Biosorption of Cr (VI) ions by Ficus religiosa barks: Batch and continuous study

  • In the present research, Ficus religiosa Bark (FRB) is used as an adsorbent for the removal of heavy metal Cr (VI) ions. This Ficus religiosa Bark was characterized by Scanning Electron Microscope, Fourier transform infrared Spectroscopy, Thermo Gravimetric Analyzer and the results showed that activated adsorbent have high adsorption capacity and withstand even in high temperature. Batch and Continuous experiments were conducted to determine the effect of various parameters such as pH, contact time, adsorbent dose and initial metal concentration. The biosorption followed pseudo first order kinetic model. The adsorption isotherms of Cr (VI) on Ficus religiosa fitted well with the Temkin model. In Batch study, maximum biosorption capacity of Cr (VI) was found to be 37.97 mg g-1 (at optimal pH of 2, adsorbent dosage of 0.3 grams and concentration of Cr (VI) is100 mg L-1). The Continuous mode of study shows that 97% of Cr (VI) ion removal at a flow rate of 15 ml min-1. From the results, selected Ficus religiosa Bark has the higher adsorption capacity for the removal of Cr (VI) ions from wastewater.

  • This study, analyzed the general and minerals composition of kamut and investigated its effect on blood components in mice fed a high fat diet. The content of each general component of kamut was as follows: $11.02{\pm}0.75%$ water, $13.16{\pm}1.28%$ crude protein, $1.85{\pm}0.19%$ crude fat, and $1.97{\pm}0.13%$ ash. The leptin level was the highest in the HF group($30.00{\pm}0.00ng/mL$) when compared to the control group. There was a significant decrease of $23.65{\pm}5.54ng/mL$ in the HFK group when compared to the HF group (p<0.05). The blood LDL-cholesterol concentration was the lowest in the control group at $10.00{\pm}2.00mg/dL$. The level was highest in the HF group at $28.00{\pm}0.00mg/dL$ when compared with the other groups (p<0.05). The aspartate transaminase (AST) level was significantly higher in the HFK group ($179.33{\pm}173.88U/L$) than in the control ($61.00{\pm}12.73U/L$) and HF groups ($132.00{\pm}0.00U/L$). According to the results of this study, the consumption of kamut lowers the blood LDL-cholesterol level more than the consumption of wheat flour. Additionally, kamut contains antioxidant substances such as selenium and zinc, which are thought to contribute to vascular health and thus aid in maintaining good health. Therefore, it is necessary to develop a variety of health foods using kamut; it should be used as a functional food for the maintenance of good health.

두 종 미세 녹조류의 연속배양을 통한 바이오매스 생산성 비교 (Comparison of Biomass Productivity of Two Green Microalgae through Continuous Cultivation)

  • In the present study, the biomass productivity of two green microalgae (Chlorella sp. and Dunaliella salina DCCBC2) were assessed in a 12 L tubular photobioreactor under optimum culture conditions. In the batch culture optimization process, the Chlorella sp. biomass was obtained as 1.2 g/L under atmospheric air as a sole $CO_2$ source and other culture conditions as follows: light intensity, temperature, pH, $NH_4Cl$ and $K_2HPO_4$ were 100 ${\mu}E/m^2/s$, $27^{\circ}C$, 7.0, 20.0 mM and 2.0 mM, respectively. On the other hand, 2.9 g/L of D. salina DCCBC2 biomass production was observed under the following conditions: light intensity, temperature, pH, $KNO_3$ and $K_2HPO_4$were 80 ${\mu}E/m^2/s$, $27^{\circ}C$, 8.0, 3.0 mM and 0.025 mM, respectively. At 1% $CO_2$ supply to the reactor, the Chlorella sp. production was reached 1.53 g/L with 25% increment under the same operating conditions. In addition, the maximum D. salina DCCBC2 biomass was observed as 3.40 g/L at 3% $CO_2$ concentration. Based on the aforementioned optimized conditions, the dilution rate and maximal biomass productivity of Chlorella sp. and D. salina DCCBC2 in the continuous cultivation were 0.4/d and 0.6 g/L/d and 0.6/d and 1.5 g/L/d, respectively.

