The effect of chemically oxidized intermediates of Polynuclear Aromatic Hydrocarbon (PAH) compounds on the degradation of the parent PAHs was characterized and evaluated for the context of cooxidation. These results may be interpreted in the context of cooxidation mechanism whereby high recalcitrant PAH compounds are biodegraded in the soil and suggest a potential tool for bioremediation of PAHs contaminated soils and protection of groundwater.

Polynuclear aromatic hydrocarbon (PAH) compounds are highly carcinogenic chemicals and common groundwater contaminants that are observed to persist in soils. The adherence and slow release of PAHs in soil is an obstacle to remediation and complicates the assessment of cleanup standards and risks. Biological degradation of PAHs in soil has been an area of active research because biological treatment may be less costly than conventional pumping technologies or excavation and thermal treatment. Biological degradation also offers the advantage to transform PAHs into non-toxic products such as biomass and carbon dioxide. Ample evidence exists for aerobic biodegradation of PAHs and many bacteria capable of degrading PAHs have been isolated and characterized. However, the microbial degradation of PAHs in sediments is impaired due to the anaerobic conditions that result from the typically high oxygen demand of the organic material present in the soil, the low solubility of oxygen in water, and the slow mass transfer of oxygen from overlying water to the soil environment. For these reasons, anaerobic microbial degradation technologies could help alleviate sediment PAH contamination and offer significant advantages for cost-efficient in-situ treatment. But very little is known about the potential for anaerobic degradation of PAHs in field soils. The objectives of this research were to assess: (1) the potential for biodegradation of PAH in field aged soils under denitrification conditions, (2) to assess the potential for biodegradation of naphthalene in soil microcosms under denitrifying conditions, and (3) to assess for the existence of microorganisms in field sediments capable of degrading naphthalene via denitrification. Two kinds of soils were used in this research: Harbor Point sediment (HPS-2) and Milwaukee Harbor sediment (MHS). Results presented in this seminar indicate possible degradation of PAHs in soil under denitrifying conditions. During the two months of anaerobic degradation, total PAH removal was modest probably due to both the low availability of the PAHs and competition with other more easily degradable sources of carbon in the sediments. For both Harbor Point sediment (HPS-2) and Milwaukee Harbor sediment (MHS), PAH reduction was confined to 3- and 4-ring PAHs. Comparing PAH reductions during two months of aerobic and anaerobic biotreatment of MHS, it was found that extent of PAHreduction for anaerobic treatment was compatible with that for aerobic treatment. Interestingly, removal of PAHs from sediment particle classes (by size and density) followed similar trends for aerobic and anaerobic treatment of MHS. The majority of the PAHs removed during biotreatment came from the clay/silt fraction. In an earlier study it was shown that PAHs associated with the clay/silt fraction in MHS were more available than PAHs associated with coal-derived fraction. Therefore, although total PAH reductions were small, the removal of PAHs from the more easily available sediment fraction (clay/silt) may result in a significant environmental benefit owing to a reduction in total PAH bioavailability. By using naphthalene as a model PAH compound, biodegradation of naphthalene under denitrifying condition was assessed in microcosms containing MHS. Naphthalene spiked into MHS was degraded below detection limit within 20 days with the accompanying reduction of nitrate. With repeated addition of naphthalene and nitrate, naphthalene degradation under nitrate reducing conditions was stable over one month. Nitrite, one of the intermediates of denitrification was detected during the incubation. Also the denitrification activity of the enrichment culture from MHS slurries was verified by monitoring the production of nitrogen gas in solid fluorescence denitrification medium. Microorganisms capable of degrading naphthalene via denitrification were isolated from this enrichment culture.

PAHs are aromatic hydrocarbon compunds with two or more benzene rings. Because they are mostly toxic to human life, they need to be converted to non-toxic compunds or to be degraded completely. This work aims to degrade PAHs (phenanthrene and pyrene) using Bacillus bacteria covered on cork or sawdust. The results show that media effect on phenanthrene was negligible whereas biodegradation ability of sawdust carrying the bacteria was better than that of biofilm-covered cork when pyrene was tested. PAH removal was also affected by soil moisture content with $45\~55\%$ of the optimal content.

