The objective of this study was to investigate the effects of the packaging method on the lipid and protein oxidation, and color in aged top round from Hanwoo (Korean native cattle) for 14 d at $4^{\circ}C$. Catalase activity was the highest (p<0.05) in vacuum packaging (VP) treatment during storage, and was higher (p<0.05) in 50% Ox-MAP and 50% Ox-MAP+vacuum skin packaging (VSP) treatments than in other treatments at d 14. Superoxide dismutase activity was higher (p<0.05) in VP, 50% Ox-MAP, and 50% Ox-MAP+VSP treatments than in other treatments at d 14. During storage, total antioxidant activity was the highest (p<0.05) in VP treatment and was higher (p<0.05) in 50% Ox-MAP+VSP treatment than in 80% Ox-MAP treatment. TBARS value was the lowest (p<0.05) in VP treatment during storage and was lower (p<0.05) in 50% Ox-MAP and Ox-MAP+VSP treatments than in 80% Ox-MAP and Ox-MAP treatments, respectively. Carbonyl content was the lowest (p<0.05) in VP treatment from 10 d. From 7 d, the $a^*$ value was the highest (p<0.05) in VP treatment and was higher (p<0.05) in 50% Ox-MAP and 50% Ox-MAP+VSP treatments than in other treatments. The $b^*$ value was the highest (p<0.05) in VP treatment from 3 d, and was higher (p<0.05) in 80% Ox-MAP+VSP, 50% Ox-MAP, and 50% Ox-MAP+ VSP treatments than in 80% Ox-MAP treatment at d 14. Therefore, VP improved the oxidation and red color stabilities in stored-aged top round compared with Ox-MAP. In addition, 50% Ox-MAP improved the lipid oxidation and red color stabilities compared with 80% Ox-MAP, and its inhibitory effect on lipid oxidation was enhanced by combination with VSP.
Transcriptional activation domains are known to be inherently "unstructured" with no tertiary structure. A recent NMR study, however, has shown that the transactivation domain in human p53 is populated with an amphipathic helix and two nascent turns. This suggests that the presence of such local secondary structures within the overall "unstructured" structural framework is a general feature of acidic transactivation domains. These pre-existing local structures in p53, formed selectively by positional conserved hydrophobic residues that are known to be critical for transcriptional activity, thus appear to constitute the specific structural motifs that regulate recognition of the p53 transactivation domain by target proteins. Here, we report the results of a NMR structural comparison between the native human p53 transactivation domain and an inactive mutant (22L,23W$\rightarrow$22R,23S). Results show that the mutant has an identical overall structural topology as the native protein, to the extent that the amphipathic helix formed by the residues 18T 26L within the native p53 transactivating domain is preserved in the double mutant. Therefore, the lack of transcriptional activity in the double mutant should be ascribed to the disruption of the essential hydrophobic contacts between the p53 transactivation domain and target proteins due to the (22L,23W$\rightarrow$22R,23S) mutation.
An 18-week growth trial was conducted in flow-through aquarium system to evaluate the practical dietary protein sources for juvenile abalone (Haliotis discus hannai). Three replicate groups of the abalone averaging 0.11g were fed one of ten diets containing casein, white fish meal (WFM), meat meal (MM), feather meal (FM), blood meal (BM), soybean meal (SM), corn gluten meal (CGM), cotton seed meal (CSM), Undaria powder (UP), or wheat flour (WF) as a dietary protein source. In addition, these dietary protein sources were cmpared with algae such as raw Undaria or dried Laminaria. Weight gain of abalone fed the diets containing casein, WFM, SM, CSM, or UP was significantly higher (P<0.05) than those of abalone fed other diets, and this value of abalone fed FM, BM, CGM, or algae was lower than other groups. Shell length, shell width, body wt./shell length ratio, and body wt./shell width ratio of abalone fed casein, WFM, SM, CSM, UP, and WF were also highe (P<0.05) than those of other groups. There were no significant difference (P>0.05) in moisture and protein contents of soft body among all diets. The data obtained in this study indicate that each of the casein, WFM, SM, CSM or UP is good dietary protein source for juvenile abalone.