Development of a Dynamic System Simulating Pig Gastric Digestion

  • The objective of this study was to develop a model for simulating gastric digestion in the pig. The model was constructed to include the chemical and physical changes associated with gastric digestion such as enzyme release, digestion product removal and gastric emptying. Digesta was collected from the stomach cannula of pigs to establish system parameters and to document the ability of the model to simulate gastric digestion. The results showed that the average pH of gastric digesta increased significantly from 2.47 to 4.97 after feed consumption and then decreased 140 min postprandial. The model described the decrease in pH within the pigs' stomach as $pH_t=5.182e^{-0.0014t}$, where t represents the postprandial time in minutes. The cumulative distribution function of liquid digesta was $V_t=64.509e^{0.0109t}$. The average pepsin activity in the liquid digesta was 317Anson units/mL. There was significant gastric emptying 220 min after feed consumption. The cybernetic dynamic system of gastric digestion was set according to the above data in order to compare with in vivo changes. The time course of crude protein digestion predicted by the model was highly correlated with observed in vivo digestion (r = 0.97; p = 0.0001), Model prediction for protein digestion was higher than that observed for a traditional static in vitro method (r = 0.89; p = 0.0001).

Pichia pastoris에서 Aspergillus ficuum 유래 Acetyl Xylan Esterase 유전자의 과발현 (High-Level Expression of Aspergillus ficuum Acetyl Xylan Esterase Gene in Pichia pastoris,)

  • Aspergillus ficuum 유래 acetyl xylan esterase(AXEase) 유전자(AXE)를 Pichia pastoris에서 과발현ㆍ분비 생산하기 위해 AOXI promoter와 mating factor $\alpha$-1 분비신호서열 하류에 AXE를 연결한 염색체 삽입 발현계(pPICZ$\alpha$C-AXE, 4.6 kb)를 구축하였다. 이것을 SacI으로 절단한 뒤 P. pastoris의 염색체 DNA 5'AOX1 부위에 삽입시켰다. 형질전환된 P. pastoris 균주를 메탄을 배지에서 플라스크 회분배양한 결과, 배양 36시간 때의 건조균체농도는 6 g-DCW/1, AXEase 총 발현량은 77 unit/ml이었다. 최적화된 methanol과 histidine 공급방법을 채용한 유가배양시 균체농도는 97 g-DCW/1, AXEase 총발현량은 930 unit/m1로 크게 증가하였다. 효소활성의 90% 이상은 배양 상등액에 존재하였으며, 상등액 단백질의 80%이상이 AXEase 단백질(33.5 kDa)였다. 이러한 결과는 9.8 g/l의 AXEase 단백질을 배양 상등액으로 대량 분비ㆍ생산할 수 있음을 의미한다.

Study on Changes in Racehorses' Metabolites and Exercise-related Hormones before and after a Race

  • Physiological changes in thoroughbred racehorses during the race were investigated by measuring concentrations of metabolites and exercise-related hormones before and after a race. The conversion point from anaerobic to aerobic exercise during the race was estimated subsequently. Blood samples were taken from the jugular vein of 53 thoroughbreds at different times -three h before and 45 min after- for measuring the concentrations of glucose, non-esterified fatty acids (NEFA), lactate, uric acid, ammonia, insulin, adrenocorticotrophin (ACTH) and cortisol according to the race distance. In accordance with the race distance, each metabolite increased in concentration compared with the level before the race. The level of glucose, in particular, increased from $56.18{\pm}3.20$ mg/dl before the race to $148.82{\pm}8.82$ mg/dl after the race for horses that raced 1,400 m, showing a significant increase of 165% (p<0.001). The concentration of NEFA rose from $76.77{\pm}5.59$ uEq/L to $335.85{\pm}35.39$ uEq/L, up 337% (p<0.01) after a 1,400 m race. Exercise-related hormones also showed similar changes. The level of insulin dropped the most in horses that raced 1,400 m, by 42%, from $0.97{\pm}0.18$ to $0.56{\pm}0.05\;{\mu}g/L$ (p<0.5); however, ACTH and cortisol jumped significantly at 1,800 m, from $20.17{\pm}2.12$ to $551.45{\pm}91.33$ pg/ml (p<0.5) and $1.13{\pm}0.16$ to $5.66{\pm}0.45\;{\mu}g/dl$ (p<0.01), respectively, representing the highest increase. Therefore, based on the changes in glucose, NEFA and insulin levels before and after the race, it was concluded that the race distance of 1,400 m represents the point where racehorses make a conversion from anaerobic to aerobic exercise.

