식품 중 폴리염화비페닐 위해평가 (Risk Assessment of Polychrorinated Biphenyls (PCBs) through Food Intake for the Korean Population)

  • 본 연구는 어류를 포함한 식품에서 PCBs 오염도 조사하고, 우리나라 전체 국민이 식품을 통해 노출되는 PCBs 수준 및 안전성을 평가하였다. 국내유통 식품 중 PCBs 오염도 실태조사를 위해 다소비 다빈도 식품 28개 품목을 선정하고 검체 382건을 구매하였고, 식품 중 PCBs 오염도 조사를 한 결과, 62종 PCBs의 검출량은 N.D.$-182.4{\mu}g/kg$ (평균 $5.4{\mu}g/kg$)이였고, 본 연구에서 조사된 PCBs의 오염도는 다른 문헌보다 상대적으로 낮거나 유사한 수준이었다. 노출 및 위해성 평가를 위해 식품별 섭취를 통한 PCBs 노출량을 산출한 후, 일본에서 설정한 TDI 값 대비 위해도를 평가하였다. 식품 섭취를 통한 전체국민의 평균 PCBs 노출량은 9.54 ng/kg b.w./day로 TDI 대비 0.19% 수준이었다. 따라서 국내 유통 식품을 통한 전체국민의 PCBs 평균 노출수준은 인체노출안전기준보다 낮은 위해도를 보임으로써, 우리나라 국민은 식품에 존재하는 PCBs에 대하여 건강상 위해 수준이 낮은 것으로 사료된다.

폐지와 폐목재에서의 잔류성 유기오염물질의 농도 및 배출원 추정 (Levels of Persistent Organic Pollutants in Waste Paper and Waste Lumber and Evaluation of their Sources)

  • 본 연구는 산업 현장에서 나오는 폐지, 폐목재에 함유되어 있는 17종 PCDD/Fs, 12종 Co-PCBs, 16종 PAHs 농도 수준과 이들의 발생원 추적을 위해 수행되었다. 농도 수준은 폐지와 폐목재 순으로 PCDD/Fs는 9.69~176.77 pg/g-dry, 0.14~0.25 pg/g-dry, Co-PCBs는 109.95~4097.25 pg/g-dry, 28.23~59.88 pg/g-dry, PAHs는 9.30~52.18 ng/g-dry, 0.82~1.82 ng/g-dry로 조사되었으며, 모든 대상물질은 폐목재보다 폐지에서 더 높은 농도로 검출되었다. PCDD/Fs 중 OCDD가 가장 높은 농도로 검출되었으며, 소각로 배출가스 중 다이옥신과 유사한 패턴을 보였다. Co-PCBs 이성질체 분포 패턴은 상용 PCB 제품의 이성질체 분포패턴과 유사하게 나타나 이로 인한 오염으로 판단되었고, PAHs의 경우 특정 화합물의 농도비를 이용하여 추정한 결과 폐기물 소각과 같은 연소와 관련이 있음을 알 수 있었다.

환경호르몬 2,2',5,5'-Tetrachlorobiphenyl의 신경세포 독성에 대한 인삼의 방어효과 (Protective Effect of Korean Ginseng on Cytotoxicity Induced by 2,2',5,5'-Tetrachlorobiphenyl in Human Neuronal SK-N-MC Cells)

  • Polychlorinated biphenyls(PCBs) are large scale industrial chemicals which are using in diverse applications. The goal of this study was to determine if exposure to 2,2',5,5'-tetrachlorobiphenyl (PCB 52) leads to an increase in the production of active oxidants, and subsequently promotes apoptosis of neuronal SK-N-MC cells. Reactive oxygen species (ROS) formation was examined in SK-N-MC cells after treatment of PCB 52 by concentrations and incubation times, respectively. It showed that the rate of ROS production in the cells was increased in a does-dependent manner to 45 min, followed by a return towards control levels after 120 min treatment. We also examined the association of PCB-induced apoptosis with the modulation of biomakers of oxidative damage to lipids (malondialdehyde [MDA]) in SK-N-MC cells. Increased MDA was observed in a dose-dependent manner in groups treated with 10, 15, and 20 figJ me of PCB 52 for 24 h. After treatment of PCB 52, the cells did not show any significant change in the rate of Cu/Zn-superoxide dismutase (Cu/Zn-SOD) activity. Whereas, the cells had a two-fold greater rate of change in catalase activity at 20 ㎍/㎖ of PCB 52 for 24 h when compared to control group. Korean Ginseng is one of the most important crude drugs which has been used as a traditional Oriental medicine. We next investigated protective effect of extracts of ginseng on cytotoxicity induced by PCB 52 in SK-N-MC cells. Pretreatment of SK-N-MC cells with 25-200 μg/ml of ginseng were reduced cell death in a dose-dependent manner in PCB 52-treated cells. To examine the sensitivity of beta-catenin to ginseng, the protective effect of a range of ginseng concentrations was examined in SK-N-MC cells treated with PCB 52. The result demonstrated that ginseng efficiently blocked PCB 52 inducible beta-catenin proteolysis in a concentration dependent manner. The ROS formation was also measured in the presences of extract of ginseng and superoxide dismutase (inhibitor of oxygen free radical production). The both SOD (400 U/ml) and ginseng (200 μg/ml) significantly inhibited RDS generation in PCB 52-treated group.