Single C-reactive protein (CRP) molecules, which are non-specific acute phase markers and products of the innate immune system, were quantitatively detected on a gold-nanopatterned biochip using evanescent field-enhanced fluorescence imaging. The $4{\times}5$ gold-nanopatterned biochip (spot diameter of 500 nm) was fabricated by electron beam nanolithography. Unlabeled CRP molecules in human serum were identified with single-molecule sandwich immunoassay by detecting secondary fluorescence generated by total internal reflection fluorescence (TIRF) microscopy. With decreased standard CRP concentrations, relative fluorescence intensities reduced in the range of 33.3 zM-800 pM. To enhance fluorescence intensities in TIRF images, the distance between biochip surface and CRP molecules was optimally adjusted by considering the quenching effect of gold and the evanescent field intensity. As a result, TIRF only detected one single-CRP molecule on the biochip the first time.

We constructed the fibroin H-chain expression system to produce enhanced cyan fluorescent proteins (ECFP) in transgenic silkworm cocoon. Fluorescent cocoon could be made by fusing ECFP cDNA to the heavy chain gene and injecting it into a silkworm. The ECFP fusion protein, each with N- and C-terminal sequences of the fibroin H-chain, was designed to be secreted into the lumen of the posterior silk glands. The expression of the ECFP/H-chain fusion gene was regulated by the fibroin H-chain promoter. The use of the 3xP3-driven EGFP cDNA as a marker allowed us to rapidly distinguish transgenic silkworms. The EGFP fluorescence became visible in the ocelli and in the central and peripheral nervous system on the seventh day of embryonic development. A mixture of the donor and helper vector was micro-injected into 1,020 Kumokjam, bivoltin silkworm eggs. We obtained 6 broods. The cocoon was displayed strong blue fluorescence, proving that the fusion protein was present in the cocoon. Accordingly, we suggest that the ECFP fluorescence silk will enable the production of novel biomaterial based on the transgenic silk.
A series of feeding experiments were conducted to study the possible utilization of blood meal as a dietary protein source in growing common carp, Cyprinus carpio. Diets were formulated on isonitrogenous and isocaloric basis of $40\%$ crude protein and 3,640 kcal/kg diet : diet 1 (100 FMP, control), $100\%$ fish meal Protein (FMP) : diet 2 (25 BMP), $75\%$$FMP+25\%$ blood meal protein (BMP) : diet 3 (50 BMP), $50\%$$FMP+50\%$ BMP : diet 4 (75 BMP), $25\%$$FMP+75\%$ BMP : d;et 5 (100 BMP), $100\%$ BMP. As the dietary protein sources, $34.2\%$ of animal protein were supplied by FMP and/or BMP, and approximately $65.8\%$ of plant protein were used. In the first experiment, weight gain and feed efficiency were improved with increased level of blood meal protein in the diets. Weight gain and feed efficiency from fish fed diets 4 and S were significantly higher (P<0.05) than those from fish fed diets 1, 2 and 3. The second experiment was designed as a cross-over study to prove the first experiment's results. This cross-over study shows that weight gain from fish fed diet 5 is greater than that from fish fed diet 1. The third experiment was conducted to compare palatability between diet 1 (100 FMP) and 5 (100 BMP). The data from this palatability study indicated that the palatability of diet 5 was lower than that of diet 1 initially, however, the palatability of iiet 5 was improved and not worse than that of diet 1 within a week. Therefore, these findings may suggest that the fish meal can be replaced with blood meal completely in growing common carp diets.

Objective: The purpose of present study was to investigate the effects of condensed molasses fermentation solubles (CMS) on lactation performance, rumen fermentation, nutrient digestibility, and serum parameters of dairy cows. Methods: A total of 75 healthy Holstein cows with the same parity (milk production = 35±2.5 kg, body weight = 570±28 kg) were randomly selected and divided into 5 groups. One group served as control group (CON; no CMS), whereas the other 4 groups were CMS1 (accounted for 1% of the diet), CMS2 (2%), CMS3 (3%), and CMS4 (4%). All cows were fed regularly three times each day at 0800, 1600, and 2400 h. Cows received diet and water ad libitum. The experiment lasted for 60 days. Results: Results showed that the dry matter intake, milk yield, and protein of CMS2 were maximum and higher (p<0.05) than CMS4. The ruminal pH was observed less than 6 in CMS3 and CMS4 groups. No noticeable difference of microbial protein was found between CON and CMS2 groups, while the microbial protein in these groups was higher (p<0.05) than CMS3 and CMS4 groups. The apparent digestibility of dry matter, organic matter, and crude protein in CMS2 group was higher (p<0.05) than CMS3 and CMS4 groups. Compared to CMS3 and CMS4 groups, the CMS2 group increased (p<0.05) the serum concentrations of immunoglobulin G and immunoglobulin M on d 60. Conclusion: Therefore, it is practicable that CMS substitutes for a part of concentrates in lactating cows' diets, but higher addition of CMS (more than 3% of the diet) could decrease production performance of dairy cows as seen in the present study.

