melanin production by cyclic AMP-dependent pathway and NFkB pathway may play an important role in cyclic AMP-dependent melanin production in B16 melanoma cells.

This study is expected to provide some basic data on how to apply more economical and efficient eco-friendly factors to reduce the carbon emissions. It has been scrutinized and analyzed the environmental factors of passive eco-friendly elements along with active eco-friendly elements as more efficient factors for energy conservation based on the case of a newly constructed broadcasting building with the green building certification in Sangam-dong DMC area. The first analysis was the energy consumption trends in Korea and the necessity of energy saving. Secondly, it was examined the energy consumption in the current status of the broadcasting building. Thirdly, it was looked into the correlation between the eco-friendly design application factors applied to the broadcasting building and the energy consumption types. As a result of the analysis, the application of fan and feed pump inverter along with ventilation system of waste heat recovery ventilation system were more meaningful in accordance with the economic feasibility of broadcasting buildings rather than economical effect of passive elements such as rooftop garden, reinforcement of building insulation. Also, the application of lighting control had the economical feasibility. Therefore, when it is intended to change the green building certification energy related evaluation items of the similar broadcasting building types, it is necessary to change the direction of adding weight to the more economical active element items in the future.

This study is basically about the changing appearance of the big pavilion, Yeongnamnu in Miryang city belonging to the government guesthouse of Joseon dynasty. And the purpose of this study confirms that even if the appearance of the building has not changed, the function of the building changes because the reason why people think about the building is also different. First, looked at the appearance of architecture and the function of architecture that changes with the times. In the course of the research, the changes of pavilion were traced by dividing it into five from Kory? dynasty which was first established to the present. Then, tried to find out the change patterns by analyzing important events related to buildings or places in each period. As a result, the following facts were confirmed. 1. Because of the Buddhist repression policy in Joseon Dynasty, the Yeongnamnu changed from the temple pavilion to the government pavilion. 2. Throughout the Joseon Dynasty, Yeongnamnu was the representative scenic of Miryang City. 3. In the late Joseon Dynasty, Yeongnamnu was famous as a place where a woman named Arang devoted her life in order to preserve his fidelity. 4. Since 1910, Yeongnamnu has become an important cultural asset, and space has been widely used as a venue for various public events. 5. From 1945 to 1999, Yeongnamnu was used mainly for the events of Arang Remembrance and Miryang Culture Festival. And the inner space of Yeongnamnu was rarely used because people thought preservation is important.

The purpose of this study is to reveal the meaning of Le Corbusier's works focused on Maison Guiette. This House was completed in 1926, 4 year later than the work of the Citrohan and also, this house is the only remaining mixed use Dwelling in Belgium. It offers a fine example of the early architectural concepts of Le Corbusier's. Despite these important facts, the meanings of the ideas of this house have not been paid attention as much as other houses constructed at the same period, such as Maison La Roche, Maison Stein de Monzie, Villa Savoye, etc. In his book, 'Oeuvre Complete 1910-1929', Le Corbusier mentioned as follows: "A special solution imposed by Belgian housing developments characterized by a dimension of 6 meters of frontage and a very great depth. The staircase serving the various floors is like the ladder of Jacob climbs Charlie Chaplin in the "Kid" and This house is an architectural experiments of his ideas. And this study aims at analysing the relationship between Maison Guiette and spaces on the basic of Le Corbusier's 'Five Points of a New Architecture' which had a great effects on architectural history and became a fundamental elements for the change into the Modern housing. By analysing the points in regard with the Features of Architectural Concept Applied in Maison Guiette, The Five Points are the background of the interpenetration of the interior and the exterior. To attain this, this research appraises each points in detail in regards with Maison Guiette. The study of Maison Guiette will be an important architectural subject to clarify the meaning of his later works and ideas.

This study examined the apartment sale in lots system from 1960s to 1970s in Seoul from a historical perspective, with the focus on the concept and changes of pre-sale in lots system. By tracing back historical context and application of the system in that period through the analysis of the advertisements for apartment sale, we found out the hidden side of the apartment pre-sale in lots system from the public's point of view, and its impact and meaning on the formation of our current residential culture. In the 1960s and 1970s, the apartment sale in lots system based on the pre-sale in lots system formed strong seller's market and it has been the main reason for imposing the wrong residential awareness that led the public to recognize housing as a means of property growth. Even now, the current apartment sale in lots system remains to be a supplier-oriented one, and lots of people tend to regard housing as investment method. That is, this mechanism is not only still working but one of the fundamental causes of impoverishing our residential culture. It is necessary to reconsider the pre-sale in lots system and the apartment sale in lots system in relation to the public of housing.