정적 초임계유체 방식에 의한 토양 중의 PCBs 추출 (Static Supercritical Fluid Extraction of PCBs from Soil Matrix)

  • 토양오염표준공정시험법과 정적 초임계 $CO_2$ 추출방식을 사용하여 환경오염물질로 알려져 있는 토양 중의 PCBs를 분석하였다. 토양오염표준공정시험법에 의한 PCBs의 평균회수율은 25-35%의 범위였으며 표준편차는 10%를 초과하였다. 이와는 대조적으로, 정적 초임계 $CO_2$ 추출방식에 의한 PCBs의 평균회수율은 토양오염표준공정시험법보다도 2-2.5배 높았으며 표준편차는 7% 이내였다. 이러한 결과들은 정적 초임계 $CO_2$ 추출방식이 토양오염표준공정시험법에서 공인된 시료전처리과정을 대체할 수 있을 가능성을 제시하고 있다. $40^{\circ}C$의 일정한 온도에서 점차적으로 초임계 $CO_2$의 압력을 1130 psi에서 1196 psi로 증가시킴에 따라 토양으로부터 PCB congener의 회수율도 역시 증가하는 경향을 보였으나, 같은 추출조건(같은 온도와 압력)에서 평형시간(5분과 60분)은 회수율의 어떠한 영향도 주지 않았다. 마지막으로, 각 PCB congener의 회수율은 같은 추출조건에서 PCB의 분자량이나 구조(coplanar versus non-coplanar)와는 관계없이 거의 비슷한 수준의 값을 보였다.

수유기간에 따른 모유 중 Indicator PCBs 분석을 통한 영유아의 인체노출평가 (Estimation of Indicator PCBs Exposure in Infants via Breast-Feeding Period)

  • Indicator PCB의 농도 수준은 2006년에 세계보건기구(WHO) 프로토콜에 따라 수집된 132개의 모유(초유, 출산 후 30일째 모유, 출산 후 60일째 모유)로 분석하였다. Indicator PCBs의 농도는 고분해능 가스크로마토그래피/고분해능 질량분석기를 이용한 동위원소희석법으로 측정되었으며, 이 방법은 국제적 경향을 비교 평가하기 위한 정확하고 정밀한 데이터를 제공한다. 또한, 모든 분석 결과를 토대로 평균일일섭취량(ADIs)이 계산되었다. 총 indicator PCBs의 중앙값은 14 ng/g lipid로, 다른 나라에서 검출된 농도보다 현저하게 낮았으며, PCBs 138, 153, 180은 분석한 지표 이성질체 합의 약 65%를 차지하였다. 가장 기여율이 높은 PCB 153은 총 indicator PCBs 농도와 매우 높은 상관관계를 나타내었다(p<0.001). PCBs 농도는 산모의 아이수가 증가할수록 확연히 감소하였고, 산모의 PCBs 축적과 배출에 있어 출산경력과 수유기간은 중요한 인자로 나타났다. 6개월 모유수유 기간 동안 총 indicator PCBs의 평균일일섭취량의 중앙값은 $0.077{\mu}g/kg{\cdot}bw/day$이었다. 초산부 모유 중 PCB 118의 농도는 1998년에 비해 25~35% 정도 감소하였으며, 이러한 경향은 지속될 것이라는 것을 보여준다.