Plant responses to nano and micro structured carbon allotropes: Water imbibition by maize seeds upon exposure to multiwalled carbon nanotubes and activated carbon

  • Multiwalled carbon-nanotubes (MWCNT) and micro-structured carbon, such as biochar or activated carbon (AC), have been seen to significantly increase the growth indices of certain plant species such as maize (Zea mays L.). Seed imbibition is the stage where environmental factors that affect water transport across the seed coat barrier, make a large impact. This work explores the effect on water imbibition by maize seeds when the aqueous environment surrounding the seed is diluted by small concentrations (10 and 20 mg/l) of pristine MWCNT (p-MWCNT), carboxylate functionalized MWCNT (COO-MWCNT) and AC. The degree of sensitivity of the process to (i) large structural changes is seen by utilizing the nano (the MWCNT) and the micro (the AC) allotropic forms of carbon; (ii) to small changes in the purity and morphology of the p-MWCNT by utilizing 95% pure and 99% pure p-MWCNTs of slightly differing morphologies; and (iii) to MWCNT functionalization by using highly pure (97%) COO-MWCNT. Water imbibition was monitored over a 15 hour period by Near Infrared Thermography (NIRT) and also by seed weighing. Seed surface topography was seen by SEM imaging. Analysis of the NIRT images suggests rapid seed surface topological changes with the quantity of water imbibed. While further work is necessary to arrive at a conclusive answer, this work shows that the imbibition phase of the maize seed is sensitive to the presence of MWCNT even to small differences in the purity of the p-MWCNT and to small differences in the physicochemical properties of the medium caused by the hydrophilic COO-MWCNT.

Effects of Dietary Vitamins C and E on Egg Shell Quality of Broiler Breeder Hens Exposed to Heat Stress

  • A feeding trial was conducted to determine whether dietary vitamin C (200 mg/kg) and vitamin E (250 mg/kg) prevent any drops in egg shell quality under heat stress in broiler breeder hens. One hundred and sixty molted Ross broiler breeders were housed randomly in an individual cage at 83 weeks of age. Four dietary treatments with forty hens and four replications per treatment were control (no additional vitamins), vitamin C-, or vitamin E-supplemented and combined supplementation of the two vitamins. After a tenday-adaptation period at 25$^{\circ}C$, the ambient temperature was kept at 32$^{\circ}C$ for a three-week-testing period. Egg production dropped dramatically over week but it did not show a significant change among treatments (p<0.05). However, egg quality parameters such as egg weight, specific gravity, shell thickness, SWUSA, puncture force and shell breaking strength from the birds fed the diet with the combined vitamins C and E were significantly improved over those of the control group during the heat stress period (p<0.05). The hens fed the vitamin C diet improved tibia breaking strength (37.16 kg), statistically higher than the birds fed the control and the vitamin E diets (p<0.05). The hens fed the control diet showed higher serum corticosterone levels, a mean of 5.97 ng/ml, than those of the other treatments (p<0.05). The heat stress resulted in elevated heterophils and decreased lymphocytes in serum, increasing the H/L ratios for all the treatments. However, the increases in H/L ratios were alleviated by feeding the diets containing vitamin C alone or together with vitamin E, although there were no significant differences in the ratio between the two groups (p<0.05). In conclusion, vitamins C (200 mg/kg) and/or E (250 mg/kg) supplemented to the diets for broiler breeder hens could prevent drops in egg shell quality and tibia bone strength under highly stressful environmental temperatures.