With microtiter plate, the assay method was developed for detecting the fungicidal activity of new compounds against spore germination, spore adhesion and mycelial growth of Colletotrichum sp. JC24 cal1Sing red pepper anthracnose. Also, the effects of some commercialized fungicides on fungal development like above mentioned were investigated by measuring the optical density of mycelia grown into wells of microtiter plate. For the standardization of assay method, some factors, such as the treatment of MTT and/or propanol, inodulum density and incubation period, affecting on mycelial optical density were investigated. For obtaining precise and consistent mycelial optical density, it was necessary the treatment of MTT for 12 hrs and propanol for 1 hr. inoculum density adjusted to 1×105 spores/mL and incubation period for 36 hrs at 25∘C. For fungicidal activities, 6 protective fungicides, 6 ones inhibiting sterol biosynthesis, and one inhibiting respiration were used in this study. While mancozeb, chlorothalonil and dithianon among 6 protective fungicides inhibited strongly spore germination, adhesion, and mycelial growth at 6.25μg/mL, propineb, iminoctadine and fluazinam inhibited intermediately spore germination and mycelial growth at 100μg/mL. Washing above 3 fungicides with new PD broth, their activity against spore adhesion decreased. With hexaconazole, tebuconazole and myclobutanil, the tendency of the activity against fungal differentiation of the early infection stage was similar to the latter group of protective fungicides, showing the decrease of the inhibitory activity against spore adhesion by washing 2 hrs after incubation. However, kresoxim-methyl inhibited spore adhesion distinctly, depending on the applied concentrations. Based on these results, it might be able to assess the fungicidal activity of many compounds against spore germination, adhesion and mycelial growth by the use of microtiter plate in vitro. Using the assay developed in this report, it was possible to investigate the inhibitory activity of some commercialized fungicides, too.
Both high glucose and glucose degradation products (GDP) have been implicated in alterations of peritoneal membrane structure and function during long-term peritoneal dialysis (PD). The present study examined the role of GDP including methylglyoxal (MGO), acetaldehyde, and 3,4-dideoxyglucosone (3,4-DGE) in HPMC activation with respect to membrane hyperpermeability or fibrosis. The role of reactive oxygen species (ROS) and activation of protein kinase C (PKC) in GDP-induced HPMC activation were also examined. Using M199 culture medium as control, growth arrested and synchronized HPMC were continuously stimulated by MGO, acetaldehyde, and 3,4-DGE for 48 hours. Vascular endothelial growth factor (VEGF) was quantified as a marker of peritoneal membrane hyperpermeability and fibronectin and heat shock protein 47 (hsp47) as markers of fibrosis. Involvement of ROS and PKC was examined by the inhibitory effect of N-acetylcystein (NAC) or calphostin C, respectively. MGO significantly increased VEGF (1.9-fold), fibronectin (1.5-fold), and hsp47 (1.3-fold) secretion compared with control M199. NAC and calphostin C effectively inhibited MGO-induced VEGF upregulation. Acetaldehyde stimulated and 3,4-DGE inhibited VEGF secretion. Fibronectin secretion and hsp47 expression in HPMC were not affected by acetaldehyde or 3,4-DGE In conclusion, MGO upregulated VEGF and fibronectin secretion and hsp47 expression in HPMC, and PKC as well as ROS mediate MGO-induced VEGF secretion by HPMC. This implies that PKC activation and ROS generation by GDP may constitute important signals for activation of HPMC leading to progressive membrane hyperpermeability and accumulation of extracellular matrix and eventual peritoneal fibrosis.
This study examined the possibility on the dietary value and cryopreservation of the marine microalga, Isochrysis galbana. Four cryoprotectants, dimethyl sulfoxide (DMSO), ethylene glycol (EG), glycerol (Gly) and 1.2-propanedial (PD) were tested at the concentrations of 1.0, 1.5, 2.0, 2.5 and 3.0 M respectively. The highest survival rates were obtained with 1.5 and 2.0 M of four cryoprotectants yielding a survival rate of 80%. Cell concentration of 30×104cell/ml at the initial point of the experiment was increased to 365×104cell/ml in 1.5 M of dimethyl sulfoxide, 298×104cell/ml in 1.5 M of ethylene glycol, 512×104cell/ml in 1.5 M of glycerol, and 385×104cell/ml in 1.5 M of 1.2-propanedial after five days, respectively. In dietary value experiment, survival rate and growth were not significantly different.

Various additives were added in small amounts on Ni/YSZ anode of SOFC (solid oxide fuel cell) in order to improve reactivity and to inhibit deactivation due to coke deposition during methane reforming using a low mole ratio steam (H2O/CH4=1.5) at 800∘C. Ni/YSZ catalysts added with various perovskites did not show any improvement but exhibited a gradual decrease in the methane conversion. K-doped Ni/YSZ showed a steady increase and maintenance of the conversion up to 42 hours, after which there was an abrupt deactivation of catalyst owing to potassium loss by volatilization. Addition of 5% of K2Ti2O5 on Ni/YSZ showed a stable maintenance of the conversion without K loss, and was able to prevent coke formation during a long time operation. Deactivation of catalyst during the reaction was mainly caused by the accumulation of graphidic carbon on the catalyst surface.

Three sex pheromone components (cis-8-dodecenyl acetate (Z12Ac), trans-8-dodecenyl acetate (E12Ac), cis-8-dodecenol (Z12OH)) of the Oriental fruit moth, Grapholita molesta, were chemically synthesized. Especially to increase the composition of cis-stereoisomer, a triple bond intermediate was hydrogenated at −20∘C with catalytic Pd/BaSO4. The resulting product consisted of the acetates with a stereoisomer ratio in 92:8 (Z:E). The biological activity of the synthesized pheromone compounds was analyzed both in male responses and orientation disruption. The indoor pheromone effect was determined by male flight behavior showing wing movement in response to lure. Different mixtures of the synthetic pheromone components were prepared by mixing acetate and alcohol components in 100:0, 99:1, and 90:10 (g/g) and tested with a comparison of a standard commercial pheromone lure. The highest pheromone effect was observed in only acetate mixture (100:0) and the effect was reduced with the addition of the alcohol component. This indoor pheromone effect could be observed in field monitoring trial, in which 100:0 mixture showed the highest trap catches. Orientation disruption assay was conducted indoor by using a cage, in which the center had a commercial lure on sticky plate and the four candidates were placed at 6 cm away from the central lure on each of four directions. Test males were released to the arena during overnight (12 h) and then the caught males on the sticky plate were counted. The synthesized pheromone as well as the commercial pheromone showed 100% orientation disruption. However, the orientation disruption effect was reduced with decrease in the number of the surrounding disrupting pheromone baits. These results clearly suggest that the synthesized sex pheromone of G. molesta is biologically active and can be used for field mating disruption.