대도시에 거주하는 여성의 모유 중 Indicator PCBs와 식이습성 및 인구통계학적 인자간의 관계 (The Relationship among the Indicator PCBs in Breast Milk and Dietary Habits and Demographic Factors in Women Living in Urban Areas)

  • In this study, breast milk levels of indicator polychlorinated biphenyls (PCBs) were estimated, and statistics drawn, for 22 Koreans in the general population, aged 26-38, who had resided in metropolitan areas for more than 5 years without occupational exposure to organochlorine pollutants. Concentrations of indicator PCBs were measured using the isotope dilution method with a high resolution gas chromatograph/high resolution mass detector, which provided accurate and precise data for investigation of trends, and international comparisons. The geometric mean of total indicator PCB levels was 22.3 ng/g lipid, which is significantly lower than the level in individuals from European countries. Overall, the geometric mean and 95th percentile of the most abundant congener PCB 153 were 8.04 ng/g lipid and 16.4 ng/g lipid, respectively. PCB congeners 138, 153 and 180 together accounted for about 75% of the indicator congeners analyzed. Breast milk total indicator PCB concentrations were significantly associated with age and parity, but not with body mass index (BMI), rate of body weight increase, or smoking habits. The geometric mean level of PCB 153 in breast milk from mothers with a pre-pregnant BMI < $21\;kg/m^2$ was significantly higher than in pre-pregnant women with a BMI of ${\geqslant}\;21\;kg/m^2$ (p<0.05). Women who ate more fish, meat, and milk products did not have significantly higher total indicator PCB levels than those who ate less of these products, but a significant difference in PCB 153 levels was demonstrated between the subjects consuming higher (11.45 ng/g lipid) and lower (6.79 ng/g lipid) amounts of fish (p<0.05), after adjusting for confounders. These results suggest that age, parity and fish intake are the important factors affecting the concentrations of indicator PCBs in these subjects.

  • A practical and efficient disposal method for chemical dechlorination of PCBs (polychlorinated biphenyls) in transformer oil was evaluated. The transformer oil containing PCBs was treated by the PEG 600 (polyethylene glycol 600) and potassium hydroxide (KOH) along with different reaction temperatures(25, 50, 100 and $150^{\circ}C$) and times(30, 60, 240 and 480 min). The best disposal efficiency of PCBs in transformer oil was attained under the experimental conditions of PEG 600 (2.5 w/w%)/KOH (2.5 w/w%)/$150^{\circ}C$/4 hrs, showing completely removal of all PCBs containing 3-9 chlorines on two rings of biphenyl. In studying the reaction of PEG/KOH with PCBs, it confirmed that the process led to less chlorinated PCBs through a stepwise process with the successive elimination of chlorines.

일본 요코하마 대기 중 PCBs의 가스-입자 분배 (Gas-Particle Partitioning of PCBs in Ambient Air, Yokohama Japan)

  • This study was aimed at estimation of gas-particle partitioning of polychlorinated biphenyls (PCBs) in ambient air. The samples were collected at urban site in Japan from March 2002 to January 2003. The concentration of total PCBs (from 4 CB to 10 CB) and TEQ (Toxic equivalent) ranged from 62 to $247\;pg/m^3$ and from 2 to $14\;fgTEQ/m^3 $, respectively. The average contribution $(\%)$ of gas phase to total PCBs concentration was above $80\%$, which suggests that in the atmosphere PCBs predominantly existed in the gas phase. The weak correlations between total PCBs concentration and temperature was found. However this result was due to a typhoon during summer and raining during sampling period. The gas-particle partition coefficient (Kp) was obtained as a function of temperature. The partition ratio of gaseous and particulate phase PCBs can be estimated for an arbitrary temperature. The plot of gas/particle partition coefficient (log Kp) vs. sub-cooled liquid vapor pressure $(log\;P_L)$ had reasonable correlations for individual samples but the slope varied among the samples (coefficients of determination for log Kp versus log $P_L$ plot were> 0.76 $(p<0.0001)$, except for 3 samples). As a result, the variations in the slope among the sampling period may be due to change of temperature, raining during sampling period and wind in this study.

  • Polyurethane foam-disk passive air samplers (PAS) were deployed in a southern area of Korea for three months. The target compounds were 12 coplanar polychlorinated biphenyls (PCBs). The congener profiles measured in this study were the same as those in ambient air and emission gas from the incinerator. A gradient of the total PCBs in different regions (industrial>residential>rural) was observed, suggesting the industrial complex may be an important source of coplanar PCBs. In general, only gas-phase compounds are mainly sequestrated by PAS. In order to estimate the concentration of particle-phase PCBs, a gas/particle partition model was used. A combined result (gas+particle-phase PCBs) was compared with previous results, indicating that the level of coplanar PCBs in our study area is comparable to those in other urban sites in the world. The validation of this method for estimating the total concentration is required through additional backup studies.
