Improved T-Vector for the Cloning of PCR DNA Using Green Fluorescent Protein

  • A new GFP-based T-vector for cloning of PCR products was developed by using a green fluorescent protein (GFP) as a mafker. In order to facilitate the DNA inserts, multiple restriction sites, SP6 and T7 RNA polymerase promoter sites, were introduced close to the PCR DNA insertion site of a pCRGv vector. The XcmI-digested pHNT plasmid can be used to clone a 3' A-overhanged PCR DNA amplified by Taq DNA polymerase. A potential method of easing some difficulties from its use along with its cost savings proveded by this vector are likely to lead to the replacement of other T-vectors for PCR DNA cloning.

  • In order to the investigate epidemiological characteristics of methicillin-resistant Staphylococcus aureus (MRSA), 31 strains of Staphylococcus aureus were isolated from the equipments of two hospitals in Chonbuk. And their antimicrobial resistance patterns against 7 kinds of antimicrobial agents and the identification of MRSA by polymerase chain reaction (PCR) were studied. Seven strains among 10 strains of methicillin resistant Staphylococcus aureus showed 554 bp DNA which was a part of mecA gene in PCR analysis.

  • Methicillin-Resistant Staphylococcus aureus (MRSA) strains are resistant to a wide range of antibiotics and are a major cause of nosocomial infections. Accurate and rapid typing of MRSA is needed to implement effective infection control measures. Arbitrarily Primed PCR (AP-PCR) is a very useful method in rapid typing. AP-PCR is not necessary information about target DNA sequence because this is basically DNA amplification and could be useful in epidemiological typing by classified band pattern. In this study, MRSA were isolated and identified from ICU, Neu, IM and Ped environments and investigated molecular typing by AP-PCR. Ped, the MRSA pattern determines the la, IIa type, 1M is Ib type, Neu is IIa type and ICU determines the IIa, lIb types. All MRSA in this study were typeable by AP-PCR, which was easy to perform and reproduce with evidence of MRSA for purposes of nosocomial infection control.

RT-PCR Detection of Citrus Tristeza Virus form Early Satsuma Nandarin and Yuzu in Cheju Island

  • Citrus tristeza virus (CTV) was identified form CTV-infected early satsuma mandarin (Citus unshiu) and yuzu (C.junos) by RT-PCR. The total RNAs were isolated from citrus bark and seaf tissues infected with CTV and reverse transcription was followed with primers designed for amplifying CTV coat protein gene. DNA fragments 738 bp were amplified by RT-PCR and these products were colned for sequence analysis. Based on the sequence analysis, this PCR product has 97% sequence homology to CTV (T-385) CP gene isolated from USA. RT-PCR assay for CTV detection was more sensitivity than ELISA assay which was done with anti-CTV CP antibody. This is the frist report about CTV identification in Cheju island Korea.

  • Turnip mosaic virus)TuMV-Ca) was isolated from a Chinese cabbage showing severe mosaic and black necrotic spots symptoms in Korea. The virus was identified as a strain of TuMV by its host range test, particle morphology, serology, double stranded RNA analysis. For detection of the virus, reverse transcription and polymerase chain reaction(RT-PCR) was performed with a set of 18-mer TuMV-specific primers to amplify a 876 bp DNA fragment The virus was rapidly detected from total nucleic acids of virus infected tissues as well as native viral RNA of purified virion particles by RT-PCR. Detection limit of the viral RNA by RT-PCR was 10 fg.

  • Vibrio cholerae O1 serogroup Wave 3 El Tor strains are presently prevalent worldwide. The Wave 3 El Tor strains contain a TLC:RS1:CTX array on chromosome 1, and no element is integrated on chromosome 2. A multiplex PCR optimized to identify the TLC:RS1:CTX array of Wave 3 strains has been developed in this study. By using eight primers, the multiplex PCR can identify the characteristic CTX and RS1 array of Wave 3 strains from various arrays of strains belonging to other Waves. The four amplified DNA fragments of Wave 3 strains have been cloned in a vector, which could be used as a positive control for the multiplex PCR. This multiplex PCR and the positive control set could be useful tools for rapid recognition of Wave 3 El Tor strains.

Occurrence of canine brucellosis in Korea and polymorphism of Brucella canis isolates by infrequent restriction site-PCR

  • In this study, occurrence of canine brucellosis was surveyed in kennels, indoor dogs and stray dogs in Korea, and infrequent restriction site-polymerase chain reaction (IRS-PCR) was applied to analyze DNA polymorphism of Brucella canis (B. canis) isolates. Among a total of 501 dogs tested, B. canis antibodies by both rapid screening agglutination with 2-mercaptoethanol (2-ME RSAT) and immunochromatographic assay were detected in only 14.1% of kennel dogs. There were no seropositive cases in indoor dogs and stray dogs. DNA polymorphism was observed in 16 B. canis isolates by the IRS-PCR. Sixteen isolates were tested with primers, PsalA, PsalC, PsalG and PsalT, and different primers produced different DNA patterns. In regard to the IRS-PCR pattern of 16 isolates, 9 (56.3%) belonged to the IRS-PCR type I. The remaining 7 were differentiated as type II, III and IV. An application of the primer PsalC provided discrimination between B. canis isolated in 2005 and others.

  • Molecular typing of Bacillus anthracis has been extremely difficult due to the lack of polymorphic DNA markers. Aiming to develop a DNA marker specific for Bacillus anthracis and to be able to discriminate this species from Bacillus genus, we applied the random amplified polymorphic DNA (RAPD)-PCR. We have identified B. anthracis from various Bacillus species. The analysis performed by RAPD clearly demonstrated substantial genetic variations among Bacillus species. This type of analysis is an easy, quick and highly discriminatory technique that may help in diagnosis of anthrax.

  • In this work, thermal design of a PCR chip for LOC is systematically conducted. From the numerical simulation of a PCR chip based on the finite volume method, how to control the average temperature of a PCR chip and the temperature difference between the denaturation zone and the annealing zone is presented. The average temperature is shown to be controlled by adjusting heat input and a cooler as well as a heater is shown to be necessary to obtain three individual temperature zones for polymerase chain reaction. To reduce the time required, a heat sink for the cooler is not included in the calculation domain for the PCR chip and heat sink design is conducted separately by using a compact modeling method, the porous medium approach.

  • 본 연구에서는 돼지 로타바이러스의 혈청형 A, B 및 C를 동시에 진단할 수 있는 RT-PCR 기법을 개발하였다. 각각의 혈청형에 특이적인 primer를 이용하여 RT-PCR 기법으로 분변시료에서 직접 바이러스 동정을 실시한 결과, 23건의 로타바이러스 감염 분변 시료에서 13건이 혈청형 A, 3건이 혈청형 B, 2건이 혈청형 C로 나타나 국내에서도 A, B 및 C형의 돼지 로타바이러스가 공히 발생하고 있음을 확인하였으며, 발생분포는 외국의 예와 유사하였다. 이 RT-PCR기법은 돼지 로타바이러스의 주요 혈청형인 A, B 및 C형의 동시감별진단법으로 이용될 수 있을 것으로 판단된다.