The effect of in-situ chemical oxidation on the indigenous soil microorganisms (total microbes and PAH-degrading microbes) and contaminant removal were investigated. Field soil contaminated with diesel in gas station was collected and the soil was treated from 0 to 900 minutes by in-situ ozonation as chemical remediation. The treated soil samples were incubated with supplying oxygen during the 9 weeks to understand the characteristics of microbes regrowth, damaged by ozone. The sharp decrease of aromatic fraction and TPH was observed within 60 minutes of ozone application and aromatic fraction and TPH then slowly decreased. The phenanthren-degrading bacteria were the most sensitive to ozonation, because 1 hour of ozonation reduced the microbes from 10$^{6}$ CFU/g-soil to below detection limits.
Polycyclic aromatic hydrocarbons (PAHs) are ubiquitous in the environment. They are highly toxigenic and carcinogenic. Probiotic bacteria isolated from fermented foods were tested to check their ability to degrade and/or detoxify PAHs. Five probiotic bacteria with distinct morphologies were isolated from a mixture of 26 fermented foods co-cultured with benzo(a)pyrene (BaP) containing Bushnell Haas minimal broth. Among them, B. velezensis (PMC10) significantly reduced the abundance of BaP in the broth. PMC10 completely degraded BaP presented at a lower concentration in broth culture. B. velezensis also showed a clear zone of degradation on a BaP-coated Bushnell Haas agar plate. Gene expression profiling showed significant increases of PAH ring-hydroxylating dioxygenases and 4-hydroxybenzoate 3-monooxygenase genes in B. velezensis in response to BaP treatment. In addtion, both live and heat-killed B. velezensis removed BaP and naphthalene (Nap) from phosphate buffer solution. Live B. velezensis did not show any cytotoxicity to macrophage or human dermal fibroblast cells. Live-cell and cell-free supernatant of B. velezensis showed potential anti-inflammatory effects. Cell-free supernatant and extract of B. velezensis also showed free radical scavenging effects. These results highlight the prospective ability of B. velezensis to biodegrade and remove toxic PAHs from the human body and suggest that the biodegradation of BaP might be regulated by ring-hydroxylating dioxygenase-initiated metabolic pathway.

Ethanol washing with distillation as a cleanup process of polycyclic aromatic hydrocarbon(PAH)-contaminated soil was investigated in this study. A multistage ethanol washing with distillation process was applied to three different types of soil, i.e., sandy soil, alluvial soil, and clay with the initial concentration of benzo(a)pyrene 10 mg/kg, benz(a)anthracene 250 mg/kg, and pyrene 100 mg/kg soil. Ethanol was selected as washing solvent because of its high PAH removal efficiency, low cost, and non-toxicity comparing to the other solvent such as isopropyl alcohol and sodium dodecyl sulfate. The satisfactory results (i.e. lower than benzo(a)pyrene 1 mg/kg, pyrene 10 mg/kg, benz(a)anthracene 25 mg/kg, which are the Canada or the Netherlands soil standard) for three types of soils were obtained by at most five-six times washing. It was suggested that organic content in soil decreased the removal efficiency by ethanol washing.
Due to increasing marine pollution there is a great possibility that seaweed is contaminated with polycyclic aromatic hydrocarbons (PAHs). To investigate the effect of processing on PAM removal from Porphyra tenera (laver) contaminated PAH, laver was contaminated with fluoranthene known to have a strong photoinduced toxicity, followed by washings and drying, which are usual processes for dried laver preparation. Sample at each step was collected and its toxicity was evaluated using cultured animal cells as well as analyzing PAH contents. Fluoranthene level in laver was significantly lowered by sequential washings with sea water and distilled water, but not by drying. Fluoranthene content in raw laver right after contamination was 221 ppm and decreased to 130 ppm by washings with seawater plus distilled water while its level was not lurker lowered by drying process. Cytotoxicity and photoinduced cytotoxicity in mammalian cells were significantly elevated in laver extracts containing fluoranthene. Cellular arylhydrocarbon hydroxylase (AHH), one of the biomarkers for cellular accumulation of PAH, was greatly induced by laver extract contaminated with fluoranthene. These results suggest that photoinduced toxicity and AHH activity can be used to monitor contamination of seafood by PAHs. Fluoranthene accumulated in laver was efficiently removed by sequential washings with seawater and tap water for 24 hrs and 12 hrs, respectively.

Surfactant-enhanced electrokinetic (EK) process was investigated to remove polycyclic aromatic hydrocarbons (PAHs) from low-permeable soils. Phenanthrene and kaolinite were selected as a representative PAH and a model soil, respectively. A nonionic surfactant Tergitol 15-S-12 was applied to improve the solubility of phenanthrene and sodium chloride was used as an electrolyte at the various concentrations from 0.001 to 0.1M. The addition of electrolyte affected both the removal efficiency and operation cost. When electrolyte was introduced, the electrical potential gradient became low and thus power consumption was reduced. However, as electrolyte concentration increased, the electroosmotic flow also decreased, so the removal efficiency of contaminant decreased. Therefore, the removal efficiency and power consumption should be considered simultaneously to determine the iptimum surfactant concentration, so a relatively lower concentration of electrolyte than certain value is desired.

Removal of phenanthrene by electrokinetic (EK) method combined with Fenton-like process was studied in a model system. Sand and phenanthrene were selected as a model soil and a representative PAH. Sand was contaminated at the concentration of 500 mg phenanthrene/kg dry sand. Bentonite and kaolinite were inserted into the space between reservoir and contaminated soil. When hydrogen peroxide supplied to a soil system from the anode reservoir was transported through the soil by EK process, the Fenton-like reaction was occurred by naturally existing iron minerals in soil. When hydrogen peroxide was supplied into the system, it showed higher removal efficiency than when just water was used. Maximum removal efficiency of phenanthrene was 81.2 % for 7 days.