Fundamental Study on Solvent Sublation Using Salphen and Its Application for Separative Determination of Trace Ni(II), Co(II) and Cu(II) in Water Samples

  • A solvent sublation using salphen as a ligand was studied and applied for the determination of trace Ni(II), Co(II) and Cu(II) in water samples. The fundamental study was investigated by a solvent extraction process because the solvent sublation was done by extracting the floated analytes into an organic solvent from the aqueous solution. The salphen complexes of Ni(II), Co(II) and Cu(II) ions were formed in an alkaline solution of more than pH 8 and then they were extracted into m-xylene. It was known that the each metallic ion formed 1 : 1 complex with the salphen and the logarithmic values of extraction constants for the complexes were 3.3 5.1 as an average value. Based on the preliminary study, the procedure was fixed for the separation and concentration of the analytes in samples. Various conditions such as the pH of solutions, the influence of $NaClO_4$, the bubbling rate and time of $N_2$ gas, and the type of organic solvent were optimized. The metal-salphen complexes could be extracted into m-xylene from the solution of more than pH 8, but the pH could be shifted to acidic solution of pH 6 by the addition of $NaClO_4$. In addition, the solvent sublation efficiency of the analytes was increased by adding $NaClO_4$. The recovery of 97-115% was obtained in the spiked samples in which given amounts of 0.3 mg/L Ni(II), 0.8 mg/L Co(II) and 0.04 mg/L Cu(II) were added.

신양천 하천수정화 연못시스템의 처리수준 및 연못반응 (Treatment Level and Reactions of a Treatment Pond System Purifying Sinyang Stream Water)

  • Treatment level and pond reactions of a pond system were examined from May to October 2002. The system was constructed in July 2000 for purifying water of Sinyang stream that flows into Koheung Estuarine Lake located in the southern part of the Korean Peninsula. The system was composed of a primary and a secondary pond in series and established on the rice field near the lake. Water pumped from the stream was funneled into the primary pond, whose effluent was discharged into the secondary pond by gravity flow. Effluent from the secondary pond was funneled into wetlands. About 130 $m^3$/day of water was pumped into the primary pond and detention time of the primary and secondary pond was about 2 days. DO from the surface to the 1.0 m depth of the primary and secondary pond was in the rage of 5.2 to 11.0 mg/L and 4.3 to 0.7 mg/L, respectively. DO at the bottom layer of the primary pond was 0 mg/L and that of the secondary pond ranged 3.0~4.7 mg/L. The primary pond functioned as a facultative pond and the secondary as an aerobic one. The temperature difference between the surface and bottom layers of the ponds in August was about $2.5^{\circ}C$ and that in May and October was about $1.0^{\circ}C$. Thermocline was observed in the primary pond during the high ambient temperature of August. The sludge depth of the primary pond in May, August, and October was 2.4, 1.9, and 2.2 cm, respectively. That of the secondary pond was 1.2, 1.0, and 1.1 cm, respectively. SS, $BOD_5$, T-N, and T-P concentrations in influent averaged 16.64, 6.71, 6.21, and 0.23 mg/L and those in effluent from the primary pond averaged 11.48, 4.97, 4.81, and 0.17 mg/L, respectively. The removal rates of the primary pond for SS, $BOD_5$, T-N and T-P were 31%, 26%, 22%, and 24%, respectively. Average concentrations of SS, $BOD_5$, T-N, and T-P in effluent from the secondary pond were 9.81, 4.07, 4.03, and 0.14 mg/L, respectively and the abatement rates of the secondary pond for SS, $BOD_5$, T-N and T-P were 20%, 12%, 13%, and 15%, respectively. SS, $BOD_5$, T-N and T-P concentrations in effluent from the primary pond were significantly low(p=0.001) when compared with those from the secondary one.