This study aimed to investigate the association between plant protein intake and health-related quality of life (HRQoL) in Korean individuals aged 50 years or older. Using the 2016-2018 Korea National Health and Nutrition Examination Survey data, 7,956 participants (3,434 men and 4,522 women) were included in the study. HRQoL was measured using the Euro-quality of life five-dimension (EQ5D), composed of physical function, self-care, daily activities, pain/discomfort, and anxiety/depression problems. The EQ5D estimates were converted into an EQ5D index score. The association between daily intake of plant proteins and HRQoL was evaluated using regression analysis. The intake of total plant protein, legume nut seed (LNS), and fermented bean (FERMB) proteins were all positively associated with HRQoL in both men and women (p < 0.01). Of EQ5D's five dimensions, physical function and daily activities were most commonly associated with plant protein intake. Compared to LNS protein or FERMB protein intake, total plant protein intake showed a better association with HRQoL. In conclusion, a significant association was observed between plant protein intake and HRQoL. It suggests that adequate intake of plant protein might be helpful for the maintenance or improvement of HRQoL in Korean adults.

Twenty four early lactating Nili-Ravi buffaloes, eight in each group, were used in a Randomized Complete Block Design to evaluate the influence of varying ruminally degradable protein (RDP) to ruminally undegradable protein (RUP) ratio on feed intake, digestibility, N balance, milk yield and its composition, conception rate and days open. Three experimental diets were formulated to contain RDP:RUP of 50:50, 66:34 and 82:18 and were denoted as HRUP, MRUP and LRUP, respectively. Dry matter (DM) intake was higher (p<0.05) in buffaloes fed HRUP diet than in those fed MRUP and LRUP diets. Dry matter digestibility was higher (p<0.05) in buffaloes fed LRUP diet than in those fed HRUP and MRUP diets. Linear increase was observed in DM digestibility with increasing RDP:RUP while Neutral detergent fiber digestibility remained unaltered in buffaloes fed HRUP and MRUP diets, however, it was higher than in those fed LRUP diet. Crude protein digestibility remained unaltered across all treatments. Milk and 4 percent fat corrected milk (4% FCM) yield was higher (p<0.05) in buffaloes fed HRUP diet than those fed MRUP and LRUP diets. Linear decrease in milk yield was observed with increased RDP:RUP. Milk protein and fat yields were higher (p<0.05) in animals fed HRUP diet than those fed MRUP and LRUP diets. Milk protein percent in animals fed HRUP diet was higher than in those fed LRUP diet, whereas it did not differ with those fed MRUP diet. Percent of fat, total solids, solid not fat and lactose remained unaltered across all diets. Nitrogen balance was higher in buffaloes fed HRUP diet than in those fed other diets. Increasing the RDP:RUP resulted in a linear decrease in N balance. The blood urea nitrogen and milk urea nitrogen were lower (p<0.05) in buffaloes fed HRUP diet than those fed MRUP and LRUP diets. The blood pH remained unaltered across all treatments. Days open did not differ significantly. Conception rate was higher in buffaloes fed HRUP diet than those fed MRUP and LRUP diets. The findings of the present study indicate that feeding high (50% of the total crude protein) ruminally undegradable protein diet not only increased nutrient intake and milk yield but also improved conception rate in early lactating buffaloes.

Protein unfolding is a key step in several cellular processes, including protein translocation across some membranes and protein degradation by ATP-dependent proteases. C1pAP protease and the proteasome can actively unfold proteins in a process that hydrolyzes ATP, These proteases catalyze unfolding by processively unraveling their substrates from the attachment point of the degradation signal. As a consequence, the ability of a protein to be degraded depends on its structure as well as its stability. An ${\alpha}$-helix is easier to unravel than a ${\beta}$-strand. In multidomain proteins, independently stable domains are unfolded sequentially. The steric constraints imposed on substrate proteins during their degradation by the proteasome were investigated by constructing a model protein in which specific parts of the polypeptide chain were covalently connected through disulfide bridges. The cross-linked model proteins were fully degraded by the proteasome, but two or more cross-links retarded the degradation slightly. Our results suggest that the pore of the proteasome allows the concurrent passage of at least three stretches of a polypeptide chain, and also explain the limited degradation by the proteasome that occurs in the processing of the transcription factor NF-KB, and also implicate difficulty in degradation of amyloidal aggregates by the proteasome
