Hypothermia Regulates Endoplasmic Reticulum (ER) Stress through the X-box Binding Protein-1 (XBP1) Gene Expression in PC12 Cells

  • Endoplasmic reticulum (ER) stress induces unfolded protein response (UPR) via inositol-requiring enzyme 1 (IRE1) activation, which sends a molecular signal for X box-binding protein 1 (XBP1) mRNA splicing in the cytosol. IRE1 endoribonuclease activity induces cleavage of XBP1 mRNA. The XBP1 mRNA is then ligated by an uncharacterized RNA ligase and translated to produce spliced XBP1 by 23 nt removed in which contains the PstI restriction enzyme site. The splicing of XBP1 mRNA can be detected by semiquantitative RT-PCR, and then splicing of XBP1 is a useful tool to measure the genetic variability in ER stress. In this study, we have estimated IRE1-dependent splicing of XBP1 mRNA under conditions of various hypothermia. The results indicated that hypothermia regulated ER stress. This study demonstrated that hypothermia is closely related to ER stress and may be useful for early diagnosis of ER-associated disease.

  • Beta-amyloid peptide (A${\beta}$) is considered to be responsible for the pathogenesis of the Alzheimer's disease. Several lines of evidence support that A${\beta}$-amyloid-induced cytotoxicity is mediated through the generation of reactive oxygen species (ROS). Agents that are able to scavenge excess ROS may be useful as protecting or reducing agents for development or progress of AD. Green tea extract has been known to have antioxidant property. Our previous studies also demonstrate that green tea extract protected ischemia/reperfusion-induced brain injury by reduction of cell death through scavenging of oxidative damages of macromolecules.   • FM방식의 악기음원합성은 해당 악기의 음색과 음정에 맞는 FM의 기본 주파수와 변조 계수를 정하여 신호 파형을 생성하는 것으로, 다른 가, 감산방식 또는 PCM방식 등에 비해 비교적 간단한 구조로 다양한 악기음원합성이 가능하다. 따라서 현재 사용되고 있는 개인용 컴퓨터에 부착되는 사운드합성 카드에는 FM방식 음원합성기술이 적용되고 있다. 본 논문에서는 FM방식 음원합성기술을 이용하여 실시간 악기음원합성이 가능한 논리회로를 설계하는데 관한 연구를 기술한다. 본 연구에서는 소프트웨어 프로그래밍에 의해 FM방식 음원합성기의 구조를 설계하고 주요 블록의 최적 변수 값을 실험하였다. 논리회로 설계 및 회로검증은 향후 주문형반도체(ASIC:Application Specific Integrated Circuit)제작을 위해 기존의 표준셀 라이브러리와 주문형반도체 전용 설계시스템을 사용하였다. 회로검증 결과는 간이 평가보드를 제작하고 PC와 접속시켜 생성된 악기음을 직접듣는 주관적 평가방법으로 최종 확인하였다.

Anti-stress effects of Gastrodia elata on catecholamine pathway in rat

  • Enzymes involved in catecholamine synthesis are present in the highest concentration in the adrenal medulla, however they were found also in other, mainly nervous tissues. Increased transcription of genes for catecholamine biosynthetic enzymes is an important mechanism to increase the capacity for epineprine/norepinephrine biosynthesis with stress. Gastrodia elata(Chinese name: Tienma), are very important Chinese herbal medicines used for the medical treatment of headaches, migraine, dizziness, epilepsy, rheumatism, neuralgia, paralysis and other neuralgic and nervous disorders. Immobilize stressed rat markedly increased tyrosine hydroxylase (TH) mRNA and dopamine-${\beta}$-hydroxylase (DBH) mRNA transcriptior level more than control group. But treated Gastrodia elata extracts in immobilized stressed rat slightly increased TH mRNA and DBH mRNA transcription level more than normal group. In addition, we are obtained identical results in PC12 cell line. Decrease of transcription level of TH mRNA and DBH mRNA is indicating that Gastrodia elata have a anti-stress effects which decrease the transcription level of TH and DBH mRNA on catecholamine biosynthesis pathway.
